Alkoxyethanols [CmH2m+1·(OC2H4)·OH] (conveniently abbreviated to CmE1) are a subgroup of the alkyl poly(ethylene glycol) mono-ethers. They are small relatives of a large number of non-ionic detergents. The thermodynamic properties of their mixing in water have been shown to be good sources of information about the interactions of both hydrophilic and hydrophobic groups with water. Theses alkoxyethanols, or glycol ethers, are the ether derivatives of ethylene glycol where one hydroxyl functional group is substituted for an alkoxy functional group. Throughout the world there exist both predator and prey. This distinction is apparent though sometimes misleading. Take for example marine snails of the genus Conus that are present across the oceans of the southern hemisphere [1]. These snails are slow moving animals that appear more prey than predator. However, they have evolved into effective predators through the development of venom consisting of biologically active peptides. The venom is loaded into a hollow harpoon that the snail injects into the intended prey: fish, worms, or other snails [2]. The structures and properties of amyloid fibrils are of considerable interest due to their associations with numerous neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease and transmissible spongiform encephalopaties (prion diseases). Understanding fibrillogenesis at a molecular level requires detailed structural characterization of amyloid fibrils. However amyloid fibrils are difficult objects to study due to their non-crystalline and insoluble nature. These properties make the application of classical tools of structural biology, such as X-Ray crystallography and solution Nuclear Magnetic Resonance spectroscopy, impractical for structural characterization of protein fibrils.

Arsenic exists as many different chemical forms including inorganic, methylated and dietary species. The toxicity of these species varies: some are highly toxic and can cause adverse health effects in many parts of the body; others are considered relatively non-toxic. Monitoring arsenic exposure is usually accomplished by its direct measurement in biological fluids. Urine is the specimen of choice for assessing arsenic exposure, because of its short biological half-life in blood. There have been very few studies of arsenic species in blood, yet such research can provide valuable information on arsenic distribution and its metabolism in the body.

The Hauser's annulation and Hauser and Xu's route for C-aryl glycoside synthesis were successfully employed to prepare the linear tetracyclic anthraquinone substituted C-aryl glycoside, galtamycin as well as bis C-aryl glycosides. Synthetic studies on angucycline C104 extended the utility of both methods to structures with angular tetracyclic framework.

An investigation of cause was started in the 1990s at a bleached Kraft mill (BKM), located in Saint John NB, Canada. The 5th effect chemical recovery condensates waste stream (derived from wastewater generated during the pulping process) was identified as having the greatest potential for affecting mummichog (Fundulus heteroclitus), compared to the rest of the inmill waste streams. A solid phase extraction (SPE) protocol was utilized to isolate the causative chemicals in the condensate stream. The acidity constants, relative stabilities, and tautomeric equilibrium constants of some 1,2,4-triazole derivatives were determined using the density functional theory (DFT) with the B3LYP method and 6-311G(d,p) basis set. The integral equation formalism version of the polarizable continuum model (IEFPCM) was used in the calculations of the aqueous phase. The calculated tautomeric equilibrium and relative stabilities values revealed that the 4H-1,2,4 triazole form for all studied molecules was favored over the 1H-1,2,4 triazole form. Protonation processes indicated the predominance of the 1H-1,2,4 triazole form over the 2H-1,2,4 triazole form. The essential oils of mosses [Tortula muralis Hedw. (Pottiaceae), Homalothecium lutescens (Hedw.) H. Rob. (Brachytheciaceae), Hypnum cupressiforme Hedw. (Hypnaceae), and Pohlia nutans (Hedw.) Lindb. (Mniaceae)] were investigated by means of GC-FID/MS techniques. The major components were nonanal (18.3%) and tetradecanol (4.3%) in the oil of T. muralis, nonanal (36.8%) and tricosane (6.5%) in the oil of H. lutescens, nonanal (12.5%) and 2E-tetradecen-1-ol (6.9%) in the oil of H. cupressiforme, and nonanal (7.8%) and 2E-tetradecen-1-ol (7.1%) in the oil of P. nutans. Eleven mineral and trace elements (Mg, Ca, K, Fe, Mn, Al, Zn, Cu, Cd, Ni, and Pb) were determined in the Thymus species growing in Turkey. The total concentrations were measured in samples mineralized with concentrated nitric acid and hydrogen peroxide in a microwave system. The determination of mineral and trace elements in the leaves of the Thymus species was carried out by flame and graphite furnace atomic absorption spectrometry. The main problem associated with metal hexacyanoferrates (MHCFs) is their instability at high pH values. We synthesized a new Prussian Blue analogue, Nile-Blue hexacyanoferrate (NBHCF), that remains stable in 0.2 M KOH. A carbon paste (CPE) chemically modified electrode (ME) containing NBHCF was prepared as a stable electrochemical sensor for measuring hydrazine. A detailed characterization of the electrochemical and electrocatalytic behavior of NBHCF was performed using cyclic voltammetric, chronoamperometric, differential pulse voltammetric (DPV) and hydrodynamic amperometric methods. The bacterial SEC pathway is commonly used for secretion of heterologous proteins in E. coli by fusing them to transported proteins to facilitate downstream processing. While some proteins are translocated very efficiently, some reside in the cytoplasm. In this work, maltose binding protein (MBP) was fused to 3 cytoplamic enzymes from Thermus thermophilus (serine protease, 251 residues; glucose isomerase, 381 residues; pullulanase, 718 residues) to study the protein transport from the cytoplasm by quantifying the distribution of activities in different cellular compartments. Pullulanase activity was harvested exclusively in the periplasm; however, glucose isomerase activity was harvested exclusively in the cytoplasm. …

Some pyridazine derivatives carrying urea, thiourea, and sulfonamide groups were synthesized and evaluated for their antimicrobial activity against gram-positive and gram-negative bacteria, and fungi by using broth microdilution. The structures of these new compounds were confirmed by ^1H-NMR, mass spectrum, and elemental analysis. The synthesized compounds exhibited generally promising inhibitory activity against S. aureus (MIC ranging from 2 to 4 \mu g/mL) and E. coli (MIC ranging from 4 to 16 \mu g/mL). Moreover, all compounds showed antifungal activity against both C. albicans and C. parapsilosis, with a MIC value of 8 \mu g/mL.

The liver is known to accumulate many trace elements; thus it is very important to determine their level in a sensitive way. The objective of this study was to develop a simple method for the simultaneous determination of trace elements in cow liver in which the elimination of interference can be accomplished. It was observed that Cd, Pb, Zn, and Cu ions could not be determined correctly in the presence of Se(IV), because of interference of Se. Chitosan (CS) based microparticles for controlled release of theophylline (THP) were obtained by ionic gelation. The microparticles were characterized by laser diffraction analysis (particle size), scanning electron microscopy (SEM), FTIR spectroscopy, and thermogravimetric analysis. The average diameter of the CS-THP microparticles was 182 \mu m. The FTIR spectra confirmed that new hydrogen bonds had formed between the carbonylic group belonging to THP and the amino group of CS, while the thermogravimetric analysis showed a modified stability in the obtained particles and confirmed the presence of an interaction between the drug and CS. Full details of an enantioselective total synthesis of 8-O-4'-neolignan perseal B are presented for the first time. The synthesis was achieved in 8 steps from vanillin and involved the asymmetric dihydroxylation reaction using AD-mix-\alpha to give the key intermedium (1S,2S)-1-(4-(benzyloxy)-3-methoxyphenyl)propane-1,2,3- triol, and the Mitsunobu reaction between phenylpropanoid and vanillin formed perseal B. The synthetic method of perseal B exhibits a new route for 8-O-4'-neolignan.

A new series of di and tri-organotin(IV) compounds containing germanium with the general formula R_{4-n}SnL_n was synthesised by reaction of 3-triphenylgermanyl-3,3-dimethylpropionic acid with organotin(IV) oxides and organotin(IV) chloride in 2:1 and 1:1 molar ratio, respectively, where R~=~CH_3, C_2H_5, n-C_4H_9, n-C_8H_{17}, and C_6H_5 n = 1 or 2 and L~=~triphenylgermanyl acid anion (GeC_{23} H_{23} O_2^-). These compounds were characterised by elemental analyses, FT-IR, multinuclear NMR (^1H, ^{13}C, ^{119}Sn), and mass spectrometry. A series of 3-hydroxy-6-methyl-2-[(substitutedpiperidine-1-yl) methyl]-4H-pyran-4-one structured compounds were synthesized by reacting 5-hydroxy-2-methyl-4H-pyran-4-one with suitable piperidine derivatives using Mannich reaction conditions. Antibacterial activities of the compounds for E. coli ATCC 25922, S. paratyphi ATCC BAA-1250, S. flexneri ATCC 12022, E. gergoviae ATCC 33426, and M. smegmatis ATCC 14468 were assessed in vitro by the broth dilution method for determination of minimum inhibitory concentration (MIC). In addition, their inhibitory effects over DNA gyrase enzyme were evaluated using a DNA gyrase supercoiling assay. Using glucose oxidase as a model enzyme, a novel rhodanine derivative chemiluminescence biosensor for the determination of glucose was formed based on multilayer-enzyme membrane as receptor, which was assembled via layer-by-layer assembly of sol-gel and glucose oxidase-gold nano-particles inside a glass tube. Compared with the traditional chemiluminescence biosensor, the proposed biosensor had some remarkable advantages, such as good selectivity of substrate, good response performance, good stability, good sensitivity, and longer service life. The linear range was from 1.0 \times 10^{-6} to 5.0 \times 10^{-2} mol L^{-1} (r = 0.9991). The detection limit was 5.0 \times 10^{-7} mol L^{-1}. In this …

A series of organosilicon(IV) and organotin(IV) complexes having the formulae R_2MCl(L^1), R_2MCl(L^2), R_2MCl(L^3), R_2MCl(L^4), R_2M(L^1)_2, R_2M(L^2)_2, R_2M(L^3)_2, and R_2M(L^4)_2, where M = Si and Sn, and R = CH_3, were synthesized with Schiff bases 5-mercapto-4-(pyridine-2-carboxalideneamino)-s-triazole (HL^1), 4-(furfuralideneamino)-5mercapto-s-triazole (HL^2), 4-(2-chlorobenzylideneamino)-5-mercapto-s-triazole (HL^3), and 4-(4-flourobenzylideneamino)-5-mercapto-s-triazole (HL^4). These complexes were characterized by elemental analysis, molar conductance, and spectroscopic studies, including UV, IR, ^1H, ^{13}C, ^{29}Si, and ^{119}Sn NMR spectroscopy. On the basis of these studies, the resulting complexes are proposed to have trigonal bipyramidal and octahedral geometries.

This article describes the preparation and characterization of anticancer drug-loaded poly(lactide-co-glycolide) (PLGA) microparticles. PLGA microparticles loaded with doxorubicin HCl (DOX) were prepared via o/w emulsion solvent evaporation. The release characteristics, encapsulation efficiency, size, and morphology of the PLGA microparticles were also determined. A cytotoxicity test was performed by using Glioma RG2 cancer cells to investigate the cytotoxicity of DOX-loaded PLGA microparticles. The DOX-loaded PLGA microparticles had an average diameter of 500 \pm 9 nm. The DOX encapsulation efficiency and drug loading were 22.75% and 0.78%, respectively. A series of new 4-(3,5-disubstitue-4H-1,2,4-triazol-4-yl)- 5-(thiophen-2-yl-methyl)-2H-1,2,4-triazol-3(4H)-ones (3a-c) were obtained by reaction N'-1-ethoxy-2-thiophen-2-yl-ethylydene hydrazino carboxylic acid ethyl ester (1) and 4-amino-4H-1,2,4-triazoles (2). 4-(3,5-disubstitue-4H-1,2,4-triazol-4-yl)-2-(2-oxo-2-arylethyl)-5- (thiophen-2-yl-methyl)-2H-1,2,4-triazol-3(4H)-ones (4a-e) and ethyl 2-(4-(3,5-disubstitue-4H-1,2,4-triazol-4-yl)-5-oxo-3-(thiophen-2-ylmethyl)- 4,5-dihydro-1,2,4-triazol-1-yl)acetates (6a-c) were obtained by reaction of compounds 3 and bromoacetophenon derivatives and bromo ethylacetate, respectively. Compounds 5a-e were synthesized from the reaction of corresponding compounds 4a-e with NaBH_4. Compounds 7a-c were obtained by the reaction compounds 6 and LiAlH_4. Seventeen new compounds were synthesized and characterized by elemental analyses, IR, ^1H-NMR, and ^{13}C-NMR spectral data. Some new unsymmetrical and optically active 2,5-disubstituted 1,3,4-oxadiazoles 5a-j were efficiently synthesized by cyclization reaction of diacylhydrazides 4a-j. The synthesis of the title compounds was achieved by the reaction of acyl hydrazides 3a-b and N-phetaloyl-L-amino acids 1a-e in the presence of the phosphoroxy chloride (POCl_3) as an anhydrous reagent.

A pH optical sensor was developed based on the use of a mixture of Malachite Green Oxalate and Bromocresol Green indicators immobilized in a triacetylcellulose membrane, which had been previously hydrolyzed. Requirements of the pH indicators were investigated, including the effects of the coupling pH, indicator ratio, and concentrations of the indicators. The pH sensor performed with a very fast response (10 s) and long term stability with no significant leaching of the dyes. Quorum sensing bacteria produce and release chemical signal molecules (like N-acyl homoserine lactones, AHL) that increase in concentration as a function of cell density. The responses cover a large spectrum of process such as the virulence in Staphylococcus aureus, competence for DNA-uptake in Bacillus subtilis and Streptococcus pneumoniae, sporulation in Bacillus subtilis, conjugal plasmid transfer in Enterococcus faecalis, and bacteriocin production in lactic acid bacteria. Atmospheric aerosol particles play an important role in climate and human health. Despite their importance, the mechanisms of their formation are still poorly understood. Ion-induced nucleation may play a role in the process, but details are lacking about how the nucleation rate depends on the size, charge, and number of ions in the cluster. The fundamental physical processes of mid infrared laser ablation in the context of laser desorption mass spectrometry were investigated. Understanding the mechanisms of infrared laser desorption and ablation can lead to improvements in these techniques and expand their applications. Particles were generated from glycerol irradiated at atmospheric pressure using a tunable infrared laser at wavelengths between 2.6 and 3.8 µm. The wavelength dependence of size distributions of ablated particles was measured. After brief overviews of low-abundance cell selection techniques in chapter 1 and circulating tumor cells in chapter 2, this dissertation initially focuses on the development of aptamer incorporated high-throughput microfluidic techniques to select rare circulation prostate cancer cells (LNCaP) directly from whole blood with subsequent quantification of these rare cells using a non-labeling approach. Then, I extended the technology to environmental samples in an effort around time, sensitivity, and portability of traditional groundwater assessment. As a model bio- pathogen, E. coli O157:H7 was chosen due to its toxicity and its adverse impact on recreational waters. Low-abundance (<100 cells mL-1) E. coli O157:H7 cells were isolated and enriched from environmental water samples using a microfluidic chip that its capture beds were covalently decorated with E.coli O157:H7 specific polyclonal antibodies. The selected cells were enumerated using RT-qPCR technique. Finally, I have integrated HTMSU with electrokinetic enrichment microfluidic unit for performance of single recombinant low-abundance CTC cell-based assay. A series of analytical processes were carried out, including immunoaffinity selection of rare CTCs, quantification of selected cells via conductivity impedance and electrophoretic enrichment of selected cells for PCR/LDR/CE interrogation for detection of low-abundance point mutations in genomic DNA.

Dissolution of chitosan in ionic liquids was accomplished. It has been shown that 1-butyl-3-methylimidazolium acetate (BMIMAc) is a better solvent than 1-butyl-3-methylimidazolium chloride (BMIMCl). Dissolution of chitosan in BMIMCl required a prior regeneration of chitosan from 1% acetic acid solution. In the case of BMIMAc, both dried chitosan and regenerated chitosan from acetic acid solution have been dissolved in a relatively short amount of time. While concentrations up to 2 wt% of chitosan in BMIMCl could be obtained, concentrations of 10 wt % of chitosan in BMIMAc were realized.
