Diepoxybutane (DEB), a known industrial carcinogen, reacts with DNA primarily at the N7 position of deoxyguanosine residues and creates interstrand cross-links at the sequence 5'-GNC. Since N7-N7 cross-links cause DNA to fragment upon heating, quantative polymerase chain reaction (QPCR) is being used in this experiment to measure the amount of DEB damage (lesion frequency) with three different targets-mitochondrial (unpackaged), open chromatin region, and closed chromatin region. Initial measurements of DEB damage within these three targets were not consistent because the template DNA was not the limiting reagent in the PCR. Follow-up PCR trials using a limiting amount of DNA are …

A series of tri- and diorganotin(IV) compounds with the general formulae R_{3}SnL and R_{2}SnL_{2} (where R = Me, n-Bu, Ph and L = 2-[N-(2,6-dichloro-3-methylphenyl)-amino]benzoate) were synthesized. These compounds were characterized by different instrumental methods such as infrared, multinuclear NMR (^{1}H, ^{13}C, ^{119}Sn) and mass spectrometry. The spectroscopic investigation illustrates that the carboxylate group acts as a bidentate in solid state and as a monodentate in solution. Biological screening tests reveal that the investigated compounds have potential as antibacterial agents.

The 7-hydrazino-3,4-diphenyl-2H-pyrazolo-[3,4-d]-pyridazine 3, obtained in 2 different ways consisting of the reactions of anhydrous hydrazine with 4-benzoyl-5-phenyl-2,3-dihydro-2,3-furandione 1 or 3,4-diphenyl-2H-pyrazolo-[3,4-d]-pyridazin-7(6H)-one 2, was converted into hydrazone derivatives 4 by the reactions of 3 with various salicylaldehydes.

In this study, the reaction between Co(III)-EDTA and hypophosphite ion, catalyzed by Pd(II), was chosen as the indicator reaction. The inhibition kinetics of this catalytic reaction were investigated in the presence of iodide ion and the possibility of its analytical application was evaluated. Catalyzers other than PdCl_2 (Pt, Au and Ni salts) were assayed for the indicator reaction and it was observed that these catalysts have no effect on the reaction. The important variables that affected the reaction rate were investigated and the optimum conditions giving maximum sensitivity were established. Inmunofluorescence staining of murine NIH3T3 fibroblasts grown at high density shows that conventional nucleoside diphosphate (NDP) kinases A and B localize to a sensory organelle, the primary cilium. Similar results are obtained with Xenopus A6 kidney epithelial cells, suggesting that NDP kinases are a universal component of the primary cilium. The translocation of NDP kinase into primary cilia depends on size, taking place only when cilia reach a critical length of 5-6 μm. In mature cilia, NDP kinases are distributed along the ciliary shaft in a punctate pattern that is distinct from the continuous staining observed with acetylated α-tubulin, a …

This work provides the first extensive study of the redox reactivity of the pyranopterin system that is a component of the catalytic site of all molybdenum and tungsten enzymes possessing molybdopterin. The pyranopterin system possesses certain characteristics typical of tetrahydropterins, such as a reduced pyrazine ring; however, it behaves as a dihydropterin in redox reactions with oxidants. Titrations using ferricyanide and dichloroindophenol (DCIP) prove a 2e(-)/2H(+) stoichiometry for pyranopterin oxidations. Oxidations of pyranopterin by Fe(CN)(6)(3-) or DCIP are slower than tetrahydropterin oxidation under a variety of conditions, but are considerably faster than observed for oxidations of dihydropterin. The potential value of the Y chromosome for forensic applications has been recognized for some time with the current work dedicated to Short Tandem Repeat analysis and Single Nucleotide Polymorphism (SNP) discovery. This study examined the ability of two different SNP analysis methods to determine if they could be utilized in forensic applications and ultimately be developed into an established system for Y chromosome SNP analysis. Photodynamic therapy (PDT) has been investigated over the past three decades and is currently an approved therapeutic modality for skin cancer, the treatment of superficial bladder, early lung and advanced esophageal cancers, and age-related macular degeneration in a number of countries. In PDT, the absorption of light by a chromophore generates cytotoxic species such as reactive singlet oxygen, leading to irreversible destruction of the treated tissue. The measurement of the singlet oxygen quantum yield is an important determinant used to evaluate the efficiency of new photodynamic therapy agents developed in the laboratory, to screen potential photosensitizers in aqueous media.The singlet …

Three dimensional random walk simulations were used to model the diffusional interactions between coplanar dual disk microelectrodes. Working curves for the dependence of shielding factor, collection efficiency, and amplification factor on the size of the gap between the electrodes were developed. The simulations showed that when the gap size was ≥22 radii, the electrodes could be considered as independent and diffusionally isolated. Empirical equations for determining the gap size from simple shielding and generator-collector experiments were established. The simulation was used to model the collector electrode response for square waves of various frequencies applied to the generator electrode. Simulation results …

Poly-L-lactic acid (PLLA) is an optically active, biocompatible and biodegradable polymer that has been widely investigated as an artificial cell scaffold material. In its most crystalline form, PLLA is highly anisotropic and is one of the most piezoelectric polymers known. Conversely, amorphous PLLA exhibits little, if any, piezoelectric behavior. Compression molded PLLA films can be endowed with varying amounts of crystalline character and piezoelectricity by uniaxially stretching the polymer in a hot air bath. Understanding the precise crystalline architecture of PLLA that results from tensile drawing is important for constructing cell scaffolds that have highly tailored biodegradation and cell guiding …

Nicotinic acid is shown to be comparable to dihydropyridine in its capacity to facilitate penetration of an attached antibacterial drug through dermal layers. Antibacterial drugs examined with nicotinic acid or dihydropyridine carriers were b-lactam antibiotics: methicillin, oxacillin, benzylpenicillin, penicillin F, penicillin dihydro F, propicillin, carbenicillin, penicillin K, penicillin X, and ampicillin. An oxymethyl (-O-CH2-) group is inserted as the linker between the antibiotic and the carrier group. Structure Property Correlations and multivariate methods such as regression analysis, cluster analysis, principal component analysis, discriminate analysis, self-organizing tree algorithm, and factor analysis clearly showed that nicotinic acid performs as an effective carrier …

Novel bifunctional five-membered cyclic dithiocarbonates (1,3-oxathiolane-2-thione)s were synthesized by the reactions of the corresponding bifunctional oxiranes (epoxides) with carbon disulfide at room temperature with lithium bromide as catalyst. Full characterization of these monomers was performed including elemental analysis, proton and carbon nuclear magnetic resonance (NMR) spectroscopy, gas chromatography-mass spectroscopy, and Fourier transmission infrared (FTIR) spectroscopy. The polyaddition polymerization of 1,3-oxathiolane-2-thione with 1,4-diaminobutane at room temperature resulted in a poly(thiourethane) material. The latter undergoes crosslinking due probably to the autooxidation of the product and formation of disulfide linkages. As a class, amides are of great interest in biological studies and pharmaceutical application. In this work, podocarpic acid, a natural tricyclic diterpene, derived from Podocarpus species, has been employed to form a novel family of amide derivatives which will later be studied for their potential as new drug leads. Novel amide derivatives of podocarpic acid were synthesized from podocarpic acid in three steps. The first step involved methylation with dimethylsulfate to form methyl-O-methylpodocarpate. This step was followed by iodination with iodine to give iodomethyl-O-methylpodocarpate. Finally amidation with various aliphatic amides using a copper catalyst yielded four amide derivatives of …

Emissions of polychlorinated dibenzo-p-dioxins and furans (PCDD/Fs) and polybrominated dibenzo-p-dioxins and furans (PBDD/Fs) from hazardous waste incinerators, and many other sources for combustion have been considered environmentally hazardous and a major health threat. Recently, a growing number of materials containing brominated hydrocarbons, commonly used flame retardants, have been disposed in municipal and hazardous waste incinerators. This results in the increased potential for formation of PBDD/Fs and other hazardous combustion by-products. In contrast to chlorinated hydrocarbons, the reactions of brominated hydrocarbons have been studied only minimally. This dissertation is concerned with random coil and rodlike polymer diffusion in complex solutions. The development of fluorescence photobleaching recovery, FPR, as a technique to determine polymer molecular weight distributions by measuring the diffusion distribution and converting it to a molecular weight distribution using a calibration curve is first covered. Fluorescently tagged probes were individually dissolved in polymer solutions at negligible amounts and measured with FPR to construct the calibration curve of self-diffusion vs. molecular weight. The colorimetric properties of resorcinarene solutions had not been fully investigated since von Baeyer's initial synthesis. We find that the solutions containing resorcinarene macrocycles develop color upon heating or standing. In the presence of saccharides, these solutions exhibit significant color changes which are readily observable by visual inspection. We determined that the solution color is due to macrocycle ring opening and oxidation. The optical responses to saccharides are due to complexation of the sugar with the acyclic chromophores. We have applied these mechanistic insights towards the challenging problems of the visual detection of saccharides and other important biologically analytes. In …

Hierarchical construction and characterization of core-shell composite particles of interest are presented. The research described shows interest in new types of polymers that provide chirality and responsiveness which have been ignored for so long. The general synthetic design described in this document can be readily used as a model for other systems. Core-shell composite systems of interest consist of silica or silica-coated cobalt cores grafted with a homopolypeptide shell, mostly poly(epsilon-carbobenzyloxy-L-lysine) or poly(gamma-benzyl-L-glutamate). Core particles were decorated with amino groups through a silylation reaction. A series of substituted 2-phenacyl-3-phenyl-1H-pyrrole-4-carboxylates were prepared from substituted acetophenones in 6 steps. The final condensations between a chloroenal and an aminoketone were carried out under neutral conditions in parallel to yield the series listed below. Selected pyrrole derivatives proved to be potent hypolipidemic agents lowering serum triglyceride concentrations in CF-1 male mice after 14 days of I.P. administration. One agent orally lowered serum cholesterol in Sprague-Dawley male rats at 2mg/kg/day after 14 days. The agents demonstrated a lowering of mouse serum LDL- cholesterol levels and selected compounds showed an elevation of serum HDL-cholesterol levels. The cholesterol concentrations in the …

The adiabatic bomb calorimeter has been an effective tool in facilitating heat transfer between molecules via combustion reaction. Heat is released from the substance combusted and transferred into another, usually water. The heat transferred is measured and the enthalpy of combustion determined for the combusted material. One such classic experiment is the determination of combustion enthalpy of sucrose, obtaining the calorimeter constant with benzoic acid. The results of an adiabatic combustion experiment can be taken one step further to determine the combustion enthalpy in kcal/g by a simple conversion calculation. Synthesis of new 1,2,4-tri and 1,3,4-thiadiazoles, bearing isomeric pyridyl and 1-naphthyl is reported using 1,4-disubstituted thiosemicarbazides in alkaline and acidic media, respectively. The methylthio and benzylthio derivatives of the synthesized triazoles are also reported. All of the synthesized compounds were characterized by their FT-IR, ^{1}H-NMR and mass spectral data. The antibacterial studies of some of the synthesized compounds against S. aureus} and E. coli} as MIC values are reported. None of them have important antibacterial activities.

Crystal morphologies are predicted for vitamin C (L-ascorbic acid) using attachment energy methods. The significant differences between predicted and observed morphologies when vitamin C is grown from water, isopropyl alcohol, acetone-water solution, and isopropyl alcohol-methanol solution are attributed to the effect of solvent on the growth rate of the various crystal habit faces. This effect, particularly the role of hydrogen bonding between solvent and crystal surface, is examined. Molecular modeling is combined with the results of simple experiments charting the habit shift of vitamin C as a function of the hydrogen bonding tendency of the solvent or solvent mixture. A set of 14 mono-organotin(IV) derivatives of general formulae n-BuSnClL_2 and n-BuSnL_3, where `L' are various non-steroidal anti-inflammatory drugs (NSAIDs), were synthesized and structurally characterized by vibrational, ^1H, ^{13}C and ^{119}Sn spectroscopies and mass spectrometry to propose their structure. Biological activity data (antibacterial, antifungal and cytotoxicity) of the synthesized complexes were compared with the precursors to establish the significance of the complexes.

This paper presents the first ice core measurements of methyl bromide (CH3Br). Samples from a shallow Antarctic ice core (Siple Dome, West Antarctica), ranging in mean gas dates from 1671 to 1942, had a mean CH3Br mixing ratio of 5.8 ppt. These results extend the existing historical record derived from air and Antarctic firn air to about 350 years before present. Model simulations illustrate that the ice core results are consistent with estimates of the impact of anthropogenic activity ( fumigation, combustion, and biomass burning) on the atmospheric CH3Br burden, given the large current uncertainties in the modern atmospheric CH3Br …

The research presented in this dissertation involves the synthesis, modification, characterization, and the application of polymeric surfactants in micellar electrokinetic chromatography (MEKC). Sodium undecenoyl-L-Leucinate (L-SUL) was synthesized and modified by use of alkali metal counterions, alcohols, and Triton X-102. Characterization of the modified poly-L-SUL was performed in order to elucidate the structural changes on the surfactants by use of four techniques. While surface tensiometry was used for the determination of the critical micelle concentration (cmc), fluorescence measurements were used for the determination of the polarity of the modified polymeric surfactants.
