We determined sperm concentrations in Tree Swallows (Tachycineta bicolor) by manually expressing semen samples from males during prelaying, egg-laying, incubation, and nestling periods. Sperm concentrations varied by orders of magnitude (0-109 sperm mL-1) among males. Sperm concentrations were highest during the incubation period and lowest during the prelaying period. None of the samples collected during the prelaying, egg-laying, and incubation periods were devoid of sperm. In contrast, 45% of samples collected during the nestling period lacked sperm. We have investigated the intracellular traffic of PrPc, a glycosylphosphatidylinositol (GPI)-anchored protein implicated in spongiform encephalopathies. A fluorescent functional green fluorescent protein (GFP)-tagged version of PrPc is found at the cell surface and in intracellular compartments in SN56 cells. Confocal microscopy and organelle-specific markers suggest that the protein is found in both the Golgi and the recycling endosomal compartment. Perturbation of endocytosis with a dynamin I-K44A dominant-negative mutant altered the steady-state distribution of the GFP-PrPc, leading to the accumulation of fluorescence in unfissioned endocytic intermediates. The large multidomain Kalirin and Trio proteins containing dual Rho GTPase guanine nucleotide exchange factor (GEF) domains have been implicated in the regulation of neuronal fiber extension and pathfinding during evelopment. In mammals, Kalirin is expressed predominantly in the nervous system, whereas Trio, broadly expressed throughout the body, is expressed at a lower level in the nervous system. To evaluate the role of Kalirin in fiber initiation and outgrowth, we microinjected cultured sympathetic neurons with vectors encoding Kalirin or with Kalirin antisense oligonucleotides, and we assessed neuronal fiber growth in a serum-free, satellite cell-free environment. The mifepristone-regulated transcriptional activation system provides a means to exogenously control gene expression in transformed mammalian cells in a temporal and spatial fashion. Such a promoter activation system consists of three components: The first of these is a steroidal inducer drug, mifepristone. This drug binds to the second component, a chimeric transcription factor complex, consisting of the mutant human progesterone receptor fused to the yeast GAL4 DNA-binding domain and the Herpes simplex virus protein VP 16 activation domain. Regression equations between skinfold (SKF) measurements and air displacement plethysmography (ADP) were determined 6 to 18 year old athletes (80 males / 34 females) using Pearson r correlation coefficients. Triceps and mid-calf SKF and percent body fat (%BF) by ADP were measured during an Athletic Ability Assessment at The HIT Center (Huntington, WV). A high relationship was found between SKF and %BF by ADP (males; r2 = .76, females; r2 = .74, all; r2 = .77). Data from the subjects were compared with similar correlations determined between SKF and %BF by hydrostatic weighing (HW) from another study …
