The research presented in this dissertation is focused on the construction of complex molecular structures on planar gold and silicon dioxide surfaces using a variety of surface modification techniques, along with thorough surface characterization at each modification step. The dissertation is structured into six separate chapters. In Chapter 1, an introduction to the importance and implications of molecular level surface modification, commonly employed surface modification methods, and available surface characterization techniques is presented. Chapter 2 shows applications of novel methodologies for the functionalization of gold surfaces using alkane dithiol self-assembled monolayers and thiol-ene click chemistry. The focus of my dissertation work was to study the basic mechanistic insights of gold-catalyzed reactions. Although the various mechanistic pathways of gold catalysis are better understood nowadays, numerous questions still remain unanswered concerning the nature of deactivation of the catalyst's active species, high resistance towards protodeauration, and how we can solve these problems to improve the efficiency of gold catalysis. Determination Of Thiamine In Solution By Uv-Visible Spectrophotometry: The Effect Of Interactions With Gold Nanoparticles, Michael Vincent Zielinski

The method presented here provides the foundation for a simple and selective qualitative determination of thiamine in solution. Gold nanoparticles in the presence of thiamine results in the formation of a secondary peak in the absorbance spectrum of the mixture. This peak can be used as an indicator of thiamine, which is useful for the qualitative analysis of solutions, and may provide an alternative to other methods for evaluating thiamine in blood and other biological systems. The focus of my dissertation work is to study the gold-catalyzed intramolecular and intermolecular cyclizations involving oxonium intermediates towards the application of synthetically interesting frameworks under ambient conditions and developing a rational approach for the effective catalyst design in gold catalysis. We explored the goldcatalyzed oxygen-transfer reactions of 2-alkynyl-1,5-diketones or 2-alkynyl-5-ketoesters to furnish five-membered rings bearing a quaternary carbon tethered to a carbonyl group. The detailed mechanistic investigation on the newly proposed intramolecular [4+2] cycloaddition mechanism was performed by means of isotopic experiments and quantum chemical calculations. "This thesis study focuses on synthesizing and characterizing gold-magnetite optically active magnetic nanoparticle and its conjugation with biomolecules for biomedical applications, especially magnetic fluid hyperthermia treatment for cancerous tissue. Gold nanoparticles have already displayed their potential in the biomedical field. They exhibit excellent optical properties and possess strong surface chemistry which renders them suitable for various biomolecule attachments. Studies have showed gold nanoparticles to be a perfect biocompatible vector. However, clinical trials for gold mediated drug delivery and treatment studied in rat models identified some problems.
