Effect Of Polymer Ligand Structures On Fluorescence Of Gold Clusters Prepared By Photoreduction, Luo Li, Zhen Li, Hui Zhang, Shoucun Zhang, Irfan Majeed, Bien Tan

Blue emission fluorescent Au5 clusters with maximum quantum yield of 20.1% were synthesized by a simple photoreduction method using three specially designed tridentate polymer ligands. The evolution of fluorescent Au nanoclusters (Au NCs) under UV irradiation was studied by fluorescence, UV-Vis and X-ray photoelectron spectroscopic techniques, suggesting that the fluorescence of Au NCs is size-dependent and is associated with the presence of Au(i) ions in the Au NCs. The effect of polymer structure on the fluorescent Au NCs has also been discussed. These highly fluorescent Au NCs have potential applications in the fabrication of optoelectronic devices and light emitting materials.

This review updates the recent advances and applications of three prominent synchrotron radiation techniques, microprobe X-ray fluorescence spectroscopy/imaging, X-ray absorption spectroscopy, and infrared microspectroscopy, and highlights how these tools are useful to the medicinal chemist. A brief description of the principles of the techniques is given with emphasis on the advantages of using synchrotron radiation-based instrumentation rather than instruments using typical laboratory radiation sources. This review focuses on several recent applications of these techniques to solve inorganic medicinal chemistry problems, focusing on studies of cellular uptake, distribution, and biotransformation of established and potential therapeutic agents. The properties of two forms of polyaniline (PAni) synthesised under acidic and basic conditions have been investigated both individually and as combined complexes. The PAni polymerised within alkaline media was redox inactive and non-conducting while the PAni emeraldine salt (ES) was electroactive and conducting. Raman, electron spin resonance, UV-Vis and fluorescence spectroscopies were used to monitor the changes in electronic properties of these conducting polymer composites. Solution cast films of alkaline synthesised (A-PAni) with the PAni ES resulted in an increase in the high spin polaron population suggesting that it acts as a pseudodopant. Thioflavin T (ThT) dye fluorescence is used regularly to quantify the formation and inhibition of amyloid fibrils in the presence of anti-amyloidogenic compounds such as polyphenols. However, in this study, it was shown, using three polyphenolics (curcumin, quercetin and resveratrol), that ThT fluorescence should be used with caution in the presence of such exogenous compounds. The strong absorptive and fluorescent properties of quercetin and curcumin were found to significantly bias the ThT fluorescence readings in both in situ real-time ThT assays and single time-point dilution ThT-type assays. In drug discovery, the occurrence of false positives is a major hurdle in the search for lead compounds that can be developed into drugs. A small-molecular-weight compound that inhibits dengue virus protease at low micromolar levels was identified in a screening campaign. Binding to the enzyme was confirmed by isothermal titration calorimetry (ITC) and nuclear magnetic resonance (NMR). However, a structure-activity relationship study that ensued did not yield more potent leads. To further characterize the parental compound and its analogues, we developed a high-speed, low-cost, quantitative fluorescence quenching assay. Frequency domain fluorescence lifetime imaging microscopy (FLIM) has been used in combination with laser scanning confocal microscopy to study the cellular uptake behavior of the antitumor drug doxorubicin (DOX) and micellar-encapsulated DOX (PLyAd-DOX). The endocytosis uptake process of PLyAd-DOX was monitored over 72 hours using confocal microscopy, with a maximum fluorescence recorded at incubation periods around 24 hours. The micellar structure was not found to release the encapsulated DOX during the time course of imaging. FLIM revealed single lifetime distributions of PLyAd-DOX during accumulation in the cytoplasm.
