Silver-gold nanocages have attracted considerable research interest recently due to their excellent performance in the fields of biomedicine and photocatalysis. These applications oftentimes manipulate at elevated temperatures and therefore impose demands on the thermal stability of the cage structures. To better understand this subject, in this work, we systematically evaluated the thermal stability of two nanocages with different wall thicknesses of 3.8 nm and 13 nm, both in the solution-phase (diethylene glycol) and solid-phase (in-situ STEM). The results revealed that the nanocages with thicker walls exhibited better thermal stabilities in both phases. By monitoring and analyzing the morphology changes of …

Background & objectives: Primary Microcephaly (MCPH) is a rare neurogenetic disease, manifesting congenitally reduced head circumference and non-progressive intellectual disability (ID). To date, twenty-eight genes with biallelic mutations have been reported for this disorder. The study aimed for molecular genetic characterization of Pakistani families segregating MCPH.
Methods: We studied two unrelated consanguineous families (family A and B) presenting >2 patients with diagnostic symptoms of MCPH, born to asymptomatic parents. We employed whole-exome sequencing (WES) of probands to find putative causal mutations. Chitosan has been studied as a non-viral vector capable of efficient gene delivery due to its favorable properties such as biodegradability, biocompatibility, and is non-toxic to mammalian cells. Incorporating electrostatic interactions in non-viral vectors enhance the vector permeability. This work focuses on two parts. Firstly, cationic chitosan derivatives were synthesized using quarternary ammonium and phosphonium groups. This was achieved by coupling carboxylic acid ligands with these quarternary groups to chitosan through an amide bond. I herein report the preparation and characterization of PEG-b-PLA block copolymer micelles and PEG-b-PLA/PLA mixed micelles as potential “plastic antidotes” for a toxic peptide, melittin. PEG-b-PLA micelles have been used in a number of biomedical applications for their biodegradability and biocompatibility. Along these lines, I investigated the colloidal stability and the peptide absorption properties of PEG-b-PLA/PLA mixed micelles. Erythropoietin (EPO) is a cytokine hormone known for initiating red blood cell proliferation by binding to its homodimer receptor (EPOR)2 in the bone marrow. Recent progress in neurobiology has shown that EPO also exerts robust neurotrophic and neuroprotective activity in the CNS. It is widely thought that EPO’s neurotrophic activity is centrally involved in its antidepressant and cognitive enhancing effects. However, EPO’s potent erythropoietic effects prevent it from being used in the clinic to treat psychiatric disorders. A chemically engineered non-erythropoietic derivative of EPO, carbamoylated EPO (CEPO), produces psychoactive effects without activating hematopoiesis. However, CEPO is expensive to produce and …

Two features make plasma a valued source of health information: First, plasma contains secreted proteins from all tissues in the body and can, therefore, provide information about the health status of all tissues. Second, plasma is available in large quantity using a minimally invasive procedure that is well tolerated. The status of plasma as a valued source of health information is matched by the difficulty in extracting proteome information from the samples. Using state of the art instrumentation a standard analysis of plasma yields quantitative information on approximately 300 proteins. Autophagy is the cellular degradation process in which cellular contents are encapsulated by double-membrane vesicles, autophagosomes, and delivered to the vacuole to be degraded and recycled. This process is important for cell health and homeostasis. There are approximately 32 different autophagy-related proteins involved. Atg10 is an enzyme that may affect overall autophagic activity by changing autophagosome size and/or number. Our goal is to find mutants that cause a significant, partial loss in Atg10’s activity by mutating some residues near the active site and testing the functionality of these mutants by performing western blots and enzymatic assays. Autophagy is an essential recycling process that occurs within eukaryotic cells, however, the individual functions of the current thirty-two known autophagic proteins are not yet entirely understood. At this time, it is known that the autophagic protein Atg7 works upstream of both Atg3 and Atg10. Atg3 is affected by Atg7 and allows for the attachment of Atg8 and the lipid PE which is part of the autophagosome membrane. The Atg10 pathway is also affected by Atg7 but attaches Atg12 to Atg5 which upregulates the function of Atg3 and ultimately Atg8 lipidation. In the past, we have demonstrated the photophysical properties of several anthraquinone derivatives that act as a luminescent host for various environmental and biological cations. Here we extend our expertise in photochemistry to the class of chemicals containing the pyridoxal moiety. Several ligands were synthesized using eco-friendly, simple, one-step reactions via Schiff base and condensation reactions. The optical properties of these ligands were examined with a series of cations. The export of mRNA out of the nucleus is a crucial step for eukaryotic gene expression. The export of mRNA transcripts is aided by Mex67, which allows export through the nuclear pore complex doorways in the nuclear envelope. Once out of the nucleus, a protein known as Dbp5, bound to ATP, Gle1, and Nup42 aids in the directionality of mRNA export by helping remove Mex67 from the mRNA strand. Following interaction with RNA, Dbp5 then hydrolyzes ATP so that it unbinds the mRNA, allowing for enzyme recycling. Filariasis, caused by a family of parasitic nematodes, affects millions of individuals through-out the tropics and is a major cause of acute and chronic morbidity. Current drugs are largely used in mass drug administration programs aimed at controlling the spread of disease by killing microfilariae, larval forms of the parasite responsible for transmission from humans to humans through insect vectors with limited efficacy against adult parasites. Although these drugs are effective, in some cases there are toxic liabilities. Over the past decade, Aza-Bodipy dyes (ADPs), a derivative of the extensively studied BF2-dipyrromethene (BODIPY), have become a growing field of research due to their unique photophysical properties, high yielding synthesis, and relative stability. They possess a unique spectral window of 600 to 750 nm which is the red to near-IR region, evidenced by the blue color of many of these dyes. Alzheimer's disease (AD) is commonly characterized by a loss of cognitive function due to the deterioration of neuronal synapses from the presence of senile amyloid beta-42 (Aß42) plaques. Evaluating cognitive deficits caused by Aß42 using human cortical neurons poses a challenge due to sourcing difficulties, and the use of animal models to assess drug efficacy creates biological hurdles from lack of species translatability. Fire debris analysis is a forensic science discipline that determines if an ignitable liquid residue is present or absent in a fire debris sample. Currently, fire debris analysis results in categorical statements based on qualitative data, not the quantitative evidentiary value of data. This dissertation includes two major projects. The first project investigated the great potential of polymer-derived ceramics (PDCs) as lithium-ion battery anode materials with good cycling stability and large capacity. SiCNO ceramic nanoparticles were produced by pyrolysis of polysilazane nanoparticles synthesized via an oil-in-oil emulsion crosslinking. The SiCNO nanoparticles had an average particle size of around 9 nm and contained graphitic carbon, Si3N4, and SiO2 domains. The electrochemical behavior of SiCNO nanoparticles anode was investigated to evaluate the Li-ion storage performance and understand its mechanism of Li-ion storage. A comprehensive two-dimensional thin layer chromatography (2D-TLC) has been developed for the separation of agarwood oil. The system employed a single stationary phase of silica gel 60 and two different mobile phases from the set of 9:1 (v/v) hexane/ethyl acetate, 1:1 (v/v) hexane/dichloromethane, dichloromethane, and acetonitrile. The best mobile phase system was found to be dichloromethane and 9:1 (v/v) hexane/ethyl acetate in the first and second dimensional separations, respectively. This showed the greater separation results with the orthogonality (A0) of 0.6636 based on the asterisk approximation which separated 14 spots of the agarwood sample compared with 9 spots separated by …

Voltage gated sodium ion channels are implicated in cardiac diseases, seizures, etc., and they play a role in maintaining ionic homeostasis in cells. Computational studies use prokaryotic model because they are simpler but function similarly to human voltage gated sodium ion channels. This study uses molecular dynamics (MD) to study three specific questions regarding voltage gated sodium ion channels of Magnetococcus marinus. The first question in this study is how the free energy of sodium diffusion compares to that of calcium ion diffusion. Fully and accurately reconstructing changes in oceanic productivity and carbon export and their controls is critical to determining the efficiency of the biological pump and its role in the global carbon cycle through time, particularly in modern CO2 source regions like the eastern equatorial Pacific (EEP). Here we present new high-resolution records of sedimentary 230Th-normalized opal and nannofossil carbonate fluxes and [231Pa/230Th]xs ratios from site MV1014-02-17JC in the Panama Basin. The mass of well-preserved calcite in planktonic foraminifera shells provides an indication of the calcification potential of the surface ocean. Here we report the shell weight of 8 different abundant planktonic foraminifera species from a set of core-top sediments along the Mid-Atlantic Ridge. The analyses showed that near the equator, foraminifera shells of equivalent size weigh on average 1/3 less than those from the middle latitudes. The carbonate preservation state of the samples was assessed by high resolution X-ray microcomputed tomographic analyses of Globigerinoides ruber and Globorotalia truncatulinoides specimens. The series of functionalized porphyrins was designed and synthesized by varying para-substituents on meso-position of porphyrin ring as ether, ester, and urea with three different long alkyl chains (3-5, 7-9, 11). Afterwards, metalation of porphyrin derivatives by some 3d metals to serve as soft materials, liquid crystal, and axial ligands were introduced to provide spin-crossover property. In this work, polydimethylsiloxane (PDMS), a well-known material for microfluidic devices, was used to fabricate a digital magnetofluidic device. The use of PDMS material allows for easy fabrication and flexibility. The superhydrophobic property of PDMS surface was easily achieved using a one-pot method by reduction between chloroauric acid and sodium formate. The superhydrophobic property of PDMS surface was investigated using water contact angle (WCA) and contact angle hysteresis, which were found to be 163.9° and 0°, respectively, indicating the superhydrophobic surface. The “magic spot” alarmones (pp)pGpp, previously implicated in Clostridioides difficile antibiotic survival, are synthesized by the RelA-SpoT homolog (RSH) of C. difficile (RSHCd) and RelQCd. These enzymes are transcriptionally activated by diverse environmental stresses. RSHCd has previously been reported to synthesize ppGpp, but in this study, we found that both clostridial enzymes exclusively synthesize pGpp. While direct synthesis of pGpp from a GMP substrate, and (p)ppGpp hydrolysis into pGpp by NUDIX hydrolases, have previously been reported, there is no precedent for a bacterium synthesizing pGpp exclusively. With the increased occurrence of wildfires around the world, interest in the chemistry of pyrogenic organic matter (pyOM) and its fate in the environment has increased. Upon leaching from soils by rain events, significant amounts of dissolved pyOM (pyDOM) enter the aquatic environment and interact with microbial communities that are essential for cycling organic matter within the different biogeochemical cycles. Heck C-C cross-coupling reaction has been widely employed in synthetic organic chemistry because of its high efficiency and simplicity, however the conventional Heck C-C cross-coupling reaction conditions involve the use of an expensive toxicant phosphine-ligated palladium complexes, and high temperature. To address these drawbacks, a highly efficient, homogeneous photocatalyst based on BODIPY for the Heck C-C cross-coupling reactions of aryl iodide and methyl acrylate in DMF solvent under inert conditions has been developed. Human C-terminal binding proteins (CtBP1 and CtBP2) are transcriptional coregulators of multiple genes in the human genome, including tumor suppressor genes (e.g., Bik, PTEN, BRCA1, and E-cadherin) as well as oncogenes (e.g. MDR1 and Tiam1). Both homologues of CtBP are overexpressed in many types of cancer, including breast cancer (92%), ovarian cancer (83%), colorectal cancer (64%), hepatocellular carcinoma (60%), gastric cancer, prostate cancer, and pancreatic adenocarcinoma. Further, expression levels of CtBP correlate with worse prognostic outcomes and more aggressive tumor features because it promotes proliferation, epithelial-mesenchymal transition, and cancer stem cell self-renewal activity.

Supramolecular solvent-based dispersive liquid-liquid microextraction technique has been developed as a preconcentration tool for the determination of trace level of Pb2+ and Cd2+. Dodecanol dispersed in tetrahydrofuran has been utilized as a supramolecular-solvent system for the extraction of analytes prior to their quantitative determination with graphite furnace atomic absorption spectrophotometer.
