Chronic stress increases generalization of fear to non-threatening cues, a key symptom in numerous psychiatric mood disorders. However, the mechanisms through which stress impacts safety learning remain poorly understood. To probe the relationship between stress and safety learning, this dissertation employed multiple behaviors, in conjunction with in-vivo multisite physiology during explicit safety and fear discrimination learning. In Chapter 1, I outline the role of chronic stress in driving neurological adaptations that result in generalized fear and highlight how this occurs because of impaired safety cue encoding. This study utilized a novel design to investigate the sensitivity of D2 dopamine receptors to modulating compounds through multiple exposures over early development of zebrafish larvae. Zebrafish were dosed for 30 minutes from 5-8 days post fertilization (dpf) with 16µ/mol of either a D2 antagonist, haloperidol, or a D2 agonist, quinpirole hydrochloride. Two other groups were then dosed with these compounds from 9-12dpf. The effects of D2 receptor modulation were measured by analyzing motor activity on measures of movement distance, frequency, and velocity. Results indicated that larvae dosed with haloperidol on 5dpf had increased activity after the first dosage, but …

Elephants have shown remarkable olfactory capabilities. Their sense of smell impacts their foraging choices, behavior, and ultimately, survival. Being able to detect a target odor can allow elephants to locate specific resources, identify threats, and find receptive conspecifics. Previous studies have shown that elephants can consistently detect target odors, but have not identified the limits of this detection. Thus, to investigate the extent of elephants’ odor detection capabilities, we tested Asian elephants in a two-step odor discrimination task. This study examines two personality traits: exploration and neophobia, which could influence human-elephant conflicts. Thirty-one semi-wild elephants were tested over two trials using a custom novel puzzle tube containing three tasks and three rewards. Our studies show that elephants do vary significantly between individuals in both exploration and neophobia.

This paper focuses on the ability of the mimic octopus Thaumoctopus mimicus to be operantly conditioned to an auditory stimulus. The octopus is known to be the most advanced of the invertebrates and has learning abilities that are comparable to vertebrates in spite of their differences in brain structure. These animals have been shown to react to visual and tactile stimuli and can be operantly conditioned to perform behaviors to obtain a food reward. The developing brain is susceptible to extensive neurotoxicity following exposure to sedative/anesthetic drugs (SADs). Every year hundreds of thousands of children around the world are exposed to SADs with no viable non-neurotoxic agents approved for clinical use. Allopregnanolone (AlloP) has well-established sedative effects in adults and neonates. AlloP and many SADs produce sedation/anesthesia through allosteric modulation of GABAA receptors, which is one of two principal mechanisms behind SAD-induced neurotoxicity. Evidence suggests AlloP has the unique capacity to regulate key apoptotic factors in adults and is widely involved with critical stages of neurodevelopment, indicating this neurosteroid might serve as a …

Objective: Novel drug 419 was examined to see the effect it has in vivo mice and rats on alcohol consumption, nicotine locomotor sensitization, and conditioned place preference (CPP) models regarding behavioral tests on dopamine transporter activity.
Methods: Mice and rats were used to see how they react to the drug 419 and control vehicle, in each of the models. The animals were assessed to pre- and post- drug administration of novel drug 419. We examined each model to see the association between how drug 419 will help with treating drug abuse.

Traumatic Brain Injury (TBI) is defined as an impact to the head, penetration of the skull, or rapid deceleration of the skull, resulting in an alteration of brain function or neurological deficit. Cognitive deficits are common following TBI and often go unresolved due to a lack of effective treatments. These deficits often perseverate into the chronic post injury phase, so the development of rehabilitative strategies is imperative. In recent years both philosophers and scientists have asked whether or not our current kinds of mental disorder—e.g., schizophrenia, depression, bipolar disorder—are natural kinds; and, moreover, whether or not the search for natural kinds of mental disorder is a realistic desideratum for psychiatry. In this dissertation I clarify the sense in which a kind can be said to be “natural” or “real” and argue that, despite a few notable exceptions, kinds of mental disorder cannot be considered natural kinds. Furthermore, I contend that psychopathological phenomena do not cluster together into kinds in the way that paradigmatic natural kinds (e.g., chemical …

Mentalization, the ability to reflect upon and revalue emotions and thoughts, is of increasing interest to clinicians and researchers alike due to its predictive and moderating effects for adaptive behavior and positive outcomes. Mentalized affectivity, reflecting upon and revaluing one’s emotions and emotional process, has only recently been made measurable through the Mentalized Affectivity Scale (Greenberg et al., 2017). The present study sought to investigate the unexplored relationship between mentalization, more specifically mentalized affectivity, selfacceptance, and self-criticism. Risky choice can be defined as choice for a larger, uncertain reinforcer over a smaller, certain reinforcer when choosing the smaller alternative maximizes reinforcement. Risky choice is studied using various procedures in the animal laboratory; one such procedure is called probability discounting. There are many variables that contribute to risky decision-making, including biological and pharmacological determinants. The present study assessed both of these variables by evaluating dose-response effects of d-amphetamine on risky choice of Lewis (LEW) and Fischer 344 (F344) rats. Captive facilities housing chimpanzees are required to provide adequate care and provisions such as dietary, social, and environmental enrichment to promote the psychological well-being of the apes in their care. Chimpanzees are social creatures and changes in groups as well as relocation to a facility with new social partners, can impact each individual chimpanzee’s welfare. By tracking each chimpanzee’s activity budgets, managers can assess welfare and make improvements or adjustments if necessary. The model of basal ganglia function proposed by Albin, Young and Penney (1989) describes two anatomically independent motor pathways, the direct and indirect. However, under normal conditions striatal dopamine (DA) is required for the expression of motor behavior, and DAergic control of the two pathways (via D1 and D2 receptors, respectively) is dependent on co-activation. We tested for a possible breakdown of D1/D2 synergism using transgenic R6/1 mice bearing the human huntingtin allele (Htt). Bottlenose dolphins are an important species of interest because they possess a variety of abilities that are relatively rare in the animal kingdom, one being complex acoustic communication. Signature whistles - distinctive calls that are unique for each individual – are one of the most studied call types, but we know little about how these calls are used in various contexts, such as during an introduction. Ruffed lemur (Varecia spp.) color vision research was conducted using a multidisciplinary approach: psychophysics, genetic analysis, technology, and animal training. The behavioral manifestation of Varecia spp. trichromacy was shown using a touchscreen apparatus (SMARTA). Trichromats performed better than dichromats when discriminating red from green (G2 = 78.10, p < 0.001).

Feather plucking (FP) is a maladaptive behavior observed in captive avian species. This self-injurious behavior results in damage to and removal of feathers and skin tissue, resulting in animal welfare and financial consequences. The etiology and maintenance of FP have been hypothesized through medical and environmental processes, yet a definitive solution has not been found. The current study investigated the environmental variables maintaining the FP of a Black Vulture (Coragyps atratus), as well as evaluated a function-based treatment for this behavior. This study was conducted to investigate whether the number of traumatic events an individual has previously experienced influences that individual’s physiological and psychological reactions when exposed to a negative affective stimulus followed by a mindfulness-based stress reduction (MBSR) intervention. Twenty-eight participants were placed into intact quasi-experimental groups based on their scores on the Traumatic Life Events Questionnaire (TLEQ). The negative affective stimulus consisted of a series of photos bearing negative affective valence. Contemporary professional applied animal behavior management employs a diagnostic and treatment approach for unwanted pet behavior that incorporates ethology-based causal factors. As in behavior therapy for humans, behavioral assessment includes descriptive functional analysis when possible to determine relevant contingency variables. But this information is then considered in the context of an appropriate motivational classification, such as social or prey-directed motivation or fear-motivated, and treatments are designed accordingly. A different view is that behavior can be treated effectively without a presumption of the motivation through analysis of the manifestation of the behavior itself. This study examined the effects of proton and gamma radiation on a dopamine-dependent sensory gating mechanism: prepulse inhibition of the startle reflex. The purpose of this research was to describe potential hazards of proton radiation to the central nervous system in long-term space fight and for proton therapy patients. Results from previous research indicate that startle behavior is modified by proton and iron radiation. Learned helplessness is a psychological concept that describes the subsequent escape-avoidance behavior of experimental subjects who are exposed to uncontrollable stressors. Subjects after treatment are unable to respond in situations where escape is possible. Two major theorists, Seligman and Weiss, have proposed explanations concerning the phenomena. Seligman feels that the subject is unable to respond due to the fact that the exposure to an uncontrollable stressor has caused him to learn to be "helpless". Twelve adult male rats were given free access to a two bottle free-selection of tap water or a 10% ethanol/tap water solution in the home cage for a period of 140 consecutive days with the alcohol consumption data in the form of mg absolute ETOH/g subject weight collected daily. This study incorporated three stages. Phase I was a 40 day period of alcohol acclimation with alcohol consumption during the second 20 days constituting the baseline consumption rate. State-Dependent Learning As A Function Of The Temporal Relationship Between Noncontingent Footshock And Electroconvulsive Shock, T. Scott Shutt

Rats were given a noncontingent footshock followed at various intervals by electroconvulsive shock. Twentyfour hours later they were trained on a non-shock passive avoidance task and tested for retention 72 hours later. When the interval between NCFS and ECS was short the animals showed an amnesia which reduced as the interval was lengthened. An interval of .5 seconds produced the most pronounced amnesia and intervals greater than 10 seconds produced virtually no amnesia. The results were consistent with a state dependent retrieval failure hypothesis.

Forty-eight rats were divided into 8 groups; four were maintained under normal conditions and the other four under sensory deprivation and tested at o, 3, 6, and 9 days after condition institution. The response was placing the head through a hole in the operant chamber and the stimulus (0, 1.2, 4, or 12 Vac) was contingent upon the response. Analysis ot variance disclosed significant differences (p<.01) between the deprived and the non-deprived groups at days 6 and 9; and a significant interaction between deprivation condition and time of test. No differences were shown between the stimulus intensities indicating that the stimulus did not have a reinforcing effect.

The Effects Of Cs Habituation And Duration Of The Directional Modification Of The Cardiac Response In The Guinea Pig (Cavia Procellus), Ray Charles Larson

The present experiment examined the effects of CS habituation and two different levels of CS duration (60 versus 6 sec) on the directional modification of the conditioned cardiac response in the guinea pig. The results indicated that the only reaction to the CS, regardless of prior habituation or CS duration, was a decelerative conditioned HR response. However, habituation to a CS prior to classical aversive conditioning was found to attenuate the magnitude of the bradycardia CR that developed. The bradycardia CR was discussed in terms of being a cardiac component of the orienting response.
