Neurobiological Mechanisms Of Cognitive Processing Therapy For Post-Traumatic Stress Disorder: A Brain Network Approach, Tessa Vuper

Psychotherapy research is increasingly targeting both psychological and neurobiological mechanisms of therapeutic change. This trend is evident in and applicable to post-traumatic stress disorder (PTSD) treatment research given the high nonresponse rate of individuals with PTSD who undergo cognitive-behavioral therapy (CBT). A review of the literature investigating neurobiological mechanisms of CBT in PTSD reveals inconsistent results that fail to fully support dual process or learning models of CBT effects in the brain. However, network-based models of psychopathology provide a new framework from which to understand both mental disorder symptoms and therapeutic mechanisms. The developing brain is susceptible to extensive neurotoxicity following exposure to sedative/anesthetic drugs (SADs). Every year hundreds of thousands of children around the world are exposed to SADs with no viable non-neurotoxic agents approved for clinical use. Allopregnanolone (AlloP) has well-established sedative effects in adults and neonates. AlloP and many SADs produce sedation/anesthesia through allosteric modulation of GABAA receptors, which is one of two principal mechanisms behind SAD-induced neurotoxicity. Evidence suggests AlloP has the unique capacity to regulate key apoptotic factors in adults and is widely involved with critical stages of neurodevelopment, indicating this neurosteroid might serve as a …

Kappa opioid receptors are colocalized with dopamine receptors and are frequently associated with depression, anxiety, stress, and drug use and abuse. KOR antagonism has widely been considered anti-depressive and anxiolytic in animal models while agonism is pro-depressive and anxiogenic. However, recent findings suggest that SalvA, a natural kappa agonist derived from a plant in the mint family, can reduce depressive- and anxiety-like behavior in rats. The current study investigated the effects of chronic mild stress on behavior, attenuation by SalvA, and sex differences. 52 Long-Evans rats, 26 males and 26 females, were exposed to six weeks of CMS. The feeling of learned helplessness has been associated with prolonged stress and trauma. Additionally, many previous studies have examined the relationship between stress and decreased feelings of control, such as self-efficacy and locus of control. However, these forms of control have been primarily self-reported. The present study aimed to investigate the relationships among learned helplessness, chronic stressors, and self-agency using a computer-based task. We also measured heart rate variability (HRV) during the self-agency task to assess psychophysiological correlates of these variables. Clinical research has revealed aberrant activity and connectivity in default mode (DMN), frontoparietal (FPN), and salience (SN) network regions in major depressive disorder (MDD). Recent functional magnetic resonance imaging (fMRI) studies suggest that variability in brain activity, or blood oxygen level-dependent (BOLD) signal variability, may be an important novel predictor of psychopathology. Racial disparities in physical and mental health are ongoing and well-documented problems in the United States. Black Americans, compared to White Americans, have higher risk of obesity, heart disease, cancer, depression, and substance abuse. Research suggests prejudice and discrimination play a role in racial health disparities. Everyday discrimination is considered a chronic, psychosocial stressor that impacts the health of Black Americans. A biopsychosocial approach states there are various factors that contribute to the pathway from discrimination to disease and proposes complex relationships that explain effects of racial discrimination on health.
