Publications

Inertial kinetics and kinematics have substantial influences on human biomechanical function. A new algorithm for Inertial Measurement Unit (IMU)-based motion tracking is presented in this work. The primary aims of this paper are to combine recent developments in improved biosensor technology with mainstream motion-tracking hardware to measure the overall performance of human movement based on joint axis-angle representations of limb rotation. This work describes an alternative approach to representing three-dimensional rotations using a normalized vector around which an identified joint angle defines the overall rotation, rather than a traditional Euler angle approach. Furthermore, IMUs allow for the direct measurement of …

Development And Application Of A Microfluidic Platform For Quantifying Intra-Tumoral Compressive Stress., Zachary P. Fowler

Electronic Theses and Dissertations

Cancer progression is linked to the emergence of aberrant mechanical signaling in the tumor microenvironment. Modulation of extrinsic signals, such as ECM stiffness and composition, have been thoroughly explored. However, the development of solid stresses within the tumor remains poorly understood. To address this, we have developed a microfluidic platform that generates deformable alginate microbeads that allow for the quantification of compressive stresses generated within a growing glioblastoma (GBM) tumorsphere. PDMS microfluidic devices were fabricated via SU-8 mold with channels ranging from 10µm-40µm in diameter. Fluorescently labeled sodium alginate underwent a cross-linking reaction within the device to generate monodisperse beads …

A Novel Biomaterial Platform For Cardiac Tissue Engineering, Andrew House

Dissertations

Heart disease is one of the leading causes of death in the world. With a growing demand for in vitro cardiac models that can recapitulate the complex physiology of the cardiac tissue, there is a desire to create novel therapies and treatments. However, due to a lack of fundamental understanding of the heart's inflammatory response to myocardial infarction, the development of new therapies is both time consuming and costly. In this work, a novel approach to fabricating a dynamic hydrogel cardiac platform is reported. This platform better mimics the dynamic changes occurring in the native cardiac tissue microenvironment during cardiac …

Integrative Physiology-Coupled Pilot-Centered Flight Simulation, Shawn C. Harrison

Electrical & Computer Engineering Theses & Dissertations

Maintaining the pilot’s physiological performance envelope within the limits of human capabilities may be crucial for avoiding hazardous physiological episodes in fighter aircraft that compromise safety. The main physiological episode of interest is impaired pilot respiration, better known as hypoxia caused by a high fraction of inspired oxygen (FiO2) at high altitudes and variation in accelerative gravitational forces (g-forces). Integrated into fighter aircraft is an Onboard Oxygen Generating System (OBOGS) developed to mitigate the necessity of gaseous and liquid oxygen cannisters [1]. OBOGS act as a life support in hypoxic environments by providing oxygen-rich air, thereby oxygenating the …

Engineering Of Ideal Systems For The Study And Direction Of Stem Cell Asymmetrical Division And Fate Determination, Martina Zamponi Aug 2022

Engineering Of Ideal Systems For The Study And Direction Of Stem Cell Asymmetrical Division And Fate Determination, Martina Zamponi

Biomedical Engineering Theses & Dissertations

The cellular microenvironment varies significantly across tissues, and it is constituted by both resident cells and the macromolecules they are exposed to. Cues that the cells receive from the microenvironment, as well as the signaling transmitted to it, affect their physiology and behavior. This notion is valid in the context of stem cells, which are susceptible to biochemical and biomechanical signaling exchanged with the microenvironment, and which plays a fundamental role in establishing fate determination and cell differentiation events. The definition of the molecular mechanisms that drive stem cell asymmetrical division, and how these are modulated by microenvironmental signaling, is …

Oxygen levels in vivo are autonomously regulated by a supply–demand balance, which can be altered in disease states. However, the oxygen levels of in vitro cell culture systems, particularly microscale cell culture, are typically dominated by either supply or demand. Further, the oxygen microenvironment in these systems is rarely monitored or reported. Here, a method to establish and dynamically monitor autonomously regulated oxygen microenvironments (AROM) using an oil overlay in an open microscale cell culture system is presented. Using this method, the oxygen microenvironment is dynamically regulated via the supply–demand balance of the system. Numerical simulation and experimental validation of …

Cfd Analysis Of Subcutaneous Deposition And Dispersion Of Insulin In Adipose Tissue, Ryan C. Lubbers Aug 2021

Cfd Analysis Of Subcutaneous Deposition And Dispersion Of Insulin In Adipose Tissue, Ryan C. Lubbers

Masters Theses

Drug delivery is the most important factor of many therapies, but a lack of technology and research have led to a very generalized understanding of drug kinetics. Insulin pump therapy for the treatment of Type 1 Diabetes depends on precise delivery of the hormone into the adipose region between the dermis and underlying muscle. The kinetics of insulin within the adipose tissue environment is not well understood and varies greatly case to case. The use of computation fluid dynamics (CFD) models to study insulin kinetics in relation to influential factors will lead to a better understanding of the characteristics of …

Subclinical Atherosclerosis Quantified Through Cumulative Shear Measurement, Margaret Lynne Papka Aug 2021

Subclinical Atherosclerosis Quantified Through Cumulative Shear Measurement, Margaret Lynne Papka

Master's Theses

With the high mortality rate of cardiovascular disease, it is important to study the early signs. The early detection of cardiovascular disease can lead to saved lives. Currently the most prevalent detection methods are the Framingham Risk Score and the carotid intima media thickness, both of which are insufficient. The necessary tool for early detection requires a uniform quantification system. The stimulus leading to endothelial dysfunction, the most significant predictor of a major adverse cardiovascular event (MACE)—and subsequently subclinical atherosclerosis—is reduced shear stress. Increased surface relative roughness affects the flow profile transition from laminar to turbulent resulting in reduced shear …

Optimization Of Mechanical Properties At The Infarct Border Zone, Kaushik Sunder Jul 2021

Optimization Of Mechanical Properties At The Infarct Border Zone, Kaushik Sunder

LSU Master's Theses

A myocardial infarction (MI) occurs every 39 seconds in the United States. As of 2016, an estimated 550,000 new and 200,000 recurrent episodes of myocardial infarction (MI) occur each year in the United States was. Infarction is often distinguished by a considerable decrease in wall thickness and an increase in heart size or left ventricular volume. This behavior happens when there is a prolonged lack of blood flow, usually due to damage or clotting, which causes cardiomyocytes to die (CMs). The development of non-contractile infarct scarring is the primary pathophysiology of MI. While the infarct scar prevents myocardial rupture during …

Modeling The Effect Of Igg Subclasses And Specificity In The Translocation Of Monoclonal Antibodies Across The Placental Barrier, Sayuri Tais Miyamoto Magnabosco

ENGS 88 Honors Thesis (AB Students)

Infections are responsible for over half a million neonatal deaths every year (Lawn et al., 2014). Thus, there is huge interest in leveraging maternal immunization against infectious diseases to grant fetal protection during its development through the vertical transferring of IgG antibodies, the only Ig subclass that can significantly cross the placental barrier. Studies about vertical immunization rely on in-vitro models to extrapolate physiological conditions of the human placenta. The BeWo Transwell model (Bode et al., 2006) presents itself as a reliable model to mimic the transplacental transport mechanism of antibodies (Ellinger et al., 1999; Poulsen et al., 2009) …

Establishing A Mechanistic Link Between Disturbed Flow And Aneurysm Formation In A 3d Cerebral Bifurcation Model, Nesrine Bouhrira

Theses and Dissertations

The effect of disturbed flow profiles on the endothelium have been studied extensively in systemic vasculature, but less is known about the response of the blood-brain barrier (BBB) to these flow regimes. Here we investigate the effect of steady and pulsatile disturbed flow on the integrity of the BBB using a three-dimensional, perfusable bifurcation model consisting of a co-culture of endothelial cells with mural and glial cells. Experimental flow patterns predicted by computational fluid dynamics mimic in vivo flow regimes, specifically the presence of a recirculation zone immediately downstream of the bifurcation reveal periodic changes in the instantaneous shear stress …

Correlates between EEG spectral bands and the subjective ratings were analyzed to identify statistically significant frequencies and electrode locations across participants. Model feasibility and …

Analysis Of Center Of Pressure And Center Of Mass To Determine Patterns Of Postural Stability In Neurotypical Subjects, Natalie Tipton Aug 2020

Analysis Of Center Of Pressure And Center Of Mass To Determine Patterns Of Postural Stability In Neurotypical Subjects, Natalie Tipton

Masters Theses

The body’s postural control mechanism is responsible for responding to perturbations of balance and keeping the body upright. One of the main ways that this is completed during quiet standing, where both feet are planted on the ground, is through center of pressure oscillations. In these oscillations, the center of pressure circles around the center of mass, constantly counteracting any lean that exists in the body. These oscillations can be recorded with floorembedded force plates and center of mass can be recorded with a motion capture system. In this research, these signals were recorded for stances with feet together and …

Development And Validation Of A Synchronous Model To Simulate Skeletal Muscle Damage Due To Spaceflight Ionizing Radiation And Microgravity, Lori M. Caldwell

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Astronauts traveling or researching in space are subject to a harsh environment that includes high levels of ionizing radiation and microgravity. Ionizing radiation is dangerous to overall health because it causes double-stranded DNA breaks, increases the production of free radicals or reactive oxygen species, and leads to an enhanced risk of cardiovascular disease and cancer over a person’s lifetime. Similarly, microgravity is hazardous to astronaut health because humans require gravity to maintain a healthy heart and muscular and vascular systems.

Both ionizing radiation and microgravity have been studied individually in previous research, but there is a lack of information on …
