Investigating The Effects Of The Plasticizers Bisphenol A And Di (2-Ethylhexyl) Phthalate On Dopaminergic Neurons In A C. Elegans Parkinson's Disease, Amanda Valera
Undergraduate Honors Theses
Parkinson's disease (PD) is a neurodegenerative disorder characterized by loss of dopamine (DA) containing neurons in the substantia nigra of the brain. Though the cellular pathways that lead to PD are varied and not well understood, elevated levels of reactive oxygen species (ROS) and inhibition of the mitochondrial electron transport chain enzyme complex I have been associated with PD pathogenesis. Environmental factors such as the plasticizers bisphenol A (BPA) and di(2-ethylhexyl) phthalate (DEHP) may also be involved in PD pathogenesis. In this research, three strains of the nematode Caenorhabditis elegans (C. elegans), a wild-type N2 strain, a ROS-sensitive nnt …

Catalytic Activity Of Titania And Titania-Containing Nanomaterials: And, Spatial Distribution Of Silica Surface Groups Using A Tethered Amine Catalyst, Ramkumar Samala
Masters Theses
Titania and titania-containing nanomaterials are semiconductor photocatalysts and are widely used to degrade (i.e. oxidize) organic matter. The adsorption and catalytic properties of highly dispersed titania prepared using sulfate and pyrogenic methods, and fumed containing mixed oxides, were studied using catalytic decomposition of methylene blue primarily by kinetic methods. Commercial ultrafine titania (PC-100, PC-105 and PC-500), fumed Ti 02, and the mixed oxide of 80% Silica/20% Titania (ST20) were studied. Catalytic activity as measured by methylene blue decomposition kinetics was highest per gram for non-treated ultrafine titania PC- 500 which has the highest SsET and smallest particle size. However this …

Characterization Of Heterogeneities And Domains In Aquatic And Sedimentary Organic Matter By 1 H Spin Diffusion: Potential For Elucidating The Formation Mechanisms, Jingdong Mao, Xiaoyan Cao
Chemistry & Biochemistry Faculty Publications
Although the information on domains and heterogeneities of natural organic matter (NOM) can provide insights into its formation mechanisms, the appropriate solid-state NMR technique for measuring them is still lacking. The traditional technique requires mobility differences in NOM whereas NOM components are primarily rigid. We introduced a new 1H spin diffusion technique, 1H-13C two-dimensional heteronuclear correlation (2D HETCOR) NMR with 1H spin diffusion, for characterization of domains and heterogeneities in aquatic and sedimentary organic matter. It was achieved by collecting a series of 2D HETCOR spectra with a variable mixing time, tm', and …

Polarographic Determination Of Anticancer Drug Vitamin K_3 With Direct And Indirect Methods/Application To Serum And K_3 Injection Solution, Ümmi̇han Taşkoparan Yilmaz, Güler Somer
Turkish Journal of Chemistry
In this work, direct and indirect methods for determination of vitamin K_3 were developed using differential pulse polarography. The reaction between sulfite and the quinone (Q) form of vitamin K_3 was used for indirect determination, since the sulfite peak at -0.7 V is sharp and very reproducible in 0.1 M HAc NaAc (pH = 5.5). It was found that at pH 4.5-5.5, this reaction was quantitative and very fast when the temperature was 45 °C. For its direct determination, on the other hand, vitamin K_3 was standardized by the indirect method using standard sulfite as the reducing agent. The calibration …

A series of [(4-nitro-1H-imidazol-1-yl)methyl]-1,2,4- triazoles and 1,3,4-thiadiazoles were prepared and evaluated for their activity against sensitive and resistant H. pylori strains. Study of the structure-activity relationship of these series of compounds indicated that the type of nitroimidazole moiety and the pendent group on the heteroaryl analog dramatically impact the anti-H. pylori activity. In triazole series, compound 5d, containing a 4-methyl phenyl group on the triazole ring, was the most potent compound tested against both metronidazole-sensitive and -resistant strains at a concentration of 8 \mu g.

Chemistry Faculty Research and Scholarship
A model system for the molybdenum cofactor has been developed that illustrates the noninnocent behavior of an N-heterocycle appended to a dithiolene chelate on molybdenum. The pyranopterin of the molybdenum cofactor is modeled by a quinoxalyldithiolene ligand (S(2)BMOQO) formed from the reaction of molybdenum tetrasulfide and quinoxalylalkyne. The resulting complexes TEA[Tp*MoX(S(2)BMOQO)] [1, X = S; 3, X = O; TEA = tetraethylammonium; Tp* = hydrotris(3,5-dimethylpyrazolyl)borate] undergo a dehydration-driven intramolecular cyclization within quinoxalyldithiolene, forming Tp*MoX(pyrrolo-S(2)BMOQO) (2, X = S; 4, X = O). 4 can be oxidized by one electron to produce the molybdenum(5+) complex 5. In a preliminary report of …

The number of medical applications of electrochemistry has grown in recent years due to the increased applications of electrochemical concepts to various systems of the organism. This includes electrochemically controlled hemosorption detoxification, where the removal of toxicants is controlled by changing the potential of the hemosorbent. It is important to avoid Faradaic processes in the course of hemosorption, which can lead to the addition of electrochemically modified toxicants to blood. The probability of their occurrence should depend on the open-circuit potential of the activated carbon. In order to elucidate the identification of Faradaic reactions, a model system was investigated. The …

The Thorgren Basin, located in Valparaiso, IN, was created as a component of the stormwater management system for the city. The basin’s primary function is to collect stormwater from two separate regions of the Salt Creek Watershed after large storm events, that is then discharged through a single outlet. In its current state the basin has limited infiltration capabilities due to a lack of vegetation and pervious surfaces. The Save the Dunes Conservation Fund, a local non-profit group, provided a cost share opportunity to naturalize Thorngren basin. The goal of the summer 2011 project was to create a monitoring strategy …
Preparation And Crystal Structures Of Chiral And Non-Chiral Mixed Ligand Copper Complexes Containing N-Methyl Imidazole And Various N-Phthaloylalanines, Hiral Patel
Theses, Dissertations and Culminating Projects
A quasiracemate is the cocrystallization of two similarly shaped but chemically distinct molecules of opposite handedness. There has been extensive work done in the field of organic quasiracemates by Dr. Kraig A. Wheeler. Limited work has been done in inorganic quasiracemates. The goal of this study was to prepare various metal based enantiomers, racemates, and quasiracemates using A-phthaloylalanines, and TV-methylimidazole as ligands bound to copper arranged in a centrosymmetric pattern. In this study, we developed a method to prepare A-phthaloylalanines and its copper complexes. Three complexes have yielded crystal structures: a racemate, bis(7V-methylimidazole) bis(D,L-3-chloro-7V-phthaloylalanato)copper(II); a quasiracemate, bis(A-methyhmidazole)(L-3-chloro-iV-phthaloylalanato)(D-3-methyl-7V-phthaloylalanato) copper(II); and an …

Nitrous acid (HONO) is an important precursor of hydroxyl radical and plays an important role in the terrestrial boundary layer photochemistry. However, there are still many unknowns regarding HONO chemistry such as formation mechanisms and distributions in the troposphere, especially in rural and remote environments. In this research, we presented the first vertical concentration profiles of HONO in the troposphere, measured on a small aircraft platform. The HONO mixing ratios ranged from 4 to 17 pptv in the free troposphere and from 8 to 74 pptv in the rural boundary layer. The HONO distribution patterns were strongly correlated to the …
Single Cell Analysis For The Characterization Of Cell Populations Using A Live Cell Array, Maureen Ann Walling
Single Cell Analysis For The Characterization Of Cell Populations Using A Live Cell Array, Maureen Ann Walling
Legacy Theses & Dissertations (2009 - 2024)
In the past decade, the shift from whole cell population analyses towards single cell measurement methods and techniques is based on experimental results that reveal significant levels of non-genetic heterogeneity in clonal cell populations. This heterogeneity manifests in multiple aspects of cell activity and is, in part, a result of stochastic noise in processes leading to gene expression, namely transcription and translation. The growing understanding of this occurrence has led to the development of methods to monitor and analyze heterogeneity for a more thorough description of cell populations and overall activity.

Legacy Theses & Dissertations (2009 - 2024)
Polynuclear copper(I) carboxylates exhibiting rich luminescent properties and a remarkable structural diversity have been the main focus of this investigation. In spite of the well-developed methods of copper(I) carboxylates synthesis, only a handful of complexes have been crystallographically characterized prior to our work. Notably, they all demonstrate different structural arrangements in the solid state. This dissertation describes the work that has been carried out towards the synthesis of new copper(I) carboxylates, the study of their molecular and solid-state structures, and their solid-state photoluminescent (PL) properties. In this work, we have prepared a series of new copper(I) carboxylates of the general …

The receptor for advanced glycation end products (RAGE) is a multiligand cell surface macromolecule that plays a central role in the etiology of diabetes, inflammation, and neurodegeneration. The cytoplasmic domain of RAGE, ctRAGE, is critical for RAGE-dependent signal transduction. As the most membrane proximal event, mDia1 binds to ctRAGE and is essential for RAGE ligand-stimulated phosphorylation of AKT and cell proliferation/migration. We show that ctRAGE contains an unusual alpha-turn that mediates the mDia1-ctRAGE interaction and is required for RAGE dependent signaling. The results establish a novel mechanism through which an extracellular signal initiated by RAGE ligands regulates RAGE signaling in …

Synthesis And Characterization Of Diallylic Polysulfanes And Study Toward The Synthesis Of Thiarubrine E, Kai Wang
Legacy Theses & Dissertations (2009 - 2024)
Garlic oil contains diallyl polysulfanes as major components, which have been used as environmentally benign bird repellants and nematicides. While previous studies found that the biological activity of polysulfanes increases with numbers of sulfur atoms, higher diallyl polysulfanes are currently unavailable. A simple approach was developed, using liquid sulfur as a reagent, to the synthesis of diallyl polysulfanes containing chains of more than 20 sulfur atoms. This method was successfully applied to other diallylic disulfides and to garlic oil to give new families of polysulfanes. Some diallyl polysulfanes were isolated in a pure form by preparative high performance liquid chromatography …
Homologous And Heterologous Crystallin Protein Interactions As The Molecular Basis Of Inherited Cataract, Priya Ranjan Banerjee
Homologous And Heterologous Crystallin Protein Interactions As The Molecular Basis Of Inherited Cataract, Priya Ranjan Banerjee
Legacy Theses & Dissertations (2009 - 2024)
Age-related cataract is the most common cause of blindness worldwide. Nearly fifty percent of Americans above the age of 75 are diagnosed with this disease (http://www.nei.nih.gov/eyedata/pbd_tables.asp), and surgical intervention is the sole method of treatment at present (http://www.nei.nih.gov/healthyvision/objective/cataracts.asp). In the developing world, even this treatment is not readily available. These are compelling reasons to search for better treatments to delay, prevent or arrest cataract formation. Recent evidence suggests that age-related cataracts also have a genetic component. Therefore, determining the mechanisms underlying genetic cataracts with a known association to a protein-mutation is one important strategy towards understanding the molecular basis for …

Biophysical Investigations Of The Molecular Basis Of Cataract Associated With The R76s Mutation In Human Gammad-Crystallin, Vurghun Ahmadov
Legacy Theses & Dissertations (2009 - 2024)
Cataract disease results when the eye lens becomes opaque and scatters a significant part of the incoming light into the eye. The lens contains very high concentrations of the lens proteins, called crystallins, which are present at concentrations comparable to those found in protein crystals (about 400-600 mg/mL). Chemical modifications of the crystallins, such as oxidation and deamidation, or genetic mutations are known to result in increased light-scattering in vitro, and are implicated in cataract formation in vivo. Here we present the in vitro work on a mutant protein of human gammaD crystallin (HGD), namely R76S (i.e. Arg 76 to …
