Extraction Of Challenging Forensic Samples Using The Microgem Dna Extraction Kit, Falyn R. Vega

In forensic science, DNA extraction can be a tedious and resource-intensive process. Extraction with Proteinase K is an industry standard but has its drawbacks, such as requiring multiple ionic detergents and washing steps. MicroGEM has developed a new enzyme called forensicGEM that is temperature-dependent and compatible with mesophilic enzymes, offering complete DNA extraction in about 20 minutes in a single tube, limiting contamination, loss of sample, and working time. ForensicGEM can extract DNA from highly degraded samples, potentially leading to more complete STR profiles. Prior to isolating DNA, it is necessary to remove any biological materials that may be present on commingled remains. Manual maceration of the outer surface is time-consuming. This study will address this issue by developing a simple method for processing bone samples prior to DNA isolation. A liquid-based technique was applied to macerate bone fragments by incubating bone fragments in an enzyme solution, removing potential contaminants, followed by a bone tissue disruption. Semi-Quantitative Detection Of Signature Peptides In Body Fluids By Liquid Chromatography Tandem Mass Spectrometry (Lc-Ms/Ms), Tebah N. Browne

This study covers a modified semi-quantitative approach for liquid chromatography mass spectrometry (LC-MS/MS) signature peptide detection for body fluid identification. Peptide concentrations were measured based on synthetic peptide standards. Samples were processed with varying trypsin digestion and purification protocols, including a three-hour trypsin digestion and Microcon membrane filtration. The Microcon filtration method generates polymerase chain reaction (PCR) compatible DNA and peptide fractions that can be analyzed without any further purification. Preliminary validation tests covered stains on different substrates, semen/ saliva mixtures, minimum sample volume, and repeatability. In this work we demonstrate the power of compound-specific isotope analysis (CSIA) to analyze proteinaceous biological materials in three distinct forensic applications, including: 1) linking necrophagous blow flies in different life stages to their primary carrion diet; 2) identifying the harvesting area of oysters for food authentication purposes; and 3) the ability to predict biometric traits about humans from their hair.

In the first application, we measured the amino-acid-level fractionation that occurs at each major life stage of Calliphora vicina (Robineau-Desvoidy) (Diptera: Calliphoridae) blow flies. Adult blow flies oviposited on raw pork muscle, beef muscle, or chicken liver. Larvae, pupae …

Numerous terrorist groups in the Middle East have been recently making detonators for IEDs (Improvised Explosive Devices) using items as basic as flip flops. Because of the semi-porous nature of flip flops, military forensic response teams have been unable to find an effective method through which they can identify, process, and lift latent fingerprints from the surface of a flip flop. The lampricide 3-trifluoromethyl-4-nitrophenol (TFM) has been used for decades to control invasive sea lamprey (Petromyzon marinus) populations in the Great Lakes, normally with little harm to other fish populations. However, due to the nature of lampricide treatments and the chemical properties of TFM, adverse effects to non-target fishes and non-target mortality occasionally occur. However, investigations of non-target mortality can be complex, particularly if fish deaths are not noticed immediately, due to decomposition of the carcasses. The following document is a capstone thesis project focusing on the unique experiences of graduate forensic coaches through lens of liminality, a performance theory used to describe a sense of being between social identities when going through a rite of passage. The author contends that this liminal experience has unique characteristics which are important to consider in the context of identity and forensic culture. In order to gather data, the author utilized qualitative, semi-structured interviews with current graduate forensic coaches. In forensic casework analysis it is necessary to obtain genetic profiles from increasingly smaller amounts of biological material left behind by perpetrators of crime. The ability to obtain profiles from trace biological evidence is demonstrated with so-called ‘touch DNA evidence’ which is perceived to be the result of DNA obtained from shed skin cells transferred from donor to an object or person during physical contact. Introduction:Estimation of facial soft tissue appearance from human skeletal remains is often necessary in forensic identification. This process has been referred to as facial reconstruction or facial approximation and is a branch of forensic facial anthropology. Original methods for facial approximation originated in nineteenth century Europe and consisted of artists shaping clay over skull models using average soft tissue depths measured in cadavers. For decades college students have been competing in forensic activity (forensics), spending countless hours researching, writing, and performing speeches. Forensic scholars have never created an overarching definition of what it means to be successful in forensics. A survey was created and sent out on the individual events listserv, asking competitors and coaches to define success. Analysis reveals both competitors and coaches believe success in forensics is a combination of competitive achievement, building communication skills, and personal growth.
