Multispecies Genomic Sex Identification Using Ddx3 Gene Polymorphisms, Jessica Felts Aug 2023

Multispecies Genomic Sex Identification Using Ddx3 Gene Polymorphisms, Jessica Felts

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

PCR sex determination assays must be reliable and cost effective due to the frequent and integral use of these assays in biological research and the animal production industry. Thus, the design of proof of a primer pair with a built-in control is warranted to not only bypass the extra cost of a multiplex reaction, but also to prevent anomalous results that have been documented with other primer pairs.

The objective of this study was to design primer pairs with built in PCR amplification control to identify sex in Equus caballus (domestic horse), Homo sapiens (humans), Macaca mulatta (rhesus macaque), and …

Anthropogenic Factors Affecting Common Raven Occurrence And Depredation Of Artificial Nests Within Greater Sage‐Grouse Habitat In Southern Utah, Zoë S. Moffett Aug 2023

Anthropogenic Factors Affecting Common Raven Occurrence And Depredation Of Artificial Nests Within Greater Sage‐Grouse Habitat In Southern Utah, Zoë S. Moffett

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Certain species of wildlife are more generalist and adaptive than others. These species often flourish when supported by human activities that provide additional food and habitat for them. The common raven (Corvus corax; hereafter raven) is one such species; their populations have risen and spread throughout the Intermountain West. As generalist scavengers and predators, ravens have been found to pose a severe threat to several threatened or sensitive species, including the greater sage-grouse (Centrocercus urophasianus; hereafter sage-grouse). The purpose of my research was to determine the specific anthropogenic and habitat factors that may increase the threat …

A Mechanistic Examination Of Interspecific Competition Between Wild And Domestic Herbivores, Courtney Check Aug 2023

A Mechanistic Examination Of Interspecific Competition Between Wild And Domestic Herbivores, Courtney Check

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Large herbivores, such as mule deer and cattle have similar life histories and likely compete for resources. However, quantifying the extent to which these species compete and the specific resources they compete for has proved challenging. My research examines if cattle influence deer abundance and behavior due to competition for forage, competition for shade, and/or by affecting the predation risk of deer. Using a grid of autonomous trail cameras, I was able to determine if cattle abundance influences local deer abundance in relation to specific resources and habitat features. Using GPS data from collared deer, I was also able to …

Treatment Of Yellow Fever Virus With The Ns4b Inhibitor Bdaa And Effects On Rna‐Sensing Innate Immune Pathways In Hamster And Mouse Models, Abbie E. Weight Aug 2023

Treatment Of Yellow Fever Virus With The Ns4b Inhibitor Bdaa And Effects On Rna‐Sensing Innate Immune Pathways In Hamster And Mouse Models, Abbie E. Weight

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Yellow fever (YF) is an acute and often severe disease cause by yellow fever virus (YFV). Although there is an effective vaccine available to prevent YF, there are no antiviral drugs approved to treat the disease, which has a considerable disease burden in endemic areas of South America and Africa. BDAA is an experimental antiviral treatment which has shown efficacy against YFV both in cell culture and when administered before infection in an animal model of disease. BDAA targets the YFV protein NS4B and has two reported mechanisms of action: the primary mechanism of action is the direct inhibition of …

Evaluation Of Heat Stress And Dietary Amino Acid Density On Nutrient And Energy Partitioning In Broilers, Jean-Remi Teyssier Aug 2023

Evaluation Of Heat Stress And Dietary Amino Acid Density On Nutrient And Energy Partitioning In Broilers, Jean-Remi Teyssier

Graduate Theses and Dissertations

The simultaneous increase in the global demand for animal-based protein products and the rise in temperature associated with global warming pose important challenges to the poultry industry. Heat stress (HS) occurs when an animal’s heat production (HP) surpasses its capacity to dissipate heat into the surrounding environment, ultimately leading to reduced feed intake (FI) and negative impacts on performance. A better understanding of bird responses to elevated temperatures is required to propose effective nutritional strategies that mitigate the adverse effects of HS. Therefore, this dissertation aimed to characterize the nutritional and physiological mechanisms explaining the loss in performance and carcass …

Investigation Of Consumer Freezing Practices, Condition And Duration On Palatability Of Beef, Elizabeth Sue Neal Aug 2023

Investigation Of Consumer Freezing Practices, Condition And Duration On Palatability Of Beef, Elizabeth Sue Neal

Graduate Theses and Dissertations

The objective of these studies was to determine: the impact of extended periods of frozen storage duration and packaging type on palatability traits of cooked beef steaks from two muscles and the impact of freezing type and storage duration on objective and subjective measures of ground beef palatability traits. In study 1, no differences were elicited from any of the possible interactions (P= 0.95) for SSF or CL: package × muscle × freezing duration, package × muscle, package × duration or muscle × duration. However, freezing duration did impact both SSF (P < 0.01) and CL (P < 0.001). Generally, for both SSF and CL steaks that were frozen for 9-months elicited both the highest SSF values as well as the greatest percentage of CL among all treatments. In comparison, EM was impacted by the interaction (P = 0.059) of packaging type  muscle  freezing duration. Samples from OW GM stored for 9-months elicited the highest percentage of EM while fresh OW LL samples resulted in the lowest. Additionally, expressible moisture was impacted by the interaction (P = 0.047) of muscle  freezing duration. Among all treatments, GM steaks that were frozen for 9-months elicited the highest EM values and fresh LL samples the lowest EM values. Contrastingly, there was no significant difference observed in expressible moisture for the interaction (P  0.18) of package type  freezing duration or for the interaction (P = 0.70) of package type  muscle. Generally, fresh OW GM steaks resulted in the greatest concentration of lipid derived volatile compounds such as aldehydes, hydrocarbons and alcohols which contrasted sensory ratings of trained panelists that indicated oxidized and refrigerator-stale ratings increased as storage time increased for OW steaks. In study 2, The interaction of freezer treatment × storage duration impacted gumminess (P = 0.05), a TPA attribute. In greater detail, samples stored in RF for 6-months resulted in the greatest gumminess values (P < 0.001), while those stored in the CF for 12-months elicited the lowest (P < 0.001). Similarly, flavor development was also impacted by the interaction (P = 0.05) of freezer treatment × storage duration. Three lipid derived compounds were of greatest concentration among RF patties stored for 1-month. In contrast, the interaction of freezer treatment × storage duration elicited no impact on consumer ratings, SF or TBARS. Nonetheless, frozen storage duration impacted TPA, flavor development, consumer ratings, SF and TBARS as a main effect (P < 0.05), especially in regard to tenderness and juiciness. Moreover, beef flavor development, tenderness and juiciness are impacted by freezing duration, muscle, freezer type and packaging, however these factors are not necessarily independent of one another. Furthermore, the retail display period is a critical period for fresh beef steaks, especially when packaged in aerobic conditions. For optimal eating experience, beef products should be stored for extended periods in a vacuum packaging, in a designated freezer that is not opened frequently to allow for optimal air flow regulation and minimize freezer burn to improve tenderness and juiciness.

Dry-Aged Beef Flavor Development, And The Effect Of High Levels Of Vitamin-E On Beef Color Stability, Nicolas Herrera Aug 2023

Dry-Aged Beef Flavor Development, And The Effect Of High Levels Of Vitamin-E On Beef Color Stability, Nicolas Herrera

Department of Animal Science: Dissertations, Theses, and Student Research

Palatability and consumer purchasing are essential factors of fresh meat. To meet consumer expectations during purchasing and consumption, fresh beef must maintain desirable cherry-red color at retail and achieve desirable cooked flavor. Dry-aged beef popularity has grown, and identifying methods to ensure quality dry-aged meat is necessary to maintain demand. To understand dry-aged palatability, this dissertation addressed effects of moisture loss and aging time, fundamental factors of dry-aged beef, using bone-in and boneless strip loins. Study two investigated flavor development using cookery methods and internal temperatures in boneless and bone-in dry-aged beef. In study one, increased dry-aging time (creation effect) …

Identification And Assessment Of Lameness In Sows Through The Utilization Of NuTrack And Gaitfour Systems, Lexi M. Ostrand

Department of Animal Science: Dissertations, Theses, and Student Research

Lameness is a financial concern for producers and a welfare concern for the animals. Utilizing both the GAITFour and NUtrack systems, gilts’ natural tendencies and gait were analyzed to better understand the onset of lameness and what indicators can be identified in a commercial setting. Beginning at 20 weeks of age, video on replacement gilts was collected for 9 consecutive days and processed using the NUtrack System to identify individual gilts’ distance travelled (m), avg speed (m/s), angle rotated (degrees), and time standing (s), sitting (s), eating (s), and laying (s). NUtrack is a deep learning-based multi-object …

Consequences Of Cow–Calf Production With Limited Perennial Forage Grazing, Hannah Fae Speer Aug 2023

Consequences Of Cow–Calf Production With Limited Perennial Forage Grazing, Hannah Fae Speer

Department of Animal Science: Dissertations, Theses, and Student Research

Conversion of perennial grasslands to cropland and rising land prices prompts a need to evaluate alternative beef cow-calf production systems. A 3-yr study was conducted comparing beef cow-calf performance between two August-calving cow systems: perennial forage and corn residue grazing; or summer drylot, fall grazing of late-summer-planted cover crop, and corn residue grazing. Cow body condition score differed between systems, but condition of cows in both systems never fell below adequate. There were no differences in pregnancy rates between systems. Calf weaning weights differed between systems each year; differences are attributed to feedstuff quantity, quality, and type available to calves …

Mirna Regulation Of Skeletal Muscle Hypertrophy, Maslyn Greene Aug 2023

Mirna Regulation Of Skeletal Muscle Hypertrophy, Maslyn Greene

All Dissertations

Muscle tissue is vital to animal survival and economically essential to livestock producers. Reduced muscle growth can occur from various reasons, including genetics, nutrition, and the pre and postnatal environment. Understanding the mechanisms behind muscle growth can aid in developing strategies to increase muscle mass. Several methods to increase muscle mass in livestock animals are currently being pursued, including nutrition, hormonal therapy, and genetic selection. One newly developing avenue is using miRNA technologies to change the expression of protein-coding genes. miRNA are a type of RNA that regulate over half of protein-coding gene expression. Here we evaluate muscle characteristics such …

Exploring Biotic And Abiotic Constraints On Competition For Signal Space Within Birdsong Communities, Joanna M. Sblendorio Aug 2023

Exploring Biotic And Abiotic Constraints On Competition For Signal Space Within Birdsong Communities, Joanna M. Sblendorio

Dissertations

With the acceleration of climate change and the conversion of natural ecosystems to human-dominated landscapes, there is concern for the persistence of biodiversity amid novel biotic and abiotic factors. An important outcome of shifting species distributions and environmental disturbance is the formation of novel assemblages. For animals that vocalize for mate attraction and resource defense, the introduction of new species into communities may alter availability of acoustic space for signaling and affect detection of signals by conspecifics. Thus, to minimize signal overlap, species should partition signal space and thereby inhabit well-defined acoustic niches. Yet mixed evidence exists regarding signal space …

Between 2018 and 2020, over 100 wild turkey fecal samples were collected from the Eastern and Central thirds of the United States, where commercial turkey production is uncommon. We hypothesized that anticoccidial-sensitive Eimeria spp. would be present in wild turkey fecal samples. Samples containing Eimeria spp. oocysts were amplified in vivo. If propagation was successful, the samples were PCR-speciated and subjected to anticoccidial sensitivity testing (AST) for key members of both ionophore and chemical categories of anticoccidial drugs. The purpose of this study was to isolate Eimeria spp. relevant to commercial turkey production that possessed sensitivity to monensin, …

Occupancy And Activity Patterns Of Nine-Banded Armadillos (Dasypus Novemcinctus) In A Suburban Environment, Brett Degregorio, Matthew R. Mcelroy, Emily P. Johansson

Biological Sciences Faculty Publications and Presentations

The geographic range of the nine-banded armadillo (Dasypus novemcinctus) has rapidly been expanding within the United States for the last 150 years. One of the factors contributing to this astounding range expansion is the species’ ability to survive in and colonize human-dominated areas. Despite the fact that armadillos live alongside humans in numerous towns and cities across the Southeastern, Southcentral, and now Midwestern United States, we know relatively little about the behavior and ecology of armadillos in human-developed areas. Here, we used motion-triggered game cameras in over 115 residential yards in the rapidly developing Northwest corner of Arkansas …

An Ecological Survey Of East Texas Salamanders Across The Camp Tyler Outdoor Field School In Smith County, Texas, Justin C. Hunt

Biology Theses

Amphibians are a unique class of organisms with a very long and storied evolutionary history of survival. Many modern amphibian clades occupy several vital ecological roles within their native freshwater environments. One of these roles, typically includes functioning as an ecological indicator species, whereby the presence of stable and diverse populations of many amphibian species, including salamanders, within a freshwater ecosystem have long been considered ecological indicators of good habitat quality and stable ecosystem health. Similarly, salamanders also function as important members of their local food webs and act as valuable mediators of complex trophic hierarchies to facilitate nutrient cycling …

Are Shrubs A Necessary Component Of The Habitat For Ammospermophilus Nelsoni In The San Joaquin Desert?, David J. Germano, Larry R. Saslaw

Western North American Naturalist

The San Joaquin antelope squirrel (Ammospermophilus nelsoni; SJAS) is a state-listed Threatened species in California that evolved in the shrublands of the San Joaquin Desert. Due to livestock overgrazing and fires, much of the desert has lost its shrub cover, which would be a conservation concern if shrubs are a necessary component of the habitat for squirrels. We established two 64-trap plots on the Lokern area of Kern County, California: one covered by saltbush scrub (Atriplex spp.) and another nearby without shrubs. We trapped quarterly starting in January 2019 and ending in November 2021 (12 trap sessions). …

Western North American Naturalist

Puma (Puma concolor) is a wide-ranging, large felid species, and site-specific research on its diet is important for local management. Like the diets of other large felids, puma diets may differ between sex due to size dimorphism and between seasons due to changes in prey vulnerability and availability. We investigated the influence of sex and season on puma diet in southwest New Mexico in terms of prey species and size categories. Pumas (10 males, 6 females) were tracked with GPS collars for an average of one year per individual between February 2008 and July 2020. Puma location was …

Background Rumen microbes break down complex dietary carbohydrates into energy sources for the host and are increasingly shown to be a key aspect of animal performance. Host genotypes can be combined with microbial DNA sequencing to predict performance traits or traits related to environmental impact, such as enteric methane emissions. Metagenome profiles were generated from 3139 rumen samples, collected from 1200 dual purpose ewes, using restriction enzyme-reduced representation sequencing (RE-RRS). Phenotypes were available for methane (CH4) and carbon dioxide (CO2) emissions, the ratio of CH4 to CH4 plus CO2 (CH4Ratio), feed efficiency (residual feed intake: RFI), liveweight at the time …

Waterbody Size Predicts Bank- And Boat-Angler Efforts, Derek S. Kane, Kevin L. Pope, Keith D. Koupal, Mark A. Pegg, Christopher J. Chizinski, Mark A. Kaemingk Jul 2023

Waterbody Size Predicts Bank- And Boat-Angler Efforts, Derek S. Kane, Kevin L. Pope, Keith D. Koupal, Mark A. Pegg, Christopher J. Chizinski, Mark A. Kaemingk

Nebraska Cooperative Fish and Wildlife Research Unit: Staff Publications

Bank- and boat-angler efforts are logistically difficult and costly to estimate, preventing landscape-scale estimates that are required to address current and future challenges (e.g., climate change, invasive species) for inland recreational fisheries. Using a large Nebraska, USA, recreational fishery dataset (N = 67 waterbodies), we demonstrate that waterbody size can be used to predict bank- and boat-angler efforts across a heterogeneous landscape of extra small (< 104 ha) and large (> 647 ha) waterbodies. Bank and boat anglers respond to waterbody size, however these relationships appear to be unique between the two angler types. Boat-angler efforts increased as a function of waterbody size, whereas bank-angler …

Crop-Livestock Diversification And Efficiency In Agriculture, Jay Parsons, Maroua Afi Jul 2023

Crop-Livestock Diversification And Efficiency In Agriculture, Jay Parsons, Maroua Afi

Cornhusker Economics

Diversification is a familiar strategy for managing risk in agriculture. It can take several forms including growing more than one crop or operating a farm with both crop and livestock enterprises. As with other strategies for managing risk, diversification comes with a unique set of costs. Managing a farm with multiple enterprises creates additional overhead as well as additional demands on management, labor, land, capital, and other resources. Diversification adds complexity to an operation and too much complexity can lead to inefficiencies.

Evaluating Vaccine Management Strategies For Edwardsiella Ictaluri Infections In Zebrafish (Danio Rerio), Brandy Malbrough Jul 2023

Evaluating Vaccine Management Strategies For Edwardsiella Ictaluri Infections In Zebrafish (Danio Rerio), Brandy Malbrough

LSU Master's Theses

Aquaculture is a globally important industry that faces significant challenges due to infectious diseases, which can result in substantial financial losses. In 2011, a new strain of Edwardsiella ictaluri emerged as a major pathogen affecting zebrafish in the ornamental fish industry, leading to high mortality rates and posing a severe threat to the sector. Vaccines provide potential benefits for disease prevention in aquaculture, particularly live-attenuated vaccines (LAVs), which induce strong host immune responses. Bacterial attenuation for vaccine development has evolved from serial passage to more targeted genetic modification techniques. However, regulatory constraints limit the availability of licensed LAVs for aquaculture. …
