Extended metal atom chains (EMAC) are multimetallic complexes that feature a linear metal chain with metal-metal bonds surrounded by stabilizing ligand molecules. When EMACs are formed with paramagnetic metal ions, unique magnetic properties can be derived from direct metal to metal communication. The challenge is that EMAC complexes are challenging to synthesize due to the inherent weakness of most M-M bonds and the specificity of the ligands required to stabilize EMACs. Peptide nucleic acids (PNAs) are oligonucleotide analogues in which the sugar-phosphate backbone has been replaced by a pseudopeptide skeleton. Since PNAs use the natural nucleobases (Adenine, Thymine, Cytosine, Uracil, and Guanine) found in either DNA and/or RNA, they are able to hybridize according to Watson-Crick base-pairing to form duplexes. PNA is a promising therapeutic agent because they can function as antigene or antisense chemical agents. To further enhance their utility, we aim to incorporate a photoswitchable moiety using azobenzene. Here, we report the results of the synthesis and purification of a photoswitchable 11 mer PNA along with initial characterization efforts.

The hydroamination of dimaleimides with diamines has been accomplished to access novel polymers. Various dimaleamides and dimaleimides were synthesized from commercially available diamines and maleic anhydride. The resulting dimaleamides were then ring-closed to their respective dimaleimides either directly in the presence of acid or via the synthesis of mixed amide-esters which then ring close in the presence of acid under heat. Hydroamination of the dimaleimides with piperazine was accomplished in bulk at room temperature. The synthesis of palladium nanoparticles (Pd NPs) using materials-directed peptides is a novel, nontoxic approach which exerts a high level of control over the particle size and shape. This biomimetic technique is environmentally benign, featuring nonhazardous ligands and ambient conditions. Nanoparticles are extremely reactive catalysts, boasting a large surface-to-volume ratio when compared to their bulk counterparts. The hydroamidation reaction is an attractive methodology for the construction of larger functional molecules. In our research, a baseline reactivity was established for the hydroamidation reaction using a set of N-alkyl and N-aryl (trifluoro)acetamides with methyl acrylate and acrylonitrile, various bases and varying reaction conditions. The combination of alkenes, acetamides, and trifluoroacetamides with base was crucial for reaction rate. N-aryl trifluoroacetamides exhibited near quantitative conversions in neat alkene with Diazabicycloundecene (DBU) as base. The less acidic N-aryl acetamides performed much faster in the presence of KOtBu and conversions of >95% were accomplished regularly but contained a large presence …

Structure activity relationship (SAR) studies are performed in order to identify the core structure that is responsible for the biological activity of an organic molecule. Recently, we have synthesized a drug prototype which contains several functional groups, such as an alcohol, an ester, a fluorine, and an aromatic ring. While studying in vivo toxicity of this molecule in zebrafish (Danio rerio) embryo, we observed that it has a unique biological activity that causes a sudden inactivity in embryo movement. In this study, we examine the dietary effects of 2-aminoanthracene (2AA), a polycyclic aromatic hydrocarbon (PAH), in rats, specifically as it relates to oxidative stress. Oxidative stress refers to the imbalance of reactive oxygen species (ROS) and antioxidants in living systems; it has been shown to contribute to type-1 diabetes. Vegetable oil-based polymers are a popular alternative due to their low-cost and low toxicity. The carbon-carbon double bonds in the oil are ideal reactive sites for a free radical polymerization. Cellulose is a naturally occurring polymer. Modifying cellulose prior to making composites with tung oil allows a hydrophilic material to mix better with a hydrophobic resin. The introduction of fluorine uniquely affects the biological properties of organic molecules such as making them more bioavailable, lipophilic and metabolically stable, and possibly increase the strength of a compound’s interactions with a target protein. Approximately 30% of agrochemicals and 20% of pharmaceuticals contain fluorine, including commonly prescribed drugs such as Lipitor, Lexapro and Prozac. Hemoglobin based oxygen carriers (HBOCs) hold promise as an effective emergency treatment of severe traumatic brain injuries (TBI). In the latest generation of HBOCs, polynitroxyl-pegylated hemoglobin (PNPH), cell-free hemoglobin is modified with TEMPO and PEG which reduce the toxicities associated with earlier generations of HBOCs. In our efforts to optimize the economic and therapeutic impacts of PNPH’s we have synthesized polydimethylaminoethyl methacrylate (poly-DMAEMA) under controlled living conditions via reverse addition-fragmentation chain transfer (RAFT) polymerization. The iodine-initiated hydration of internal alkynes is a novel, green, and inexpensive synthetic pathway in comparison with the commonly used transition metal catalyzed reactions. Therefore, research into the selective recognition of anions and heavy metal cations has acquired much attention. One approach involves the use of chemosensors. In an attempt to create more effective chemotherapeutic compounds, the naphthoquinone adduct, 12,13-dihydro-N-methyl-6,11,13-trioxo-5H-benzo[4,5]cyclohepta [1,2 b]naphthalen-5,12-imine (hereafter called TU100) was synthesized. Inspired by its unique and novel mechanism of action, a series of structural derivatives were synthesized to explore structure-activity relationships. The analogues exhibited different cytotoxicity profiles, revealing the indicated regions are involved in cell death induction. Furthermore, the analogues had dramatically different effects on cellular ATP production, suggesting different molecular targets. Synthesis, biological activity, and SAR study of these analogues will be revealed.

During a traumatic brain injury (TBI) sustained in combination with hemorrhagic shock the brain undergoes anoxic conditions resulting from swelling and blood loss. Traditional intravenous solutions are used to replace blood volume, but are unable to replace the blood’s oxygen carrying capacity. A proposed treatment based on isotonic solutions containing hemoglobin-based oxygen carriers (HBOC’s) which were intended to provide readily available oxygen was developed. However, cell-free hemoglobin was shown to have excessive oxidative potential. Polynitroxylated Pegylated Hemoglobin (PNPH) was developed to address this shortcoming.
