Successful memory involves not only remembering information over time, but also keeping memories distinct and less confusable. The computational process for making representations for similar input patterns more distinct from each other has been referred to as "pattern separation." In this work, we developed a set of behavioral conditions that allowed us to manipulate the load for pattern separation at different stages of memory. Thus, we provide experimental evidence that a brain-derived neurotrophic factor (BDNF)-dependent pattern separation process occurs during the encoding/storage/consolidation, but not the retrieval stage of memory processing. The automated touchscreen operant chamber for rats and mice allows for the assessment of multiple cognitive domains within the same testing environment. This protocol presents the location discrimination (LD) task and the trial-unique delayed nonmatching-to-location (TUNL) task, which both assess memory for location. During these tasks, animals are trained to a predefined criterion during ∼20-40 daily sessions. In LD sessions, touching the same location on the screen is rewarded on consecutive trials, followed by a reversal of location-reward contingencies. An increasingly popular method of assessing cognitive functions in rodents is the automated touchscreen platform, on which a number of different cognitive tests can be run in a manner very similar to touchscreen methods currently used to test human subjects. This methodology is low stress (using appetitive rather than aversive reinforcement), has high translational potential and lends itself to a high degree of standardization and throughput. Recognition memory accuracy is harmed by prior testing (a.k.a., output interference [OI]; Tulving & Arbuckle, 1966). In several experiments, we interpolated various tasks between recognition test trials. The stimuli and the tasks were more similar (lexical decision [LD] of words and nonwords) or less similar (gender identification of male and female faces) to the stimuli and task used in recognition testing. Not only did the similarity between the interpolated and recognition tasks not affect recognition accuracy but performance of the interpolated task caused no interference in subsequent recognition testing. A few empirically supported principles can account for much of the thematic content of waking thought, including rumination, and dreams. (1) An individual’s commitments to particular goals sensitize the individual to respond to cues associated with those goals. The cues may be external or internal in the person’s own mental activity. The impact of Parkinson’s disease (PD) on rule-guided behavior has received considerable attention in cognitive neuroscience. The majority of research has used PD as a model of dysfunction in fronto-striatal networks, but very few attempts have been made to investigate the possibility of adapting common experimental techniques in an effort to identify the conditions that are most likely to facilitate successful performance. It is well known that emotionally salient events are remembered more vividly than mundane ones. Our recent research has demonstrated that such memory vividness is due in part to the subjective experience of emotional events as more perceptually vivid, an effect we call emotion-enhanced vividness, or EEV. The present study built on previously reported research in which fMRI data were collected while participants rated relative levels of visual noise overlaid on emotionally salient and neutral images. An integrative model of narrative identity builds on a dual memory system that draws on episodic memory and a long-term self to generate autobiographical memories. Autobiographical memories related to critical goals in a lifetime period lead to life-story memories, which in turn become self-defining memories when linked to an individual's enduring concerns. Self-defining memories that share repetitive emotion-outcome sequences yield narrative scripts, abstracted templates that filter cognitive-affective processing. The life story is the individual's overarching narrative that provides unity and purpose over the life course. This thesis examines the nature of long-term absolute pitch memory—an ability traditionally assumed to belong only to absolute pitch (AP) possessors—by testing for evidence of this memory for “standard” frequencies in musicians without AP. Standard frequencies, those based on the equally tempered system with A = 440 Hz, are common in the sonic environment of the Western college musical education, and thus could have the opportunity to penetrate listeners’ long-term memories. To determine what factors are important for minimizing interference effects in spatial memory, Clark’s Nutcrackers, Nucifraga columbiana were tested for their spatial memory for two serial lists of locations per day. In this experiment two unique landmark sets were either different between List 1 and List 2 or the same. We found that Nutcrackers were most susceptible to interference when the landmark sets were the same. This study suggests that repeatedly testing animal memory in the same room, with the same cues, can hamper recall due to interference.

Increasingly memory deficits are recognized in Parkinson's disease (PD). In PD, the dopamine-producing cells of the substantia nigra (SN) are significantly degenerated whereas those in the ventral tegmental area (VTA) are relatively spared. Dopamine-replacement medication improves cognitive processes that implicate the SN-innervated dorsal striatum but is thought to impair those that depend upon the VTA-supplied ventral striatum, limbic and prefrontal cortices. Our aim was to examine memory encoding and retrieval in PD and how they are affected by dopamine replacement. Mild cognitive impairment (MCI) is associated with early memory loss, Alzheimer's disease (AD) neuropathology, inefficient or ineffective neural processing, and increased risk for AD. Unfortunately, treatments aimed at improving clinical symptoms or markers of brain function generally have been of limited value. Physical exercise is often recommended for people diagnosed with MCI, primarily because of its widely reported cognitive benefits in healthy older adults. However, it is unknown if exercise actually benefits brain function during memory retrieval in MCI. Genetic variations in the large-conductance, voltage- and calcium activated potassium channels (BK channels) have been recently implicated in mental retardation, autism and schizophrenia which all come along with severe cognitive impairments. In the present study we investigate the effects of functional BK channel deletion on cognition using a genetic mouse model with a knock-out of the gene for the pore forming α-subunit of the channel. We tested the F1 generation of a hybrid SV129/C57BL6 mouse line in which the slo1 gene was deleted in both parent strains. Readers recruit information from both general world knowledge and episodic memory during reading comprehension. The present experiment used eye tracking to investigate the time-course of how these two sources of memory interact. Participants read passages describing scenarios in which an actor performs a role that was either scriptually appropriate or inappropriate. Half the passages containing the inappropriate role-filler were preceded by an episodic justification for this scriptural violation. Using the same paradigm, Cook and Myers (2004) found context had an early influence on the integration of the role-filler, but world knowledge showed a later effect in the post-target region. The …
