Quantitative Analysis Of The Proteomic Selectivity Of Acidic Reductive Alkylation Of Peptides, Connor Jewell May 2023

Quantitative Analysis Of The Proteomic Selectivity Of Acidic Reductive Alkylation Of Peptides, Connor Jewell

Honors Scholar Theses

Proteome quantification is a complicated process which takes on many methods, usually involving peptide derivatization and complex mass spectral analysis. Scores of techniques have been utilized to address issues of incomplete quantification. The implementation of IPTL labeling, or isobaric peptide terminal labeling, is a recent addition to the literature, which exploits the supposedly selective labeling of N-termini of tryptic peptides from the C-termini of lysine residues due to differences in basicity. This method has been shown to improve quantitation by reducing spectral complexity and provide more accurate and reproducible results on peptide abundance. Investigations into the selectivity of terminal derivatization …

The Concentration Of Heavy Metals In Lincoln Nebraska's Tap Water, Evan Neuhaus Mar 2023

The Concentration Of Heavy Metals In Lincoln Nebraska's Tap Water, Evan Neuhaus

Honors Theses

Heavy metals are commonplace in nature, and are typically found in scarce quantities in all drinking sources. However, in high enough concentrations, they can have a myriad of negative health effects. This study seeks to find the concentrations of such metals in the Lincoln water supply utilizing the resources of the Water Center at the University of Nebraska at Lincoln. To test for discrete concentrations of trace elements, a method of using Inductively Coupled Plasma Mass Spectrometry (ICP-MS) was implemented. ICP-MS uses plasma to ionize and atomize a sample which is then filtered into a series of cones into a …

Evaluation And Improvement Of Capillary Microextraction Of Volatiles Coupled To Gas Chromatography-Mass Spectrometry For The Analysis Of Ignitable Liquid Residues In Fire Debris, Michelle Nikole Torres

FIU Electronic Theses and Dissertations

A key aspect of fire debris analysis is the ability to extract the remnants of an ignitable liquid from a matrix with a high degree of reliability and sensitivity. Although there are several robust, standardized methods, there is no single technique universally applicable to casework. In this work a novel extraction technique – Capillary Microextraction of Volatiles (CMV) – has been applied, for the first time, for ignitable liquid residue (ILR) extraction. A 20-minute dynamic sampling laboratory protocol from traditional 1 L paint cans was established and optimized based upon ASTM guidelines. The development of new adsorption phases for CMV …

Elemental Analysis Of Adhesive Tapes By Laser-Based Methods, Claudia Martinez Lopez Oct 2018

Elemental Analysis Of Adhesive Tapes By Laser-Based Methods, Claudia Martinez Lopez

FIU Electronic Theses and Dissertations

Adhesive tapes are a common type of evidence involved in violent crimes and national security threats. This research evaluated the utility of LA-ICP-MS and LIBS for the characterization of the trace elemental signature in adhesive tapes for forensic comparisons. LA-ICP-MS and LIBS methods were developed, for the first time, for the qualitative and quantitative analysis of adhesive tapes.

The backings of 90 black electrical tapes, previously characterized by conventional techniques (physical examination, IR, Py-GC-MS, and SEM-EDS), were analyzed by LA-ICP-MS to evaluate the ability of the technique to discriminate samples originating from different sources and to associate pieces of tapes …

Measurement Of Antioxidant Activity Toward Superoxide In Natural Waters, D. Whitney King, Emma Berger, Zachary Helm, Eleanor Irish, Kenneth Mopper

Chemistry & Biochemistry Faculty Publications

Antioxidants are a class of molecules that provide a protective function against reactive oxygen species (ROS) in biological systems by out competing physiologically important molecules for ROS oxidation. In natural waters, the reactivity of antioxidants gives an estimate of oxidative stress, and may determine the reactivity and distribution of reactive oxidants. We present an analytical method to measure antioxidant activity in natural waters through the competition between ascorbic acid, an antioxidant, and MCLA, a chemiluminescent probe for superoxide. A numerical kinetic model of the analytical method has been developed to optimize analytical performance. Measurements of antioxidant concentrations in pure and …

The Creation And Evaluation Of Novel Canine Training Aids For Cocaine Using Molecularly Encapsulated Sol-Gel Polymers And An Investigation Of Canine Field Accuracy, Michelle Cerreta Mar 2015

The Creation And Evaluation Of Novel Canine Training Aids For Cocaine Using Molecularly Encapsulated Sol-Gel Polymers And An Investigation Of Canine Field Accuracy, Michelle Cerreta

FIU Electronic Theses and Dissertations

Biological detectors, such as canines, are valuable tools used for the rapid identification of illicit materials. However, recent increased scrutiny over the reliability, field accuracy, and the capabilities of each detection canine is currently being evaluated in the legal system. For example, the Supreme Court case, State of Florida v. Harris, discussed the need for continuous monitoring of canine abilities, thresholds, and search capabilities. As a result, the fallibility of canines for detection was brought to light, as well as a need for further research and understanding of canine detection. This study is two-fold, as it looks to not only …

The Importance Of Context: An Exploration Of Factors Influencing The Adoption Of Student-Centered Teaching Among Chemistry, Biology, And Physics Faculty, Travis J. Lund, Marilyne Stains

Chemistry Department: Faculty Publications

Background: Research at the secondary and postsecondary levels has clearly demonstrated the critical role that individual and contextual characteristics play in instructors’ decision to adopt educational innovations. Although recent research has shed light on factors influencing the teaching practices of science, technology, engineering, and mathematics (STEM) faculty, it is still not well understood how unique departmental environments impact faculty adoption of evidence-based instructional practices (EBIPs) within the context of a single institution. In this study, we sought to characterize the communication channels utilized by STEM faculty, as well as the contextual and individual factors that influence the teaching practices …

Development Of Presumptive And Confirmatory Analytical Methods For The Simultaneous Detection Of Multiple Improvised Explosives, Kelley L. Peters Nov 2014

Development Of Presumptive And Confirmatory Analytical Methods For The Simultaneous Detection Of Multiple Improvised Explosives, Kelley L. Peters

FIU Electronic Theses and Dissertations

In recent years, there has been a dramatic increase in the use of improvised explosive devices (IEDs) due to ease of synthesis and improved controls placed on commercial/military explosives. Commonly used materials for IED preparations include fertilizers and industrial chemicals containing oxidizers such as ClO3-, ClO4-, and NO3-, as well as other less stable compounds, such as peroxides. Due to these materials having a wide range of volatility, polarity, and composition, detection can be challenging, increasing the amount of time before any analytical information on the identity of the explosive can …

The Activity Of Silicon Carbide Particles In Al-Based Metal Matrix Composites Revealed By Scanning Electrochemical Microscopy, David O. Wipf, L. Díaz-Ballote, L. Veleva, M. A. Pech-Canul, M. I. Pech-Canul

College of Arts and Sciences Publications and Scholarship

Scanning electrochemical microscopy (SECM) is used to image variations in electrochemical activity over the surface of an aluminum-based metal matrix composite (MMC) in contact with buffered or unbuffered neutral solutions. The composite consists of an Al - 13.5% Si - 9% Mg alloy matrix and reinforcing silicon carbide particles (SiCp). Feedback mode SECM imaging using ferrocenemethanol as a redox mediator in 0.1 M NaCl solution and in buffer solution (pH 6.8) revealed that the SiC particles are electrochemically active. The data suggest that the electronic conductivity at these sites is higher than that of the Al2O …

An Experiment Using Time-Based Detection In Flow Injection Analysis, Mary K. Carroll, Julian Tyson

Chemistry Department Faculty Publication Series

We present a laboratory experiment, suitable for implementation in analytical chemistry courses at the undergraduate level, that demonstrates the utility of time-based measurements with doublet peaks in flow injection analysis. The measurements are taken with an instrument that incorporates a simple solid-state detector, which has as its source a light-emitting diode (LED) and as its detecting element a photodiode (PD). The system studied here is a simple acid-base titration. The experiment could be adapted to any number of chemical reactions that result in a product that absorbs light of the LED wavelength significantly.

Atomiser, Source, Inductively Coupled Plasmas In Atomic Fluoresence Spectrometry (Asia): A Study Of Chemical And Ionisation Interference Effects, Stanley Greenfield, M. S. Salman, Julian Tyson

Chemistry Department Faculty Publication Series

The effects of phosphate, aluminium, sodium and potassium on the atomic fluorescence of calcium at 422.7 nm and the ionic fluorescence at I:393.4-396.8 nm have been studied. When the operating conditions are optimised for maximum fluorescence signal from a solution containing no interferents, interference effects are observed which may be interpreted in terms of stable compound formation, ionisation suppression and fluorescence quenching. These effects may be removed by optimising the operating parameters for minimum interference.
