In a mass casualty medical evacuation after a bioaerosol (BA) dispersal event, a decontamination (DC) method is needed that can both decontaminate and prevent biological particle (BP) re-aerosolization (RA) of contaminated clothes. However, neither the efficacy of current DC methods nor the risk of BP RA is greatly explored in the existing literature. Thermostable fluorescent proteins, such as thermal green protein (TGP) and yellow thermal protein (YTP), could be used as biosensors to monitor cellular activity and as a fusion tag to monitor a protein of interest. The use of fluorescent proteins can sometimes be limited in certain organelles with low pH and in thermophilic organisms. This research aims to improve the thermal stability of TGP and YTP. Dipyrromethanes are useful mediator structures which can be used as a part of other molecules such as bis-porphyrins and their derivations. Various methods have been developed for their synthesis. This study presents the synthesis of a new bis- dipyrromethane, 7,12-bis(4-(di(1H-pyrrol-2-yl)methyl)phenyl)benzo[k]fluoranthene, using the Lewis acid catalyzed reaction between a new dialdehyde and pyrrole at room temperature. The UV spectroscopic and fluorometric properties of the final product and precursors were determined. The discovery of a method by which proteins of interest can selectively be labeled with a probe of choice intracellularly is a longstanding goal in chemical biology research. Conventional labeling techniques have utilized large domain tags but despite the development of small labeling molecules there have been no short peptide sequences known to covalently label a small molecule without the aid of an enzymatic process or metal chelation. In this dissertation, novel fluorescent materials, which are referred to as a group of uniform materials based on organic salts (GUMBOS) are synthesized, characterized, and studied for different applications. GUMBOS are solid state organic salts having melting points from 25°C to 250°C. Similar to ionic liquids, GUMBOS are highly tunable and can be designed for specific applications by incorporating multiple-functions into a single ion pair by selection of cation-anion combinations. Three luminescent cytidine analogues have been synthesized in order to perform SNP analysis by fluorescence spectroscopy. Herein is described the synthesis of 6-pyrenylpyrrolocytidine (PypdC), its photophysical characterization, and its subsequent incorporation into oligodeoxynucleotides (ODNs). The behavior of PypdC in ODNs is described as well as fluorescence intensity changes with respect to the match and mismatch cases. In order to obtain a greater understanding of pyrene’s interaction with pyrrolocytidine, a congener, pyrenyl ethynyl cytidine (PyEtdC) was synthesized. The congener was photophysically studied and prepared for oligo synthesis. Finally, exploration into indole substituted pyrrolocytidines (pdCs) was undertaken to explore new pdC structures.

The investigation of polypeptide composite particles, PCPs, is described. The production of mesoporous and polycolloid architectures is also addressed. PCPs were prepared by both growing form and grafting to method. They consist of a core made of silica, silica-coated magnetite or silica-coated cobalt. Some of the particles were covalently labeled with fluorescein isothiocyanate. Several polypeptides were chosen for attachment on the core: poly(γ-stearyl-L-glutamate), PSLG, poly(Nε-carbobenzyloxy-L-lysine), PCBL and poly(o-benzyl-L-tyrosine), PBTY. Attachment of a well-characterized alkyne-end terminated PSLG and PCBL to an azide- functionalized particle yielded PCPs with a desired sparse coverage. In 1978, Lee found an increase and then a large decrease in the diffusion coefficient of poly(lysine) as the concentration of salt is decreased (Lin, S. C.; Lee, W. I.; Schurr, J. M. Biopolymers 1978, 17, (4), 1041-1064). Since the “slow mode” discovery, many have studied it without finding a fully satisfactory answer. Additional dynamic light scattering (DLS) measurements have been the main experimental technique used for reinvestigating the slow mode decay. Three marking techniques were tested to determine their applicability for mosquitofish. Tetracycline drugs and DC Ar administered in the diet successfully marked laboratory-cultured mosquitofish, but exposure to direct sunlight in outdoor tanks resulted in the rapid disappearance of the marks. Preliminary data on fluorescent marks from a polystyrene pigment in a melamine-sulfonamide-formaldehyde resin forced into the dermal ,tissue with compressed air are more promising.

A t an optimal deliver pressure of 7.3 m Hg (140 p.s.i.) and spraying time of 15 sec., marking percentage is maximized and fish mortality is minimized. Mark retention time was up to 80 days in …
