Novel Analogues Of 2-Pyrazoline: Synthesis, Characterization, And Antimycobacterial Evaluation, Ahmet Özdemi̇r, Gülhan Turan Zitouni, Zafer Asim Kaplancikli

Fourteen new 1-[(N,N-disubstitutedthiocarbamoylthio)acetyl]- 3-(2-thienyl)-5-aryl-2-pyrazoline derivatives were synthesized by reacting 1-(chloroacetyl)-3-(2-thienyl)-5-aryl-2-pyrazolines and appropriate sodium salts of N,N-disubstituted dithiocarbamoic acids in acetone. The structures of the synthesized compounds were confirmed by UV, IR, ^1H-NMR, and FAB^+-MS spectral data. The compounds were evaluated for in vitro antimycobacterial activity against Mycobacterium tuberculosis H37Rv using the BACTEC 460 radiometric system and BACTEC 12B medium. The preliminary results showed that all of the tested compounds were inactive against the test organism.

The electrocatalytic behavior of hydroxylamine was studied on a glassy carbon electrode modified by electrodeposition of quinizarine, using cyclic voltammetry, chronoamperometry, and rotating disk voltammetry as diagnostic techniques. Cyclic voltammetry showed that the catalytic current of the system depends on the concentration of hydroxylamine. The magnitude of the peak current for quinizarine increased sharply in the presence of hydroxylamine and proportional to hydroxylamine concentration. The diffusion coefficient of hydroxylamine and the catalytic rate constant for the catalytic reaction of quinizarine with hydroxylamine were also estimated using a rotating disk electrode experiment. In this study, (hydroxyimino)(2-phenyl(1,2,3,4-tetrahydroquinazolin- 2-yl))methane (H_2L^1) and (hydroxyimino)(2-(2-thienyl)(1,2,3,4- tetrahydroquinazolin-2-yl))methane (H_2L^2) were synthesized by the condensation of 2-(hydroxyimino)-1-phenylethan-1-one (INAP) and 2-(hydroxyimino)-1-(2-thienyl)ethan-1-one (INAT) with 2-aminobenzylamine (2-ABA). Complexes of these ligands with Co^{3+} were prepared with a metal:ligand ratio of 1:2. The ligands and their complexes were elucidated on the basis of elemental analyses, AAS, FT-IR, ^1H- and ^{13}C-NMR spectra, mass spectra, magnetic susceptibility measurements, and molar conductivity.

Chemical fate models can be used to estimate the transformation reactions and the partitioning behavior of pharmaceuticals and personal care products (PPCPs) in sewage treatment plants (STPs). Modeling the fate of PPCPs in a STP requires parameters such as rate constants or partition coefficients that are often unique to a specific STP, and that may exhibit significant temporal variability. Chemical fate models provide both a valuable test of our understanding of the fundamental underlying mechanisms regarding the fate of PPCPs in STPs, as well as a means of providing both qualitative and quantitative estimates for PPCP removal. To determine chromophoric dissolved organic matter (CDOM) sources in Southern California coastal waters, optical properties of a river outlet and adjacent tidally flushed salt marshes were monitored (dry season; June July 2001). Average absorption coefficients doubled at ebb vs. flood tides (4.8 +/- 1.5 vs. 2.1 +/- 0.9 m(-1); 300 nm), suggesting significant salt marsh CDOM inputs into coastal waters. Average spectral slopes were not statistically different for any sites or tides (0.010 +/- 0.002 nm(-1)), consistent with salt marsh CDOM dominating coastal waters. Three-dimensional fluorescence excitation-emission matrices (EEMs) at ebb tide showed contributions from terrestrial, protein, and marine humic-like …

Crystals of the title compound [systematic name: (3R,6R,7S,8aR,9R,12aR)-7-hydroxy-3,6,9-trimethyloctahydro-3,12-epoxy[1,2]dioxepino[4,3-i]isochromen-10(3H)-one], C15H22O6, were obtained from microbial transformation of artemisinin by a culture of Cunninghamella elegans. The stereochemistry of the compound is consistent with the spectroscopic findings in previously published works. A weak O—H⋯O hydrogen bond occurs in the crystal structure, together with intermolecular C—H⋯O hydrogen bonds.

We have developed a new imaging method for SECM employing fast scan anodic stripping voltammetry (ASV) to provide sensitive and selective imaging of multiple chemical species at interfaces immersed in solution. A rapid CV scan (100 V/s) is used along with a short preconcentration time (300-750 ms) to allow images to be acquired in a normal SECM time frame. A Hg-Pt film electrode is developed having an equivalent Hg thickness of 40 nm that has good sensitivity at short preconcentration times and also retains thin-film behavior with high-speed voltammetric stripping. Material properties of industrial polymer blends are of great importance. X-ray tomography has been used to obtain spatial chemical information about various polymer blends. The spatial images are acquired with synchrotron X-ray tomography because of its rapidity, good spatial resolution, large field-of-view, and elemental sensitivity. The spatial absorption data acquired from X-ray tomography experiments is converted to spatial chemical information via a linear least squares fit of multi-spectral X-ray absorption data. A fiberglass-reinforced polymer blend with a new-generation flame retardant is studied with multi-energy synchrotron X-ray tomography to assess the blend homogeneity. A diverse set of human diseases is associated with the misfolding of proteins into insoluble fibrillar structures that have predominantly β-sheet secondary structure. Thus it is imperative to elucidate the structural elements that contribute to β-sheet formation and stability. Peptides that autonomously form β-sheets are ideal models to study principles of protein folding and design. Gellman and coworkers first introduced an automonously folded β-hairpin (H-Arg-Tyr-Val-Glu-Val-Yyy-Xxx-Orn-Lys-Ile-Leu-Gln-NH2) that remained monomeric up to ~1 mM. Rapid rotation of guanine base derivatives about Pt–N7 bonds results in fluxional behavior of models of the key DNA intrastrand G–G cross-link leading to anticancer activity of Pt(II) drugs (G = deoxyguanosine). This behavior impedes the characterization of LPtG2 models (L = one bidentate or two cis-unidentate carrier ligands; G = guanine derivative). The objective of this study is to understand the types of conformers formed as L is systematically varied. Catechol, hydroquinone and Phenol are major constituents of the mainstream tobacco smoke. The toxicity of tobacco has been attributed to the ability of catechol and hydroquinone to undergo endogenous or exogenous redox cycling to form semiquinone type radicals responsible of Reactive Oxygen Species (ROS) formation. ROS such as hydroxyl radicals can cause severe oxidative stress on biological tissues and can provoke severe signaling pathways leading to cardiovascular and pulmonary dysfunctions and carcinogenesis. In situ atomic force microscopy (AFM) characterizations and lithography can be applied to investigate the orientation, reactivity and stability of protein molecules adsorbed on nanostructures of self-assembled monolayers at near-physiological conditions. Automated nanografting was used to fabricate regular arrays of nanopatterns of ù-functionalized n-alkanethiols with designated terminal chemistries. After writing nanopatterns, protein binding occurs selectively on carboxylate-terminated nanopatterns via covalent bonds that are formed using N-ethyl-N'(dimethylaminoporpyl)-carbodiimide and N-hydroxysuccinimide activation. The amine groups of lysine residues of proteins bond covalently to nanopatterns of carboxylate-terminated alkanethiol self-assembled monolayers, to form a robust surface attachment for sustained contact-mode AFM imaging during biochemical reactions. …

Chapter I introduced the background of phthalocyanines including its physical properties and various synthetic methods of free-base and metallo-phthalocyanines. The background of PDT and BNCT was introduced and the objective of ideal sensitizers was described. Chapter II described the synthesis and characterization of a series of pyridiloxy-substituted zinc phthalocyanines with different length of alkyl groups. Pyridiloxy-substituted silica phthalocyanines bearing different associated axial ligands, were alkylated with methyl or PEG chains. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry was used for off-line analysis of bioaerosols. The overall goal of the work is to develop methods and instrumentation to minimize sample treatment and maximize sensitivity and selectivity for bioaerosol analysis. This work is divided into three parts: sample preparation, test bioaerosol analysis, and instrument development. Untreated silicon wafer substrates were used as an alternate to conventional stainless steel targets and proved to be excellent substrates for large molecule analyses as well as small molecule analyses with excellent spot-to-spot and shot-to-shot reproducibility, which is useful for high-throughput and automated sample analysis. This dissertation explores the development of analytical methods for studies of CVD biomarkers and related biomolecular indicators. Initially, spectroscopic studies were conducted to investigate the chemical reactivity of homocysteine (Hcy), an independent CVD risk factor and serological biomarker. Consequently, we proposed an alternate theory for in vivo Hcy clearance based on spontaneous pyridoxal tetrahydrothiazine (PT) formation from Hcy and pyridoxal. The validity of PT-assisted Hcy clearance was further evaluated by use of capillary electrophoretic methods, which allowed rapid monitoring of protein oligomerization in PT-protein reaction mixtures. "In the quest of building mechanically strong materials with low density and high porosity, polymer crosslinked aerogels stand as the most promising nano-engineered examples. Covalent attachment of polymers and bridging of skeletal nanoparticles of typical aerogels is demonstrated by using surface initiated polymerization (SIP) with a bidentate free-radical initiator structurally related to azobisisobutyronitrile (AIBN) and confined on mesoporous silica surfaces. Different monomers were introduced in the mesopores and upon heating at 70⁰C, all mesoporous surfaces throughout the entire skeletal framework were coated conformally with a 10-12 nm thick polymer layer indistinguishable spectroscopically from the respective commercial bulk materials. Polychlorinated biphenyls are a ubiquitous environmental contaminant that can be found today throughout the world in soils and sediments, lakes and rivers, and flora and fauna. PCBs have percolated throughout the food chain, so that almost every human being has a detectable amount of the contaminant within their blood stream. Nanoparticles possessing multiple functionalities provide synthetic handles for varied surface chemistries, making them useful for a range of applications such as biotargeting and drug delivery. However, the combination of interfering functionalities on the same particle is often challenging. We have employed a synthetic scheme involving chemical protection/deprotection to combine interfering functional groups on the same hydrogel nanoparticle. The synthesis of amine-containing poly(N-isopropylacrylamide) nanogels was carried out via free radical precipitation polymerization by incorporating a Fmoc-protected amine poly(ethylene glycol) (PEG) macromonomer. The Fmoc group was then removed to obtain free amines, which were shown to be available for conjugation. We further …

Previous studies have demonstrated that cyclooxygenase-2 (COX-2) inhibitor NS-398 decrease aromatase activity at the transcript level in breast cancer cells. However, N-Methyl NS-398, which does not have COX-2 inhibitory activity but has very similar structure to NS-398, decreases aromatase activity and transcription in MCF-7 and MDA-MB-231 breast cells to the same extent as NS-398. This suggests that NS-398 decrease aromatase expression in breast cancer cells via other mechanism(s). The paranemic crossover (PX) is a motif for assembling two nucleic acid molecules using Watson-Crick (WC) basepairing without unfolding preformed secondary structure in the individual molecules. Once formed, the paranemic assembly motif comprises adjacent parallel double helices that crossover at every possible point over the length of the motif. The interaction is reversible as it does not require denaturation of basepairs internal to each interacting molecular unit. The dimensions of microfluidic devices closely parallel those of biological cells; thusly, they are excellent platforms for the speciation, transport, manipulation, and analysis of cells. Electrokinetic transport of Escherichia coli and Saccharomyces cerevisiae was evaluated in microfluidic devices fabricated in pristine and UV-modified poly(methylmethacrylate) and polycarbonate. The magnitude and direction of transport of the cells was dictated by the buffer composition, conduit surface chemistry, and intrinsic cellular electrical properties. This paper will give an example of a pedagogical approach taken in integrating nanotechnology into a chemistry degree course. In recent years nanotechnology has widely become part of the course content for undergraduate chemistry and physics degree curriculum. How contextualised the delivery of the subject matter may vary.
