The Influence Of Stored Product Mites On The Function Of Skin Microvascular Endothelial Cells, Jennifer Graff Curp
The Influence Of Stored Product Mites On The Function Of Skin Microvascular Endothelial Cells, Jennifer Graff Curp
Browse all Theses and Dissertations
The prevalence of allergenic diseases and sensitization to stored product mites is increasing worldwide. Stored product mites are prevalent in homes, foods, and some work environments. Stored product mites need to be further studied so we can characterize their inflammatory properties to understand their role in skin disease, and provide proper treatment to patients with mite allergies. The purpose of the study was to show the role of stored product mites in modulating the inflammatory and immune response to human dermal endothelial cells in the skin. The epidermis serves as the first line of defense between the body and the …

Seasonal Patterns Of Arthropod Diversity And Abundance On Big Sagebrush, Artemisia Trientata, M P. Stanford, Nancy J. Huntly
Biology Faculty Publications
The sagebrush biotype is the largest in the western United States. This vast sagebrush community is thought to harbor equally vast and diverse arthropod communities, but these remain little explored. Our objective was to examine the diversity, abundance, and seasonal phenology of arthropod taxa found on the dominant shrub of the sagebrush ecosystem, big sagebrush (Artemisia tridentata). We wanted to improve understanding of this little-studied arthropod assemblage that may play significant roles in the dynamics of sagebrush populations and the sagebrush ecosystem. We sampled free-living and gall-forming arthropods from a stratified random sample of sagebrush plants at the Barton Road …
Loss Of Foundation Species Increases Population Growth Of Exotic Forbs In Sagebrush Steppe, J S. Prevey, M J. Germino, Nancy J. Huntly
Loss Of Foundation Species Increases Population Growth Of Exotic Forbs In Sagebrush Steppe, J S. Prevey, M J. Germino, Nancy J. Huntly
Biology Faculty Publications
The invasion and spread of exotic plants following land disturbance threatens semiarid ecosystems. In sagebrush steppe, soil water is scarce and is partitioned between deeprooted perennial shrubs and shallower-rooted native forbs and grasses. Disturbances commonly remove shrubs, leaving grass-dominated communities, and may allow for the exploitation of water resources by the many species of invasive, tap-rooted forbs that are increasingly successful in this habitat. We hypothesized that exotic forb populations would benefit from increased soil water made available by removal of sagebrush, a foundation species capable of deep-rooting, in semiarid shrub-steppe ecosystems. To test this hypothesis, we used periodic matrix …
The Combined Influence Of The Local Environment Andregional Enrichment On Bird Species Richness, Ethan P. White, A. H. Hurlbert
The Combined Influence Of The Local Environment Andregional Enrichment On Bird Species Richness, Ethan P. White, A. H. Hurlbert
Biology Faculty Publications
It is generally accepted that local species richness at a site reflects the combined influence of local and regional processes. However, most empirical studies evaluate the influence of either local environmental variables or regional enrichment but not both simultaneously. Here we demonstrate the importance of combining these processes to understand continental scale richness patterns in breeding birds. We show that neither regional enrichment nor the local environment in isolation is sufficient to characterize observed patterns of species richness. Combining both sets of variables into a single model results in improved model fit and the removal of residual spatial autocorrelation. At …

Background Despite considerable progress in our understanding of land plant phylogeny, several nodes in the green tree of life remain poorly resolved. Furthermore, the bulk of currently available data come from only a subset of major land plant clades. Here we examine early land plant evolution using complete plastome sequences including two previously unexamined and phylogenetically critical lineages. To better understand the evolution of land plants and their plastomes, we examined aligned nucleotide sequences, indels, gene and nucleotide composition, inversions, and gene order at the boundaries of the inverted repeats. Results We present the plastome sequences of Equisetum arvense, a …

Background Tortula ruralis, a widely distributed species in the moss family Pottiaceae, is increasingly used as a model organism for the study of desiccation tolerance and mechanisms of cellular repair. In this paper, we present the chloroplast genome sequence of T. ruralis, only the second published chloroplast genome for a moss, and the first for a vegetatively desiccation-tolerant plant. Results The Tortula chloroplast genome is ~123,500 bp, and differs in a number of ways from that of Physcomitrella patens, the first published moss chloroplast genome. For example, Tortula lacks the ~71 kb inversion found in the large single copy region …

After transcription, a large number of cellular RNAs employ modifications to increase their diversity and functional potential. Modifications can occur on the base, ribose, or both, and are important steps in the maturation of many RNAs. Our lab recently showed that plant microRNAs (miRNAs) possess a 2′-O-methyl group on the ribose of the 3′ terminal nucleotide, and that this methyl group is added after miRNA/miRNA* formation. One function of this modification is to protect miRNAs from 3′ terminal uridylation by an unknown enzymatic activity. It is possible that uridylation of miRNAs triggers their degradation. Here we describe a …

Nm23-H1 is a well-known tumor metastasis suppressor, which functions as a nucleoside-diphosphate kinase converting nucleoside diphosphates to nucleoside triphosphates with an expense of ATP. It regulates a variety of cellular activities, including proliferation, development, migration and differentiation known to be modulated by a series of complex signaling pathway. Few studies have addressed the mechanistic action of Nm23-H1 in the context of these cellular processes. To determine the downstream pathways modulated by Nm23-H1, we expressed Nm23-H1 in a Burkitt lymphoma derived B-cell line BJAB and performed pathway specific microarray analysis. The genes with significant changes in expression patterns were clustered in …

Amyloid-β (Aβ) overproduction and dendrite arbor atrophy are hallmarks of Alzheimer’s disease. The RhoA GTPase (Rho) signals through Rho kinase (ROCK) to control cytoskeletal dynamics and regulate neuron structure. Hyperactive Rho signaling destabilizes neurons leading to dendritic regression that can be rescued by genetic or pharmacological reduction of ROCK signaling. To understand what effect reduced ROCK signaling has on the dendrite arbors of mice that overproduce Aβ, we administered the ROCK inhibitor fasudil to AβPP/PS1 transgenic mice. We report that increased dendrite branching occurs in AβPP/PS1 mice and that fasudil promotes …

Directed cell migration requires precise spatial control of F-actin-based leading edge protrusion, focal adhesion (FA) dynamics, and actomyosin contractility. In spreading fibroblasts, the Abl family kinases, Abl and Arg, primarily localize to the nucleus and cell periphery, respectively. Here we provide evidence that Abl and Arg exert different spatial regulation on cellular contractile and adhesive structures. Loss of Abl function reduces FA, F-actin, and phosphorylated myosin light chain (pMLC) staining at the cell periphery, shifting the distribution of these elements more to the center of the cell than in wild-type (WT) and arg—/— cells. Conversely, loss of Arg function …

Efficient mitochondrial function requires physical interactions between the proteins encoded by the mitochondrial and nuclear genomes. Co-evolution between these genomes may result in the accumulation of incompatibilities between divergent lineages. We test whether mitochondrialnuclear incompatibilities have accumulated within the Drosophila melanogaster species subgroup by combining divergent mitochondrial and nuclear lineages and quantifying the effects on relative fitness. Precise placement of nine mtDNAs from D. melanogaster, D. simulans and D. mauritiana into two D. melanogaster nuclear genetic backgrounds reveals significant mitochondrial-nuclear epistasis affecting fitness in females. Combining the mitochondrial genomes with three different D. melanogaster X chromosomes reveals significant epistasis …

The c-Abl tyrosine kinase is present in mouse brain synapses, but its precise synaptic function is unknown. We found that c-Abl levels in the rat hippocampus increase postnatally, with expression peaking at the first postnatal week. In 14 d in vitro hippocampal neuron cultures, c-Abl localizes primarily to the postsynaptic compartment, in which it colocalizes with the postsynaptic scaffold protein postsynaptic density protein-95 (PSD-95) in apposition to presynaptic markers. c-Abl associates with PSD-95, and chemical or genetic inhibition of c-Abl kinase activity reduces PSD-95 tyrosine phosphorylation, leading to reduced PSD-95 clustering and reduced synapses in treated neurons. c-Abl can phosphorylate …

The Polypeptide Syn67 Interacts Physically With Human Holocarboxylase Synthetase, But Is Not A Target For Biotinylation, Yousef I. Hassan, Hideaki Moriyama, Janos Zempleni
School of Biological Sciences: Faculty Publications
Holocarboxylase synthetase (HCS) catalyzes the binding of biotin to lysines in carboxylases and histones in two steps. First, HCS catalyzes the synthesis of biotinyl-5′-AMP; second, the biotinyl moiety is ligated to lysine residues. It has been proposed that step two is fairly promiscuous, and that protein biotinylation may occur in the absence of HCS as long as sufficient exogenous biotinyl-5′- AMP is provided. Here, we identified a novel polypeptide (Syn67) with a basic patch of lysines and arginines. Yeast-two-hybrid assays and limited proteolysis assays revealed that both N- and C-termini of HCS interact with Syn67. A potential target lysine in …

The molecular mechanism(s) underlying transition from CCR5 to CXCR4 usage of subtype C viruses remain largely unknown. We previously identified a subtype C HIV-1 infected child whose virus demonstrated CXCR4 usage along with CCR5 upon longitudinal follow-up. Here we delineated the molecular determinants of Env involved in expanded coreceptor usage. Residue changes in three positions of Env V3 domain are critical for the dual-tropic phenotype. These include: substitution of arginine at position 11, MG or LG insertion between positions 13 and 14, and substitution of threonine at the position immediately downstream of the GPGQ crown. Introducing these mutations into V3 …

Biocontrol Of Fusarium Wilt By Bacillus Pumilus, Pseudomonas Alcaligenes And Rhizobium Sp. On Lentil, Mohd Sayeed Akhtar, Uzma Shakeel, Zaki A. Siddiqui
Turkish Journal of Biology
The present study examined the effects of Bacillus pumilus, Pseudomonas alcaligenes, and Rhizobium sp. on wilt disease caused by Fusarium oxysporum f. sp. lentis and on the growth of lentil. Inoculation with F. oxysporum caused significant wilting, and reduced plant growth, the number of pods, and nodulation. Inoculation with B. pumilus together with P. alcaligenes caused a greater increase in plant growth, number of pods, nodulation, and root colonization by rhizobacteria, and also reduced Fusarium wilting to a greater degree than did individual inoculation. Use of Rhizobium sp. resulted in a greater increase in plant growth, number of pods, and …

The Effect Of 4-Chloroindole-3-Acetic Acid On Some Growth Parameters In Mung Bean Under Cadmium Stress, Mohammad Mazid, Barket Ali, Shamsul Hayat, Aqil Ahmad
Turkish Journal of Biology
Seedlings of mung bean (Vigna radiata L. Wilczek) cv. T-44 grown on a sand culture were exposed to cadmium (Cd) (100 µM) on day 10 of growth. These seedlings were subsequently sprayed with deionized water (control) or 4-chloroindole-3-acetic acid (4-Cl-IAA) (10^{-6} M) on day 15 and were sampled 30 days after sowing to assess growth and metabolite content. Growth (length, and fresh and dry mass of roots and shoots) and the level of metabolites (chlorophyll, nitrogen, phosphorus, total sugar, and soluble protein) in the leaves significantly decrease in response to Cd stress; however, the 4-Cl-IAA (10^{-6} M) treatment, both in …

Stimulatory Effects Of Ghrelin On Spontaneous Contractions In The Rat Myometrium, Alpaslan Dayangaç, Seli̇m Kutlu, Vahi̇t Konar
Turkish Journal of Biology
Ghrelin, a recently isolated hormone, has been reported to have modulatory effects on smooth muscle contractility. In this study, we investigated the effects of ghrelin on spontaneous contractions in the rat uterus in vitro. Myometrium strips were removed from Wistar rats following decapitation and placed in a jacketed tissue bath. After initiation of spontaneous contractions, control contractions were recorded for 10 min and various concentrations of ghrelin were added to the tissue bath cumulatively. The Wilcoxon signed ranks test was used for statistical analysis. Application of ghrelin augmented the spontaneous contractions in myometrial strips. The mean peak amplitudes of contractions …

The Effect Of Ascorbic Acid And H_2o_2 Treatment On The Stability Of Anthocyanin Pigments In Berries, Elham Nikkhah, Masoud Khaiamy, Reza Heidary, Ali Sorayaie Azar
Turkish Journal of Biology
Anthocyanins are natural pigments widely distributed in nature. Anthocyanin pigment molecules are a subclass of flavonoids. They are responsible for the red, purple, and blue colors observed in many flowers, fruits, and vegetables. Fruits and berries are the main sources of anthocyanins in nature. Anthocyanins are thought to contribute to the nutritive value of fruits and berries due to their antioxidative, anti-carcinogenic, anti-inflammatory, and anti-angiogenic properties. Anthocyanins can also improve the nutritional value of processed foods by preventing the oxidation of lipids and proteins. As such, identification of agents that can affect the stability of anthocyanins and the protective effect …

Germination Characteristics And Molecular Characterizations Of Some Wheat Varieties In Sulaimanyah By Ssr Marker, Nawroz Abdul-Razzak Tahir
Turkish Journal of Biology
The genetic diversity of 11 bread and durum wheat varieties was estimated using germination characteristic and molecular analysis. The phenotypic data showed highly significant differences between cultivars for germination speed, germination percentage, seedling length, fresh weight, and dry weight. Simple sequence repeats (SSRs) were used to examine the genomic diversity of wheat (Triticum aestivum L. and Triticum durum L.). Eleven wheat varieties of diverse origins were analyzed with 12 selected SSRs to provide uniform and maximum genome coverage. A total of 18 bread wheat and 20 durum wheat polymorphic allelic variants were detected, ranging from 1 to 4 per locus …

Effects Of Chromium Vi Stress On Green Alga Ulva Lactuca (L.), Di̇lek Ünal, Nuray O. Işik, Atakan Sukatar
Turkish Journal of Biology
In order to contribute to the understanding of its response to metal stress, Ulva lactuca (L.) was treated with different concentration of chromium (VI) (0.2, 0.5, 1, and 5 mM K2CrO4), and alterations in Fv/Fm ratio (maximal photochemical efficiency of PSII) and cell viability were investigated. Significant alterations of the maximal photochemical efficiency of PSII (Fv/Fm) ratio were observed in response to the increase in chromium concentration. The Fv/Fm ratio decreased in Ulva lactuca following 2 h treatment with 1 and 5 mM Cr(VI) solution. The cell viability was decreased in cells exposed to higher Cr(VI) concentrations. Thallus cell morphology …
