We report the thermal stability of wild type (WT) and 14 different variants of human copper/zinc superoxide dismutase (SOD1) associated with familial amyotrophic lateral sclerosis (FALS). Multiple endothermic unfolding transitions were observed by differential scanning calorimetry for partially metallated SOD1 enzymes isolated from a baculovirus system. We correlated the metal ion contents of SOD1 variants with the occurrence of distinct melting transitions. Altered thermal stability upon reduction of copper with dithionite identified transitions resulting from the unfolding of copper-containing SOD1 species. The current paradigm of biodiversity conservation requires the assessment of alien plant invaders, and their potential negative impacts on indigenous species and communities. Leptospermum laevigatum (Gaertn.) F. Muell. (Victorian tea tree/ Coast tea tree) is indigenous to eastern Australia. It has invaded ecosystems within its natural biogeographic range, within new ranges in Australia and overseas. The species is listed as a high priority weed in the Environmental Weed Strategy for Western Australia. However, the basis for its listing has been casual observation rather than focussed research. A reconnaissance of pharmaceutical chemicals in urban streams of the Tualatin River basin was conducted in July 2002 in an effort to better understand the occurrence and distribution of such compounds, and to determine whether they might be useful indicators of human-related stream contamination. Of the 21 pharmaceutical chemicals and metabolites tested, only 6 (acetaminophen, caffeine, carbamazepine, codeine, cotinine, and sulfamethoxazole) were detected in filtered stream samples from 10 sites. The concentrations of most of the detected compounds were relatively low (less than 0.05 microgram per liter). The most frequently detected compounds were cotinine (a nicotine metabolite, 8 of 10 …

The threat to biodiversity posed by increasing rates of anthropogenic habitat fragmentation necessitates an understanding of the consequences of spatial pattern for natural communities. Reduction of patch size, loss of habitat, changes in the quantity and proportion of habitat edge, and reduced connectivity among habitats can all shape ecological processes and faunal behavior. Seagrass habitats provide a natural model system for examining spatial inf1uences on marine fauna, but separating the effects of habitat structure, environmental conditions, and spatial pattern is difficult because shoot density, percent cover, and hydrodynamic regime often co-vary with patch size. The relationship between wetland polycyclic aromatic hydrocarbon (PAH) concentrations and adjacent land use was assessed using surface sediments, sediment cores and global information systems (GIS) in the Elizabeth River, Virginia, an urbanized subestuary of the Chesapeake Bay. Wetlands adjacent to parking lots and petroleum industrial sites exhibited the highest PAH concentrations of all land uses in surfaces sediment and historically. Automobile and coal are the dominant sources of PAHs in wetland sediments historically as determined by relative concentration, fingerprinting and source specific compounds. Organic Matter Cycling In The York River Estuary, Virginia: An Analysis Of Potential Sources And Sinks, Leigh Mccallister

A study of the organic matter (OM) sources and biogeochemical and physicochemical sinks was undertaken in the York River estuary, Virginia. The reactivity of dissolved organic carbon (DOC) was enhanced from ∼25--68% by the combined effects of exposure to natural sunlight and bacterial decomposition. In contrast, sunlight exposure decreased the bioreactivity of DOC in the higher salinity lower York by a factor of five. The combined effects of photochemical and bacterial processing were found to modify both the bioavailability and metabolic fate of OM (e.g. respiration vs. biomass). Immune responses against cardiac myosin and group A streptococcal M protein have been implicated in the pathogenesis of rheumatic heart disease. Although cardiac myosin is known to produce myocarditis in susceptible animals, it has never been investigated for its role in production of valvular heart disease, the most serious sequelae of group A streptococcal infection in acute rheumatic fever. In our study, cardiac myosin induced valvulitis in the Lewis rat, and epitopes responsible for production of valvulitis were located in the rod region. Rethinking Mechanistic Explanation, Stuart Glennan
Scholarship and Professional Work - LAS
Philosophers of science typically associate the causal-mechanical view of scientific explanation with the work of Railton and Salmon. In this paper I shall argue that the defects of this view arise from an inadequate analysis of the concept of mechanism. I contrast Salmon's account of mechanisms in terms of the causal nexus with my own account of mechanisms, in which mechanisms are viewed as complex systems. After describing these two concepts of mechanism, I show how the complex-systems approach avoids certain objections to Salmon's account of causal-mechanical explanation. I conclude by discussing how mechanistic explanations can provide understanding by unification.

Surficial sediments and sediment cores were collected from two distinct depositional regimes of the York River subestuary of Chesapeake Bay to document seasonal inputs, spatial variability, and longer-term (>40 years) fate of total organic carbon (TOC), lipid biomarker compounds and polycyclic aromatic hydrocarbons (PAHs). These regimes included biological mixing in the lower York and episodic mixing at the mid river site. Compounds were selected to represent a range of chemical reactivities, biological and anthropogenic sources, and modes of entry to the environment. Oxytetracycline (OTC) is used to chemically mark calcified structures in walleye Stizostedion vitreum fry and fingerlings. Current visual methods for detection of these marks are subjective, and biologists are unable to quantify the amount of OTC in a sample. The goal of this study was to determine if high pressure liquid chromatography (HPLC) is a feasible and superior alternative for detecting and quantifying OTC in otoliths of fry-marked and spines of fingerlingmarked walleyes. This research develops a model to identify indicators of potential suitable spawning habitat for river herring, Alosa pseudoharengus and A. aestivalis, using watershed and stream-reach metrics. The results of icthyoplankton samples collected from thirty-four streams feeding into the Rappahannock River below the Embree Dam at Fredericksburg indicate where river herring spawning occurred. Watershed and stream-reach metrics were either measured in the field or derived from digital data in a GIS. Benthic macroinvertebrate analysis was used to compare habitat quality among sites. To date, most research on the interaction of nonequilibrium, atmospheric pressure plasma discharges with bacteria has concentrated on the germicidal effects. Therefore, published results deal mainly with killing efficacy and little attention is given to physical mechanisms and biochemical pathways and their potential alterations when cells of microorganisms are exposed to the plasma. In this letter, an attempt to investigate the effects of plasma exposure on the biochemical pathways of bacteria is presented. Fertigation is the technique of using soluble fertilisers to supply essential nutrients to vegetables through sprinkler or trickle irrigation systems, or by means of a boom spray. It is ideally suited to most horticultural areas in Western Australia, especially the sandy soils of the Swan Coastal Plain that have a poor capacity to retain nutrients.
Before and after planting, nutrients are often applied to vegetables on soils of the Swan Coastal Plain by applications of poultry manure* or solid fertilisers. After planting, fertigation is a safe and effective method of applying top-dressings of nutrients to crops, especially after the rows …

Going-to-the-Sun Road (GTSR) is one of the premier road facilities in the world. As the only cross-mountain route through Glacier National Park, the road brings nearly two million visitors a year into the heart of Montana wilderness. How to meet this demand for travel while protecting the nature of the visitor experience and the pristine setting for which the experience exists is a challenge of on-going complexity.
