The percent of people with mental health disorders in the United States have skyrocketed over the past decade. With stigma surrounding the discussion of mental health and the symptoms associated with it, often those that are suffering do not receive sufficient treatment, as they might with a physical illness. Polycyclic Aromatic Hydrocarbon (PAH) from crude oil and other sources is one of the most serious environmental problems. PAH has an adverse effect on human health, water bodies and agricultural products. Removal of PAH through the physical and chemical methods are expensive and not environmentally friendly. Bioremediation, which is the use of microorganisms to degrade environmental pollutants like naphthalene and anthracene and render them less toxic, provides a cheaper and safer method. Triple negative breast cancers (TNBC) are closely related to basal-like cancers and classified based on their molecular signatures and their progenitor cell type. TNBCs lack the presence of three common types of receptors known to fuel breast cancer growth: estrogen receptors (ER), progesterone receptors (PR), and human epidermal growth factor receptors 2 (HER2neu). TNBC represent 10-20% of all molecular breast cancer subtypes. Even though genomic and transcriptome analyses show that many of the molecular signatures associated with TNBC are not related to ethnicity, clinicians and researchers find that African American (AA) TNBC women have higher mortality rates compared to Caucasian …

Breast cancer is the second leading cause of cancer death among American women. Traditional therapies, such as chemotherapy and radiation therapy, target proliferative cells within the tumor bulk. However, these treatments do not provide long-term disease survival because they fail to eliminate a small population of long-lived tumor initiating cells known as breast cancer stem cells (CSCs). Two features of CSCs are that they slowly divide and are more resistant to conventional anti-cancer treatments due to altered cellular machinery. A previous study conducted in our laboratory demonstrated V-Myb Avian Myeloblast Viral Oncogene Homolog Like 1 (MYBL1) gene over-expression in triple negative breast cancer (TNBC) compared to normal, some luminal, and a subpopulation of other TNBC. The MYBL1 gene belongs to the Avian myeloblastosis virus (MYB) family and is classified as proto-oncogene that functions as a strong transcription factor. The MYBL1 gene is related to cancer progression which involves dysregulation of cell cycle signaling, apoptosis and differentiation processes. Narrow, deep, and elaborate dental fissures are widely believed to increase the predisposition of enamel to develop carious lesions as these surfaces are thought to be ideal for cariogenic bacteria to adhere to. Therefore, dentists prescribe sealants as a preventative measure for patients with such fissures to avoid tooth decay. Yet to date, there is no objective data supporting the notion that fissure morphology actually affects caries susceptibility, nor is there a quantifiable, clinically practical method to characterize fissure patterns expected to increase risk of caries disease.

In the electron transport chain (ETC), Cc transfers electrons from cytochrome bc1 to cytochrome c oxidase (CcO), which helps build the electrochemical gradient that drives ATP synthase. Transcription factors recognize and bind to DNA sequences within these regulatory regions and affect said regulatory elements. The extent and specific circumstances regarding transcription factors are long-standing questions in the field. In this thesis, we wanted to investigate how transcription factors work in a particular case of enhancers, known as intragenic enhancers. The centrifuge force microscope (CFM) is one such technique that allows for high-throughput single-molecule pulling experiments. The circadian rhythm affects behavior and physiology in many animal species. Our previous work in mice showed that within the hippocampus, a subordinate circadian oscillator, astrocytes change their proximity to excitatory synapses. Here we obtain 3D reconstructions of biocytin-filled hippocampal astrocytes processed with protein retention expansion microscopy and imaged using two-photon laser scanning microscopy. Our findings identify the subtle changes in astrocyte morphologies that encode information about time-of-day in hippocampal astrocytes. These findings shed light on fundamental mechanisms that allow the function of hippocampal circuits to adapt to different times of the day. This will help us understand how structural …

Gram-negative bacteria account for the majority of antibiotic resistant infections. Regulated modifications within the cell envelope can promote resistance to clinically important antibiotics. The increased prevalence of antibiotic treatment failure highlights the importance of investigating the underlying resistance mechanisms. Of particular concern is resistance to last-line antimicrobials, such as polymyxin E (colistin). Colistin is a cationic antimicrobial lipopeptide that binds the negatively charged lipid A domain of lipopolysaccharide, a glycolipid enriched in the Gram-negative outer membrane, to perturb the envelope and lyse the cell. Sex determination is a critical element of successful vertebrate development, suggesting that sex chromosome systems might be evolutionarily stable across lineages. For example, mammals and birds have maintained conserved sex chromosome systems over long evolutionary time periods. Other vertebrates, in contrast, have undergone frequent sex chromosome transitions, which is even more amazing considering we still know comparatively little across large swaths of their respective phylogenies. Few studies have explored the evolutionary mechanisms that maintain adaptive immunogenetic diversity in nature. We took advantage of museum samples to test for evidence of parasite-mediated fluctuating selection at MHC Class II-ß loci in an endemic island reptile. The Saban anole Anolis sabanus is commonly infected with three species of malaria (Plasmodium). Proportions of each parasite species detected in anole blood samples fluctuated over space and time, suggesting competitive interactions between parasites or differences in vector ecology. Our analyses of parasite prevalence and MHC Class II-ß allelic variation found that malaria infection was not associated with patterns of …

This project investigated the importance of stresses and the immune system response of hydrogen peroxide formation within the model system Physcomitrella patens, as well as in Mnium cuspidatum. I examined the role of two hormones, abscisic acid and jasmonic acid, along with the role that drought-like conditions play in the specified defense response of hydrogen peroxide production. Climate change is inducing changes in the distributions of many species, causing range shifts and habitat loss as well as facilitating invasions. It is a broad contributor to global amphibian decline, already causing mass extinctions and extirpations of amphibian populations, a phenomenon which is expected to continue. The understudied direct-developing frog Eleutherodactylus cystignathoides is a notable outlier, having rapidly expanded its distribution in recent years. This is of particular interest given that other eleutherodactylids, such as E. coqui and E. planirostris, have already shown propensity for rapid dispersal and invasion.
