Symbionts, or organisms that form close and persistent associations with hosts, are ubiquitous across the tree of life, and symbioses are among the most common interspecific interactions in the natural world. Symbionts play a fundamental ecological and evolutionary role across multiple levels of biological organization – from individuals to ecosystems – and can have strong effects on global biodiversity. However, many aspects of these relationships are poorly understood including symbiont genetic diversity (inter- and intraspecific), host specificity, and the mechanisms underlying their functional nature or any context-dependent patterns. The research presented in this dissertation focuses on microbial roles in biogeochemical cycling in Antarctic aquatic environments. The major objective of my research was to examine the impact of biotic and abiotic pressures on nutrient cycling in microbially dominated systems. I used three perennially ice covered lakes in the McMurdo Dry Valleys (MDVs) along with subglacial lakes Whillans and Mercer as natural case studies. The MDVs are located in Victoria Land, East Antarctica and have been studied since 1993 as part of the McMurdo Long Term Ecological Research Project. BackgroundNanocarrier-based systems have cultivated significant improvements in prostate cancer therapy. However, the efforts are still limited in clinical applicability, and more research is required for the development of effective strategies. Here, we describe a novel nanoliposomal system for targeted apoptotic gene delivery to prostate cancer.MethodsPoly (2-ethyl-2-oxazoline) (PEtOx) dioleoyl phosphatidylethanolamine (DOPE) nanoliposomes were conjugated with the prostate-specific membrane antigen (PSMA)-targeting peptide GRFLTGGTGRLLRIS (P563) and loaded with BikDDA, a mutant form of the proapoptotik Bik. Brace root architecture is a critical determinant of maize's stalk anchorage and nutrition uptake, influencing root lodging resistance, stress tolerance, and plant growth. To identify the key microRNAs (miRNAs) in control of maize brace root growth, we performed small RNA sequencing using brace root samples at emergence and growth stages. Metals and metalloids (hereafter, metal(loid)s) in plant-based foods are a source of exposure to humans, but not all metal(loid)-food interactions are the same. Differences exist between metal(loid)s in terms of their behavior in soils and in how they are taken up by plants and stored in the edible plant tissue/food. Thus, there cannot be one consistent solution to reducing toxic metal(loid)s exposure to humans from foods. Prochlorococcus is a globally abundant cyanobacterium that can help reveal the ecology of microorganisms in the ocean. Specifically, mortality by zooplankton such as appendicularians is globally important and Prochlorococcus can help reveal some of the choices made around feeding for this ubiquitous filter feeder. Two ecotypes of Prochlorococcus were introduced to appendicularians to determine if appendicularian feed selectively between these ecotypes. Further, a single ecotype of Prochlorococcus was fed to appendicularians over a variety of life stages to determine if retention rate changed with appendicularian life stage. Several key ecological advantages and life history traits of this species jointly impact growth rates and survivorship of native freshwater turtles where sympatry occurs. In this paper, we introduce the numerical strategy for mixed uncertainty propagation based on probability and Dempster–Shafer theories, and apply it to the computational model of peristalsis in a heart-pumping system. Specifically, the stochastic uncertainty in the system is represented with random variables while epistemic uncertainty is represented using non-probabilistic uncertain variables with belief functions. The mixed uncertainty is propagated through the system, resulting in the uncertainty in the chosen quantities of interest (QoI, such as flow volume, cost of transport and work). Traumatic brain injuries (TBIs) are a common form of head injury, with mild TBIs (mTBIs) making up 75-90% of the TBI severity scale. It’s been suggested that repetitive mild TBIs (rmTBIs) may enhance effects of Alzheimer’s Disease (AD). With dementia linking TBI and AD together, we investigated the memory hub of the brain - the hippocampus. Astrocytes and microglia are two glial cells that respond to neuroinflammation and therefore imaged in hippocampal regions of the 3xTg-AD mouse brain. Royal catchfly (Silene regia) is a perennial prairie forb known to grow in rocky, alkaline soils within tallgrass prairies of the eastern United States. S. regia is recorded from twelve states, including Georgia, and is now confined to fragmented habitats due to the loss of 95% of its natural environment. The species is now listed as critically imperiled in the state of Georgia. Synanthropic bats live in close proximity to humans and domestic animals, creating opportunities for potential pathogen spillover. We explored environmental correlates of occurrence for a widely distributed synanthropic African bat, Mops pumilus—a species associated with potential zoonotic viruses—and estimated current and future environmental suitability in the Taita Hills region and surrounding plains in Taita–Taveta County in southeast Kenya. Two-photon voltage imaging has long been heralded as a transformative approach capable of answering many long-standing questions in modern neuroscience. However, exploiting its full potential requires the development of novel imaging approaches well suited to the photophysical properties of genetically encoded voltage indicators. We demonstrate that parallel excitation approaches developed for scanless two-photon photostimulation enable high-SNR two-photon voltage imaging. We use whole-cell patch-clamp electrophysiology to perform a thorough characterization of scanless two-photon voltage imaging using three parallel illumination approaches and lasers with different repetition rates and wavelengths. Traumatic Brain Injury (TBI) is a major cause of morbidity and mortality that afflicts millions of people in the United States annually. TBI mice had significantly increased expression of the pro-inflammatory cytokines and decreased expression of tight junction proteins CLDN5, CLDN12, and OCLD in brain lesion tissue. TBI mice exhibited a significant accumulation of FITC in the middle and distal part of the small intestine compared to sham mice. TBI mice exhibited an increase in pathogenic bacteria Enterobacteriaceae and a decrease in beneficial bacteria lactobacillus. Determining space use for species is fundamental to understanding their ecology, and tracking animals can reveal insights into their spatial ecology on home ranges and territories. Recent technological advances have led to GPS-tracking devices light enough for birds as small as ~30 g, creating novel opportunities to remotely monitor fine-scale movements and space use for these smaller species. Microbial communities comprising bacteria, viruses, fungi, and protists live within and on the surfaces of animal hosts. These microbial communities exist in symbiosis with the host, and heavily influence host physiology, development, health, and fitness. Gut-dwelling microbes (i.e., gut microbiota) contribute to host neurodevelopment through a bidirectional Microbiota-Gut-Brain (MGB) axis. Myxomycetes (plasmodial slime molds) are eukaryotic protist predators that are associated with wood, leaf litter, and soil in forests, where they feed on bacteria, protozoans, and (to a more limited extent) fungi. The health of crop plants is essential because they represent a primary food source for humans. Human serum albumin (HSA) nanoparticles have been widely used as versatile drug delivery systems for improving the efficiency and pharmaceutical properties of drugs. The present study aimed to design HSA nanoparticle encapsulated with the hydrophobic anticancer pyridine derivative (2-((2-([1,1'-biphenyl]-4-yl)imidazo[1,2-a]pyrimidin-3-yl)methylene)hydrazine-1-carbothioamide (BIPHC)). The synthesis of HSA-BIPHC nanoparticles was achieved using a desolvation process. Atomic force microscopy (AFM) analysis showed the average size of HSA-BIPHC nanoparticles was 80.21 nm. The percentages of entrapment efficacy, loading capacity and production yield were 98.11%, 9.77% and 91.29%, respectively. Many species of zooplankton exhibit the predator avoidance behavior called diel vertical migration (DVM). During typical DVM, zooplankton ascend to surface waters during the night and descend to deeper water during the day where predation by visual predators is reduced due to lower light levels (Goldman and Horne 1983; Wetzel 1983). The Role Of Micrornas In Lung Inflammation After Alcohol And Burn Injury, Connor O'Neal Guzior

Acute lung injury (ALI) and the subsequent development of acute respiratory distress syndrome (ARDS) are among the leading causes of post-burn morbidity and mortality. MiRNAs are small non-coding RNA molecules that are known to regulate gene expression and help regulate lung homeostasis. In this study, we profiled four anti-inflammatory miRNAs (miR-146a, miR-150, miR-194, and miR-671) in lung tissue following an alcohol and burn injury. We found that miR-146a and miR-671 were significantly downregulated in lungs one day after combined injury, leading to increased lung inflammation and pathology. Circular RNA (circRNA) molecules have critical functions during brain development and in brain-related disorders. Here, we identified and validated a circRNA, circHTT(2,3,4,5,6), stemming from the Huntington’s disease (HD) gene locus that is most abundant in the central nervous system (CNS). We uncovered its evolutionary conservation in diverse mammalian species, and a correlation between circHTT(2,3,4,5,6) levels and the length of the CAG-repeat tract in exon-1 of HTT in human and mouse HD model systems. The mouse orthologue, circHtt(2,3,4,5,6), is expressed during embryogenesis, increases during nervous system development, and is aberrantly upregulated in the presence of the expanded CAG tract. …

Emerging infectious diseases threaten wildlife globally. While the effects of infectious diseases on hosts with severe infections and high mortality rates often receive considerable attention, effects on hosts that persist despite infection are less frequently studied. To understand how persisting host populations change in the face of disease, we quantified changes to the capture rates of Eptesicus fuscus (big brown bats), a persisting species susceptible to infection by the invasive fungal pathogen Pseudogymnoascus destructans (Pd; causative agent for white-nose syndrome), across the eastern US using a 30-year dataset. Bird song is a classic example of a sexually selected trait, but much of the work relating individual song components to fitness has not accounted for song typically being composed of multiple, often-correlated components, necessitating a multivariate approach. We explored the role of sexual selection in shaping the complex male song of house wrens (Troglodytes aedon) by simultaneously relating its multiple components to fitness using multivariate selection analysis, which is widely used in insect and anuran studies but not in birds.   • Animal space use and spatial overlap can have important consequences for population-level processes such as social interactions and pathogen transmission. Identifying how environmental variability and inter-individual variation affect spatial patterns and in turn influence interactions in animal populations is a priority for the study of animal behaviour and disease ecology. Environmental food availability and macroparasite infection are common drivers of variation, but there are few experimental studies investigating how they affect spatial patterns of wildlife.

When plants die, neighbours escape competition. Living conspecifics could disproportionately benefit because they are freed from negative intraspecific processes; however, if the negative effects of past conspecific neighbours persist, other species might be advantaged, and diversity might be maintained through legacy effects. We examined legacy effects in a mapped forest by modelling the survival of 37,212 trees of 23 species using four neighbourhood properties: living conspecific, living heterospecific, legacy conspecific (dead conspecifics) and legacy heterospecific densities. This thesis compares the status of bronze artefacts from two archaeological sites in Dubai (UAE): the Sarouq Al-Hadid site (Stone Age to Early Islamic Period) and the Al- Qusais site (Late Bronze Age to Early Iron Age). The main objective of this research is to understand the two sites' environment and their influence on the corrosion of archaeological bronze artifacts found at the sites. This study uses mCed methods to comprehensively understand the corrosion behavior of bronze objects, depending on their burial environment.
