The diffraction-limited spot size of synchrotron-based IR microscopes provides cell-specific, spectrochemical imaging of cleared leaf, stem and root tissues of the model genetic organism Arabidopsis thaliana, and mutant plants created either by T-DNA insertional inactivation or chemical mutagenesis. Spectra in the wavelength region from 6 to 12 microns provide chemical and physical information on the cell wall polysaccharides of mutants lacking particular biosynthetic enzymes (‘‘Cellulose synthase-like’’ genes). In parallel experiments, synchrotron IR microscopy delineates the role of Arabidopsis cell wall enzymes as susceptibility factors to the fungus Erysiphe cichoracearum, a causative agent of powdery mildew disease. Austrochlamys heardensis (Fleming) is recorded from a boulder of Late Pliocene (3.62–2.5 Ma) volcaniclastic sandstone dredged 70 km east-north-east of Heard Island, the third record of the species. The collection is much larger than the original described by Fleming and includes left valves which are described for the first time. The species is compared with A. anderssoni (Hennig) from Cockburn Island and ‘Chlamys’ mawsoni Fletcher from Îles Kerguelen. The source rock accumulated in fully marine, highly current-affected conditions. The collection is dominated by right valves, possibly because left valves are more susceptible to breakage and the effects of current winnowing. …

The present pilot study is a histological evaluation of mandibular condyles taken from minipigs that underwent either traditional mandibular advancement (bilateral sagittal split osteotomy), or a distraction osteogenesis procedure to advance the mandible. One pig received a bilateral sagittal split osteotomy with an immediate mandibular advancement of 14 mm, while another pig underwent distraction osteogenesis consisting of a gradual advancement of 2 mm per day to advance the mandible the same distance. The control animal received no surgery.

Our lab has used an optical coherence microscope (OCM) to assess both the structure of tissue-engineered corneal constructs and their transparency. Currently, we are not able to resolve cells versus collagen matrix material in the images produced. We would like to distinguish cells in order to determine if they are viable while growing in culture and also if they are significantly contributing to the light scattering in the tissue. A motion-sensitive en-face-scanning 3-D optical coherence microscope (OCM) has been designed and constructed to study critical events in the early development of plants and animals. We describe the OCM instrument and present time-lapse movies of frog gastrulation, an early developmental event in which three distinct tissue layers are established that later give rise to all major organ systems. OCM images constructed with fringe-amplitude data show the mesendoderm migrating up along the blastocoel roof, thus forming the inner two tissue layers. Motion-sigma data, measuring the random motion of scatterers, is used to construct complementary images that indicate the presence of Brownian …

Characterizing the genome-wide dynamic regulation of gene expression is important and will be of much interest in the future. However, there is currently no established method for identifying differentially expressed genes in a time course study. Here we propose a significance method for analyzing time course microarray studies that can be applied to the typical types of comparisons and sampling schemes. This method is applied to two studies on humans. In one study, genes are identified that show differential expression over time in response to in vivo endotoxin administration. There is a growing literature demonstrating the effectiveness of using computer environments to assist students’ in visualizing science and mathematics concepts. However, with many of these computerized learning environments, students do not have the option of manipulating the environment. Instead, they are presented with pre-made visualizations. Enabling students to display their understanding through multiple representational forms is more interesting. In our peer-led, peer-review environment, students generate a complex, literature-based, multimedia text on which their final examination is based. The connectivity of marine populations and the degree to which they are considered open or closed has important implications for the ecology, management, and resilience of commercially harvested species. Larval exchange is a primary determinant of the level of connectivity between populations, and this thesis examines the intrinsic and extrinsic factors influencing larval transport and the distribution of larvae of the American lobster (Homarus americanus). The potential for larval transport is directly proportional to the planktonic larval duration. Our field data suggest development times in situ were up to three times faster than previous laboratory development times, indicating that potential …

The Borana pastoral system has come under increasing pressure as human populations grow and per capita availability of resources declines. Livestock exhibit large, periodic die-offs that threaten wealth accumulation and food security. Several types of interventions may improve risk management here. For example, there may be opportunity for some pastoralists to diversify their livelihoods. Here we report on a community-based process involving pilot projects begun since 2000. We have embraced Participatory Rural Appraisal (PRA) and Action Research (AR) as tools for investigation and empowerment of local people. Percent litter, average litter depth, percent bare ground, vegetation density, vegetation canopy, and distance to an ecotone boundary were measured and compared to small mammal capture locations in the Las Vegas Wash. Neotoma lepida appear to be greatly dependent upon the foliage litter of T. ramosissima. Alternatively, ecotone boundaries appear to be the most important factor affecting Dipodomys merriami distributions. Peromyscus eremicus distributions may be predicted based on vegetation density and increased canopy during shorter, winter torpor periods. Chaetodipus penicillatus were consistently associated with dense, seed-bearing vegetation, although C. penicillatus is extremely opportunistic and vegetation density should not limit their …

High speed and/or high efficiency separations can be realized using small particles (~ 1 µm) in liquid chromatography (LC). However, due to the large pressure drop caused by small particles, conventional LC pumping systems cannot satisfy the pressure requirements needed to drive the mobile phase through the column. Use of ultrahigh pressure, elevated temperature, or both can overcome these pressure limitations and allow the use of very small particles for high speed and/or high efficiency separations.

The FMP focuses on increasing the state’s efforts to collect data on the resource and the fishery it supports through both fishery-dependent and fishery-independent studies. Native Monterey pine (Pinus radiata) forests occur in five populations on the west coast of North America. High severity fire has been reported to be the main disturbance agent that initiates seedling establishment for this species. To investigate the impacts of fire in this ecosystem, age structure and fire history data were collected from the native Ano Nuevo Monterey pine forest near Santa Cruz California. Stump cross-sections were removed from 10 randomly placed openings, and each was dated to determine tree age and fire history. Average mean fire return intervals were 11.2-20.1 y. In summary, primary measures are aimed at the sustainability of dusky and whiskery sharks, with dusky sharks being of greater concern as their status is more critical. Point Quobba is a popular site for locals and tourists for beach activities, fishing and snorkelling, and as a place for children to learn to swim and gain an awareness of the marine environment. The marine life and habitats of the area are of considerable scientific and recreational interest and are highly valued in the local community. However, the area is at risk from a high level of use and from conflict between users - which is due to the area’s proximity to popular tourism sites, the boat ramp, camping and settlement areas.

This paper presents a new approach to estimating mixture models based on a recent inference principle we have proposed: the latent maximum entropy principle (LME). LME is different from Jaynes' maximum entropy principle, standard maximum likelihood, and maximum a posteriori probability estimation. We demonstrate the LME principle by deriving new algorithms for mixture model estimation, and show how robust new variants of the expectation maximization (EM) algorithm can be developed. We show that a regularized version of LME (RLME), is effective at estimating mixture models. It generally yields better results than plain LME, which in turn is often better than …

The genetic study of certain quantitative traits in growth curves as a function of time has recently been of major scientific interest to explore the developmental evolution processes of biological subjects. Various parametric approaches in the statistical literature have been proposed to study the quantitative-trait-loci (QTL) mapping of the growth curves as multivariate outcomes. In genetic studies, the variation in genotypes may not only affect different inheritance patterns in qualitative traits, but may also affect the age-at-onset as quantitative trait. In this article, we use standard cross designs, such as backcross or F2, to propose some hazard regression models, namely, the additive hazards model in quantitative trait loci mapping for age-at-onset, although the developed method can be extended to more complex designs. With additive invariance of the additive hazards models in mixture probabilities, we develop flexible semiparametric methodologies in interval regression mapping without heavy computing burden.
