Sickle Cell Anemia & Associated Neurological Complications: A Literature Review, Rodjmar Joseph
Sickle Cell Anemia & Associated Neurological Complications: A Literature Review, Rodjmar Joseph
Graduate Student Dissertations, Theses, Capstones, and Portfolios
Sickle cell anemia, an inherited disorder, causes red blood cells to contort into a disk or sickled shape becoming hard and sticky and obstructing blood flow. As the most common hemoglobinopathy, over 100,000 Americans in the US are affected by sickle cell disease. Sickle cell anemia is an inheritance of the abnormal sickle cell gene genetically transferred from both parents. Sickle cell anemia can be easily diagnosed in DNA through a blood test or genetic screening. With a 1 in 4 chance of inheriting copies of the sickle cell gene from both parents if both biological parents are carriers, the …

Cultivation Of Industrial Hemp On And Near Airports: Implications For Wildlife Use And Risk To Aviation Safety, Bradley F. Blackwell, Page E. Klug, Lee A. Humberg, Zachary T. Brym, Bryan M. Kluever, Jennifer M. Edwards
Human–Wildlife Interactions
Land-use planning on and near airports should consider possible revenue from land covers, associated maintenance costs, and potential for land covers to attract vertebrate species recognized as hazardous to aviation safety. The U.S. Federal Aviation Administration has expressed interest in recent attention given to industrial hemp (Cannabis sativa L.; hemp) as a revenue-producing land cover that might be cultivated on or near airports. Our purpose was to better understand the potential production value of hemp as well as its possible role in affecting aviation safety if cultivated on or near airports. Our objectives were to: (1) review the literature …

Anthropogenic Influences On The Decline, Restoration, And Eco-Evolutionary Dynamics Of Lake Superior’S Coaster Brook Trout, Austin Johnson
Anthropogenic Influences On The Decline, Restoration, And Eco-Evolutionary Dynamics Of Lake Superior’S Coaster Brook Trout, Austin Johnson
Dissertations, Master's Theses and Master's Reports
The coaster brook trout is a life history variant of the brook trout (Salvelinus fontinalis) that is characterized by either lake residency or migration between stream and lake habitats. Coaster brook trout were once widespread throughout Lake Superior and its tributaries, but populations declined sharply in the late 19th and early 20th centuries. Historically, brook trout were a popular target of recreational and subsistence fishing in the Lake Superior basin, and it has been hypothesized that angling pressure combined with multiple forms of industrial development are what drove the coaster brook trout’s decline. In the mid …
Effects Of Mindfulness And Stress Management On Neurocardiovascular And Psychological Outcomes, Aditi P. Vyas
Effects Of Mindfulness And Stress Management On Neurocardiovascular And Psychological Outcomes, Aditi P. Vyas
Dissertations, Master's Theses and Master's Reports
There has been a higher prevalence of developing anxiety due to frequent episodes of stress among adults in recent years. Chronic anxiety can contribute to the prevalence of elevated blood pressure and hypertension. High anxiety and stress also contribute to overactivation of the sympathetic nervous system which can be quantified by increased muscle sympathetic nerve activity (MSNA). Sympathetic overactivation can lead to vasoconstriction and loss of arterial elasticity. Anxiety, MSNA, blood pressure, and arterial stiffness are all interconnected, thus studying these relationships is crucial to understand the underlying mechanisms for prevention and treatment of cardiovascular disease (CVD). Non-pharmacological and mind-body …

Targeted Browsing With Goats For Eastern Redcedar (Juniperus Virginiana L.) Control, Alanna M. Hartsfield
Electronic Theses and Dissertations
As eastern redcedar (ERC) (Juniperus virginiana L.) grassland encroachment progresses, all potential control methods should be explored in the interest of Great Plains grassland health and longevity. Targeted browsing with goats has been proven as an effective control method on some juniper species; however, little is known about its ability to control ERC. These studies intend to mend knowledge gaps of how targeted browsing with goats control ERC by causing tree death without chemicals or machinery. The first study is two 3x3 Latin squares comparing protein-supplemented diets. The second study is a randomized complete block design of five 0.224 ha …

Coronavirus disease 2019 (COVID-19) pandemic has been an important global interest that affected millions of people, and it requires a deep investigation of the disease immunology for developing further therapeutic applications. Adoptive T cell therapy promises to address T cell-dependent immune dysregulation in COVID-19 patients by the generation of specific T cell clones against virus-specific antigens. Additionally, targeting B cell-dependent protection through COVID-19 vaccines, which have been developed in the recent year, possessed sufficient prevention for spreading the virus, since the cases and deaths related to COVID-19 tend to decrease after the vaccination. However, adoptive cell therapies are now encouraging …

The multifunctional BAG-1 (Bcl-2 athanogene-1) protein promotes breast cancer survival through direct or indirect interaction partners. The number of the interacting partners determines its cellular role in different conditions. As well as interaction partner variability, the amount of BAG-1 protein in the cells could cause dramatic alterations. According to previous studies, while the transient silencing of Bag-1 enhanced drug-induced apoptosis, deletion of BAG-1 could induce stemness properties and Akt-mediated actin remodeling in MCF-7 breast cancer cells. Considering the heterogeneity of breast cancer and the variability of BAG-1 -mediated cell response, it has become essential to determine microRNA (miRNA) functions in …

Regulatory Roles Of An Atypical Ubiquitin Ligase Ube2o In Orphans Of Multi-Protein Complexes For Degradation, Yi Lv, Feiyue Xing
Turkish Journal of Biology
UBE2O as an atypical ubiquitin-conjugating enzyme possesses an E2-E3 hybrid enzyme activity. It can regulate substrate levels or transcriptional activities by cooperating with other E3 ubiquitin ligases or forming homomeric complexes displaying intrinsic E2 and E3 activities. UBE2O controls the quality of cell proteome including protein degradation, modification, transport and location. Recent studies reveal that UBE2O plays a vital role in intracellular protein ubiquitination processes by regulating BMP/SMAD, TRAF/ NF-κB, mTOR/HIF1a and IL-1ß/IRAK4 signaling pathways, c-Maf stability and BAP1 subcellular location, which is proposed as a quality control supervisor of multiprotein complexes for degradation. Its abnormality leads to a variety …

Cancer is a disease in which abnormal cells grow uncontrollably and invade other tissues. Several types of cancer have various subtypes with different clinical and biological implications. Based on these differences, treatment methods need to be customized. The identification of distinct cancer subtypes is an important problem in bioinformatics, since it can guide future precision medicine applications. In order to design targeted treatments, bioinformatics methods attempt to discover common molecular pathology of different cancer subtypes. Along this line, several computational methods have been proposed to discover cancer subtypes or to stratify cancer into informative subtypes. However, existing works do not …

Human SARS coronavirus 2 (SARS-CoV-2) causes the current global COVID-19 pandemic. The production of an efficient vaccine against COVID-19 is under heavy investigation. In this study, we have designed a novel multiepitope DNA vaccine against SARS-CoV-2 using reverse vaccinology and DNA vaccine approaches. Applying these strategies led to reduce the time and costs of vaccine development and also improve the immune protective characteristics of the vaccine. For this purpose, epitopes of nucleocapsid, membrane glycoprotein, and ORF8 proteins of SARS-CoV-2 chose as targets for B and T-cell receptors. Accordingly, DNA sequences of selected epitopes have optimized for protein expression in the …

Accumulating evidence shows that gut microbial dysbiosis may represent a risk factor for Parkinson's disease (PD). Exercise has a positive effect on microbiota in general. The effect of aerobic exercise training (AET) on the gut microbial environment in PD remains to be explored. Here, we performed the 16S rRNA gene sequencing on feces from sham operated-mice (sham), PD mice model, and mice receiving AET (AET). Results indicated that AET had no remarkable effect on species richness and bacterial diversity of PD mice. The relative abundance of the Bacteroidetes was reduced, while Firmicutes, Actinobacteria, Lactobacillaceae, Streptococcaceae, Lactobacillus, Streptococcus, Lactococcus, Lysinibacillus, Pelomonas, …

Diabetes resulting from insufficient insulin secretion or insulin resistance (IR) is a highly prevalent metabolic disease. Since microRNAs have been linked with elevated IR, the current research hypothesized that miR-124-3p has a role in IR and the establishment of IR and type 2 diabetes (T2DM). The study aimed to explore the molecular mechanisms of miR-124-3p which influence IR leading to T2DM establishment. HepG2 cells were cultured in vitro, and palmitic acid (PA) was used to construct the IR cell model. In the IR model, transfection of miR-124-3p or phosphoglucomutase 1 (PGM1) linked plasmids were transfected into HepG2 cells. RT-qPCR was …

Type 2 diabetes mellitus (T2D) constitutes 90% of the diabetes cases, and it is a complex multifactorial disease. In the last decade, genome-wide association studies (GWASs) for T2D successfully pinpointed the genetic variants (typically single nucleotide polymorphisms, SNPs) that associate with disease risk. In order to diminish the burden of multiple testing in GWAS, researchers attempted to evaluate the collective effects of interesting variants. In this regard, pathway-based analyses of GWAS became popular to discover novel multigenic functional associations. Still, to reveal the unaccounted 85 to 90% of T2D variation, which lies hidden in GWAS datasets, new post-GWAS strategies need …

Majority of 37 human aminoacyl tRNA synthetases have been incriminated in diverse, mostly recessive, genetic diseases. In accordance with this, we uncovered a novel homozygous valyl-tRNA synthetase 1 (VARS1) gene variant, leading to p.T1068M mutation. As in the previously reported VARS1 mutations, the affected individual harboring p.T1068M was experiencing a neurodevelopmental disorder with intractable seizures, psychomotor retardation, and microcephaly. To link this phenotypic outcome with the observed genotype, we structurally modeled human VARS1 and interpreted p.T1068M within the spatial distribution of previously reported VARS1 variants. As a result, we uncovered that p.T1068M is clustered with three other pathogenic …

Background/aim: Despite great progress in complex gene regulatory mechanisms in the dynamic tumor microenvironment, the potential contribution of long noncoding RNAs (lncRNAs) to cancer cell metabolism is poorly understood. Death receptor 5 antisense (DR5-AS) is a cisplatin inducible lncRNA whose knockdown modulates cell morphology. However, its effect on cell metabolism is unknown. The aim of this study is to examine metabolic changes modulated by cisplatin and DR5-AS lncRNA in HeLa cells. Materials and methods: We used cisplatin as a universal cancer therapeutic drug to modulate metabolic changes in HeLa cervix cancer cells. We then examined the extent of metabolic changes …
The Role Of Slu7 In Spliceosome Function In Saccharomyces Cerevisiae, Sovira A. Chaudhry
The Role Of Slu7 In Spliceosome Function In Saccharomyces Cerevisiae, Sovira A. Chaudhry
Theses and Dissertations
The removal of introns from pre-messenger RNA via splicing in the nucleus is an essential step in gene expression. In recent decades, significant effort has been made to understand the chemical mechanisms of splicing reactions and the composition of the spliceosome that catalyzes them. Yet very little is known about the exact function of many splicing factors and small proteins within the spliceosome. Slu7, a pre-mRNA splicing factor, is essential during the second catalytic step of splicing because it interacts with several proteins and stabilizes the active site conformation. It has been found that Slu7 is involved in 3' splice …

As human population increases in the years to come so too will the agriculture business. We already know that the use of chemicals such as fertilizers and other agriculture chemicals aid to promote accelerated crop growth so that we can harvest the most food for the most people. There is less research on the effects that the runoff of these chemicals have on wetland environments that are downstream from farmland. I would like to explore how some of these agricultural chemicals, like ammonium, will affect the growth of different types of wetland graminoids. I will be looking at Poa polustris, …

The oral behavior of drinking includes interactions between the tongue, jaw, and liquid being ingested to provide hydration to the animal. Different mechanisms may be used to ingest liquid such as lapping, licking, or sucking. The objective of the study was to compare the mechanism of lapping across marsupial species through comparison of the length of gape cycle, and the amplitude of jaw pitch and tongue protraction between the two marsupial species, Didelphis virginiana and Monodelphis domestica. I hypothesized that there would be no differences in any of the variables for the lapping mechanism between species. Previous work demonstrates …

Effect Of Glaucoma On Melanopsin Retinal Ganglion Cells In Mice, Abigail Mcmullen
Williams Honors College, Honors Research Projects
Within the mammalian retina, there are melanopsin-containing ganglion cells. These intrinsically photoreceptive cells help control behavioral aspects of vision as well as to aid in circadian rhythms. In this project, it will be determined whether or not glaucoma has an effect on these cells, and what that effect is, from different points in the disease. Glaucoma is an eye disease that damages the retinal ganglion cells and eventually causes them to die, also it can be linked to irregular circadian rhythm. Many techniques will be used to prepare this project but computer imaging will be the primary method for determining …

Asian clams (Corbicula spp.), zebra and quagga mussels (Dreisenna spp.) have invaded and spread throughout North American surface waters. Corbicula and Dreisenna species bio foul aquatic systems, occupy benthic substrates and degrade environments through shell deposition. I explored how Dreissena and Corbicula invasions affect benthic fish and macroinvertebrate communities, and examine how their impacts differ between urban and rural systems, and temperate and tropical climates. Macroinvertebrate and fish communities were evaluated at sites with increasing shell densities in the Rouge, and Huron rivers (MI, USA) using the United States Environmental Protection Agency’s Rapid Bioassessment Protocols (RBP). Urban and rural macroinvertebrate …
