Preweanling Exposure To Selective Catecholamine Transporter Inhibitors Differentially Effects Morphine-Induced Antinociception In Adulthood, Joseph Marco Valentine

The purpose of this project is to determine how early methylphenidate exposure alters opioid receptor functioning in adulthood. To this end, we will administer selective monoamine transporter inhibitors for dopamine and norepinephrine (GBR-12904 and atomoxetine) during preweanling period and assess opioid function in adult rats by testing morphine-induced antinociception using the hot-plate and tail-flick tasks. In order to assess the effects of methylphenidate use on brain functions during development, investigations have been conducted in rats using ages roughly analogous to early and late childhood.

This study sought to determine the individual and combined contribution of adolescent and adult nicotine exposure on METH self-administration and METH-seeking behavior in male rats. Nicotine is a commonly abused substance among adolescents and is believed to be a "gateway" to other drugs of abuse. In support of the gateway hypothesis, cigarette smoking during adolescence is related to increased methamphetamine (METH) use in adulthood.

The purpose of this study was to determine if Fischer 344 (F344) rats, a strain which naturally shows deficits in social communication during adolescence, could be used as a model of the social communication deficits observed in autism. Thus, the goal was to determine if F344 rats would also show fewer emissions of isolated-induced ultrasonic vocalizations (an early onset form of social interaction) as compared to SD rats. Behavioral sensitization is defined as an increase in behavioral responding that occurs after repeated administration of a psychostimulant. Behavioral sensitization is an important component of the addiction process that is mediated by different brain regions. In this thesis, lidocaine was used to determine those brain areas mediating one-trial behavioral sensitization in preweanling rats.

The purpose of this thesis was to: a) investigate the effects of intrastriatally administered N-ethoxycarbonyl-2ethoxy-1, 2-dihydroquinoline (EEDQ) on nonselective dopamine agonists (NPA) -induced locomotor activity in preweanling and adult rats; b) determine which dopamine receptor subtype (D1- or D2-like) is responsible for modulating EEDQ's paradoxical behavioral effects in preweanling rats; and c) examine the magnitude of EEDQ-induced D1- and D2-like receptor inactivation in both adult and preweanling rats. Subjects were 252 male and female rat pups of Sprague-Dawley descent (Charles River Laboratories, Hollister, CA, USA), born and raised at California State University, San Bernardino (CSUSB).

The purpose of this thesis was to determine the persistence of one-shot context-specific and context-independent behavioral sensitization by administering a single pretreatment injection of cocaine or saline in either the home cage or a novel test chamber on PD 19 or PD 79. In order to examine the persistence of conditioned activity, adult rats were given an injection of saline after 1, 3, or 5 abstinence days. Preweanling rats were given an injection of saline after only 1 abstinence day.

This experiment examined the effects of environmental enrichment on performance in an attention-dependent learning task in rats with selective lesions of the cholinergic nucleus basalis magnocellularis (NBM). The findings demonstrate that environmental enrichment can enhance attention-dependent learning performance in normal animals as well as attenuating NBM lesion-induced impairments otherwise seen following rearing in standard conditions.

Major depression is a common problem in adolescents. Unfortunately, one of the most effective antidepressants in adults, the selective serotonin reuptake inhibitor (SSRI) paroxetine, is not clinically effective in pediatric populations. There are many variables that increase the risk of suicide including genetic, social, psychological, and biological factors, making the evaluation of the effect of SSRIs on suicidal behavior difficult to assess in human adolescents. The goal of the present investigation was to compare the effects of paroxetine and fluoxetine in adolescent rats on markers associated with depression and suicidal behavior in humans.

Methylphenidate is the preferred treatment for attention-deficit hyperactivity disorder in preschool-aged children, even though few human studies have assessed the long-term consequences of early methyphenidate use. In rats, exposure to methylphenidate during a developmental period that approximates early childhood increases morphine-induced antinociception in adulthood. However, because morphine activates three distinct opioid receptors it is unknown which receptor is resonsible for this methylphenidate effect. The purpose of the present study was to assess the long-term effects of early methylphenidate exposure on DAMGO- and U50, 488-induced antinociception, a mu-opioid and kappa-opioid selective agonist.

Infusions of the nonselective dopamine agonist R( - ) propylnorapomorphine (NPA) into the dorsal striatum of preweanling rats atteuates kappa opioid-mediated locomotor activity. The purpose of this thesis was to ascertain which dopamine receptor subtype (D1 or D2) is responsible for this effect. It was hypothsized that a) D1, but not D2, receptors in the dorsal striatum underlie NPA's ability to attenuate the kappa opioid-mediated locomotor activity of preweanling rats, and b) microinjectng SKF38393 (a D1 agonist), and not quinpirole (a D2 agonist), would attenuate kappa opioid-mediated locomotion.

The purpose of the current study was to determine if rat pups treated with amphetamine (AMPH) during a critical time of hippocamal development would exhibit deficits in spatial learning and memory when tested in the Morris water maze during adolescence. Exposure to amphetamine during early postnatal development induces long-term reductions in protein kinase A (PKA) activity. Because PKA activity is known to regulate the production of brain derived neurotrophic factor (BDNF), and reductions in BDNF are associated with memory deficits, it was hypothesized that early exposure to amphetamine would lead to declines in both BDNF levels and memory performance. The purpose of the study was to investigate the role of metabotropic glutamate receptors (mGluR) in the ontogeny of amphetamine-induced behavioral sensitization. Eleven-day-old rat pups were given five daily bilateral infusions of the mGluR antagonist, (RS)-methyl-4-carboxyphenylglycine (MCPG) followed by a systemic injection of amphetamine and locomotor activity was measured. It was hypothesized that rats receving amphetamine pretreatment and an amphetamine challenge would exhibit a significant increase in activity, indicating short-term behavioral sensitization. As predicted, repeated amphetamine administration during the pretreatment phase produced progressively enhanced locomotor activity, indicating the development of behavioral sensitization. Biconditional Discrimination Learning In Rats With 192 Igg-Saporin Lesions Of The Nucleus Basalis Magnocellularis, Michael Ryan Kitto

The experiment tested the hypothesis that 192 IgG-saporin lesions of the nucleus basalis magnocellularis (NBM) in rats would impair performance in a biconditional visual discrimination task, which requires configural association learning. Experiment used 22 male Long-Evan rats (Harlan Sprague-Dawley). Behavioral testing was conducted in two identical T-mazes. Rats were randomly assigned to either a bilateral 192 IgG-saporin lesion group (n = 10) or to a control group (n = 12). Results support the hypothesis that NBM is critically involved in configural but not simple association learning and suggest that NBM may be involved more generally in cognitive flexibility.

The experiment tested the hypothesis that the acetylcholine (ACh) release in the rat auditory cortex is greater in rats undergoing auditory classical conditioning compared to rats in a truly random control paradigm where no associative learning takes place and that this is mediated by prefrontal afferent projections influencing the nucleus basalis magnocellularis (NBM), which in turn modulates ACh release in neocortex. The neurochemical effects of partial D2-like agonists (i.e., terguride) to alter striatal DOPA accumulation under high and low dopaminergic tone was examined in preweanling rats. The results indicate that terguride has agonist-like (quinpirole-like) effects under a low dopaminergic tone and antagonist-like (haloperidol-like) effects under a high dopaminergic tone during the preweanling period.

Examines whether the reinforcing properties of drugs of abuse were altered in adulthood by methylphenidate, more commonly known as Ritalin. Subjects were 108 rats of Sprague-Dawley descent (Harlan). Methylphenidate, or saline was administered daily to the subjects from the postnatal period (11-20 days old). The rats preference for morphine during early adulthood was measured using conditioned place preference. The number of dopamine D₂ receptors was measured in each rat and the correlation between receptor number and morphine preference was determined. In order to assess flexibility in acquiring and using conflicting response rules, rats with selective lesions of the NBM or sham-lesion controls were subjected to serial reversal training in a simple operant discrimination paradigm. The NBM lesion group did not differ from the control group in acquisition of the original rules; the NBM lesion group required more time to master the changes in rules in the first reversal, but not in subsequent reversals.

It was hypothesized that 192 IgG-saporin lesions of the basal forebrain cholinergic system (BFCS) would disrupt differential reinforcement of low rate (DRL) learning in an uncued DRL task, but would not impair acquisition and performance in the cued version of the task. Results suggest that BFCS lesions impair vigilance to the external cues despite continued practice in the cued DRL, whereas continuous attention to internally produced cues recovers with extended practice in the uncued DRL.

The purpose of this thesis was to determine the neuronal circuitry mediating U50,488-induced locomotion in preweanling rats. To this end, preweanling rats received bilateral electrolytic lesions of the ventromedial thalamus or superior colliculus and, two days later, the same rats received a challenge injection of U50,488. It was predicted that bilateral lesions of the ventromedial thalamus or superior colliculus would attenuate the U50,488-induced locomotor activity of 18-day-old rats.

The ability of the indirect dopamine agonist, amphetamine, to produce behavioral sensitization was assessed in adult D₁A-deficient and wild-type mice. It was originally predicted that : 1) dopamine (DA) D₁-like receptors are necessary for the occurrence of short- and long-term amphetamine-induced behavioral sensitization, 2) DA D₁-like receptors are necessary for environmental conditioning factors associated with amphetamine-induced behavioral sensitiazation, and 3) DA D₅ receptors are required for amphetamine-induced behavioral sensitization. Locomotor activity and sterotyped sniffing were assessed in each of three experiments.
