Early detection of genetic mutations is important for control of diseases such as cancer and Alzheimer's. Early detection requires methods that detect small amounts of mutated DNA in very large amounts of normal or wild type DNA. One method to detect mutated DNA is the ligase detection reaction (LDR). Since its inception LDR has evolved greatly from a simple detection reaction after PCR amplification to PCR/RE/LDR, a scheme which uses nucleoside base analogs in PCR to convert wild type sequences to sequences containing restriction endonuclease (RE) sites which can then be cleaved leaving only mutant sequences for detection by LDR. …

The visual determination of saccharides has been of interest for well over a century. Sugars are a relatively challenging class of compounds to analyze. They exhibit great structural similarity as well as transparency in the visible region. The sensing of specific saccharides could aid the monitoring of disease or industrial fermentation products. Herein I present new methods allowing for simple and rapid visual detection of sialic acid, fructose and a homologous series of prototypical neutral oligosaccharides, the linear maltodextrins, containing up to seven glucose residues. The percentage composition of Al13, [AlO4Al12(OH)24(H2O)12]7+, in water treatment coagulants is an important criterion in the development and use of polymeric coagulants. Polymeric coagulants are generally used in cold climates or with highly turbid waters. Size exclusion chromatography–flame atomic absorption spectrometry (SEC-FAAS) can separate Al13 and monomeric Al within 6 min. Biotin is an essential growth factor found in all living cells. It functions as a cofactor for a group of coenzymes that catalyze transcarboxylation, decarboxylation, and carboxylation reactions. Acetyl CoA carboxylase catalyzes the first committed and regulated step in fatty acid synthesis in which malonyl CoA is the product. The biotin-dependent enzyme is found in all animals, plants, and bacteria. We studied the kinetic and structural aspects of acetyl CoA carboxylase, and in 1999 we reported the bioactivity and total synthesis of a unique first generation biotin-derived inhibitor of acetyl CoA carboxylase, 1(BP1). Using a direct ultraviolet second-derivative spectroscopy method, three peroxynitrite preparative methods were investigated for the nitrite and nitrate present either as impurities or produced during peroxynitrite decomposition: (I) ozonation of azide, reaction of hydrogen peroxide with (II) isoamyl nitrite and (III) nitrous acid. The oxidation of morpholine by peroxynitrite in the presence and absence of added carbonate gives N-nitromorpholine and N-nitrosomorpholine. Nitration and nitrosation of morpholine are catalyzed by low levels of CO2; however, excess CO2 dramatically reduces the yields of nitrosation but not nitration, and the combined yields of the products are about the same under …

New 5-alkyl and 3-(2,4-dimethylphenyl) substituted 1,3,4-oxadiazole-2-thione derivatives were synthesized by the ring closure reactions of various acylhydrazides with carbon disulphide. Mannich bases for some of these compounds were also synthesized by condensation with benzaldehyde and primary amines. All new compounds were characterized by spectral data. Most of them were tested for their antibacterial and antituberculostatic activity. Molecular orbital calculations at the HF/6-31G** level were also performed to determine the optimized geometrical structures. Results indicated electron delocalization over several atoms of the ring. Cyproheptadine hydrochloride (CPH), methdilazine hydrochloride (MDH) and promethazine theoclate (PMT) were determined in their pure state and in pharmaceutical formulations by a simple spectrophotometric method. The determination was based on the formation of a charge-transfer complex between chloranilic acid as a \pi-acceptor and the studied drugs as n-donors in an acetonitrile-chloroform mixture. The spectra, various experimental parameters, the stoichiometry and the stability of the complexes formed were investigated. The complexes formed were found to absorb at 520 nm. Beer's law is obeyed in the concentration ranges 25-125, 20-100 and 25-150 \mu g ml^{-1}, for CPH, MDH and PMT respectively. The …

Pleurotus sapidus} basidiospores immobilized onto Ca-alginate beads were used for the removal of cadmium and mercury ions from aqueous solutions. The biosorption of Cd(II) and Hg(II) ions on the alginate beads and both immobilized live and heat inactivated fungal mycelia of Pleurotus sapidus} was studied from aqueous solutions in the concentration range of 30-500 mg L^{ - 1}. The biosorption of Cd(II) and Hg(II) ions by the alginate and both live and heat inactivated immobilized preparations increased as the initial concentration of the ions increased in the medium. This study provides a new approach for the synthesis of pyridine and benzene templated potential peptidomimetics. By the attachment of various amino acid side chains in different combinations on the pyridine template at the 2- and 3- positions and on the benzene core at 1,2,4 positions, different peptidomimetics may be obtained. Arg-10, Leu-11 and Tyr-13 amino acid side chain mimics and their precursors were attached to the pyridine and benzene core with a special arrangement to produce potential peptidomimetics of omega-conotoxin MVIIA.

The most promising technologies to remove perchlorate from water are ion exchange and biological reduction. Although successful, ion exchange only separates perchlorate from water; it does not eliminate it from the environment. The waste streams from these systems contain the caustic or saline regenerant solutions used in the process as well as high levels of perchlorate. Biological reduction could be used to treat the regenerant waste solutions from the ion exchange process. A treatment scheme, combining ion exchange and biodegradation, is proposed to completely remove perchlorate from the environment. A review is given of the experimental determination of electron affinities (EAs). Also considered is the ability of computational chemistry methods to predict electron affinities. The development and present status of computational chemistry techniques for the prediction of atomic molecular EAs is also reviewed in a historical context. Through comparisons to experimental EAs and results from other theoretical methods, analysis of DFT EAs is given. In the context of other theoretical methods, the successes and failures of DFT EA predictions are detailed.

Multiphoton-absorption processes have recently attracted growing interest because of the potential impact on a wide spectrum of applications, ranging from data storage to photodynamic therapy. From the materials standpoint, particular interest is centered in designing chromophores that exhibit a large two-photon cross-section. The current research is focused on establishing synthetic routes that lead to the formation of derivatives of N,N-diphenyl-(7-benzothiazol-2-yl)-9,9-diethyl-9H-fluoren-2-ylamine which contain meta functionalization. A bromoethoxy functionalized chromophore, N-phenyl-N-(3-(2-bromoethoxy)phenyl)-7-(benzothiazol-2-yl)-9,9-diethyl-9H-fluoren- 2-ylamine , was synthesized and used as a polymer pendent while a vinylbenzyloxy chromophore, N-phenyl-N-(3-(4-vinylbenzyloxy)phenyl)-7- (benzothiazol-2-yl)-9,9-diethyl-9H-fluoren-2-ylamine, was synthesized as a hydrosilation substrate. 6-Thioguanine (65G) has been used for the past thirty years as an anti-cancer agent in maintenance chemotherapy for childhood acute lymphoblastic leukemia as well as acute myeloid leukemia. Despite its long-standing clinical use, little is understood of the molecular basis of the cytotoxicity associated with ?sG. Research has shown that ?sG is incorporated into DNA during replication after which it may exert its cytotoxicity by inducing rapid mutation or by affecting DNA-protein interactions. 65G has also been shown to impair HIV replication by inhibiting HlV-l reverse transcriptase and to inhibit human telomerase activity. HMG1 is a nonhistone chromosomal protein that binds preferentially to some types of globally modified DNA. HMG1 has been implicated in the cellular response to the anti-cancer agent cisplatin, functioning to block excision repair of specific distorted platinated DNA lesions and resulting in enhanced cytotoxicity. We have investigated whether other DNA-binding agents also produce lesions that are recognized by HMG1. Specifically, we used polyacrylamide gel shift assays to monitor the potential HMG1 binding of diepoxybutane interstrand cross-links, which have been suggested to induce DNA bending. The traditional format of electrophoresis, the slab gel, is quickly being replaced by capillary and microdevice platforms, which offer improvements in cost, resolution, speed, quantitation and automation in genetic analyses. These techniques also employ a variety of separation matrices while the slab gel is limited to agarose and polyacrylamide. The research presented here explores the use of these electrophoretic formats for the detection of single nucleotide mutations using two genetic models associated with human disease. The gene encoding for the methionyl aminopeptidase from the hyperthermophilic archaeon Pyrococcus furiosus (PfMetAP-II; EC 3.4.11.18) has been inserted into a pET 27b(+) vector and overexpressed in Escherichia coli. The new expression system resulted in a 5-fold increase in purified enzyme obtained from a 5 L fermentor growth. The as-purified PfMetAP-II enzyme, to which no exogenous metal ions or EDTA was added, was found to have 1.2 equiv of zinc and 0.1 equiv of iron present by ICP-AES analysis. This enzyme had a specific activity of 5 units/mg, a 60-fold decrease from the fully loaded Fe(II) …

During neurotransmitter release, the neuronal SNARE proteins synaptobrevin/VAMP, syntaxin, and SNAP-25 form a four-helix bundle, the SNARE complex, that pulls the synaptic vesicle and plasma membranes together possibly causing membrane fusion. Complexin binds tightly to the SNARE complex and is essential for efficient Ca2+-evoked neurotransmitter release. A combined X-ray and TROSY-based NMR study now reveals the atomic structure of the complexin/SNARE complex. Complexin binds in an antiparallel α-helical conformation to the groove between the synaptobrevin and syntaxin helices. This interaction stabilizes the interface between these two helices, which bears the repulsive forces between the apposed membranes. These results …

C.I. Acid Orange 7 (15510) is the sodium salt of p-[(2-hydroxy-1-naphtalenyl)azo] benzenesulfonic acid. This azo dye is commonly known as Orange II and has been used to dye soaps, shoes, wood and paper (Ramchandani et al., 1994). In the United States and Europe, this dye is not allowed in foodstuffs. Some new triorganotin carboxylates containing germanium with general formulae (R^{1}GeCHR^{2}CH_{2}COO) Sn(CH_{2}C(CH_{3})_{3})_{3} where R^{1} = N(CH_{2}CH_{2}O)_{3} and C_{6}H_{5}, R^{2} = C_{6}H_{5}, p-C_{6}H_{4}OCH_{3} and o-/p-C_{6}H_{4}F were synthesized and characterized by elemental analysis, infrared, multinuclear (^{1}H, ^{13}C, ^{119}Sn) NMR and mass spectrometry. Antibacterial activity of these compounds was evaluated against the standard drugs ampicilline and amoxicilline, and found to be more active in some cases than the standard drugs.

Systems formed by the pyrene derivatives 3-hydroxypyrene and 1-aminopyrene with pyridine were studied. The thermodynamic and kinetic parameters of the systems were calculated using the changes in the absorption and emission spectra which occur due to hydrogen bond or complex formation. The decrease of the formation rate constant in the series methylcyclohexane, benzene, and o-chlorotoluene may be due to a more difficult formation of the hydrogen bond complex due to the increase of the dielectric constant in the solvent series.

A new series of imidazole nucleosides was synthesized via direct condensation and construction of heterocyclic moiety from glycosyl amine templates. The characterization of the compounds was accomplished by elemental analysis, ^{1}H-NMR, ^{13}C-NMR, IR, UV-VIS spectral data and optical rotation.

Application Of Quantitative Structure-Retention Relationships (Qsrr) To A Set Of Organic Bromo And Nitrile Derivatives, Gustavo P. Romanelli, Juan Carlos Autino, Eduardo A. Castro

The retention times are studied for two sets of 1-bromo-2-aryiloxyetanes and 3- aryloxypropiononitriles derivatives by means of Quantitative Structure-Retention Relationships (QSRR}). Five quantum mechanical molecular descriptors are used to calculate the regression equations. The fitting polynomials are computed in several-variable at first, second, and third order equations. Results are only significant when resorting to several-variable formulae which seems to point out the rather complex physical chemistry nature of the property under study.

An Indirect Lectin Affinity Bioassay For Uromodulin, Zeki̇ Topcu

Uromodulin is a monomeric glycoprotein excreted in large amounts in urine. Most of the assay methods for quantifying uromodulin rely on the availability of antibodies to this glycoprotein. This study reports a new bioassay for uromodulin without requiring the presence of antibodies. The bioassay is based on the lectin affinity of the glycoprotein and the known interaction between uromodulin and the tumor necrosis factor. The measuring range of the bioassay described in this paper is between 0.13 and 1.25 \mu g/ml, which makes it a potential method for clinical use.

A simple, convenient and accurate indirect titrimetric method and a rapid, sensitive and precise indirect spectrophotometric method for the determination of six phenothiazine drugs using chloramine-T as an oxidant are described. The titrimetric method is based on the oxidation of the drugs in sulphuric acid medium by a known excess of chloramine-T and iodometric determination of the unreacted chloramine-T. In the spectrophotometric method, the unreacted oxidant is made to react with metol in the presence of an aromatic amine to produce a purple-red product with the absorption maximum at 520 nm. The phenolic \beta-diketones I prepared by modified Baker-Venkataraman rearrangement were converted to the flavones II in acidic medium which on treatment with aqueous ethylenediamine/propylenediamine gave diazepine derivatives III. After coupling with acetobromo sugars in the presence of mercuric cyanide and nitromethane, deacetylation in methanolic ammonia yielded nucleosides V. The structures of all the intermediates and final products were confirmed with the help of modern spectroscopic techniques.

The research presented in this dissertation involves the synthesis, characterization, and the use of novel surfactants, including both micelles and vesicles, as pseudostationary phases in micellar capillary electrophoresis (MCE) for the separation of achiral and chiral compounds. Separation of environmental pollutants such as 2 to 6-ring polycyclic aromatic hydrocarbons (PAHs) was achieved using poly(sodium undecylenic sulfate). A baseline separation of all 16 PAHs was possible for the first time in MCE by a single-surfactant system. Short peptides containing Cα,α-dipropylglycine (Dpg) at alternating sequence positions were synthesized and examined for conformational behavior. Peptide assembly was performed using Fmoc-solid-phase chemistry where the coupling with PyAOP could be significantly enhanced at elevated temperature. Circular dichroism (CD) and NMR conformational studies revealed that incorporation of Dpg residues induced folded structures into peptides. It was observed that Dpg residues adopted helical conformation in a helix-promoting sequence. The resulting helical structure was comprised of consecutive β-turns whose structure was stabilized by salt bridge in aqueous solution. The stable isotope compositions of organic carbon (OC) and nitrogen, contents of OC and nitrogen, and C/N ratios for four short cores, which were recovered from Msansani Bay (WO 57), off Msimbazi River (WO 76) and Dar es Salaam harbour (WO 62 and WO 74), are used to document temporal variation in the sources and diagenetic alteration of organic matter (OM).
