Effects Of Temperature And Ph On The Growth And Composition Of The Sporophyte And Gametophyte Generations Of Allomyces Arbusculus Butler (1911), Lawrence Wayne Lenz

Pure cultures of the sporophyte and gametophyte generations of Allomyces arbusculus Butler (1911) were grown under varied conditions of temperature and pH. On solid media the gametophyte did not grow at 10 and 15 C; the sporophyte began slow growth after a long lag period at 15 C. There were no significant differences in growth between the sporophyte and gametophyte at 20 and 25 C. Increasing colony areas occurred for both generations at temperatures between 30 and 40 C, with the largest amount of growth for either generation at 35 C.

During bacteriological studies of the Logan City Sewage Oxidation Ponds, Logan, Utah, a group of bacteria was discovered which could lyse killed cells of Escherichia coli. The present research effort was an attempt to determine whether two isolates from this earlier study might play a role in the reduction of coliforms and other bacteria in the oxidation ponds. It was also desired to characterize partially the lytic enzymes produced by the two isolates and to determine as far as possible the organisms' identity.

Experiments on the incorporation of 14C-protoporphyrin IX into chlorophyll indicate that ferrous iron may be involved in reactions between protoporphyrin IX and chlorophyll. Experiments using both health and chlorotic tobacco tissue for incorporation studies show that incorporation of protoporphyrin IX into chlorophyll was activated by Fe2+.

S-adenosylmethionine-magnesium protoporphyrin IX methyltransferase (E.C.2.1.1.11) which catalyzes the reaction between S-adenosylmethionine and Mg protoporphyrin IX to form S-adenosylhomocysteine and Mg protoporphyrin IX monomethyl ester was demonstrated in barley seedlings. In the epidermis of PET/Wmr mice the population of melanocytes reaches a peak and begins to decline during the first postnatal week, and has disappeared within four weeks. During this period a small number of weakly DOPA reactive miniature melanocytes are seen dispersed among the generally large, highly DOPA reactive melano­cytes typical of the animal. These miniature melanocytes appear early, are among the last to disappear, and their population remains relatively constant against the drastically changing population of typical melanocytes. Water consumption, and weight loss in response to water deprivation and saline water consumption, were studied in the Eutamias townsendii townsendii from Portland, Multnomah County, Oregon. These chipmunks occupy moist Pacific Northwest forests. The results of these studies suggest that Townsend's chipmunks are dependent on sources of drinking water, at least for normal above ground activity, that their tolerance for water deprivation is comparatively low, and that they are unable to obtain a net gain of water by drinking 0.2 M sodium chloride solutions. Chipmunks cannot drink 0.4 M NaCl solution and survive for more than a few days. Metabolic …

Dark respiration rates of the aerial shoots of Arceuthobium tsugense, obtained by manometric and IRGA techniques, show production of C02to range between 155-300 µl CO2 g-1h-1 with evidence of seasonal variation. Experiments with 14C02 indicate that the aerial shoots are capable of some photosynthetic CO2 fixation, with 10-15% of the available 14C incorporated by the plant tissue in one hour.The portions of the o aerial shoots which are most active in C02 fixation are the young terminal regions. Two bacteriophages which infect the myxobacterium Cytophaga succinicans, strains 14 and 16, were isolated from fish hatchery effluent. Each was submitted to a detailed study. These phages have a limited host range, even within the genus Cytophaga. The turbid plaques formed by these phages are similar in appearance to those produced by coliphage Lambda, and average 0.85 mm in diameter. Both phages have a latent period of 195 minutes and a rise period of an additional 165 minutes. Phage 0CS-16 has a burst size of 137 and phage 0CS-14 has a burst of 45. The present study has attempted to investigate some of the more basic questions concerning the toxicity and accumulation of mercury in the purple shore crab, Hemigrapsus nudus. This rocky intertidal denizen is found in abundance along the entire Pacific coast of the United States from Sitka, Alaska, to the Gulf of California where it dominates the mid-tide pool region. Local specimens were collected just north of the Pacific Marine Stations, Dillon Beach, Marin County, California. Recent experiments published in the literature have questioned the importance of the appearance of chromosomal puffs during larval development in Dipterans since Sciara larvae fed the vertebrate hormone cortisone apparently show no puffs without impairment of normal development. We have used Drosophila virilis larvae to test these results because distinct morphological criteria can be used to assess the developmental age of third instar larvae and predict the appearance of a specific puff. This study of the distribution of the fresh-water sponge, Meyenia fluviatilis, at the Lincoln Beach area of Utah Lake involved the following: First, sampling of the area for the presence or absence of the species. Second, sampling at two separate areas at different depth intervals to determine if the number of sponge colonies and colony size were in any way related to depth. Finally, monthly sampling at one area to determine when gemmules of M. fluviatilis were present in the sponge colonies. Quantitative Growth Relationships Of Microcoleus Vaginatus (Gom.) Ex Drouet And Alcaligenes Sp. In A Sucrose-Salts Medium, James Rexford Brownell

The blue-green alga, Microcoleus vaginatus (Gom.) ex Drouet 1968, and a bacterium, Alcaligenes sp., collected from the same habitat, were cultured together and separately in mineral salts media with and without sucrose. Growth of axenic M. vaginatus was stimulated by sucrose. In association with the bacterium in sucrose-containing media algal growth was suppressed, while in inorganic media it was stimulated. Factors involved in this algal-bacterial system probably included competition for the carbon source, changes in pH, and accumulation of cellular waste and lytic products.

A study was undertaken to compare proteins present in the gill membranes of freshwater and salt­ water adapted killifish, Fundulus heteroclitus. For this purpose, a method for the preparation or gill plasma membranes was developed. Polyacrylarnide gel electrophoresis indicated that the two types of gills possessed many membrane proteins in common. However, the freshwater adapted group possessed a single protein not found n the saltwater adapted group and the latter contained a protein not found in the freshwater adapted group.

The parasitic effects of the ichneumonid Bathyplectes curculionis (Thomson) on the larval alfalfa weevil, Hypera postica (Gyllenhal), were studied.

Results of experiments on the rates of parasitism of the four host larval instars indicated that the first three are either preferred by the parasite over the fourth instar larvae or are more susceptible to the parasite's attack. Survival of the younger weevil larvae after their exposure to female parasites was markedly poorer than that of unparasitized larvae. Premature death of host larvae was probably from both the puncturing by the parasite's ovipositor and the feeding and other activities of parasite …

Intestinal Absorption And Lipolysis Of Safflower Oil And Other Unsaturated Vegetable Oils In Rats, Robert Leslie Gregory

The intestinal lipolysis and absorption of safflower, corn, peanut, olive, cottonseed and soybean oils were studied in the rat. Oils and pancreatic lipase were injected into the rat jejunum and ileum (ligated in situ), and the amount of esterified fatty acids absorbed and free fatty acids present in the gut after 3 hours was determined.

Safflower oil was absorbed significantly less than the other oils. There was no significant difference between the absorption rates of the other oils. When the oils were subjected to porcine pancreatic lipase in vitro, safflower oil also exhibited the lowest rate of hydrolysis.

A study of the phytoplankton periodicity and its correlation with chemicophysical parameters, diel variation of the plankton, and the cyclomorphotic variation of Ceratium hirundinella was made in Shanty Hollow Lake, Warren County, Kentucky, from October, 1969, to August, 1970.

Phytoplankton samples were collected at zero and six feet with a Kemmerer sampler (31 October through 8 December) and with a ten -liter Birge-Juday plankton trap (23 December through 9 August). Water samples for the chemicophysical determinations were collected with a Kemmerer sampler. The phytoplankton samples were preserved with 5% formalin until they were enumerated. DMSO (dimethyl sulfoxide) and propham (isopropyl carbanilate) were investigated to determine their effect singly and in combination on mitoses in rye (Secale cereale) root tip cells. Root tips were prepared for observation by the paraffin and aceto-carmine squash methods. Results showed DMSO caused the following abnormalities: arrested metaphases, multipolar spindles, multinucleate cells, stickiness, shortened chromosomes, and mitotic inhibition. Cells from propham treated root tips exhibited the same abnormalities as those treated with DMSO. In addition, propham treatments resulted in polyploid cells and chromosome granulation. In combination, DMSO and propham caused all the abnormalities mentioned above except polyploidy. In …

The urolithiasis condition investigated occurred at a "hard water" hatchery in Utah after the trout were changed from a meat diet to a commercial pelleted fish feed as a sole sustaining diet in 1961. Numerous diet modifications proved unsuccessful in preventing urolithiasis at the hatchery. It was eventually determined that different commercial diets resulted in varied percentages of trout developing urolithiasis. The urolithic deposits found in the kidney tubules were amorphous and composed of calcium phosphate (apatite) and an organic matrix. Bacterial contamination of the kidneys was not an important factor in the formation of urolithiasis. Rainbow trout containing urolithic …

The oral plate sense organs of the honey bee, Apis mellifera L., have been investigated employing an electrophysiological technique which allows simultaneous stimulation and recording.

The results of the study present evidence that the four bipolar sense cells innervating the sensory papillae on the oral surface of the hypopharynx are chemoreceptors. The sensory papillae respond to cations, glucose, water, and amino acids. Evidence for the absence of a mechanosensory cell is presented. The data concerning the sensory structures agree with the current theories of chemoreception.

Genetic And Metabolic Controls For Sulfate Metabolism In Neurospora Crassa: Isolation And Study Of Chromate-Resistant And Sulfate Transport-Negative Mutants, George A. Marzluf

Mutants of Neurospora resistant to chromate were selected and all were found to map at a single genetic locus designated as cys-13. The chromate-resistant mutants grow at a wild-type rate on minimal media but are partially deficient in the transport of inorganic sulfate, especially during the conidial stage. An unlinked mutant, cys-14, is sensitive to chromate but transports sulfate during the mycelial stage at only 25% of the wild-type rate; cys-14 also grows at a fully wild-type rate on minimal media. The double-mutant strain, cys-13;cys-14, cannot utilize inorganic sulfate for growth and completely lacks the capacity to …

The experimental procedure described in this thesis consisted of a quanitative lipid analysis, a standard measurement of oxygen consumption, and determinations of fresh liver weight, and total body weight. The lipid analysis involved tissue extraction in a Soxlet apparatus (Rawat and Roy, 1958). The oxygen consumption measurements were made using a small animal respirometer, and fresh liver weights and total body weights were determined with an Ainsworth-Type-I0N analytical balance.

The objective of this study was to clarify the relationships between drift rates, population density, production rates, key environmental factors, and movements of adults within two populations of stream insects (a caddisfly--Oligophlebodes sigma and a mayfly--Baetis bicaudatus).

Samples of benthic invertebrates (424 total) were collected every 28 days at four stations on Temple Fork of the Logan River, Utah, from October, 1967 to September, 1969. Monolayer primary cultures of cells from bovine embryonic intestine (BEint), kidney (BEK), spleen (BES), and thyroid (BET) and cell line cultures of embryonic bovine trachea ( EBTr) and synovium (BESy)as well as established cell line cultures of Madin-Darby bovine kidney (MDBK), human intestine (Int 407), and Syrian hamster kidney (BHK-21) were inoculated with freshly excysted sporozoites of Eimeria alabamensis and observed for 4 days. Sporozoites penetrated all cell types during the first 24 hours in culture. Numerous intracellular sporozoites, trophozoites, and binucleate schizonts were seen in all cell cultures except Int 407.
