It is well known that solid diet supplementation in early life can significantly promote rumen development and metabolic function in young ruminants. However, the changes in the expressed proteome and related metabolism in rumen epithelium in response to a supplemented solid diet remain unclear. In this study, rumen epithelial tissue from goats in three diet regimes including milk replacer only (MRO), milk replacer supplemented concentrate (MRC), and milk replacer supplemented concentrate plus alfalfa pellets (MCA) were collected for measurement of the expression of epithelial proteins using proteomic technology (six per group). Heat stress (HS) is a growing concern in broiler production due to increasing environmental temperatures. Little is known of the overall effect of HS on innate immunity. Lipopolysaccharide (LPS), an outer membrane component of Gram-negative bacteria, is commonly used to study the inflammatory response. Streptococcus gallolyticus (SG) is a Gram-positive found as commensal gut flora in animals and humans. SG has been associated with acute mortality in poults 2 to 3 weeks of age, with lesions mainly consisting of splenomegaly, hepatomegaly, and pericarditis. The objective of this thesis was to reproduce disease in poults caused by SG recovered from clinical field isolates and evaluate different routes of administration. Three field isolates from geographically disparate areas in Arkansas and Missouri were used for the experiments contained in this thesis. Skeletal muscle hypertrophy is a complex process that involves a range of signaling pathways and transcriptional regulators. Many hormones and growth factors can activate key signaling pathways, such as the PI3K/AKT/mTOR, MAPK, and cAMP pathways, which play a crucial role in the regulation of muscle hypertrophy. Essential oils (EO) affect performance, intestinal integrity, bone mineralization, and meat quality in broiler chickens subjected to cyclic heat stress (HS). Day-of-hatch Cobb 500 male broiler chicks (n = 475) were randomly divided into four groups. In recent times, meat quality has become a key aspect of poultry production. In the current study, 2,400 sex separate broilers from 4 commercial strains were placed in 6 replications to evaluate various meat quality characteristics when grown to 2 market weights. Broilers were fed 1 of 2 diets with varying degrees of amino acid inclusion to evaluate meat quality of broiler raised on varying planes of nutrition. In this study we determined the impact of increasing dietary whole cottonseed (WCS) on rumen fermentation, nutrient digestibility, and microbial community composition. This study contributes novel information to the dairy community deepening the understanding of how including different levels of WCS can affect the rumen environment. This research was conducted in continuous culture fermenters. Treatments included a control diet without WCS, or the control diet plus 5, 10, or 15% (dry matter) WCS. Femur head necrosis (FHN), also known as bacterial chondronecrosis with osteomyelitis (BCO), has remained an animal welfare and production concern for modern broilers regardless of efforts to select against it in primary breeder flocks. Characterized by the bacterial infection of weak bone, FHN has been found in birds without clinical lameness and remains only detectable via necropsy. This presents an opportunity to utilize untargeted metabolomics to elucidate potential non-invasive biomarkers and key causative pathways involved in FHN pathology. This dissertation utilized a mixed-methods approach which illustrated the potential benefits of HAI in addressing human health concerns, focusing on specific populations within the United States. The chapters presented in this dissertation provide insight into the complex relationships between humans and animals, including the roles of companion animals in promoting human well-being. Chapter three surveys 234 Americans observing COVID-19 stay at home isolation orders and findings suggest that the presence of companion animals alleviates stress and isolation by providing attachment figures and activities on which to focus one’s energy. Driven by changes in the quality of beef and consumer health preferences, the beef industry began to advocate for “value-based marketing,” resulting in the development of grid pricing and eventually formula transactions. The present study investigated continuing exposure of sow vocalization and scent during the early weaning period on complete blood cell count. Aquatic organisms are frequently exposed to various environmental stressors. In this study, we observed the short-term (1-week) combined dose-dependent effect of Roundup, a glyphosate-based herbicide, and high temperature (30°C) on the morphology of gills and digestive glands, expression of heat shock protein-70 (HSP70), dinitrophenyl protein (DNP), nitrotyrosine protein (NTP), antioxidant enzymes such as catalase (CAT) and superoxidase dismutase (SOD) in tissues of American oyster (Crassostrea virginica). The poultry industry has significant coccidiosis and necrotic enteritis (NE) challenges, leading to high mortality and unacceptable growth without antibiotic treatment. This research explored supplementing Nigella sativa (black cumin) seed oil in poultry feed to mitigate coccidiosis and prevent or lessen NE in broilers. In vivo studies consisted of 384 and 320 Cobb 500 male broiler chicks distributed in a randomized complete block experimental design for trials 1 and 2, respectively. A rising problem in agriculture is the increase of antibiotic-resistant Salmonella cases associated with chicken eggs, which transmit infection to humans. To counter this, new approaches to combat Salmonella in chickens and desiccated on eggshells are vital in the prevention of human foodborne illness. Disrupting signaling pathways with naturally occurring compounds provides a potential novel avenue for prevention of Salmonella infections, as this would disrupt sensing of these environments and inhibit subsequent optimal gene expression. Two, 3-year studies were conducted to evaluate the value of cereal rye, winter wheat, and winter triticale as forage sources for either for silage or early spring grazing. For silage, the objective was to evaluate species and maturity effects on dry matter yield, nutritive value, and fermentation of small cereal silage. There are no major differences between wheat, rye, or triticale in terms of the relative timing of each species reached each maturity stage. A finishing experiment evaluated the effect of incremental supplemental rumen-protected lysine on beef cattle performance. All cattle were fed the same diet of high moisture corn, dried rolled corn, modified distillers grains plus solubles, corn silage, and supplement with inclusion levels of lysine ranging from 0 to 7 grams/day, in 1 g increments. Dry matter intake (P ≤ 0.04) and ADG (P ≤ 0.04) decreased linearly as supplemental lysine increased in the diet with no effect on final carcass adjusted feed efficiency (P ≥ 0.34). Play behavior is ubiquitous among young mammals and has a variety of important influences on early development. I evaluated possible influences of social play behavior on the development of temperament in juvenile Belding’s ground squirrels (Urocitellus beldingi), using the caution/boldness spectrum as a measure of temperament. I disrupted the play behavior of juvenile U. beldingi by walking toward juveniles at play until the play interactions stopped. Breast myopathies have caused significant economic losses to the poultry industry over the last decade. The induction of woody breast (WB), white striping (WS), and spaghetti meat (SM) have caused textural issues, functional issues, and a decline in consumer acceptance. Of these three myopathies, WB causes the most pronounced effects on product quality and reduces functional use. The objective was to evaluate the influence of pregnant cattle grazing endophyte-infected tall, toxic, fescue (E+) during gestation on innate immune response of male offspring to an endotoxin challenge. Crossbred multiparous cows (n = 36) were bred to Red Angus sires and housed in replicated E+ or novel endophyte-infected, non-toxic, fescue (NE+) pastures prior to breeding and throughout gestation. From this calf-crop, a subset of post-weaned steers (n = 8 E+; n = 8 NE+) were selected for a lipopolysaccharide (LPS) challenge. Herein, we present a dataset based on the RNA-Seq analysis of liver tissue from bovine female fetuses at day 83 of gestation. The findings were reported in the main article, “Periconceptual maternal nutrition affects fetal liver programming of energy- and lipid-related genes”[1] . These data were generated to investigate the effects of periconceptual maternal vitamin and mineral supplementation and rates of body weight gain on the transcript abundance of genes associated with fetal hepatic metabolism and function. Wild sloth bears (Melursus ursinus) display seasonal diet preferences for insects and fruit during the dry and monsoon season respectively. To potentially utilize as a method for reducing abnormal behaviors and improving animal welfare, the purpose of the study is to survey zoos with captive sloth bears and determine if zoo-housed individuals display seasonal diet preferences. From May 18th, 2022 to March 2nd, 2023 data was collected on Priya, a sloth bear from Idaho Falls Zoo, and her preference for fruit and insects. Although traditional ryegrass pastures are still commonly used, they require intense management and input and do not perform well during the summer months. Multispecies systems have been recognised as more sustainable, needing less fertiliser and being tolerant to drought and heat. While data on monoculture system fat composition exist, information for multispecies is scarce.
