Should Advanced Maternal Age Be A Deterrent For Attempting A Pregnancy?, Sarina Spira

The Science Journal of the Lander College of Arts and Sciences

In the developed world, the trend toward women of advanced age bearing children is very prevalent and seems likely to continue. For a variety of reasons, women are delaying pregnancy until 35 years and older when they suddenly seem keenly aware that their biological clocks are ticking. A review of various studies obtained from the Proquest and EBSCO databases indicates that there are definitely certain risks associated with advanced maternal age (AMA). As females age, the reduced number and quality of their remaining eggs contribute to reduced fertility. Congenital anomalies involving the number of chromosomes in the embryo increases as …

Is Rapamycin An Effective Anti-Aging Drug?, Basya Aboud

The Science Journal of the Lander College of Arts and Sciences

Scientific and pharmaceutical advancements have revolutionized medicine. Many once- debilitating or fatal diseases can now be managed with medication. However, the search for a cure for the inevitable aging diseased state remained futile until recently. Rapamycin has been studied for its possible longevity effects, providing promising results for the development of anti-aging therapies. This paper evaluates the benefits and risks of rapamycin use. While rapamycin cannot be supported as a safe anti-aging drug, rapamycin studies have elucidated parts of the aging pathway, providing a breakthrough for anti-aging research.

Determinants Of Holliday Junction Formation And Resolution During Budding Yeast Meiosis, Marina Bykova Jan 2020

Determinants Of Holliday Junction Formation And Resolution During Budding Yeast Meiosis, Marina Bykova

ETD Archive

In the process of meiosis, 4 haploid cells are produced from 1 diploid cell in two rounds of division. During prophase I of meiosis I, double strand breaks (DSBs) are created by the Spo11 protein, some of which are later repaired by homologous recombination into crossovers. During homologous recombination, structures called Holliday Junctions are formed, which later are resolved into crossovers to allow homologous chromosomes to separate. Ifthey are not resolved, the cell arrests and cannot progress further in meiosis. I have investigated three proteins known or suspected to be involved in Holliday Junction formation and resolution: the transcription factor …

Hiding In Plain Sight: The Globally Distributed Bacterial Candidate Phylum Pauc34f, Michael L. Chen, Eric D. Becraft, Maria Pachladaki, Julia M. Brown, Jessica K. Jarett, Josep M. Gasol, Nikolai V. Ravin, Duane P. Moser, Takuro Nunoura, Gerhard J. Herndl, Tanja Woyke, Ramunas Stepanauskas

Biology

Bacterial candidate phylum PAUC34f was originally discovered in marine sponges and is widely considered to be composed of sponge symbionts. Here, we report 21 single amplified genomes (SAGs) of PAUC34f from a variety of environments, including the dark ocean, lake sediments, and a terrestrial aquifer. The diverse origins of the SAGs and the results of metagenome fragment recruitment suggest that some PAUC34f lineages represent relatively abundant, free-living cells in environments other than sponge microbiomes, including the deep ocean. Both phylogenetic and biogeographic patterns, as well as genome content analyses suggest that PAUC34f associations with hosts evolved independently multiple times, while …

Toward A Symbiotic Perspective On Public Health: Recognizing The Ambivalence Of Microbes In The Anthropocene, S. Sariola, Scott F. Gilbert Jan 2020

Toward A Symbiotic Perspective On Public Health: Recognizing The Ambivalence Of Microbes In The Anthropocene, S. Sariola, Scott F. Gilbert

Biology Faculty Works

Microbes evolve in complex environments that are often fashioned, in part, by human desires. In a global perspective, public health has played major roles in structuring how microbes are perceived, cultivated, and destroyed. The germ theory of disease cast microbes as enemies of the body and the body politic. Antibiotics have altered microbial development by providing stringent natural selection on bacterial species, and this has led to the formation of antibiotic-resistant bacterial strains. Public health perspectives such as “Precision Public Health” and “One Health” have recently been proposed to further manage microbial populations. However, neither of these take into account …

The Effects Of Sleep Deprivation On Mental Health And Neurological Disorders, Gabriella Victoria Fernandez De Salvo Jan 2020

The Effects Of Sleep Deprivation On Mental Health And Neurological Disorders, Gabriella Victoria Fernandez De Salvo

Honors Undergraduate Theses

Sleep deprivation is defined as an individual sleeping less than 6 hours per night. It is a common behavior amongst individuals who live in industrialized nations.Along with industrialization, its prevalence is also expected to rise with age. Despite this, there has been a shift in recent years with sleep deprivation increasing in the youth, likely related to the increased use of technology during the nighttime. Sleep deprivation and chronic sleep disruption are behaviors commonly observed amongst patients suffering from neurological and psychiatric disorders, like Alzheimer’s disease, Parkinson’s disease, depression, and anxiety. Many of these patients suffer from sleep disorders like …

Developing And Testing A Cognitive Field Test For Lead-Exposed Wild Songbirds, Samantha Glowacki Jan 2020

Developing And Testing A Cognitive Field Test For Lead-Exposed Wild Songbirds, Samantha Glowacki

Senior Honors Theses and Projects

Anthropogenic impacts drive global changes that can negatively impact wildlife. For example, using lead pipes in Flint, MI has resulted in contaminated drinking water that is likely to have moved into the environment through irrigation. Juvenile American robins (Turdus migratorius) captured in watered parks of Flint have blood-lead levels considered dangerous to brain functioning. The purpose of this study was to develop and test a feeder puzzle for assessing the cognitive effects of lead in wild urban songbirds. We predicted that songbirds could learn how to feed from a feeder puzzle, and if they visited the feeder in groups, then …

Larval Performance Of A Major Forest Pest On Novel Hosts And The Effect Of Stressors, Donnie Lee Peterson, Don Cipollini Jan 2020

Larval Performance Of A Major Forest Pest On Novel Hosts And The Effect Of Stressors, Donnie Lee Peterson, Don Cipollini

Biological Sciences Faculty Publications

Novel hosts lacking a coevolutionary history with herbivores can often support improved larval performance over historic hosts; e.g., emerald ash borer [Agrilus planipennis (Fairmaire) Coleoptera: Buprestidae] on North American ash (Fraxinus spp.) trees. Whether trees are novel or ancestral, stress on plants increases emerald ash borer preference and performance. White fringetree [Chionanthus virginicus (L.) Lamiales: Oleaceae] and olive [Olea europaea (L.) Lamiales: Oleaceae] are closely related non-ash hosts that support development of emerald ash borer to adulthood, but their relative suitability as hosts and the impact of plant stress on larval success has not been well …

Developmental Effects Of Terpenes On Vanessa Cardui At Varying Temperatures, Mari Irving Jan 2020

Developmental Effects Of Terpenes On Vanessa Cardui At Varying Temperatures, Mari Irving

Honors Undergraduate Theses

Secondary metabolites are organic compounds produced by plants that are not essential in growth, development, or reproduction. While they are not strictly necessary to complete the plant life cycle, secondary metabolites play key roles in functions like defense against herbivory and the moderation of symbiotic relationships. Terpenes are a major class of secondary metabolites present in all plants, and this class is hypothesized to have diversified in response to specific plant-herbivore interactions. These complex interactions are further complicated by changes in abiotic conditions experienced seasonally. Herbivory is a major biotic interaction that plays out across broad temporal and spatial scales …

Massard Prairie Restoration And Soil Microbiome Succession, Jeffrey M. Shaver, Emily S. Bellis, Chizuko Iwaki, Jake Qualls, Jay Randolph, Jeremiah Smith

Journal of the Arkansas Academy of Science

We have initially sequenced soil microbial DNA from 4 restored and 4 virgin tallgrass prairie soil samples from Ben Geren Park and Massard Prairie (Fort Smith, AR), respectively. As expected, the soil microbiomes are distinct, with several lineages of nitrogen-fixing bacteria more common in virgin tall grass prairie. However, we predict that as restoration of tallgrass prairie in Ben Geren Park progresses, the soil microbiome of restored prairie will more closely mirror those of the virgin prairie.

A Natural Antimicrobial From Bacillus Subtilis As A Biosanitizer For Resilient Membrane Biofilms, Pratishtha Verma Jan 2020

A Natural Antimicrobial From Bacillus Subtilis As A Biosanitizer For Resilient Membrane Biofilms, Pratishtha Verma

Electronic Theses and Dissertations

The microbial attachment and colonization on separation membranes lead to biofilm formation. Some selective isolates within the biofilm constitutive microflora acquire resistance and emerge predominant over prolonged use of the membrane. Thus, proving the cleaning and sanitization protocols to be ineffective in adequately removing resilient biofilms. This subsequently leads to selecting microbial resistance within the constitutive microflora to almost all antimicrobial treatments and hence, creates a need to develop novel alternative strategies to control biofilm formation on membrane surfaces. The first research project under this study was designed to understand the microbial interactions and emergence of predominance within biofilm constitutive …

Interaction Of Streptomyces Closely Associated With Bacillus In Nitrogen-Limiting Conditions, Aminya Baniya Jan 2020

Interaction Of Streptomyces Closely Associated With Bacillus In Nitrogen-Limiting Conditions, Aminya Baniya

Electronic Theses and Dissertations

Biological Nitrogen Fixation is the process of conversion of atmospheric dinitrogen into ammonia which is performed by symbiotic as well as free-living diazotrophic bacteria. A previous project to isolate free-living nitrogen fixing bacteria from a natural prairie yielded Streptomyces which grew on nitrogen free medium. Genome sequencing of three isolates revealed the presence of a second 16S rRNA gene sequence aligning with Bacillus, however repeated streaking only yielded Streptomyces colonies. The aim of this research was to characterize the interaction of three Streptomyces isolates that grow under nitrogen-free conditions, and appeared to be associated with Bacillus. The separation of Bacillus …

Investigating Links Between Soil Microbial Structure And Function In Three Major Plant Communities Across Temporal Scales Of Arctic Alaska, Kaj Lynoe

Electronic Theses and Dissertations

Arctic microbial systems continue to get attention today as our understanding regarding their structure and function in a changing system is paramount to C feedbacks with warming and changes in precipitation. Plant communities and microbial community processes across the Arctic landscape are central to understanding tundra ecosystem processes because environmental conditions and plant community structure drive microbial cycling of soil organic matter. Here, we want to understand how soil microbial respiration, mineralization, biomass, and community composition are linked to three Alaskan tundra plant communities, namely Shrub, Tussock, and Sedge tundra and the seasonal variability in this system. A total of …

Utilizing Rhizospheric And Bacterial Endophytes For Use As Potential Bio-Fertilizers For Sustainable Agricultural Production, Vincent Peta Jan 2020

Utilizing Rhizospheric And Bacterial Endophytes For Use As Potential Bio-Fertilizers For Sustainable Agricultural Production, Vincent Peta

Electronic Theses and Dissertations

Brassica carinata crop production offers an exciting alternative approach to the energy crisis when looking at novel ways to power our motor vehicles. Bacterial endophytes, while residing in host-plant tissues, offer a wide array of plant growth promotion benefits such as nitrogen fixation to impart nitrogen (N) nutrition, phosphate (P) solubilization to make soil-bound phosphate more mobile in soil for plant uptake, can suppress pathogenic microbes such as fungus and produce plant growth hormones to offset stress incurred by the plant. Previous literature has shown that there is large interest in studying endophytes from economically important crops such as wheat, …

Turkish Journal of Biology

A Kunitz-type trypsin inhibitor protein has been purified and characterized from seeds of Acacia nilotica L. LC-MS/MS analysis of Acacia nilotica trypsin inhibitor (AnTI) provided the N-terminal fragment of 11 amino acids which yielded 100% identity with already reported Kunitz-type trypsin inhibitor protein of Acacia confusa (AcTI) in UniProtKB database search. SDS-PAGE showed a single band of ~21 kDa under nonreduced condition and appearance of a daughter band (17 kDa) in the presence of ß-mercaptoethanol indicating the presence of interchain disulfide linkage typical for Kunitz-type trypsin inhibitors. AnTI was purified from seed extract by using a combination of anion exchange …

Environmental And Trophic Factors That Influence The Prevalence Of Gonionemus Vertens In The Northeastern United States, Matthew Rigby Jan 2020

Environmental And Trophic Factors That Influence The Prevalence Of Gonionemus Vertens In The Northeastern United States, Matthew Rigby

Theses, Dissertations and Culminating Projects

Gonionemus vertens, often referred to as the clinging jellyfish, is an invasive hydrozoan first documented in 2016 in New Jersey. Populations of the adult medusa stage bloom in May and continue to persist through the middle of July, during which time it is of particular concern from a public health standpoint. G. vertens 'clings' to sea grasses and other submerged aquatic vegetation in shallow coastal waters, which are highly utilized for recreational activities, and its sting is highly toxic, which can hospitalize victims for days with both physical and psychiatric symptoms. Encounters with highly toxic G. vertens are, unfortunately, becoming …

Construction Of A Yeast-Based Rescue System For Screening Of Effects Of Pharmacological Agents On Twik Channels, Mosi Lin Jan 2020

Construction Of A Yeast-Based Rescue System For Screening Of Effects Of Pharmacological Agents On Twik Channels, Mosi Lin

Legacy Theses & Dissertations (2009 - 2024)

Two-pore domain potassium (K2P) channels conduct leak or background K+ currents, which primarily maintain resting membrane potential and regulate cellular excitability. Among the TWIK subfamily of K2P channels, TWIK-1 (K2P1) and TWIK-2 (K2P6) channels are characterized as weak inwardly rectifying K+ channels, whereas TWIK-3 (K2P7) channels do not produce detectable currents in heterologous expression systems. Although K2P channels were first identified in the 1990s, only a few K2P-specific pharmacological agents are available. In this study, I developed a yeast-based rescue system to screen the effects of pharmacological agents on TWIK channels. First, I employed K+-uptake-deficient SGY1528 yeasts that K+ transporters …

Daily Energy Expenditure Of Florida Scrub-Jays (Aphelocoma Coerulescens) In Suburban And Wildland Landscapes, Hugh I. Ellis, Reed Bowman Jan 2020

Daily Energy Expenditure Of Florida Scrub-Jays (Aphelocoma Coerulescens) In Suburban And Wildland Landscapes, Hugh I. Ellis, Reed Bowman

Biology: Faculty Scholarship

Urbanization alters ecological systems, disturbance regimes, food webs, and a variety of other processes that can influence local flora and fauna. In birds, most studies have focused on behavioral or demographic responses to altered conditions; however, the physiological mechanisms associated with these responses have been understudied. We have previously documented shifts in foraging behavior because of the availability of human-provided food; we sought to see if this was driven by or associated with a change in field metabolic rates. We measured field metabolic rates (FMR), a measure of daily energy expenditure (DEE), of Florida Scrub-Jays (Aphelocoma coerulescens) in 2 habitats …

Boron is a naturally occurring trace element found in organic and inorganic complexes. Boron-containing compounds are required for living organisms for diverse metabolic functions, including nitrogen fixation in microorganisms, cell wall stability in plants, and bone and carbohydrate metabolism in animals. The number of studies about the effect of boron in biological model systems is very limited; so far, there has been no study on the correlation between boron and amyloid-beta toxicity. Here, we investigated the possible effects of 2 boron-containing compounds-sodium borate decahydrate and boric acid-against amyloid-beta toxicity. In our in vitro amyloid-beta toxicity model, we showed that these …

“The contamination of soil and groundwater from leachate leakage from landfill has turned, nowadays, into a global public issue. One of the main concerns about this pollutant is the potential threat to human and ecosystem health. The current research studied endophyte bacteria in association with poplar trees (Populus sp.), as a method of bioremediation of landfill leachates. The objective of the project was to identify treatment strategies that may improve plant performance (survival rates, plant fitness, and degradation efficacy) with the purpose of being implemented in phytoremediation plots, aimed to intercept and treat landfill leachate, before the contamination of watershed. …

Intermittent Fasting (If) Promotes Longevity Through Alterations Of The Mammalian Target Of Rapamycin (Mtor) And The Epigenome, Tayt Boeckholt Jan 2020

Intermittent Fasting (If) Promotes Longevity Through Alterations Of The Mammalian Target Of Rapamycin (Mtor) And The Epigenome, Tayt Boeckholt

Biology and Microbiology Graduate Students Plan B Research Projects

Many studies with a range of subjects from Nematodes to Homo sapiens have found intermittent fasting (IF) to significantly improve the cardiometabolic health of individuals, but how IF promotes longevity through epigenetic modulations remains a sparse understanding throughout the literature. The process of aging may be characterized by a loss of cellular identity sprouted from a disrupted epigenome rich with information for a cell, while also losing the ability to recycle ineffective cellular components. The nutrient-sensing kinase Mammalian Target of Rapamycin (mTOR) is disrupted during bouts of fasting which allows for the recycling of cellular components through increased autophagy. Furthermore, …

Invasive Species Shift Fungal Driven Decomposition In Midwestern Forests, Adam M. Reed

Browse all Theses and Dissertations

Midwestern forests are currently impacted by two prominent invaders, Agrilus planipennis and Lonicera maackii. The Ag. planipennis induced loss of Fraxinus spp. trees can facilitate Lo. maackii invasion, which is likely altering microbial driven forest nutrient cycling. To assess these changes in microbial processes, I conducted litter bag and culture-based decomposition experiments using leaf litter from Acer spp., Quercus spp., F. nigra, F. pennsylvanica, Lindera benzoin, and Lo. maackii. For the culture-based decomposition experiment, I inoculated six species of fungi separately onto both single species and multispecies (half Lo. maackii and half native spp.) leaf litter and measured decomposition rate, …
