ICOT 18 - International Conference on Thinking - Cultivating Mindsets for Global Citizens

With the advent of the 21st Century, scholars, politicians, scientists, educators, entrepreneurs, religious leaders, and parents have looked at the increasing rapidity of change and complexity in the world, and wondered what capabilities are necessary to embrace the challenges facing us. The P21 Partnership cites four basic “C” skills: creativity and innovation; critical thinking and problem-solving; communication; and, collaboration. However, developing the “4 Cs” relies on another attribute – curiosity, the hallmark of which is the ability to formulate good questions. Alzheimer’s Disease (AD) is the sixth leading cause of death in the United States with no known methods to cure, reverse or halt disease progression. The “metal hypothesis” states that FeII, CuII, ZnII, and AlII bind to truncated Aβ peptides and form soluble oligomers which deposit as senile plaques. These plaques play a crucial role in AD pathogenesis. For example, metallated-Aβ aggregates are hypothesized to disrupt membranes or generate a reactive oxygen species (ROS) through redox cycling in the presence of CuI/II or FeIII/II and a reducing agent. Molecular beacons are important bioanalytical probes which are most often

constructed from a single-stranded oligonucleotide which has been labeled at

opposite termini with a fluorophore and a quencher. When the fluorophore and

quencher are in close proximity, no fluorescence is observed due to FRET

(Fluorescence Resonance Energy Transfer). DABCYL (4-dimethylaminoazobenzene-

4'-carboxylic acid) has been used as a quencher in the molecular beacon to absorbs

excitation energy from a fluorophore and to dissipate the energy as heat. However,
