The main stem reservoirs on the Colorado River comprise one of the largest and most heavily used freshwater bodies in the nation. These reservoirs (Lake Powell, Lake Mead, Lake Mohave and Lake Havasu) can store up to 53,590,400 acre-feet of water at their maximum capacities. Nonetheless, local water shortages still exist in some areas of the Colorado River Basin. There is also concern that salt concentrations are approaching levels that could severely affect municipal and agricultural uses. Water shortages will become even more acute as demands for water increase with continued urban and agricultural development in the basin. Water conservation …

The Rhizobium trifolii polysaccharides from SU843 and mutants--HC843(pBR1AN), HC843, 8002--in which the sym plasmid is replaced, absent, or mutated by transposon Tn5 were isolated. The surface polysaccharides of genetically altered mutants were compared quantitatively and qualitatively with one another and with the parent strain. The interpretation of these results is discussed in relation to the function of Rhizobium surface polysaccharides in the symbiotic process. In the last step of polypeptide chain initiation in eukaryotes, the interaction of the 40S preinitiation complex eIF-2.GTP.Met-tRNAi.40S [the complex between the 40S ribosomal subunit and the ternary complex containing equimolar amounts of eukaryotic initiation factor 2 (eIF-2), GTP, and eukaryotic initiator methionyl tRNA (Met-tRNAi)] with a 60S ribosomal subunit in the presence of mRNA, cap binding protein (with "capped" messengers), ATP, and the initiation factors eIF-3, eIF-4a, -4b, -4c, and eIF-5, results in the formation of an 80S initiation complex (Met-tRNAi.80S.mRNA) with concomitant hydrolysis of GTP and liberation of eIF-2 for recycling in subsequent initiation events. Kinetic studies of the cobalt (II)-4, 4’, 4’’, 4’’’-tetrasulfophtalocyanine and cyanide system were performed under argon and in air over temperature ranges of 294-314°K and 294-324°K, respectively. Two reactions were detected under argon. One was quite rapid and was followed on the stopped-flow apparatus. The results are consistent with the perturbation of a monomer-dimer equilibrium. The second reaction was slow enough to be followed spectrophotometrically on the Cary Model 14 spectrophotometer. The second order rate constant of 0.11 M-1 sec-1 at 294°K has been attributed to the rate of substitution of the second cyanide addition to a dye-cyanide …

Spermidine-Condensed Phi X174 Dna Cleavage By Micrococcal Nuclease: Torus Cleavage Model And Evidence For Unidirectional Circumferential Dna Wrapping., Kenneth A. Marx, Thomas C. Reynolds

Spermidine-condensed phi X174 replicative form (RF) II DNA was digested with micrococcal nuclease to yield seven identifiable DNA bands forming an arithmetic fragment-length series. The DNA monomer unit length was found to be 780 +/- 80 base pairs. This result is most consistent with a proposed model for micrococcal nuclease cleavage of a DNA torus organized by the unidirectional, circumferential wrapping of B-geometry DNA. By a topological consideration, the blunt-end-rod-fusion model for torus formation [Eickbush, T. H. & Moudrianakis, E. N. strong evidence suggests that the serum ferroxidases (ceruloplasmin andferroxidase II) promote the formation of Fe-III transferrin and thereby stimulate the turnover of iron from tissue stores. Ceruloplasmin is the major ferroxidase in human serum; whereas, ferroxidase II accotmts for an increased proportion of the activity in less highly developed animals. A large fold increase in total ferroxidase II activity was observed when both human and rabbit sera were subjected to gel-filtration on sephadex G-200. This indicated that whole serum might contain a potent inhibitor of ferroxidase II. Such an inhibitor has been isolated and purified to homogeneity by a combination …

This thesis describes the development of a sensitive method for the separation and quantitation of organic N-chloramines in chlorinated tap water. 5-Dimethylaminonaphthalene- l-sulfinic acid has been synthesized and characterized. It yields highly fluorescent dansyl derivatives on reaction with N-chloramines. Conditions of the derivatization were optimized for the detection of N-chloropiperidine at concentrations in aqueous solution of 10-7 M. The technique gives a quantity of derivative which is proportional to the concentration of chloramine present over the range examined (10-7 M to 10-4 M). The PDP-11/10 minicomputer was interfaced to the Beckman 25 ultraviolet/visible spectrophotometer and this on-line computer system was applied to enzyme kinetics studies. This interface involved hardware equipment to locate, amplify, and transmit the analog signal to an analog-to-digital converter, and software to acquire and store the digital data. Data processing programs were used to calculate kinetic parameters and statistical information from the acquired data.

The Varian Cary 219 ultraviolet/visible spectrophotometer can be readily interfaced to the Apple II Plus microcomputer by hardware equipment which can be purchased from the manufacturer. Various software may also be purchased for use in enzyme …

Excessive concentrations of the eukaryotic initiation factor 2(eIF-2).stimulating protein, a factor that catalyzes the formation of binary (GTP.eIF-2) and ternary (GTP.eIF-2.initiator methionyl-tRNA) initiation complexes at physiological Mg2+ concentrations, can cause ternary complex dissociation when the Mg2+ concentration is raised from 0.5 to 5.0 mM. Stimulation of ternary complex formation and dissociation have similar (a) pH optima, (b) metal ion specificity, and (c) sensitivity to phosphorylation of the eIF-2 alpha subunit. The results suggest that ternary complex dissociation is an artifact with no physiological significance.

A study was undertaken to examine the potential for ortho cyclization associated with the synthesis of 1,2,3,4,-tetrahydroisoquinolines using the Pictet-Spengler reaction. Specif1cally, the condensation of the biogenic amines, dopamine, norepinephrine and epinephrine were carefully examined in light of current literature and the facility and extent of ortho cyclization in each case. The para isomers of these three biogenic amine condensation products were isolated and characterized, while only the ortho isomer derived from dopamine was found stable and could be isolated, and structurally identified. Hydrocarbon concentrations were determined in bulk, wet, and dry deposition samples from urban and non-urban locations surrounding lower Chesapeake Bay. Mean hydrocarbon concentrations in bulk deposition samples were over three times greater at the urban station indicating a localized source. The major hydrocarbons present were n-alkanes, polycyclic aromatic hydrocarbons (PAH), and the unresolved complex mixture (UCM). The PAH and the UCM were an indication of significant anthropogenic inputs while the odd/even distribution of the n-alkanes was an indication of biogenic sources. These findings indicated seasonal trends of highest biogenic hydrocarbon inputs in the spring and summer months and highest anthropogenic …

This report discusses the research conducted in two areas: the synthesis of potential insecticides and the investigation of vinamidinium and azavinamidinium salt chemistry. Firstly, four classes of compounds were synthesized and characterized in the investigation of new potential insecticides. These classes of compounds include a-formamidine methyl esters, amidines, diacylhydrazines and 2,5-disubstituted-1,3,4-oxadiazoles and were characterized via NMR and infrared spectroscopy. A battery calorimeter was built for the measurement of thermal energy generation of high-temperature lithium–aluminum/iron sulfide battery cells, which are under development for electric vehicle propulsion and other energy storage applications. The calorimeter was designed with a temperature range of 400˚–500˚C, a detection limit of 1 mW, and an upper limit of heat flow of 50 W. The results of measurements on 200-Ah LiAl/FeS cells were in excellent agreement with the predictions of thermodynamic calculations based on precise measurements of the total cell polarization and the temperature coefficient of the emf. A general formulation of traveling‐wave sub‐Doppler excited molecule energy transferspectroscopy is presented. The line profile analysis is applied to that determined experimentally for the R(22) ν3 HCN transition. Pn the noise equivalent power of the detector is demonstrated to be ⩽10−12 W. Finally, the technique is applied to resolve the KsR(7) ν1 transition head in NH3.

Ab initio molecular orbital methods are used to study proton transfers in the cationic heterodimer (H3NHOH2)+ as well as the symmetric homodimers (H2OHOH2)+ and (H3NHNH3)+. All calculations are carried out at the Hartree–Fock level with a 4‐31G basis set to ensure consistency. The application of electrochemical techniques to biological systems has become an attractive method for characterizing the redox and electron transfer behavior of biomacromolecules. This study focuses on the role of soluble spinach ferredoxin (Fd) in photosynthesis where it functions as an electron carrier to membrane bound species in the plant chloroplast. An electrochemical model is used where in Fd now serves as a soluble redox species which undergoes oxidation and reduction via electron transfer at an electrode. Thin polymer films obtained by plasma polymerization usually show good biocompatibility when compared to classical biomaterials such as Silastic. The thicknesses of these films (from several hundreds of Å to several μm) make them suitable for the purpose of changing the surface properties of the substrate without altering its bulk properties. Standard reference markers for accurate, reproducible synchrotron x-ray energies are obtained using a three Si crystal spectrometer. The first two crystals are in the monochromator and the third is used to obtain diffraction markers which monitor the energy. Then for any value of the glancing angle on the reference Si crystal the energy for the (333) diffraction must occur at 3/4 that of the (444) and 3/5 of that for the (555). This establishes for the first time an absolute synchrotron energy scale. Higher-order diffractions are used to determine excitation line profiles. The stability of aqueous solutions of organic N-chloramines, suspected of contaminating chlorinated water, has been studied. Two factors influence the decomposition of solutions of N-chloropiperidine and N-chlorodiethylamine: a spontaneous decomposition and photodecomposition. Since solutions of these compounds are relatively long-lived, a need for an analytical method for their identification is discussed. A new method is described which involves reaction of organic N-chloramines with arenesulfinic acid salts. The method gives high yields of stable arenesulfonamides. Several toxicological studies of N-chloropiperidine are described. The compound is mutagenic by Ames assay in Salmonella typhimurium strain TA 100 and does not require metabolic activation …

The effects of micelle forming surfactants hexadecyltrimethylammonium bromide, sodium dodecyl sulfate and igepal CA~6JO on the visible spectra of merocyanine dyes of the form 1-alkyl-4-[(3', 5'-alkyl oxocyclohexadienylidene)ethylidene]- 1,4-dihydropyridine were studied. In all cases the visible spectrum shifted to the red when the dye was incorporated into the micelles. The magnitude of the shifts was found to be dependent on the substituted groups and the positioning of the chromophore of the dye in the micelle aggregates. The shifts indicate a highly hydrated environment about the surfactant head groups and an environment equivalent to that of pure ethanol solvent at some undetermined …

Bacteria of the genus Rhizobium are capable of forming a complex nitrogen fixing (fix+) symbiotic association with leguminous plants. The surface polysaccharides are believed to be essential in the symbiotic process. Earlier, we isolated eukaryotic initiation factor 2 (eIF-2)-stimulating protein (SP) as a homogeneous complex with eIF-2 (eIF-2-SP) and showed that, in the presence of Mg2+, eIF-2-SP promotes formation of a ternary complex with GTP and eukaryotic initiator methionyl tRNA (Met-tRNAi) (eIF-2-GTP-Met-tRNAi) catalytically. We now show that SP-bound eIF-2 exchanges with eIF-2 (eIF-2 exchange). Furthermore, in the presence of Mg2+, eIF-2-SP catalyzes the exchange of eIF-2-bound [3H]GDP with unlabeled GDP or GTP (GDP exchange) and the release of [3H]GDP when the ternary complex is formed from eIF-2-[3H]GDP, GTP, and [35S]Met-tRNAi. The interfacial electrochemistry of horse heart cytochrome c was studied at metal and metal oxide electrodes. Sperm whale myoglobin reactions at metal oxide and surface modified electrodes were also investigated. Emphasis was on the measurement and mechanistic interpretation of the heterogeneous electron transfer kinetic parameters for these electrode reactions.

The motivation for this study stems from the fact that some electron transfer proteins, e.g., cytochrome c, transfer electrons heterogeneously in physiological systems. Specific electrodes may, therefore, serve as suitable models for physiological surfaces. Electrode and membrane interfaces are both electrically charged and both have oriented hydration water layers.

Polystyrene bis(salicylaldehyde)-propylene-1,3-diiminato Cobalt (II) [salen] and Polystyrene bis(2,4-pentanedione)-propylene-1,3-diiminato Cobalt (II) [BAE] were prepared stepwise from chloromethylated polystyrene. In addition, a new preparation of a Schiff base was attempted resulting in Polystyrene bis(salicylaldehyde)-isobutylene-1,3-diiminoato Cobalt (II) being produced. Optimum reaction conditions were determined with regard tot ime, temperature, reaction ratios and solvent for each step for the reaction. The ability of the above named Cobalt (II) Schiff bases to oxidize 3-methyl indole to give the corresponding o-formylaminoacetophenone was also studied. The oxyanions of vanadium, chromium, molybdenum, arsenic, and selenium are stable forms of these elements in high oxidation states which cross cell membranes using the normal phosphate and/or sulfate transport systems of the cell. Once inside the cell, these oxyanions may sulfuryl transfer reactions. Often the oxyanions serve as alternate enzyme substrates but form ester products which are hydrolytically unstable compared with the sulfate and phosphate esters and, therefore, decompose readily in aqueous solution. The interstitial water chemistry of the sediments of Las Vegas Bay and Bonelli Bay in Lake Mead has been studied as part of a comprehensive water quality study of those locations. Pore water and solid phase analyses were completed from four stations in Las Vegas Bay and two stations in Bonelli Bay. At both locations the pore water compositions and organic matter diagenesis in the sediments are dominated by sulfate reduction. The transport and intracellular distribution of iron in Ustilago sphaerogena via the native siderochrome ferrichrome A has been examined. Evidence is presented which suggests the involvement of deferriferrichrome in iron transport when the metal is supplied to the cell as ferrichrome A. This process requires both oxygen and ATP synthesis and may require a transmembrane electrochemical gradient. Once inside the cell, iron derived from ferrichrome A is rapidly distributed, with a major fraction found in the mitochondria, presumably as heme.

This study was designed to accomplish two primary objectives: (1) to compare two test methods commonly used to evaluate toxicity of inhaled air pollutants, and (2) to observe the response as measured by each of the methods, of pulmonary alveolar macrophages exposed to 2μg hexavalent chromium.

The first method evaluated featured use of intratracheal injections to simulate live inhalation exposures, and the second required exposure of macrophages cultured on petri plates. Pulmonary alveolar macrophages harvested from Long Evans rats were used.
