Cisplatin is a widely used anticancer drug with notable side effects including ototoxicity and nephrotoxicity. Macrophages, the major resident immune cells in the cochlea and kidney, are important drivers of both inflammatory and tissue repair responses. To investigate the roles of macrophages in cisplatin-induced toxicities, we used PLX3397, a U.S. Food and Drug Administration-approved inhibitor of the colony-stimulating factor 1 receptor, to eliminate tissue-resident macrophages. Mice treated with cisplatin alone had considerable hearing loss (ototoxicity) and kidney injury (nephrotoxicity). Macrophage ablation resulted in significantly reduced hearing loss and had greater outer hair cell survival. Macrophage ablation also protected against cisplatin-induced …

BACKGROUND: Down syndrome is associated with an increased risk for otitis media with effusion (OME), a childhood condition in which fluid accumulates in the middle ear, potentially leading to hearing loss. The American Academy of Pediatrics Down syndrome guidelines and the American Academy of Otolaryngology - Head and Neck Surgery OME guidelines recommend hearing testing to assess the hearing status of children with Down syndrome diagnosed with OME.

Objective: The objective of the research is to examine the association between mental health status as measured by number of mentally healthy days in the past month and deafness and binge drinking.
Methods: The Behavioral Risk Factor Surveillance System (BRFSS) study was conducted via telephone survey in 50 states, including the District of Columbia and US territories. The sample size is 402,156 participants. Univariate, bivariate, and multivariate regressions, odds ratio, and 95% confidence interval were used to measure the data.
Results: Deaf respondents have 1.56 times the odds of having poor mental health than those who are not deaf. (95% …

OBJECTIVE: Uterine leiomyoma is a common gynecological condition that negatively affects women's quality of life. Vitamin D plays an important role in tumor development and progression. However, clinical studies comparing serum vitamin D levels between women with and without uterine leiomyomas are limited and inconclusive. This study aimed to compare serum vitamin D levels in women with and without uterine leiomyomas.
METHODS: This hospital-based case-control study included 150 women who visited a gynecological clinic. The cases included 75 women with uterine leiomyoma, whereas the controls included 75 age-and parity-matched participants without uterine leiomyoma. Serum vitamin D levels were measured in …

UVM Milton Family Medicine practice has a significant patient population of elderly adults who are experiencing age-related hearing loss. Age related hearing loss is very common in people over 65 – it’s the third most common chronic physical condition in the US. However due to both social stigma and a lack of health literacy many of them are not having their hearing changes addressed with appropriate audiology assessments and hearing aids. Purpose: The aim of this rapid review was to identify and evaluate evidence exploring hearing loss as a risk factor for cognitive decline in the elderly population. Methods: A literature search was performed in three databases: CINAHL, Cochrane Central, and PubMed. The PRISMA template was used to record the search and selection process. Search criteria included older adults aged 65 and up with diagnosed or self-reported hearing loss and no previous diagnosis of dementia or Alzheimer’s disease. Traumatic brain injury (TBI) affects over 2.87 million people in the US each year. The large majority of these injuries can be classified as mild. However, evidence suggests that even mild TBIs can lead to long-lasting symptoms that greatly impact quality of life. This is particularly true when an individual sustains multiple TBIs (repetitive TBI, rTBI). Much of the damage caused by rTBI is due to secondary injury mechanisms that cause the initial injury site to spread and worsen over time. The term “dementia” includes a wide array of diseases. Millions of Americans are affected by these diseases, especially with aging. Its prevalence makes dementia a candidate for exploratory research in understanding its various etiologies and cause-effect relationships in hopes of developing treatment. For decades, sarcomeric myosin heavy chain proteins were assumed to be restricted to striated muscle where they function as molecular motors that contract muscle. However, MYH7b, an evolutionarily ancient member of this myosin family, has been detected in mammalian nonmuscle tissues, and mutations in MYH7b are linked to hereditary hearing loss in compound heterozygous patients. These mutations are the first associated with hearing loss rather than a muscle pathology, and because there are no homologous mutations in other myosin isoforms, their functional effects were unknown. Early identification, diagnosis, and intervention are crucial for individualized planning and outcomes. Children who need binaural amplification and are not managed appropriately become exposed to sound deprivation, which leads to diminished speech audibility, sound localization, and speech and language development. BACKGROUND: This study investigated the association of hearing loss and tinnitus with overall health-related quality of life (HRQoL) among long-term oropharyngeal cancer (OPC) survivors.
METHODS: This study included OPC survivors treated between 2000 and 2013 and surveyed from September 2015 to July 2016. Hearing loss and tinnitus were measured by asking survivors to rate their "difficulty with hearing loss and/or ringing in the ears" from 0 (not present) to 10 (as bad as you can imagine). Hearing loss and tinnitus scores were categorized as follows: 0 for none, 1-4 for mild, and 5-10 for moderate to severe. The primary outcome …

Peroxisome Biogenesis Disorders (PBD) and Zellweger syndrome spectrum disorders (ZSD) are rare genetic multisystem disorders that include hearing impairment and are associated with defects in peroxisome assembly, function, or both. Mutations in 13 peroxin (PEX) genes have been found to cause PBD-ZSD with ~70% of patients harboring mutations in PEX1. Limited research has focused on the impact of peroxisomal disorders on auditory function. As sensory hair cells are particularly vulnerable to metabolic changes, we hypothesize that mutations in PEX1 lead to oxidative stress affecting hair cells of the inner ear, subsequently resulting in hair cell degeneration and hearing loss. Sensorineural hearing loss (SNHL) is typically a permanent and often progressive condition that is commonly attributed to sensory cell loss. All vertebrates except mammals can regenerate lost sensory cells. Thus, SNHL is currently only treated with hearing aids or cochlear implants. There has been extensive research to understand how regeneration occurs in nonmammals, how hair cells form during development, and what limits regeneration in maturing mammals. This study explores the impact of the type and dosage of listening and spoken language (LSL) services on speech and language outcomes in children with cochlear implants or hearing aids in two LSL programs. Identical demographic variables were collected across the two programs for use in the statistical analyses. Speech and language outcomes were examined at ages 3 and 5 using standardized test measures. At age 3, significant differences in LSL outcomes existed between programs for children using cochlear implants but not for children using binaural hearing aids. Introduction: Vestibular aqueducts follow a route through the inner ear that terminates within the skull and, in most circumstances, aqueducts are narrow and bony. An enlarged vestibular aqueduct (EVA) is most commonly caused by a mutation in the SLC26A4 gene. There are other unidentified environmental and genetic causes that produce EVA as well. At the time this case was reported, no surgeries or treatments existed that could repair an EVA. Case presentation: A bilingual male child presented with congenital bilateral EVAs. Hearing loss is a known symptom among people who suffer from traumatic brain injuries. Studies have shown that sensorineural hearing loss is the most common type of hearing loss resulting from traumatic brain injury. However, it is not well understood whether the auditory pathway is affected by traumatic brain injury. In this study, we examined the auditory radiation, using diffusion tensor imaging data and probabilistic tractography. Fifty-three veterans with traumatic brain injury and forty-four veterans without traumatic brain injury are compared. It is well-documented that auditory thresholds decrease with age, which can be referred to as Age-related hearing loss (ARHL). ARHL is one of the most common conditions affecting older adults and is associated with several conditions including decreased postural stability and falls. Age-related changes to auditory function have been attributed to, through histopathological study, specific degradation of the sensory, supporting, and afferent structures of the cochlea. Adequate language exposure is crucial for speech-language development in children. Previous research suggests that decreased language exposure leads to an increased risk for speech-language developmental delays. This study aims to explore the language exposure of two arguably language-deprived environments: Neonatal Intensive Care Units (NICU) and severe-to-profound hearing loss. This is a two-part study that examines caregiver-infant interactions through the analysis of infant-directed speech (IDS). In the first part of the study, researchers transcribed and analyzed pre-recorded Language Environment Analysis (LENA) audio recordings of five (5) infants with severe-to-profound hearing loss (6-months post cochlear implantation). Children who are Deaf or Hard-of-Hearing (DHH) are at-risk for language delays and associated developmental challenges that impact academic, social, and communication skills. Technology Assisted Language Intervention (TALI) is a novel approach that focuses on using augmentative and alternative communication (AAC) as an intervention for children who are DHH. Results from a recent pilot study suggested that TALI may be a viable approach for enhancing spoken language and communication. Firefighters are susceptible to auditory dysfunction due to long-term exposure to noise from sirens, air horns, equipment, and tools used in forcible entry, ventilation, and extrication. In addition, they are exposed to ototoxic chemicals, particularly, during overhaul operations. Studies indicate that 40% of firefighters have hearing loss in the noise-sensitive frequencies of 4 and 6 kHz. Noise-induced hearing loss (NIHL) is often accompanied by tinnitus, which is characterized by ringing noise in the ears. The presence of phantom sounds can adversely affect the performance of firefighters. However, there has been limited research conducted on the prevalence of tinnitus in firefighters. …

Mutations of gap junction connexin genes induce a high incidence of nonsyndromic hearing loss. Pannexin genes also encode gap junctional proteins in vertebrates. Recent studies demonstrated that Pannexin-1 (Panx1) deficiency in mice and mutation in humans are also associated with hearing loss. So far, several Panx1 knockout (KO) mouse lines were established. In general, these Panx1 KO mouse lines demonstrate consistent phenotypes in most aspects, including hearing loss. Mutations of Connexin 26 (Cx26, GJB2), which is a predominant gap junction isoform in the cochlea, can induce high incidence of nonsyndromic hearing loss. We previously found that targeted-deletion of Cx26 in supporting Deiters cells and outer pillar cells in the cochlea can influence outer hair cell (OHC) electromotility and reduce active cochlear amplification leading to hearing loss, even though there are no gap junction connexin expressions in the auditory sensory hair cells.
