ABSTRACT

A series of bis(3,4-ethylenedioxythiophene)-thiophene derivatives (BEDOT-Ts) and bis(3,4-ethylenedioxythiophene)-benzene derivatives (BEDOT-Bs) monomers were electrochemically polymerized onto glassy carbon or ITO-coated glass electrodes. Cyclic voltammetry (CV), infrared (IR), UV-vis-NIR and photoluminescence (PL) spectroscopy were employed to investigate and characterize properties of resulting polymers. dc-conductivities of these polymers were measured by the four-point probe method.
Cyclic voltammetry studies show that the redox half potentials of PBEDOT-3BT, PBEDOT-3OT, PBEDOT-3TBS and PBEDOT-3MPT are -290, -297, -325 and -284 mV versus Ag/Ag+, respectively. The redox half potentials of PBEDOT-TOL, PBEDOT-BOB and PBEDOT-FB are -130, -147 and -327 mV vs. Ag/Ag+, respectively, and a bit higher …

Beckmann Rearrangement Of Cyclotriveratrylene (Ctv) Oxime: Tandem Beckmann-Electrophilic Aromatic Addition, Marlon R. Lutz Jr., Matthias Zeller, Daniel P. Becker

Chemistry: Faculty Publications and Other Works

The Beckmann rearrangement has been performed on the oxime of cyclotriveratrylene (CTV) with thionyl chloride affording the ring-expanded 10-membered ring amide exclusively in high yield. Modified conditions afford a helical pentacycle derived from an unusual tandem Beckmann rearrangement and electrophilic aromatic addition followed by demethylation and tautomerization.

Structure Of The Thioredoxin-Like Domain Of Yeast Glutaredoxin 3, Lydia M. Gibson, Nin N. Dingra, Caryn E. Outten, Lukasz Lebioda

Faculty Publications

Yeast glutaredoxin 3 (Grx3) is a cytosolic protein that regulates the activity of the iron-responsive transcriptional activator Aft1. This member of the monothiol glutaredoxin family contains a thioredoxin-like domain and a glutaredoxin-like domain, which both possess a monothiol active site. The crystal structure of the thioredoxin-like domain has been determined at 1.5 Å resolution and represents the first published structure of this domain for the monothiol glutaredoxin family. The loop containing the signature motif WAxxC is partially disordered, indicating a greater degree of flexibility in this region compared with classical dithiol thioredoxins with a WCGPC active-site motif.

I. Thermodynamics And Magnetism Of Cu2ocl2 Ii. Repairs To Microcalorimeter The "2"S Are Subscripts, And The Second 2 Is Preceded By A Lower Case L, Not A One, Thomas J. Parry

Theses and Dissertations

Adiabatic calorimetry provides accurate and precise specific heat (Cp) data. From this data, thermodynamic functions may be calculated. Cu2OCl2, melanothallite, became of interest as part of a study of a particular thermochemical cycle. The experimental specific heat data and the calculated thermodynamic functions are reported here. Free energies of formation, calculated from the thermodynamic functions, suggest the particular cycle of interest with this compound as an intermediate is not feasible; uncertainty as to the accuracy of CuO and CuCl2 data used in the calculations indicate further study may be necessary. Upon collection of the specific heat data, an antiferromagnetic transition …

Synthesis Towards Fulminic Acid And Its Derivatives In 1, 3-Dipolar Cycloaddition Reactions., Ophilia Ndi Toh

Electronic Theses and Dissertations

A new approach to fulminic acid cycloadditions has been developed. At reduced temperatures, fulminic acid is generated in situ and undergoes 1, 3-diploar cycloaddition reactions with dipolarophiles to form isoxazolines and/or its dimers. This procedure represents a novel, safe general method for the one-step generation of fulminic acid, which complements existing potentially explosive protocols.

This study aims to determine the distribution of particulates carrying heavy metals at selected sites on a college campus using dandelion leaves as collectors. As a comparison, sites far away from the campus surrounding Bristol Motor Speedway Car Racing Stadium were also monitored. To reduce the probability of memory effects from the long-term atmospheric deposition or absorption of metals from soil a seasonal plant, dandelion, was used to monitor the metal contents. The metals monitored are cadmium, calcium, copper, chromium, iron, lead, and zinc. Fourteen sites were monitored and samples were collected once a week initially (growing time of the …

A Radical Conjugate Addition Approach To The Total Synthesis Of Celogentin C, Steven G. Capps Aug 2008

A Radical Conjugate Addition Approach To The Total Synthesis Of Celogentin C, Steven G. Capps

Theses and Dissertations

The synthesis of five chiral DBFOX (dibenzofuran-oxazoline) ligands with either aryl or benzyl substituents will be presented. The requisite amino alcohols were obtained with high enantioselectivity either commercially (DBFOX/Bn), via Sharpless asymmetric aminohydroxylation (DBFOX/Nap, DBFOX/t-BuPh, DBFOX/Pip), or via phase-transfer catalyzed asymmetric alkylation (DBFOX/MeNap). These ligands, complexed with Mg(NTf2)2, were used as Lewis acid promoters of enantioselective radical conjugate additions to α/β-unsaturated nitro-amides/esters. A summary of these results is presented and discussed. These findings led us to believe that our initial binding model between metal, ligand, and substrate was flawed. Thus, we figured that if we started with a functionality known …

Regulators Of G-Protein Signaling, Rgs13 And Rgs16, Are Associated With Cxcl12-Mediated Cd4+ T Cell Migration, Lijin Xia Aug 2008

Regulators Of G-Protein Signaling, Rgs13 And Rgs16, Are Associated With Cxcl12-Mediated Cd4+ T Cell Migration, Lijin Xia

Theses and Dissertations

Chemokines are important chemical signals that guide lymphocyte movement within the immune system and promote the organization and functions of germinal centers (GCs) in the secondary lymphoid tissues. Previous studies have shown that GC T cells exhibit high expression of chemokine receptor 4, CXCR4, but that these cells are unable to migrate to the ligand for this receptor, the chemokine CXCL12. This “migratory paralysis” to CXCL12 was found to be correlated with the expression of two Regulators of G-protein Signaling, RGS13 and RGS16 in the GC T cells. The objective of my research was to determine whether RGS13 and RGS16 …

Molecularly imprinted polymers (MIPs) are proving to be very effective in development of synthetic recognition systems and are of great interest to those interested in the field of sensor technology. The use of MIPs is receiving considerable interest due to the ability to prepare recognition matrices that possess high substrate selectivity and specificity. Conducting polymers (CP) have proved to be an excellent tool for the preparation of nano-structured biologically selective systems. Polypyrrole (Ppy) is one such CP that is extensively used for the construction of bioanalytical sensors. Ppy has shown great promise primarily due to its biocompatibility and thermal stability …

Role Of Adenine In Thymine-Dimer Repair By Reduced Flavin-Adenine Dinucleotide, Guifeng Li, Vincent Sichula, Ksenija D. Glusac Aug 2008

Role Of Adenine In Thymine-Dimer Repair By Reduced Flavin-Adenine Dinucleotide, Guifeng Li, Vincent Sichula, Ksenija D. Glusac

Chemistry Faculty Publications

We present a study of excited-state behavior of reduced flavin cofactors using femtosecond optical transient absorption spectroscopy. The reduced flavin cofactors studied were in two protonation states: flavin-adenine dinucleotide (FADH(2) and FADH(-)) and flavin-mononucleotide (FMNH2 and FMNH-). We find that FMNH- exhibits multiexponential decay dynamics due to the presence of two bent conformers of the isoalloxazine ring. FMNH2 exhibits an additional fast deactivation component that is assigned to an iminol tautomer. Reduced flavin cofactors also exhibit a long-lived component that is attributed to the semiquinone and the hydrated electron that are produced in photoinduced electron transfer to the solvent. The …

Chemistry & Biochemistry Dissertations - Archive

Integrins are transmembrane proteins found in almost all cells. They play vital roles in many biological processes, such as angiogenesis and signal transduction, through their interactions with different extracellular matrix ligands. These interactions are fostered by forming noncovalent bonds with the ligands. Integrin involvement in angiogenic cancer metastasis has sparked the interest of many researchers in the field of oncology. Many cutting edge research works are now focused on studying the noncovalent interactions of integrins with potential drug compounds formulated from peptide mimics and bearing the putative RGD amino acid sequence through which integrins are known to bind their ligands. …

Optical And Electrochemical Molecular And Nanoscale Sensors For The Selective Detection Of Organophosphorus Based Pesticides, Chandrima De Aug 2008

Optical And Electrochemical Molecular And Nanoscale Sensors For The Selective Detection Of Organophosphorus Based Pesticides, Chandrima De

Masters Theses

Organophosphorus based pesticides are highly toxic and represent serious environmental concerns. These compounds tend to easily enter ground water and cause drastic pollution. The toxicity levels of the organophosphorus pesticides however are based on their structures, and hence methods for their selective detection are in high demand. The creation of new materials for sensing and actuation requires careful manipulation of the responsive units required to control analyte selectivity. We have successfully designed and synthesized a molecular sensor based on the family of stilbenes that not only can detect organophosphorus pesticides, but also distinguish between them. Four different pesticides have been …

High-Definition Self-Assemblies Driven By The Hydrophobic Effect: Synthesis And Properties Of A Supramolecular Nanocapsule, Bruce C. Gibb Aug 2008

High-Definition Self-Assemblies Driven By The Hydrophobic Effect: Synthesis And Properties Of A Supramolecular Nanocapsule, Bruce C. Gibb

Chemistry Faculty Publications

High definition self-assemblies, those that possess order at the molecular level, are most commonly made from subunits possessing metals and metal coordination sites, or groups capable of partaking in hydrogen bonding. In other words, enthalpy is the driving force behind the free energy of assembly. The hydrophobic effect engenders the possibility of (nominally) relying not on enthalpy but entropy to drive assembly. Towards this idea, we describe how template molecules can trigger the dimerization of a cavitand in aqueous solution, and in doing so are encapsulated within the resulting capsule. Although not held together by (enthalpically) strong and directional non-covalent …

Identification Of Two Distinct Glutaminyl Cyclase Isoforms In Drosophila Melanogaster, Amanda Thigpen Parker Aug 2008

Identification Of Two Distinct Glutaminyl Cyclase Isoforms In Drosophila Melanogaster, Amanda Thigpen Parker

Dissertations

Sequence analyses revealed two putative glutaminyl cyclase genes in Drosophila melanogaster with greater than 60% amino acid sequence identity to previously identified glutaminyl cyclase in humans. Expression, purification, and characterization of both putative proteins revealed two enzymatically active glutaminyl cyclase enzymes. Comparison of the Drosophila melanogaster glutaminyl cyclase enzymes to the published human glutaminyl cyclase revealed the enzymes were very similar. The Km values obtained were identical to those of human glutaminyl cyclase for three different substrates tested. However, the Drosophila glutaminyl cyclases demonstrated lower turnover numbers. Both Drosophila glutaminyl cyclase isoforms were inhibited by imidazole, imidazole derivatives, and metal …

Spectroscopic Study Of Compressible Mobile Phase And Stationary Phase Behavior In Chromatography, Lawrence R. Baker Jul 2008

Spectroscopic Study Of Compressible Mobile Phase And Stationary Phase Behavior In Chromatography, Lawrence R. Baker

Theses and Dissertations

Raman spectroscopy, laser-induced fluorescence, and sum-frequency generation (SFG) spectroscopy are used to investigate the behavior of compressible mobile phases and stationary phases under a variety of chromatographic conditions. Efforts to understand and optimize separations employing compressible mobile phases have been limited by a lack of understanding of the mobile phase density gradient. Mobile phase compressibility leads to gradients in linear velocity and solute retention and affects separation speed and efficiency, especially in packed columns. This work describes on-column density measurement of CO2, a common carrier fluid for SFC and SGC, in packed capillary columns using Raman microspectroscopy. On-column detection by …

Synthesis Of Α-Hydroxy And Fluoro Phosphonates And Cyclic Ether-Containing Natural Products, Nongnuch Sutivisedsak

Dissertations

A number of important classes of natural product (eg. amphidinolide C, (-)-centrolobine, etc) contain common structural features of tetrahydrofurans and tetrahydropyrans. This study involved the development of stereoselective mothods for the synthesis of tetrahydrofurans and tetrahydropyrans including the application of allylic hydroxy phosphonates and their derivatives as chiral, non racemic building blocks for the synthesis of biological active molecules. The enantiomeric excess of non racemic α-hydroxy phosphonates and carbonate derivatives prepared via the asymmetric Pudovik reaction can be improved by kinetic resolution. Both chemical and enzyme catalysts were utilized in the kinetic resolution of phosphonates. Enantioselective acylation of a variety …

A Study On The Protein Interaction With Different Platinum Compounds, Nayna Kotadia Jul 2008

A Study On The Protein Interaction With Different Platinum Compounds, Nayna Kotadia

Masters Theses & Specialist Projects

Since the discovery of anti-tumor activity of cisplatin in 1960, significant progress has been made in treating metastatic or advanced cancer with cisplatin and platinum compounds. Platinum compounds covalently bind to DNA and disrupt DNA function. They are also known to bind with amino acids like methionine, histidine and cysteine to form cisplatin-protein adducts which are responsible for most of its cytotoxicity and side effects. Recent articles on cisplatin-protein have shown that adding bulky adjuncts to cisplatin or using different platinum compounds varies the degree and extent of reaction thus possibly reducing cisplatin resistance and side effects.

Mechanistic Studies Of Specific Dna Cleavage By Pvuii Restriction Endonuclease, Xie Fuqian Jul 2008

Mechanistic Studies Of Specific Dna Cleavage By Pvuii Restriction Endonuclease, Xie Fuqian

Dissertations

PvuII restriction endonuclease is a homodimeric protein which recognizes and cleaves the palindromic sequence (CAG?CTG) in the presence of Mg(II) ions. Starting with PvuII as a model system, pKa calculations with crystallographically defined metal ligated water are applied to PD?D/ExK motif metallonucleases in order to investigate the activation of nucleophile in metal dependent DNA hydrolysis. These results establish the electrostatic contributions of the metal ions and the conserved Lys in lowering water pKa. The calculated pKa values of metal ligands have been used to simulate the pH dependence of Mg(II) binding to PvuII. The bell shaped pH-rate profile is dissected …

Differences In A Conformational Equilibrium Distinguish Catalysis By The Endothelial And Neuronal Nitric-Oxide Synthase Flavoproteins, Robielyn P. Ilagan, Mauro Tiso, David Konas, Craig Hemann, Deborah Durra, Russ Hille, Dennis J. Stuehr

Department of Chemistry and Biochemistry Faculty Scholarship and Creative Works

Nitric oxide (NO) is a physiological mediator synthesized by NO synthases (NOS). Despite their structural similarity, endothelial NOS (eNOS) has a 6-fold lower NO synthesis activity and 6-16-fold lower cytochrome c reductase activity than neuronal NOS (nNOS), implying significantly different electron transfer capacities. We utilized purified reductase domain constructs of either enzyme (bovine eNOSr and rat nNOSr) to investigate the following three mechanisms that may control their electron transfer: (i) the set point and control of a two-state conformational equilibrium of their FMN subdomains; (ii) the flavin midpoint reduction potentials; and (iii) the kinetics of NOSr-NADP+ interactions. Although eNOSr and …

Genetic Analysis Of The Role Of Smpb In Establishing The Reading Frame On Tmrna, Talina Christensen Watts Jul 2008

Genetic Analysis Of The Role Of Smpb In Establishing The Reading Frame On Tmrna, Talina Christensen Watts

Theses and Dissertations

Ribosomes translate the genetic information encoded by mRNA into proteins. Defective mRNAs can cause stalling of translating ribosomes. The molecule tmRNA (transfer-messenger RNA) rescues stalled ribosomes in eubacteria. Together with its protein partner SmpB, tmRNA mimics a tRNA by entering the ribosomal A site and linking an alanine residue to the growing polypeptide chain. The ribosome then abandons the defective mRNA template and resumes translation on tmRNA, adding ten more amino acids to the nascent polypeptide. As a result of tmRNA action, stalled ribosomes are released and recycled, the defective mRNA is destroyed, and the aborted protein product is tagged …

Ion Permeation Through Membrane Channels: Molecular Dynamics Simulations Studies, Morad Mustafa

Theses and Dissertations

Molecular dynamics simulation was used to study ion permeation through different membrane proteins embedded in a lipid bilayer (DMPC) with different saline solutions. The potential of mean force (PMF) for ion transport was obtained by umbrella sampling simulations. A revised MacKerell force field for tryptophan residues was studied using gramicidin A (gA) channel as a test model. The revised force field contribution to the Na+ PMF was consonant with the prediction from the experimental results, but in stark contrast to the prediction of the CHARMM force field, version 22, for the tryptophan side-chain. A new grid-based correction map algorithm by …

Surface Charge Effects Involved In The Control Of Stability Of Sols Comprising Uniform Cholesterol Particles, Vuk Uskoković Jul 2008

Surface Charge Effects Involved In The Control Of Stability Of Sols Comprising Uniform Cholesterol Particles, Vuk Uskoković

Pharmacy Faculty Articles and Research

The reported is a method for the preparation of stable dispersions of narrowly dispersed cholesterol crystals, which may provide an excellent model for the aggregation studies thereof. The effects of experimental parameters that include concentration, pH, ionic strength, temperature, and aging on the resulting morphologies are acknowledged. Regardless of the cause of destabilization, the multilayered aggregates of cholesterol particles are formed by “face-to-face” stacking of the original platelets, as promoted by higher surface charge density on particle sides comparing to their faces. In accordance with the crucial involvement of surface charge effects in stabilization of cholesterol dispersions produced hereby, aggregation …

A Novel Approach To The Synthesis Of Silicon Nanowires, Joo Chick Chan Jul 2008

A Novel Approach To The Synthesis Of Silicon Nanowires, Joo Chick Chan

Dissertations and Theses

Contemporary VLS-SiNW synthetic methods employ CVD and laser ablation methods that produce single crystalline SiNW; which can be doped by co-flowing appropriate dopants, such as diborane (p-type) and phosphine (n-type). Not only are these gases toxic (non-green), but also the synthetic methods used are costly requiring elaborate instrumental assembly. This thesis presents a low-cost and "green" method in synthesizing and doping of SiNW in a glass tube. It was found that by selecting the appropriate metal catalyst, precursor reactants, and the reaction conditions both SiNW and carbon nanotube (CNT) can be synthesized on metallic (aluminum and copper), semiconducting (silicon) and …
