The objective of this project was to examine the effect of fungal treatment and liquid hot water pretreatment of switchgrass combine in view of increasing glucose release. The fungal treatment consisted of incubating Pleurotus ostreatus in square switchgrass bales, at 50% moisture content for 25 days, 54 days, and 82 days. The digestibility of the switchgrass biomass was subsequently evaluated using Accelerase 1500 enzyme. Lignin is an important barrier to enzymatic hydrolysis, and it was stipulated that incubation with P. ostreatus would disrupt plant cell walls, resulting in enhanced saccharification. Cotton (Gossypium hirsutum L.) is sensitive to heat stress (HS) during reproductive development. The objective of this study was to evaluate different screening methods for identification of heat tolerance in cotton genotypes. Three growth chamber studies and four field trials were conducted from 2014 to 2017 using genotypes Arkot 9704, VH260, DP 210 B2RF and DP393. Measurements were made of membrane leakage (ML), chlorophyll fluorescence (ChlF), glutathione reductase (GR), and sucrose concentration. A doubled haploid (DH) population of barley (Hordeum vulgare L.) generated from salinity tolerant genotype CM72 and salinity sensitive variety Gairdner was studied for salinity stress tolerance at germination, seedling emergence and first leaf full expansion growth stages. Germination study was conducted with deionized water, 150 mM and 300 mM NaCl treatments. Seedling stage salinity tolerance was conducted with three treatments: control, 150 mM NaCl added at seedling emergence and first leaf full expansion growth stages. We established and elaborated on a method to enrich the membrane proteome of mature pollen from economically relevant crop using the example of Solanum lycopersicum (tomato). To isolate the pollen protein fraction enriched in membrane proteins, a high salt concentration (750 mM of sodium chloride) was used. The membrane protein-enriched fraction was then subjected to shotgun proteomics for identification of proteins, followed by in silico analysis to annotate and classify the detected proteins.

Mango (Mangifera indica) is an important, and popular fruit in Bangladesh. However, the post-harvest processing of it is still mostly performed manually, a situation far from satisfactory, in terms of accuracy and throughput. To automate the grading of mangos (geometry and shape), we developed an image acquisition and processing system to extract projected area, perimeter, and roundness features. The medicinal legume Mucuna pruriens (L.) DC. has attracted attention worldwide as a source of the anti-Parkinson’s drug L-Dopa. It is also a popular green manure cover crop that offers many agronomic benefits including high protein content, nitrogen fixation and soil nutrients. The plant currently lacks genomic resources and there is limited knowledge on gene expression, metabolic pathways, and genetics of secondary metabolite production. Identifying and characterizing alternative splicing (AS) enables our understanding of the biological role of transcript isoform diversity. This study describes the use of publicly available RNA-Seq data to identify and characterize the global diversity of AS isoforms in maize using the inbred lines B73 and Mo17, and a related species, sorghum. Identification and characterization of AS within maize tissues revealed that genes expressed in seed exhibit the largest differential AS relative to other tissues examined. Additionally, differences in AS between the two genotypes B73 and Mo17 are greatest within genes expressed in seed. We demonstrate that changes in the level …

High respiration rate of a grain indicates faster degradation of its dry matter. Proper grain management requires chronological and precise measurements of carbon dioxide evolved from grain respiration during the postharvest storage duration. Therefore the main goal of this research was to develop a new technique that evaluates temporal corn respiration rate using pressure sensors. It was based on measuring pressure drop associated with the grain respiration in a closed container and using it to calculate the grain respiration rates.

A new herbicide site of action (SOA) is needed by rice (Oryza sativa L.) producers in the Midsouth for the control of problematic and herbicide-resistant weeds. Currently, six problematic weeds are relevant to Midsouthern rice producers because of resistance to at least one SOA, if not more. Gowan Company is in the process of commercializing benzobicyclon for use in Midsouthern rice systems. Benzobicyclon, a Group 27 post-flood herbicide, controls a broad spectrum of aquatics, broadleaves, grasses, and sedges, including those currently resistant to Group 2 herbicides. Research has shown that applications of some organophosphate insecticides at planting can reduce cotton injury following applications of the soil-applied herbicide clomazone. Weed-resistance management has become a topic of concern for modern agriculture. Cost related to herbicide usage has increased greatly due to evolution and proliferation of resistant weeds. Therefore, experiments were conducted to investigate the potential for using cover crops to suppress problematic weeds in cotton as well as chemical options for cover crop dessication, and sensitivity of cover crops to residual herbicides were evaluated. No differences were observed for cereal rye biomass production and consequently weed suppression between broadcast and drilled planting methods. The gross primary production (GPP) metric is useful in determining trends in the terrestrial carbon cycle. Models that determine GPP utilizing the light use efficiency (LUE) approach in conjunction with biophysical parameters that account for local weather conditions and crop specific factors are beneficial in that they combine the accuracy of the biophysical model with the versatility of the LUE model. Maize lethal necrosis disease (MLND) is one of the most important viral diseases of maize. MLND occurs when Maize chlorotic mottle virus (MCMV) co-infects the same plant with one of several potyviruses, including Sugarcane mosaic virus, Wheat streak mosaic virus or Maize dwarf mosaic virus. Originally prevalent in the Midwest and Peru in the 1970s, the disease was called corn lethal necrosis (CLN) and was controlled through breeding and sanitation. Recently, the disease has re-emerged in East Africa and is rapidly spreading and threatening the food sources of subsistence-farming populations. The possibility of predicting plant leaf chemical properties using hyperspectral images was studied. Sixty maize and 60 soybean plants were used, and two experiments were conducted: one with water limitation and the second with nutrient limitation, with the purpose of creating wide ranges of these chemical properties in plant leaf tissues. A hyperspectral imaging system with a spectral range from 550 to 1700 nm was used to acquire plant images in a high throughput fashion (plants placed on an automated conveyor belt). Switchgrass (Panicum virgatum L.) is undergoing development as a biomass crop to support conversion of cellulosic biomass to energy. To avoid the competition of biomass with food or feed crops, most commercialization proposals suggest that switchgrass should be grown exclusively on marginal lands that are not fit for food or feed production. The objective of this study was to investigate the potential for cultivar x environment interactions that would affect the methods and approaches for breeding and evaluating switchgrass cultivars, including both upland and lowland types, for high-input versus low-input types of environments. Assessing Anther Extrusion And Its Effect On Us Hard Winter Wheat (Triticum Aestivum L.) Hybrid Seed Production, Nicholas Garst

The promise of higher grain yields as a result of the development and production of hybrid wheat (Triticum aestivum L.) has not been fully realized primarily due to the high cost of seed production. Anther extrusion is a key trait that improves pollen availability, and thus, is expected to enhance hybrid wheat seed production yields. Hard winter wheat germplasm adapted to the US Great Plains was visually assessed for anther extrusion in the field and greenhouse environments. Significant genotypic differences were detected and high broad-sense heritability was calculated (ranging from 0.62 to 0.85) for anther extrusion in the field. …
