Climate change has had devastating effects on tropical coral reefs; however, much less is known regarding how heat stress affects temperate coral. This research focuses on Astrangia poculata (Ellis and Solander, 1786) collected from Narragansett Bay, RI, during the summer and winter seasons and understanding the effect of experimental thermal extremes (i.e., 26 °C) on seasonally different populations. Photosynthetic efficiency (Fv/Fm), symbiont density (via an inverse relationship with pixel intensity), and oxidative stress via reactive oxygen species (ROS) concentrations were measured on symbiotic and aposymbiotic A. poculata. Climate change has caused drastic declines in corals. As sessile organisms, corals acclimate to environmental shifts through genome-wide changes in gene expression, epigenetic modifications, and alterations in microbiome composition. However, alternative splicing (AS), a conserved mechanism of stress response in many organisms, has been under-explored in corals. Using short-term acute thermal stress assays, we investigated patterns of AS in the scleractinian coral Acropora cervicornis during response to low (33°C), medium (35°C), and high (37°C) heat stress and subsequent overnight recovery. Our findings demonstrate reproducible dynamic shifts in AS of at least 40 percent of all genes during response to heat …

Coral reefs are vital marine ecosystems that support many different species and ecosystem services through protecting coastlines from storms and erosion, providing raw materials for medication, and facilitating diversity amongst marine species. Anthropogenic climate change is causing ocean temperatures to exceed the range of coral thermal tolerance resulting in more coral bleaching events globally. However, some corals have the capacity to withstand high temperatures through different thermal tolerance strategies. Amblyomma maculatum Koch (Acari: Ixodidae), Ixodes scapularis Say, and Dermacentor variabilis Say are three hard-bodied ticks responsible for vectoring pathogens that cause most human tick-borne diseases in the United States of America (USA). Tick surveillance is critical to elucidate high-risk areas for targeted vector control and public health interventions. Despite tick-borne diseases having a higher annual incidence compared to mosquito-borne diseases in the USA, tick traps used for surveillance are grossly under-developed compared to the suite of sophisticated mosquito traps available on the commercial market. Motivation: Here, we make available a second version of the BioTIME database, which compiles records of abundance estimates for species in sample events of ecological assemblages through time. The updated version expands version 1.0 of the database by doubling the number of studies and includes substantial additional curation to the taxonomic accuracy of the records, as well as the metadata. Moreover, we now provide an R package (BioTIMEr) to facilitate use of the database.

Plants form the basis of life on Earth. However, due to the phenomenon of plant awareness disparity, a self-perpetuating cycle whereby plants are overlooked on an individual and global level ensues. Lack of plant awareness leads to a lessened emphasis on plants in sustainability and conservation as well as biology education, resulting in negative impacts on biodiversity and food security. The severity of plant awareness disparity is dependent on cultural context, and Indigenous and/or traditional societies are less likely to experience plant awareness disparity because of they are typically more connected to plants for their everyday needs. Host microbiota may impact disease vector behavior and pathogen transmission, but little is known about associations between ectoparasites and microbial communities in wildlife reservoir species. We used Illumina metagenomic sequencing to explore the impacts of tick parasitism on the rodent fecal microbiome in both a field and laboratory setting. We found that tick parasitism on wild hosts was associated with variation in the fecal microbiota of both the white-footed deermouse, Peromyscus leucopus, and the southern cotton rat, Sigmodon hispidus. Mobulas (manta and devil rays) are large-scale ram filter feeders that separate planktonic food particles from large volumes of water with minimal clogging. This contrasts with most human-made filters that can suffer from problematic clogging requiring additional mechanisms for clearing blocked surfaces and maintaining performance. Prior studies have shown that mobulas employ a unique mechanism referred to as ricochet separation to filter feed, whereby captive vortices in filter pores cause particles to bounce off the filter surfaces and away from the filter pores. Habitat suitability commonly differs between the core and periphery of ecosystems. The longleaf pine ecosystem is recognized for its value in providing habitat to a variety of specialist and endemic species, especially herpetofauna. However, at the northernmost extent of longleaf pine (in southeastern Virginia) little is known about the denizen reptiles and amphibians. Moreover, the remaining longleaf pine ecosystem at the latitudinal ecotone is fragmented amongst a mosaic of other ecosystems. We examined herpetofauna diversity, richness, and capture rates between four focal habitat types—mature longleaf pine, restored longleaf pine, maple-gum swamp, and pocosin-bog—in the Zuni Pine Barrens of southeastern Virginia. …

Caribbean reefs face increasingly frequent and intense bleaching events, adding to the numerous other threats impacting these ecosystems. Addressing these challenges requires global action to reduce climate drivers, along with local efforts like reef restoration. Active restoration using thermotolerant coral colonies offers a potential strategy to alleviate these impacts; however, gaps remain in identifying context-specific temperature thresholds to guide colony selection and standardize thermotolerance assessment methods. This study addressed these gaps in two phases. First, by determining practical thresholds to differentiate species responses to heat stress; and second, by developing a framework to identify and prioritize resilient colonies for restoration. …

Coral mortality is occurring worldwide at an alarming rate. Despite the immense and underestimated biodiversity of reef-building corals, very few genomes are available. Further, almost all genomic resources originate from shallow water corals even though photosynthetic, symbiotic corals occur at mesophotic depths deeper than 30 m and even >100 m. We present annotated, de novo genomes for two mesophotic, scleractinian (reef-building) corals Montipora cf. grisea and Leptoseris cf. scabra from American Sāmoa, the latter being the first genome for the widespread genus Leptoseris. Immunohistochemistry (IHC) is an essential technique in biological research that allows for the visualization and localization of proteins within tissue samples. This systematic review analyzes the most effective, efficient and reproducible protocols for IHC. It focuses on three critical steps: freezing, cryosectioning, and staining of skeletal muscle tissues in mice models. A total of 41, 44, and 48 articles on freezing, cryosectioning, and staining techniques, respectively, were analyzed, from reputable sources in biology like PubMed and Web of Science. Salmon occupy a vital role in the ecosystem, economy, and indigenous culture of the Pacific Northwest of the United States, specifically within the Columbia River Basin. Of the five species of Pacific Salmon that live in the basin, Chinook salmon (Oncorhynchus tshawytscha) are the most widely commercially fished and are the largest. In recent years, Chinook Salmon populations have heavily declined from human influence. Zoonoses are infectious diseases transmitted from animals to humans. Bats have been suggested to harbour more zoonotic viruses than any other mammalian order1. Infections in bats are largely asymptomatic2,3, indicating limited tissue-damaging inflammation and immunopathology. To investigate the genomic basis of disease resistance, the Bat1K project generated reference-quality genomes of ten bat species, including potential viral reservoirs. Here we describe a systematic analysis covering 115 mammalian genomes that revealed that signatures of selection in immune genes are more prevalent in bats than in other mammalian orders. Sterile Insect Technique (SIT) is an effective method for controlling pests and has been increasing in popularity to control Aedes mosquitoes. SIT involves inundating an area with sterilized male mosquitoes to reduce egg hatch and ultimately the number of wild adults. Previous research indicates that enacting mortality early in the larval life of Aedes mosquitoes can result in counter-productive effects, due to reduced competition among developing larvae. I hypothesize that the effectiveness of SIT at reducing adult populations is modified by densitydependent mortality and density-dependent growth among larvae. The objective of this study was to evaluate the influence of maternal nutrient restriction during mid-gestation on the performance of the dam, feeding performance, carcass composition, and genetic expression in the offspring. Cows (n = 46) were randomly assigned to one of two treatments based on global nutrient intake: 1) fed to achieve and/or maintain a BCS of 5 or 6 (Positive Energy Status [PES]; n = 24) or 2) fed to lose one BCS during 99 days of mid-gestation (Negative Energy Status [NES]; n = 22). Cows in NES had greater decreases (P < 0.05) in body fat stores and muscle mass, elevated NEFA concentrations, and displayed decreased (P < 0.05) DMI, HP, and GHG emissions. There were no differences in milk components or volume produced between treatments (P > 0.05). There were no differences in …

This study aimed to discover new antibiotics produced by soil microbes. Previous studies have shown that manipulation of microbial growth conditions through variable carbon sources and growth in coculture could induce the production of novel antibiotics by soil microbes. This study expanded on these observations by testing the levels of antibiotic production when a producing strain was grown in twenty different carbon sources and cocultured with more than one hundred different coculture partners. Fanconi Anemia (FA) is a rare human genetic disease caused by mutation of any of twenty-three genes on the FA pathway, which functions in the repair of DNA damage and the maintenance of genome stability. FA is clinically characterized by congenital abnormalities and a predisposition to aplastic anemia and malignancies. Recently, progressive neurological symptoms and abnormal neurological findings on imaging have been noted in FA patients, including cerebral and cerebellar lesions, calcification, necrosis, gliosis, and associated acute and chronic neurological symptoms; this constellation of symptoms has been termed Fanconi Anemia-Associated Neurological Syndrome (FANS).

Eastern oyster aquaculture has increased steadily over the last three decades, but the industry has yet to achieve its full potential. In the Northeast USA, production is limited in part by a lack of high performing stocks suited to the geographically broad and heterogenous growing environments. Thermal gradients impact fishes differently, due to variations in ecological niches and physiological adaptations. For Largemouth Bass, I analyzed the relationships between air temperature (MATmin°C, MAT24hr°C, and MATmax°C), and growth parameters including maximum length and weight, growth rate (K), longevity, and total length-at-ages 3, 5, and 8 years (TLAge3,5,8), using least squares regression. In the early 1980s Diadema antillarum, or the long-spined urchin, was hit by a catastrophic mass mortality event across the western Atlantic and Gulf of Mexico, inclusive of the Florida Keys. The cause of this die-off event was likely triggered by a pathogen or environmental stressor that decimated native populations by up to 98%. The species has not yet fully recovered and is struggling with recruitment success which is why it may benefit from an ex-situ recovery program. - Ecological immunology posits that variation in host resistance to infection may be attributed partly to the ecological and evolutionary costs of immunity. While the deployment of immune defence is necessary to combat pathogenic infection, hosts pay energetic and other costs for activation.

House flies, Musca domestica L., are pests at livestock facilities. House fly numbers are controlled mainly by manure management and chemical controls, like use of toxic baits. Some toxic baits contain denatonium benzoate as a bittering agent, to avoid poisoning of children. House fly behavioral response to denatonium benzoate was examined, using 2 different diluents, sucrose solution or orange juice. Response of flies to quinine and sucrose octaacetate, 2 other bittering agents, was also examined in sucrose. Sucrose is the sweetener in commercially available toxic house fly baits. Biofluorescence is increasingly recognized as a potentially important form of visual communication across a wide range of taxa, yet its ecological and behavioral function remains largely untested in many terrestrial species. In the red-backed salamander (Plethodon cinereus), fluorescence is a sexually dimorphic trait most strongly expressed in males, particularly around the cloacal region and ventral side of the tail, areas that are often displayed during territorial contests and courtship. Rival males also direct behaviors like cloacal investigation toward these fluorescent regions, suggesting a potential signaling function. This study investigated the molecular mechanisms underlying chemotherapy-induced cognitive impairment (CICI) following doxorubicin and cyclophosphamide treatment in a rat model, revealing three significant alterations in hippocampal tissue: selective downregulation of heterogeneous nuclear ribonucleoprotein A1 (hnRNPA1) (21% decrease, p=0.0298), compromised astrocytic network integrity evidenced by reduced GFAP-positive area in cornu ammonis 3 (CA3) (24%, pB=0.026) and dentate gyrus (DG) (19%, pB=0.027) regions, and elevated cellular senescence marker p19Arf specifically in DG (69%, pB=0.033) and CA3 (27%, pB=0.024). The inverse relationship between hnRNPA1 reduction and p19Arf elevation suggests a potential mechanistic link between RNA dysregulation and senescence induction, with pronounced vulnerability in …

The figitid parasitoid wasps of Drosophila are gaining importance in agriculture due to their ability to kill the resilient soft-fruit pest Drosophila suzukii. Their success and infection strategies vary, and the mechanisms underlying these differences are largely unknown. Some of their venom proteins have been shown to be important in suppressing host defense. Here, we report that Ganaspis brasiliensis, a parasitoid of D. suzukii survives well on Drosophila yakuba, a host species with a strong cellular immunity, and infected flies are unable to mount a strong immune response. The lampricide, 3-trifluoromethyl-4-nitrophenol (TFM), has been used to successfully control invasive sea lamprey (Petromyzon marinus) in the Laurentian Great Lakes for over 50 years. TFM’s effectiveness is due to the relatively low capacity of sea lamprey to detoxify TFM. Water temperature has been shown to increase TFM tolerance in larval sea lamprey. However, the effects of temperature on TFM sensitivity in non-target species such as the endangered lake sturgeon (Acipenser fulvescens) are poorly understood, particularly during their early life stages. Palaeoecological data shows fluctuating salinity levels in lakes across the Great Plains due to alternating wet and dry periods over the last two millennia. Droughts caused by this natural variation may be exacerbated by anthropogenic climate change, as the Great Plains are expected to become drier over the coming decades. As conditions become drier, the evaporation of water will increase the salinity of lakes across the region. Increases in salinity may significantly impact biodiversity, including important members of lake food webs such as zooplankton. Caribou lichen species are common in mid- to late-seral boreal forests, serving as an important winter diet staple for boreal woodland caribou. However, as climate change causes boreal wildfires to increase in size, severity, and frequency, available tracts of mature forest are decreasing, diminishing the extent of mature caribou lichen stands. Caribou lichens reproduce vegetatively, so bigger and more severe fires could exacerbate their dispersal limitations, potentially extending the timeline for caribou lichen stand recovery beyond 80 years. Trials have shown that “transplanting” caribou lichen fragments into disturbed areas can lead to the establishment of caribou lichen mats; as such, …
