Biotinylated analogs of drug candidates provide useful tools for studying the drug--target protein interactions. Polyhalogenated N,N'diarylureas are potent activators of adenosine monophosphate-activated kinase (AMPK) and potentially promising agents for the treatment of cancer. Various biotinylated versions of these N,N'-diarylureas were synthesized and evaluated in AMPK-activation studies.

Titanium(II) complexes of composition TiCl_2(L)_2 [where L = 2,2'-bipyridine (bipy), 4,4'-dimethyl-2,2'-bipyridine (bpMe), 4,4'-dimethoxy-2,2'-bipyridine (bpoMe), 6,6'-dimethyl-2,2'-bipyridine (dpMe), adamantylamine (ada)] were prepared by reacting titanium tetrachloride and N-containing bulky ligands in predetermined molar ratios. The complexes synthesized were characterized by different spectroscopic techniques, viz. UV-visible, FTIR, ^1H NMR, and mass spectrometry. The stoichiometry of the complexes was established by their elemental analysis (chlorine and titanium estimation). The unit cell parameters calculated by using the powder XRD tool confirm the monoclinic unit system and P lattice type for all complexes. Synthesis, Characterization, And Biological Activity Of Coordination Polymers Derived From Pyromellitic Dianhydride, Yogesh Patel, Ritu Dixit, Hasmukh Shanubhai Patel

A new ligand, 2,5-bis(furan-2-ylmethylcarbamoyl)terephthalic acid (BFMTA), was synthesized using 1,2,4,5-benzenetetracarboxylic dianhydride (pyromellitic dianhydride-PMDA) with furan-2-ylmethanamine (2-furfurylamine). New coordination polymers of the ligand (BFMTA) were also prepared using transition metal ions Mn(II), Fe(II), Co(II), Ni(II), Cu(II), and Zn(II) metal salts. The coordination polymers and ligand were characterized by physico-chemical characteristics, magnetic susceptibilities, spectroscopic investigations, and thermogravimetry. Antimicrobial activity was tested using the agar-plate method against various strains of bacteria and spores of fungi. The results showed significantly higher antimicrobial activity of coordination polymers compared to the ligand.

"Colloidal Unimolecular Polymer (CUP) particles are a new genre of material formed by self-assembly into spheroidal particles due to the effect of hydrophilic/hydrophobic interactions of the polymer with a change in the solvent. The particle sizes were characterized by means of Dynamic Light Scattering with corrected viscosities instead of water's viscosity. The distribution of particle size agreed well with the distribution of molecular weight proving that the polymer had undergone unimolecular collapse. The rheology study shows the presence of surface water and its significant effect on the rheology. The primary and secondary electroviscous effects were also found to play roles …

"Part I. The trona is being injected to control SOx emission from coal-fired power plants. The results showed that trona ash leached significantly more As, Se, Mo, and V than the control fly ash did, especially under the natural pH condition. In addition, trona ash had a significantly greater soluble fraction than the control ash. Further investigation indicated that greater amounts of the studied anionic elements in the trona ash were associated with the soluble, trona reaction products. Moreover, the insoluble fraction of the trona ash lost its capability to adsorb these elements under the natural pH condition. The …

"Nicotine, a major alkaloid, is present in tobacco products as well as in smaller amounts in potatoes, tomatoes, and eggplants. Although it is the addictive compound in cigarettes, nicotine is believed to play a protective role in neurodegenerative disorders such as Parkinson's disease. Researchers have found that the short-term administration of nicotine may help improve the memory process by reducing oxidative stress in the brain. Our studies have shown that, by blocking Fenton's reaction, nicotine is able to inhibit free radical generation, so this research has focused on the antioxidant properties of nicotine. This review article represents a survey covering the synthetic strategies leading to 5-membered heterocycles. The reactions are subdivided into groups that cover the synthetic methods of those heterocycles, i.e. pyrroles, furans, thiophenes, pyrazoles, isoxazoles, thiazoles, and others, utilizing benzoylacetonitriles as starting precursor from 1985 up to the present. The reactions are subdivided into groups that cover the synthetic methods for those heterocycles from benzoylacetonitriles.

Ionic liquid mediated synthesis of novel heterocyclic compounds 1-[(1H-benzoimidazol-2-yl)amino]-2'-phenylspiro[azetidine- 4,4'-[4'H]chroman]-2-ones (3) and 1-[(1H-benzoimidazol-2-yl)amino]- 3-chloro-2'-phenylspiro[aze tidine-4,4'-[4'H] chroman]-2-ones (4) was accomplished by condensing substituted 2-hydrazino benzimidazole (1), flavanone (2), and acetyl chloride/chloroacetyl chloride in ionic liquid, [bmim]PF_6 with or without using catalyst in excellent yield (90%--95%). Further, compounds 3 and 4 were acylated with trifluoroacetic anhydride to give N-acylated products (5 and 10); 3 when treated with HCHO and (C_2H_5)_2NH gave Mannich bases (6) and with aldehydes afforded 3-arylidene-2-azetidinone (7). Synthesis and in vitro antimicrobial activities are reported for a series of 1,3,5-substituted 4-oxo-3,4-dihydro-5H-pyridazino[4,5-b]indole derivatives. Corresponding pyridazino[4,5-b]indoles were prepared from ethyl 3-formyl-1H-indole-2-carboxylate precursors and the functional group in question was installed with hydrazine and its derivatives. The purity and primary structures of pyridazino[4,5-b]indole were confirmed by IR, ^1 H NMR, and ^{13}C NMR spectroscopy and elemental analyses. Oligodeoxynucleotides (ODNs) containing latent electrophilic groups can be highly useful in antisense drug development and many other applications such as chemical biology and medicine, where covalent cross-linking of ODNs with mRNA, protein and ODN is required. However, such ODN analogues cannot be synthesized using traditional technologies due to the strongly nucleophilic conditions used in traditional deprotection/cleavage process.

Optical pH sensors have several advantages over traditional potentiometric pH measurement, such as high sensitivity, no need of constant calibration, easy for miniaturization and possibility for remote sensing. This dissertation is related to the studies of functionalized nanoparticles for self-assembly and as controlled drug delivery system. The whole topic is composed of two parts. In the first part, the research was conducted to design and synthesize a new type of ionic peptide-functionalized copolymer conjugates for self-assembly into nanoparticle fibers and 3D scaffolds with the ability of multi-drug loading and governing the release rate of each drug for tissue engineering. The self-assembly study confirmed that such peptide-functionalized amphiphilic copolymers underwent different self-assembly behavior. Amino bisphosphonate compounds are found in a vast array of products from water chelators to flame retardants to pharmaceuticals. Previous methods to synthesize these compounds involved the use of protic acids, which don’t provide ideal conditions for the starting materials containing acid labile functional groups in pharmaceutical compounds. The present study adopts a new synthetic route in which diethyl phosphite addition to the nitrile would be effected in the presence of a Lewis acid catalyst. In order to properly evaluate the ecological and human health risks of ZnO Manufactured nanomaterials (MNMs) released to the environment, it is critical to understand the likely transformation products in the wastewater treatment process and in soils receiving biosolids. To address this critical knowledge gap, we examined the transformation reactions of 30 nm ZnO MNMs in single component and multi-component systems, with phosphate and natural organic matter (NOM). Current wet chemical methods for biomass composition analysis using two-step sulfuric acid hydrolysis are time-consuming, labor-intensive, and unable to provide structural information about biomass. Infrared techniques provide fast, low-cost analysis, are non-destructive, and have shown promising results. Chemometric analysis has allowed researchers to perform qualitative and quantitative study of biomass with both near-infrared and mid-infrared spectroscopy. This review summarizes the progress and applications of infrared techniques in biomass study, and compares the infrared and the wet chemical methods for composition analysis. Inductively coupled plasma mass spectrometry (ICPMS) is an analytical technique that can be used to determine multiple elements and isotope ratios in a wide variety of samples. In this work, four projects are described that involve the method development using laser ablation (LA) and solution-based ICPMS and their application in environmental science and forensic (provenance) investigations. In the first project, plutonium (Pu) concentrations and 240Pu/ 239Pu atom ratios were measured in kelp, lichen, aquatic moss, and horse mussel collected on or near Amchitka Island, a former nuclear test in Alaska, and at a background site. The human immunodeficiency virus (HIV) requires a programmed -1 ribosomal frameshift for Pol gene expression. The HIV frameshift site consists of a heptanucleotide slippery sequence (UUUUUUA) followed by a spacer region and a downstream RNA stem-loop structure. Here we investigate the role of the RNA structure in promoting the -1 frameshift. The stem-loop was systematically altered to decouple the contributions of local and overall thermodynamic stability towards frameshift efficiency. No correlation between overall stability and frameshift efficiency is observed. In contrast, there is a strong correlation between frameshift efficiency and the local thermodynamic stability of the first 3-4 bp in …

A libraries from libraries combinatorial chemistry approach was employed to synthesize fluorinated derivatives of both oxindoles and isatins as potential pharmaceuticals or targeting agents for imaging purposes related to cancer or Alzheimer's disease. Synthesis for these fluorinated derivatives are described by routes involving, either: a) N-alkylation of 5-substituted isatins followed by Wolff-Kishner reduction to the corresponding oxindoles and final Knoevenagel condensation with aryl aldehydes, or; b) Wolff-Kishner reduction of the isatins followed by condensation and finishing with the N-alkylation of the aldol products. The synthesis of the arylnaphthalene lignan haplomyrtin using vanillin as a starting material is synthesized via: 1) a bromination of vanillin, 4-hydroxy-5-methoxybenzaldehyde, to produce 6-bromovanillin 2) a hydroxyl protection step of 6-bromovanillin with triisopropylsilylchloride to produce 2-bromo-4-triisopropylsilyoxy-5-methoxybenzaldehyde 3) an aldehyde protection step through a cyclic acetal formation using ethylene glycol to produce 2-(2-bromo-4-triisopropylsilyloxy-5-methoxyphenyl)-1,3-dioxolane 4) a lithium for halogen exchange of the doubly protected 6-bromovanillin leads to neucleophilic coupling with piperonal producing 1,3-benzodioxol-5-yl-[5-triisopropylsilyloxy-2-(1,3-dioxolane-2-yl)-4-methoxyphenyl]methanol and 5) an acid catalyzed intramolecular cycloaddition /Diels Alder adduct with dimethyl acetylenedicarboxylate producing dimethyl 1-(1,3-benzodioxol-5-yl)-4-hydroxy-6-methoxy-7-triisopropylsilyloxynaphthalene-2,3-dicarboxylate.

Hyperbranched poly(ethylenimine)s (HPEI) were modified with hydrophobic isobutyryl amide groups (HPEI-IBAm0.60) to cause an LCST. These modified polymers were then further substituted with hydrophobic alkyl chains (HPEI-IBAm0.60-R0.40), a mixture of alkyl chains and hydroxyethyl groups (HPEI-IBAm0.60-EtOH0.20-R0.20), and a mixture of alkyl chains and low molecular weight PEG (HPEI-IBAm0.60-PEG0.20-R0.20) to determine the effect of different functional groups on solubility behavior. A recently development thermal infrared camera system was improved by designing new components to downsize physical aspect and improve the instrument's performance. A new thermal infrared light source was developed and characterized. It is designed from a custom-made Macor backpiece that holds an alumina screw. High resistance wire is coiled around the screw. The coiled wire is covered with an alumina coating. Most light sources have a spectral emissivity similar to that of a blackbody emitter. A colloidal solution of citrate-stabilized gold nanoparticles (AuNPs) with an average size of ca. 2.6 nm has been prepared, characterized and further implemented in electro-sensing applications. This colloidal solution of AuNPs has been prepared via the reduction of NaAuCl4 with sodium tetrahydroborate (NaBH4) using trisodium citrate as a stabilizer. The optical properties of this solution have been studied with UV–Vis spectroscopy. Next, these AuNPs have been immobilized onto a polycrystalline Au (poly-Au) electrode with the assistance of benzenedimethanethiol (BDMT), which served as a binder. A novel route to the exponential trapping-time distribution within a solidlike state in water clusters is described. We propose a simple homogeneous network (SHN) model to investigate dynamics on the potential energy networks of water clusters. In this model, it is shown that the trappingtime distribution in a solidlike state follows the exponential distribution, whereas the trapping-time distribution in local potential minima within the solidlike state is not exponential. To confirm the exponential trapping-time distribution in a solidlike state, we investigate water clusters, (H2O)6 and (H2O)12, by molecular dynamics simulations. We explored the potentials of microarray printing using quill-like microcantilevers onto solid supports that are typically used in microspot printing, including paper, polymeric nitrocellulose and nylon membranes. We compared these membranes with a novel porous poly(2-hydroxyethyl methacrylate-co-ethylene dimethacrylate) support (HEMA) with narrow pore size distribution in the 150 nm range, which demonstrated advantages in pattern definition, spot homogeneity, and consistent spot delivery of different dyes (phloxine B and bromophenol blue) with diameters of several micrometres. Riboswitches are structural elements in the 50 untranslated regions of many bacterial messenger RNAs that regulate gene expression in response to changing metabolite concentrations by inhibition of either transcription or translation initiation. The preQ1 (7-aminomethyl-7-deazaguanine) riboswitch family comprises some of the smallest metabolite sensing RNAs found in nature. Once ligand-bound, the transcriptional Bacillus subtilis and translational Thermoanaerobacter tengcongensis preQ1 riboswitch aptamers are structurally similar RNA pseudoknots; yet, prior structural studies have characterized their ligand-free conformations as largely unfolded and folded, respectively.
