Q2S Enhancing Pedagogy
In the new, transformed Biology BS program for conversion from quarter to semester, multiple faculty will teach varied courses under the “Cell and Molecular” requirement for our program. Our old program required multiple core courses, leading to bottlenecks as students could not get seats in needed classes. We have designed our new program to provide students with options under core categories, providing students more flexibility which should improve time to graduation. Students must take at least ONE of the courses, although students who chose to focus on Cell and Molecular Biology should be able to learn new material in ALL …

Biological Pathways Associated With Wild And Domestic Animals, Cayla Steemer, Laura Carson, Jim Hu, Gloria Regisford
Pursue: Undergraduate Research Journal
Background: Zoonotic diseases are problematic, in that, they impact both wild and domestic animals alike. Thus, there is a need to investigate the genomes of wild and domestic. Gene ontology (GO) is a major bioinformatics initiative, whereby descriptions of gene products across the database are developed and unified to describe all species. This process is performed by biocurators, who gather, annotate, and validate information on the databases, consequently affording easy access to accurate and updated data. In this study, we investigated the biocuration of two biological processes, DNA integration, which is used for DNA coding, and the tricarboxylic acid (TCA) …

Journal of the Arkansas Academy of Science
We analyzed the energy content of seeds of common sunflowers (Helianthus annuus) obtained from the crops of scaled quail (Callipepla squamata) collected from plains-mesa sand-scrub habitat in Eddy and Lea counties, New Mexico. Seeds were removed from crops and dried for 48 hours at 60°C to remove moisture and to standardize masses. Seeds were then analyzed for gross caloric value (i.e., energy content) in an oxygen bomb calorimeter. Energy content of seeds of common sunflowers from New Mexico was greater than that of many seeds previously reported from the diet of scaled quail and other granivorous …
Characterizing The Transport Of Parasitoid Wasp Genaspis Hookeri Venom Serca Into Host Drosophila Hemocytes, Christopher Lark
Characterizing The Transport Of Parasitoid Wasp Genaspis Hookeri Venom Serca Into Host Drosophila Hemocytes, Christopher Lark
Theses and Dissertations
The female endoparasitoid wasps introduce venom accompanied by an egg into the larval hosts’ body cavity during oviposition. The venom contains a mixture of proteins that exploit the host’s biological processes, such as, regulating gene expression, disrupting immune signaling pathways, or inducing immune cell death to give a decided advantage to the incipient wasp to develop unchallenged. Drosophila possesses a highly conserved innate immune response that recognizes and neutralizes foreign microbes and macroparasites. The venom of Ganaspis hookeri (strain G1) endoparasitoid wasp has co-evolved to suppresses the humoral phase Drosophila’s innate immune response by inhibiting Ca2+ bursts needed to induce …

Group Differences In Mother-Infant Macaca Fascicularis Behavior, Parasite Load, And Body Condition Within An Anthropogenically Altered Forest, Elizabeth M.C. Coggeshall
All Master's Theses
This study aimed to establish preliminary health and behavioral data, as well as understand group variation for a large population of Macaca fascicularis individuals within an anthropogenically altered monkey forest. A parasitic analysis of 40 mother and infant individuals showed that M. fascicularis carried 13 different parasitic taxa, and that there was parasitic variation between groups. Body condition scores were determined using a newly created and adapted body condition scale from 146 sampled mother macaques. Body condition scores were significantly different between groups, specifically the pond group when compared to the three other groups. Mother-infant behavioral differences were seen between …

Accessing The Functional Capabilities Of The Tick Microbiome Through Metagenomics, Darin Lee Shrewsberry Jr
Graduate Theses, Dissertations, and Problem Reports (ETD)
The black-legged tick Ixodes scapularis is a common ectoparasite of animals and an obligate blood feeder, attaching to a host animal and taking a blood meal once per life stage (i.e. 3 times during its life). Unfortunately for the tick, blood represents a nutrient poor diet largely consisting of lipids and proteins but notably lacking in essential nutrients such a B vitamins (e.g., biotin, riboflavin, niacin) Other exclusive blood feeders such as the tsetse fly harbor symbiotic bacteria that provision B-complex vitamins to their host. I. scapularis harbors Rickettsia buchneri an endoparasitic bacterium that intriguingly possesses two copies of a …

Garcinia species are widely used for their slimming effects via increased fat burning and suppression of satiety. However, scientific evidence for the biological effects of Garcinia atroviridis (GA) is lacking. We investigated the phytochemical composition, safety profiles, and antioxidant and antiobesity effects of methanolic extracts of Garcinia atroviridis (MeGa) in obese female rats. Repeated dose toxicity studies were conducted according to the OECD guidelines. Upon sacrifice, haematological, biochemical, lipid profile, and serum-based metabolomics analyses were performed to evaluate metabolic expression changes and their related pathways. MeGa contains several phytochemical groups and GA fruit acids. MeGa was found to be nontoxic …

Hephaestin (Hp) is a trans-membrane protein, which plays a critical role in intestinal iron absorption. Hp was originally identified as the gene responsible for the phenotype of sex-linked anaemia in the sla mouse. The mutation in the sla protein causes accumulation of dietary iron in duodenal cells, causing severe microcytic hypochromic anaemia. Although mucosal uptake of dietary iron is normal, export from the duodenum is inhibited. Hp is homologous to ceruloplasmin (Cp), a member of the family of multi copper ferroxidases (MCFs) and possesses ferroxidase activity that facilitates iron release from the duodenum and load onto the serum iron transport …
Sinorhizobium Meliloti Changes Motility Phenotypes In The Presence Of Serum, Eris K. Bisal
Sinorhizobium Meliloti Changes Motility Phenotypes In The Presence Of Serum, Eris K. Bisal
Theses and Dissertations
Sinorhizobium meliloti is a nitrogen-fixing flagellated soil bacterium that engages in a mutual symbiotic relationship with the legume Medicago sativa. Using its flagella, S. meliloti is chemotactic in response to varying environmental cues thereby directed to a target, which is typically the root hairs of M. sativa. Lu et al, 2012 suggested that a signal from the plant can switch S. meliloti from a free-living flagellated microbe to a host-invading non-flagellated microbe, although this signal has not been identified. This switch is thought to occur inside curled root hairs of the plant, and after becoming non-flagellated, S. …

White fringetree is a host for the invasive emerald ash borer (EAB) despite being lower quality than black ash. Observations suggest that host trees grown in full sun are more resistant to EAB than those grown in shade, however chemical defense mechanisms and the impact of environmental stress have not been assessed. We quantified constitutive and induced defenses and other characteristics white fringetree and black ash phloem tissue grown under differential light conditions, and these traits were related to EAB larval performance. White fringetree had significantly lower constitutive and induced activities of defense associated enzymes and lignin but higher phenolic, …
Termination-Independent Role Of Rat1 In Cotranscriptional Splicing In Budding Yeast, Zuzer Hakimuddin Dhoondia
Termination-Independent Role Of Rat1 In Cotranscriptional Splicing In Budding Yeast, Zuzer Hakimuddin Dhoondia
Wayne State University Dissertations
Rat1 is a 5′→3′ exoribonuclease in budding yeast belonging to the XRN-family of nucleases. It is a highly conserved protein with homologs being present in fission yeast, flies, worms, mice and humans. Rat1 and its homolog in metazoan have been shown to function in multiple facets of RNA metabolism. In this study, we report a novel role of Rat1 in splicing of pre-mRNA in budding yeast. In the absence of the functional Rat1 in the nucleus, an increase in the level of unspliced transcripts was observed in yeast cells. Strand-specific TRO analysis revealed that the accumulation of unspliced transcripts upon …
Gut Microbiota Composition Is Correlated To Host Hummingbird Protein Requirements, Ryan C. Stanton , '20
Gut Microbiota Composition Is Correlated To Host Hummingbird Protein Requirements, Ryan C. Stanton , '20
Senior Theses, Projects, and Awards
The gut microbiome shapes and is shaped by a host animal’s physiology. Avian taxa hold physiological characteristics unique from mammals and might inform novel pressures experienced by microbial communities. Further, the symbionts’ relative abundance and their abilities to adapt to available resources are of critical importance to a holobiont’s fitness in rapidly changing climates. Therefore, wild populations of hummingbirds Selasphorus rufus and Calypte anna were studied. The two systems differ in S. rufus’s annual migrations from wintering grounds to their breeding grounds in the Pacific Northwest, whereas C. anna are resident in the latter region. Previous findings have indicated …

Changes In The Home Range Sizes Of Terrestrial Vertebrates In Response To Urban Disturbance: A Meta-Analysis, Kate O'Donnell, Javier Delbarco-Trillo
Faculty Publications – Biological Sciences
The unprecedented growth rate in human population and the increasing movement of people to urban areas is causing a rapid increase in urbanisation globally. Urban environments may restrict or affect the behaviour of many animal species. Importantly, urban populations may change their spatial movement, particularly decreasing their home ranges in response to habitat fragmentation, the presence of landscape barriers and the availability and density of resources. Several species specific studies suggest that urban animals decrease their home ranges compared with their non-urban counterparts; however, it remained unclear whether this pattern is widespread across taxa or is instead restricted to specific …

The evolution of networks is constrained by spatial properties of the environment; a characterization that is true in both biological and built networks. Hence built networks such as urban streets can be compared to biological networks to reveal differences in efficiency and complexity. This study assessed foraging networks created by the slime-mold Physarium polycephalum on proportional 3D-printed topographic maps of metropolitan city of Los Angeles, California. Rapidly-generated isomorphic solutions were found to be consistently and statistically shorter than existing roadways in system length. Slime mold also allocated resources to supporting key nodes, analogous to how heavy traffic flows through major …
Non-Invasive Method For Leptin Supplementation In Zebrafish (Danio Rerio), Regan Mcnamara
Non-Invasive Method For Leptin Supplementation In Zebrafish (Danio Rerio), Regan Mcnamara
Williams Honors College, Honors Research Projects
I tested the hypothesis that recombinant leptin protein can be introduced to zebrafish in vivo through non-invasive soaking in a solution containing the protein. One way to study various molecules’ effects in vivo is through intraperitoneal or intracerebroventricular injections during the embryonic or larval stage, which is invasive, difficult to administer, and can have a high mortality rate. 48 hours post fertilization (hpf) zebrafish were soaked in a His-tagged recombinant leptin protein solution at 10 nM and 100 nM concentrations (produced by Genscript). After soaking, zebrafish larvae were washed extensively to remove all recombinant protein on their exterior before homogenization. …

Ornamental white fringetrees in IL, IN, OH, and PA were reassessed in 2018 as a follow-up study to determine the fate of the white fringetree through the invasion wave of emerald ash borer. Attack rates decline by half from 2015-2018 and only 13% of trees were infested. Health of not reinfested trees in 2018 improved suggesting resilience against EAB attack whereas currently infested trees in 2018 displayed signs of declining health. Trees differed in their resistance to EAB attack and 41 additional ornamental and wild white fringetrees were studied in OH to determine if the anti-herbivory defense chemical, oleuropein, influenced …
To Eat Or Not To Eat: Urbanization And The Foraging Behaviors Of Michigan Passerines, Victoria T. Vo
To Eat Or Not To Eat: Urbanization And The Foraging Behaviors Of Michigan Passerines, Victoria T. Vo
Senior Honors Theses and Projects
Urbanization creates novel problems for wildlife, potentially altering the foraging behavior of passerine birds. Foraging is driven by factors like food preference and food accessibility; the foraging of urban birds may be more innovative due to frequent exposure to unfamiliar items. I hypothesized that high novelty exposure in urban birds would promote faster discovery and longer visits to novel feeders compared to rural conspecifics. Additionally, I hypothesized that birds would prioritize food quality over access regardless of habitat, but when faced with barriers, urban birds would be better at accessing food compared to rural birds because of frequent exposure to …

Small RNAs (sRNAs) associate with Argonaute (AGO) proteins in effector complexes, termed RNA-induced silencing complexes (RISCs), which regulate complementary transcripts by translation inhibition and/or RNA degradation. In the unicellular alga Chlamydomonas, several metazoans, and land plants, emerging evidence indicates that polyribosome-associated transcripts can be translationally repressed by RISCs without substantial messenger RNA (mRNA) destabilization. However, the mechanism of translation inhibition in a polyribosomal context is not understood. Here we show that Chlamydomonas VIG1, an ortholog of the Drosophila melanogaster Vasa intronic gene (VIG), is required for this process. VIG1 localizes predominantly in the cytosol and comigrates with monoribosomes and …
Abiotic Conditions, Algal Biomass & Fish Growth Rates Affect Fish Mercury Concentrations In Two Subarctic Lakes, Ari Yamaguchi
Abiotic Conditions, Algal Biomass & Fish Growth Rates Affect Fish Mercury Concentrations In Two Subarctic Lakes, Ari Yamaguchi
Theses and Dissertations (Comprehensive)
Kakisa Lake and Tathlina Lake, located in the Dehcho Region of the Northwest Territories, support important fisheries for the local Ka’a’gee Tu First Nation (KTFN). Recently, Walleye (Sander vitreus) of typical catch size in Tathlina Lake were found to have mercury concentrations above Health Canada’s commercial sale guideline of 0.5 ppm. Wild foods with elevated mercury concentrations can pose health risks to the humans who consume them, depending on consumption amounts and vulnerability factors such as age and pregnancy. Because wild fish can accumulate relatively high mercury levels and subsistence fishing contributes greatly to food security in northern …
Effects Of Abiotic Factors On Host Plant Quality And Community Structure Of The Parasitoid Guild That Attacks Asphondylia Borrichiae Midges On Borrichia Frutescens, Kevin Orta
UNF Graduate Theses and Dissertations
Asphondylia borrichiae is a gall-inducing insect that attacks its ancestral host plant Borrichia frutescens: through ovipositional mistakes it has acquired two additional hosts, Iva frutescens and Iva imbricata. Oviposition results in the formation of a gall, a tumor-like growth of plant tissue within which immature A. borrichiae develop. During development, immature A. borrichiae, are attacked by a suite of four generalist hymenopteran parasitoids. Of these, Galeopsomyia haemon and Torymus umbilicatus are facultative hyperparasitoids, more common and exhibit non-random attack patterns. In the present study, soil quality was manipulated through bi-weekly application of either ammonium nitrate fertilizer or sodium …

Diet Of Spalangia Cameroni (Hymenoptera: Pteromalidae), A Parasitoid Of Filth Flies, Elizabeth Taylor
Graduate Research Theses & Dissertations
Parasitoid wasps are often part of the rich invertebrate communities in decaying organic matter where their hosts are found. However, little is known about what adults of these parasitoids eat. Most parasitoid wasps parasitize herbaceous insects, so nectar from flowers often is readily available. The present study examined feeding by adult Spalangia cameroni Perkins (Hymenoptera: Pteromalidae), a species that burrows through manure, where it finds and parasitizes pupae of filth flies. Feeding on host fluids occurs in many parasitoid wasps, including S. cameroni. In the laboratory, these wasps also readily feed on honey or a sucrose solution, which increases their …

Reprograming Neuronal Cells By Overexpression Of Fibroblast-Specific Transcription Factors, Abdulmohsen Alanazi
Masters Theses
In mammals, a complex system of regulatory signals distinguishes tissues, structures and functions. Combinations of transcription factors and co-factors regulate activation and repression of genes that result in cellular differentiation. Whole genome arrays allow the monitoring of genomic expression in specific tissues. Fibroblast microarray studies have shown candidate genes that may be involved in fibroblast identification, including genes that express transcription factors Prrx1, Snai2 and Twist1. A previous study showed that the Prrx1 and Snai2 could reactivate a fibroblast phenotype in hybrid cells that had lost fibroblast identity. Furthermore, overexpression of these factors in liver-derived cells strongly repressed liver gene …

Species distribution models (SDMs) correlate species occurrences with environmental predictors, and can be used to forecast distributions under future climates. SDMs have been criticized for not explicitly including the physiological processes underlying the species response to the environment. Recently, new methods have been suggested to combine SDMs with physiological estimates of performance (physiology-SDMs). In this study, we compare SDM and physiology-SDM predictions for select marine species in the Mediterranean Sea, a region subjected to exceptionally rapid climate change. We focused on six species and created physiology-SDMs that incorporate physiological thermal performance curves from experimental data with species occurrence records. We …

The cattle fever tick, Rhipicephalus annulatus (Say), is an economically destructive arthropod because of its ability to vector bovine babesiosis. Cattle fever ticks can spend more than 90% of their life cycle as questing larvae, but the effect of climatic factors on their off-host behavior and survival is unclear. The goal of this study was to measure the effects of specific ecological factors on off-host larvae in nature. The study was conducted in a south Texas pasture over a 20-mo period, during which time larval populations were surveyed and ambient weather variables - relative humidity and temperatures - were recorded. …

The measurement of bulk tissue nitrogen (δ15N) and carbon isotope values (δ13C) chronologically along biologically inert tissues sampled from offspring can provide a longitudinal record of their mothers' foraging habits. This study tested the important assumption that mother-offspring stable isotope values are positively and linearly correlated. In addition, any change in the mother-offspring bulk tissues and individual amino acids that occurred during gestation was investigated. Whiskers sampled from southern elephant seal pups (Mirounga leonina) and temporally overlapping whiskers from their mothers were analyzed. This included n = 1895 chronologically subsampled whisker segments for bulk …
