Previously, we reported the effects of fescue toxicosis on developing Angus-cross steer growth, carcass, hepatic mRNA, and protein expression profiles of selected serum proteins, and blood clinical and chemical profiles, after summer-long grazing (85 days) of high endophyte (HE)- vs. low endophyte (LE)-infected fescue pastures. We now report the temporal development of acute, intermediate, and chronic responses of biochemical and clinical blood analytes determined at specified time intervals (period 1, day 0–36; period 2, day 37–58; and period 3, day 59–85). Throughout the trial, the alkaloid concentrations of the HE forage was consistently 19–25 times greater (P ≤ 0.002) …

Detection Methods And Intestinal Adherence Of Non-O157 Shiga Toxin-Producing Escherichia Coli, Zachary R. Stromberg
School of Veterinary and Biomedical Sciences: Dissertations, Theses, and Student Research
Shiga toxin-producing Escherichia coli (STEC) are enteric pathogens of humans. Cattle serve as a reservoir and harbor STEC in their intestines. Intimin-positive STEC are referred to as enterohemorrhagic E.coli (EHEC). Seven serogroups (O26, O45, O103, O111, O121, O145, and O157) account for the majority of illness due to STEC and are hereafter referred to as STEC/EHEC-7. To improve detection, enrichment broths were compared for supporting growth of STEC-7 and STEC O104:H4 (STEC-8). In pure culture, STEC enriched in trypticase soy broth (TSB) had significantly greater growth compared to TSB containing antimicrobials. In fecal samples, E. coli broth enrichment yielded growth …
Investigations Into The Cross-Infectivity Of Nematode Parasites Of Cattle And Sheep, Eva Marie Wray
Investigations Into The Cross-Infectivity Of Nematode Parasites Of Cattle And Sheep, Eva Marie Wray
Graduate Theses and Dissertations
The existence of anthelmintic resistance is not singular in parasite species, host species or geographic region. It has become a problem of worldly stature, especially when considering sheep nematodes. Recommendations have been modified from chemical-based control to multipart, managerial intervention. Compared with nematodes of cattle, the worldwide resistance of sheep nematodes to anthelmintic remedies is much more pronounced and urgent. As the resistance to essentially all anthelmintic classes approaches a cautionary level, the implementation of non-chemical controls has grown increasingly essential. Six-6 to 8 month old Holstein steer calves, six-5 month old Katahdin ram lambs and eight-5 month old Suffolk …

Mammalian gestation and pregnancy are fast evolving processes that involve the interaction of the fetal, maternal and paternal genomes. Version 1.0 of the GEneSTATION database (http://genestation.org) integrates diverse types of omics data across mammals to advance understanding of the genetic basis of gestation and pregnancy-associated phenotypes and to accelerate the translation of discoveries from model organisms to humans. GEneSTATION is built using tools from the Generic Model Organism Database project, including the biology-aware database CHADO, new tools for rapid data integration, and algorithms that streamline synthesis and user access. GEneSTATION contains curated life history information on pregnancy and …

This study examined the effects of increasing energy availability from both dextrose and lipid treatments on the proinflammatory response to LPS in Holstein steers. Steers were randomly assigned to one of three groups: saline at 0.5 ml/ kg body weight (Control) or 50% dextrose [0.5 ml/kg body weight (Dextrose) to mimic calm cattle’s response to LPS] administered immediately prior to LPS (0.5 mg/kg body weight at 0 h) or continuous lipid emulsion infusion from –1 to 6 h [Intralipid 20% (Baxter, Deerfield, IL USA); 0.5 ml/kg/hr (Lipid) to mimic temperamental cattle]. Concentrations of non-esterified fatty acids (NEFA) were greater in …

The hypothalamic gonadotropin-releasing hormone receptor (GnRHR) plays an essential physiological role in reproductive function. In this investigation, we studied genetic variation in the entire coding region of GnRHR gene using PCR-SSCP technique in 250 Bos taurus (Holstein Friesian, Jersey) and Bos indicus (Malnad Gidda, Deoni) bulls. The genetic variants in GnRHR gene were determined by PCR-SSCP technique by using 5 sets of primers. SSCP analysis of fragment 1, comprising exon 1, and fragment 3, comprising exon 2, revealed 2 SSCP patterns in all breeds under study, while fragment 2 of exon 1 and fragment 4, comprising exon 3, revealed 3 …

The group of bacteria known collectively as Salmonella has historically been a major cause of both health and economic hardships. In particular, Salmonella in bovine has been estimated to cause the economy to lose billions of dollars annually, both due to the increased morbidity and mortality rates of the cattle themselves and the indirect loss of potential profits, and also results in the spread of the bacteria to humans to ill effect. Salmonella thus can be quite taxing to agricultural communities, the animals upon which they rely, and the consumers that buy said communities’ commodities. Thus, the importance of an …

The economics of dairy cattle breeding around the world has been significantly affected by the autosomal recessive hereditary diseases of bovine leukocyte adhesion deficiency (BLAD), factor XI deficiency (FXID), bovine citrullinemia (BC), and deficiency of uridine monophosphate synthase (DUMPS). The aim of this study was to determine BLAD, FXID, BC, and DUMPS alleles in Holstein cattle raised in Burdur Province in Turkey. This study involved taking blood samples randomly from 500 head of Holstein cattle at various dairy farms in different towns in Burdur Province. Polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) analysis was employed to identify cows …

In this study, the presence of paratuberculosis was investigated by serological and molecular methods in herds of dairy cattle. Blood, milk, and stool samples of 147 cows aged 2 years old or older with chronic diarrhea were collected. A California mastitis test (CMT) was performed on milk samples. Indirect paratuberculosis enzyme-linked immunosorbent assay (ELISA) test was used for serological investigation. Polymerase chain reaction (PCR) was utilized for molecular identification of Mycobacterium avium subsp. paratuberculosis (MAP) from milk and stool samples. Immunomagnetic separation (IMS) method was used for milk and fecal samples. Eighteen (12.24%), 44 (29.93%), 36 (24.49%), 28 (19.05%), and …
