Microsatellites Reveal The Population Genetic Structure Of Yellow Perch In Minnesota, Megan Boche

When analyzing a population’s genetic makeup, looking at variation of individuals and populations is a crucial factor. The management of yellow perch will be aided by this information particularly in regard to stocking or relocation. Microsatellite loci were used to analyze the genetic population structure of yellow perch Perca flavescens within and among 29 sampling areas throughout Minnesota’s basins. Seven microsatellite DNA loci were evaluated, with heterozygosities ranging from 0.41 to 0.78. The Lower Mississippi basin samples showed higher numbers of alleles per locus and greater expected heterozygosities compared to other basins, with Lake Pepin (PEP) exhibiting the highest average …

The insect molting process is critical to growth and development and is regulated in part by the neuropeptides corazonin, eclosion hormone, and α and β bursicon. We found messages in a synganglion transcriptome from adult, female American dog ticks, Dermacentor variabilis (that do not molt), with a high similarity to the larval insect neuropeptides that control molting. The phylogenetic analysis of the tick putative neuropeptides compared to other arthropods is discussed in detail. Eggs of Schistosoma mansoni are produced by adult female worms in mesenteries of infected hosts. Eggs can cross the intestinal barrier and form granulomas in the tissue or breach and exit the host through fecal excretion. These interactions may affect the host microbiome assemblages. Given the potential for schistosomal alteration of host gut microbiome and subsequent effects on the fecal bacterial composition, it is important to conduct controlled microbiome studies on model animals. Foraging success is a crucial factor in survival and reproductive success for many animals. One mechanism that can improve an organism’s foraging success is foraging plasticity, or the organism’s ability to alter their behavior in response to environmental changes. This study aims to evaluate if the Colorado Brown Tarantula, Aphonopelma hentzi, a long-lived, sit-andwait predator will use foraging plasticity in response to increased prey encounters. The individual size distribution (ISD) is an individual based model that estimates the relative abundance of individual body sizes in a community. ISD’s slope or lambda (λ) gives insight into the communities’ food web and energy transfer. Climate change is expected to shift mean body sizes and λ as it increases individuals’ energy demands. Alterations in food webs and spatial distribution throughout the middle Missouri River basin may also have an impact on these communities λ. The adaptation of small birds to cold environments involves mechanisms for both heat conservation and heat generation. While minimizing heat loss reduces the energy needed to maintain body temperature, thermogenesis remains essential. Shivering and non-shivering thermogenesis (NST) are two likely mechanisms supporting cold adaptation in small avian species. Although sarcolipin (SLN) aids NST in small mammals, studies in birds such as Dark- eyed Juncos have shown no increase or even a decrease in SLN expression during cold exposure, suggesting limited involvement. Montane regions in the Neotropics are hotspots of endemism and biodiversity, yet their species are especially vulnerable to the impacts of climate change and geographic isolation. This thesis investigates the evolutionary and ecological dynamics of three bird species endemic to the Darién Highlands of eastern Panama: Myadestes coloratus, Chlorospingus tacarcunae, and Margarornis bellulus. Through an integrative approach combining ecological niche modeling, morphometric analysis, and genomic data from ultraconserved elements (UCEs) and mitochondrial DNA, the study explores the historical biogeography, current distribution patterns, population structure, and phenotypic differentiation of these montane taxa. Rough-skinned newts (Taricha granulosa) occur along the Pacific coast of the U.S. from central California to southwestern Alaska at elevations from zero to 2,800 m. Across that range, the timing and duration of various life history events (i.e., hatching, metamorphosis, breeding, and oviposition) are largely unknown, although general trends have been observed in a few locales. "This thesis aims to identify interactions between C. elegans’ reproductive system and global pollutants such as microplastics, HgCl₂, nicotine, and pesticides. Human exposure to these pollutants and their effect on reproduction can be analyzed using the C. elegans as a model organism. Systematic analysis of matricidal hatching, progeny production, germline apoptosis, and lifespan assays determined effects on reproduction and reproductive aging. Through my experiments, it was determined that no significant effects were produced from the selected polystyrene, polyethylene, vape juice, nicotine hemi sulfate salt, imidacloprid, or glyphosate regarding matricidal hatching or progeny production. Parasites of freshwater fish play a vital functional role in aquatic ecosystems, but the spatial and ecological determinants of their prevalence and characteristics are still poorly understood. The current study quantified the diversity, prevalence, and environmental determinants of whole parasite burdens in bluntnose minnows across multiple creeks in central Illinois. Fish sampling and the collection of stream habitat data were conducted during the summers of 2023 and 2024. Fish were examined for parasites using necropsy and histology. A total of 20,817 parasites, predominantly trematodes (Posthodiplostomum spp.), cestodes (Proteocephalus spp), and protozoans, were identified. The most prevalent and abundant …

Anthropogenic factors have been the cause of the decline of tree species across the globe. From unsustainable logging and development to disease and improper management, threats to forests have taken their toll. The American chestnut (Castanea dentata) was once an integral basal tree species in Appalachian forests, but chestnut blight drove the species to functional extinction by the mid-1900s. The recent creation of genetically modified chestnuts gives hope for future restoration of the species throughout its historical range. Oligodendrocyte progenitor cells differentiate into oligodendrocytes, which myelinate axons during development and following demyelinating injury. However, the mechanisms that drive the timing and specificity of developmental myelination are not well understood. We hypothesized that oligodendrocyte progenitor cell proliferation and differentiation would be affected by pathological neuronal activity during adolescent development when developmental myelination is occurring and that this would also impact neuron-to-oligodendrocyte progenitor cell connectivity and myelination. We used kainic acid to induce a seizure in mice, treating equal numbers of males and females, in sample sizes of at least five animals. We found that the seizures led to increased …

The typical American diet exceeds dietary fat recommendations, contributing to the rising rates of obesity. This negatively impacts fertility, pregnancy and fetal growth, and lifelong offspring health. During pregnancy, this also increases risk of co-morbidities like gestational diabetes. Despite this, lipids are not screened during pregnancy and limited research exists evaluating these co-exposures on offspring health. Our lab used a rat model of maternal high fat (HF) diet with late-gestation diabetes (DM) to demonstrate that mitochondrial dysfunction and oxidative stress incite lifelong cardiometabolic disease risk in offspring. Amphibians are at disproportionately higher risks of extinction than all other vertebrate classes, and a major causing factor is invasive predation. The American bullfrog (Lithobates catesbeianus), native to Eastern North America, is a notorious invasive species which preys on native Sierran treefrogs (Pseudacris sierra) in Northern and Central California. Promoting regeneration of native trees, like Quercus spp., is a priority for land managers given the ecological and economic importance of oak woodlands. Although direct seeding may promote recruitment of Quercus spp., the effectiveness of direct seeding may be greatly reduced in environments where the activity of granivorous rodents is high. Importantly, the activity of granivorous rodents may be highest in environments where oak restoration is most desired, such as habitats invaded by non-native woody shrubs. The development of offshore wind structures in the northeastern U.S. will contribute to renewable energy goals, but will overlap with many marine species as well as economically important fisheries. High voltage transmission cables from offshore wind farms emit electromagnetic fields (EMFs) that elasmobranchs may detect using sensory organs known as ampullae of Lorenzini. Animals face the risk of encountering pathogenic microbes while searching for nutritive resources. Assessing the interplay between the benefits of nutrition vs. the risk of infection might influence the behavioral regulation of immune processes. In Caenorhabditis elegans (C. elegans), a nematode roundworm, behavioral immunity is partially regulated by the innate immune molecule TOL-1: a homolog of vertebrate Toll-like Receptor (TLR) proteins. Climate change is intensifying permafrost thaw across terrain of the circumpolar Arctic. Increased temperatures and rainfall at northern latitudes have resulted in the expansion of thermokarst features such as retrogressive thaw slumps, which make previously frozen debris available for transport into nearby river systems. "We explored 1) the toxicological impacts of e-cigarette aerosols and 2) the decontamination efficacy of Cold Atmospheric Plasma (CAP) technology over organophosphates. In examining the effects of firsthand and secondhand e-cigarette aerosol exposure on A549 and BEAS-2B lung cells, we observed significant cytotoxicity, increased ROS levels, and epigenetic modifications, including heightened DNA methylation and downregulation of histone deacetylases (HDACs). These molecular alterations may contribute to chronic respiratory conditions, such as COPD, underscoring the health risks of e-cigarette exposure.

Coastal fish populations are threatened by multiple anthropogenic impacts, including the accumulation of industrial contaminants and the increasing frequency of hypoxia. Some populations of the Atlantic killifish (Fundulus heteroclitus), like those in New Bedford Harbor (NBH), Massachusetts, USA, have evolved a resistance to dioxin-like polychlorinated biphenyls (PCBs) that may influence their ability to cope with secondary stressors. To address this question, we compared hepatic gene expression and DNA methylation patterns in response to mild or severe hypoxia in killifish from NBH and Scorton Creek (SC), a reference population from a relatively pristine environment. Abstract: In this essay I review my 45+ year career and some of the significant changes I have observed. This journey involved research in rivers and streams, estuaries, coastal systems (including the continental shelf), and the deep sea to a depth of about 4,000 m.
