T Cell Growth Factor Receptors. Quantitation, Specificity, And Biological Relevance, Richard J. Robb, Allen Munck, Kendall A. Smith Nov 1981

T Cell Growth Factor Receptors. Quantitation, Specificity, And Biological Relevance, Richard J. Robb, Allen Munck, Kendall A. Smith

Dartmouth Scholarship

To examine directly the hypothesis that T cell growth factor (TCGF) interacts with target cells in a fashion similar to polypeptide hormones, the binding of radiolabeled TCGF to various cell populations was investigated. The results indicate that TCGF interacts with activated T cells via a receptor through which it initiates the T cell proliferative response. Internally radiolabeled TCGF, prepared from a human T leukemia cell line and purified by gel filtration and isoelectric focusing, retained biological activity and was uniform with respect to size and charge. Binding of radiolabeled TCGF to TCGF-dependent cytolytic T cells occurred rapidly (within 15 rain …

Detergent Extraction Of A Presumptive Gating Component From The Voltage-Dependent Sodium Channel, W J. Culp, D T. Mckenzie Nov 1981

Detergent Extraction Of A Presumptive Gating Component From The Voltage-Dependent Sodium Channel, W J. Culp, D T. Mckenzie

Dartmouth Scholarship

A physiologically characterized radiolabeled neurotoxin complex obtained from venom of the scorpion Leiurus quinquestriatus has been used to identify detergent-solubilized presumptive sodium channel components in sucrose gradients. This toxin-binding component is found in extracts prepared from three sources of excitable membrane but appears to be absent from similar extracts prepared from nonexcitable membrane or from Torpedo californica membrane. Procedures that destroy the physiological activity of the Leiurus neurotoxin lead to a corresponding loss of toxin binding to the putative sodium channel component. The major component recognized by the Leiurus toxin sediments at 6.5 S. Scatchard analysis of quantitative binding experiments …

Low Pressure Membrane Separation Process To Remove Heavy Metal Complexes, Dibakar Bhattacharyya, Chin-Shun Cheng Sep 1981

Low Pressure Membrane Separation Process To Remove Heavy Metal Complexes, Dibakar Bhattacharyya, Chin-Shun Cheng

KWRRI Research Reports

The overall objective of this investigation is to establish the rejection behavior of heavy metals in the presence of complexing agents, utilizing negatively charged ultrafiltration membranes. An extensive experimental investigation is conducted with Zn2+, Cd2+, Cu2+, and Cu1+ in the presence of cyanide, ethylenediamine tetraacetic acid, and oxalates, under insignificant concentration polarization condition. The rejection dependence of the heavy metals is found to be a function of feed metal concentration, metal types, complexing agent to metal feed molar ratio, pH and ionic strength. The dependence of rejection behavior of heavy metals and complexing …

Development Of An Instrument For The Evaluation Of Weight Reduction Advice In Women's Magazines, Sheryl H. Calish Jun 1981

Development Of An Instrument For The Evaluation Of Weight Reduction Advice In Women's Magazines, Sheryl H. Calish

FIU Electronic Theses and Dissertations

The purpose of this study was to develop a valid and reliable instrument to evaluate the weight reduction advice in popular women's magazines. An apparent proliferation of articles on weight reducation in the mass media and women's magazines have been critized as faddist by some professionals. Regardless of this concern, a standardized method of evaluating any diet remains noexistent.

To achieve the purpose the study was conducted in three parts: a Field Test, a Validity Study and a Reliability Study. An Article Evaluation form, which listed the negative characteristics of a faddist weight reduction article was constructed as the instrument. …

Testing The Integrity Of Liquid Containing Hermetically Sealed Containers By The Use Of Radioactive Markers, Patrick P. Deluca, L. David Butler May 1981

Testing The Integrity Of Liquid Containing Hermetically Sealed Containers By The Use Of Radioactive Markers, Patrick P. Deluca, L. David Butler

Pharmaceutical Sciences Faculty Patents

A method for detecting leaky ampules filled with a liquid such as a parenteral solution. The ampules to be tested are immersed in a solution containing a short-lived radionuclide, and a pressure differential is imposed between the solution containing the radionuclide and the parenteral solution or other liquid contained in the ampules. The ampules are then removed from solution, decontaminated to remove any solution adhering to the outer surface and pores thereof, dried, and finally examined for electromagnetic radiation, preferably gamma radiation, emanating from the interior of the ampules which would indicate a leaky condition thereof.

Isolation Of Plasma Membrane From Human Neutrophils And Determination Of Cytochrome B And Quinone Content, Edward P. Sloan, Dana R. Crawford, Donald L. Schneider May 1981

Isolation Of Plasma Membrane From Human Neutrophils And Determination Of Cytochrome B And Quinone Content, Edward P. Sloan, Dana R. Crawford, Donald L. Schneider

Dartmouth Scholarship

Analyses of plasma membrane and other subcellular fractions indicate that the primary location of cytochrome b in human neutrophils is not the plasma membrane. The procedure developed for the purification of plasma membrane from fresh human neutrophils yielded a 14-fold enrichment in the marker enzyme 5'-nucleotidase and a 10-fold enrichment in ouabain-sensitive ATPase. On sucrose density gradients, the peak density of 5'-nucleotidase activity was 1.12 g/ml, and was shifted after digitonin addition to 1.15 g/ml. Protein in the plasma membrane equalled approximately 8 percent of the whole cell protein. A b-type cytochrome was found to be present in the plasma …

Exercise And Stress Tolerance, Richard A. Dienstbier, James Crabbe, Glen O. Johnson, William Thorland, Julie A. Jorgensen, Mitchel M. Sadar, Dennis C. Lavelle

Department of Psychology: Faculty Publications

Presented in this paper is research designed in part to show that although moderate running by well-trained runners plays a significant role in reducing stress response to subsequently introduced stressors, running at the level of marathon competition alters psychological and physiological dispositions in a manner not conducive to reduced stress responses. In these demonstrations, the choice of our psychological and physiological dependent measures was guided by a larger theoretical framework concerning the relationship of exercise to temperament. We will discuss this larger view first because it provides a theoretical perspective that is useful in considering our specific hypotheses. We believe …

Synthesis And Use Of Diagnostic Radio-Pharmaceuticals Comprising Radioactive Isotopes Of Bromine With Dyes, John W. Triplett, Randall B. Smith

Pharmaceutical Sciences Faculty Patents

A process for preparing bromine-containing dyes labelled with gamma-emitting isotopes of bromine, and the product thereof which is useful as an imaging agent for the hepato-biliary system, particularly in dynamic imaging methods. The dyes prepared are from the class of triarylmethane dyes, and also from the phthalein subclass of the class of xanthene dyes, and the labelling thereof is effected with 76Br, 77 Br or 82 Br. The process for preparing these dyes involves reacting the non-brominated dye precursor with either 76 Br2, 77 Br2 or 82 Br2. This is a substitution type of …

The Isolation Of Ecdysterone Inducible Genes By Hybridization Subtraction Chromatography, Michael P. Vitek, Susan G. Kreissman, Robert H. Gross Mar 1981

The Isolation Of Ecdysterone Inducible Genes By Hybridization Subtraction Chromatography, Michael P. Vitek, Susan G. Kreissman, Robert H. Gross

Dartmouth Scholarship

We have developed a procedure for selectively enriching a mRNA population for inducible sequences. Other than the induced mRNA species, the population of mRNA in control cells is approximately the same as the mRNA population in induced cells. Cytoplasmic mRNA from control cells is bound to oligo (dT)-cellulose and used as a template for reverse transcriptase, the oligo (dT) serving as a primer. After removing the template mRNAs, the cDNA-cellulose column is used to hybridize a population of mRNAs from induced cells. The non-hybridized poly A + RNAs are greatly enriched in the inducible sequences. We have used this technique …
