Alzheimer’s, Parkinson’s and Creutzfeldt-Jakob disease are associated with inappropriate protein deposition and ordered amyloid fibril assembly. Molecular chaperones, including αBcrystallin, play a role in the prevention of protein deposition. A series of site-directed mutants of the human molecular chaperone, αB-crystallin, were constructed which focused on the flexible C-terminal extension of the protein. We investigated the structural role of this region as well as its role in the chaperone function of αB-crystallin under different types of protein aggregation, i.e. disordered amorphous aggregation and ordered amyloid fibril assembly. In this paper, an attempt has been made to investigate the effect of particle breakage on densification behaviour of ballast under cyclic loading using Discrete Element Method (DEM). Numerical simulations using PFC2D have been carried out on an assembly of angular particles with and without incorporation of particle breakage. Two-dimensional projection of angular ballast particles were simulated using clusters of bonded circular particles. Degradation of the bonds within a cluster was considered to represent particle breakage. Clump logic was used to make the cluster of particles unbreakable. Amyotrophic lateral sclerosis (ALS) is characterized by progressive motor neuron degeneration, which ultimately leads to paralysis and death. Mutation of TAR DNA binding protein 43 (TDP-43) has been linked to the development of an inherited form of ALS. Existing TDP-43 transgenic animals develop a limited loss of motor neurons and therefore do not faithfully reproduce the core phenotype of ALS. Here, we report the creation of multiple lines of transgenic rats in which expression of ALS-associated mutant human TDP-43 is restricted to either motor neurons or other types of neurons and skeletal muscle and can be switched on and off. …

For decades, Americans expressed dissatisfaction with conventional cancer treatments. I examine the appeal of biological alternatives to surgery, chemotherapy, and radiation therapy for patients with cancer. The limited effectiveness and side effects of many conventional cancer therapies led many patients to pursue a variety of unconventional therapies. To improve the quality of care for cancer patients, "mainstream" providers must better understand, and even collaborate with, alternative practitioners.

Developmental cortical malformations are a major cause of intractable seizures. Determining the location and timing of susceptibility for epileptiform activity is critical to identifying what mechanisms contribute to epileptogenesis in any model. Using the freeze lesion rat model of polymicrogyria, we have identified, in lesioned cortex, these two aspects of epileptogenesis. Previous studies have demonstrated that epileptiform activity cannot be evoked prior to postnatal day (P) 12, but the malformed cortex is more susceptible to seizures as early as P10. An increase in excitatory afferents to the epileptogenic zone occurs before the onset of network epileptiform activity. Whether or not …

The purpose of the present research was to collect qualitative data regarding twelve-step programs which was then used to inform the creation of a new measure of twelve-step engagement. Focus groups were conducted, consisting of twelve-step members, to ascertain an insider perspective on the twelve-step experience. Results showed that "successful recovery" was considered more than just time abstinent by these insiders. These findings were then used to guide the creation of the Glassboro Inventory For Twelve-Step (GIFTS), a new measure of twelve-step engagement which improves upon previous twelve-step measures. BACKGROUND: The development and progression of hepatocellular carcinoma (HCC) is significantly correlated to the accumulation of genomic alterations. Array-based comparative genomic hybridization (array CGH) has been applied to a wide range of tumors including HCCs for the genome-wide high resolution screening of DNA copy number changes. However, the relevant chromosomal variations that play a central role in the development of HCC still are not fully elucidated.

METHODS: In present study, in order to further characterize the copy number alterations (CNAs) important to HCC development, we conducted a meta-analysis of four published independent array-CGH datasets including total 159 samples.

The overall purpose of this dissertation was to examine the planning, implementation, and formative evaluation of a community-based food literacy program for youth. Article 1 provided a summary of the community-based cooking program for at-risk youth. Objectives included the provision of applied food literacy and cooking skills education augmented with fieldtrips to local farms. Eight at-risk youth (five girls and three boys, mean age = 14.6) completed the intervention. Post intervention, five of eight participants completed in-depth interviews about their experiences.

OBJECTIVE: Recent studies have suggested that cannabis use is a risk factor for developing schizophrenia. We tested the hypothesis that cannabis use increases the likelihood of psychosis-like experiences in non-clinical participants who scored highly on a measure of schizotypy. METHOD: The psychological effects of cannabis were assessed in 137 healthy individuals (76% female, mean age 22 years) using a newly developed questionnaire concerned with subjective experiences of the drug: the Cannabis Experiences Questionnaire. The questionnaire has three subscales: Pleasurable Experiences, Psychosis-Like Experiences and After-Effects. Respondents also completed the brief Schizotypal Personality Questionnaire. In a previous paper we reported preliminary evidence that cannabis use increases the likelihood of psychosis-like experiences in non-clinical respondents who scored highly on a measure of schizotypy. We now present findings from pooled data from 3 new follow-up studies comprising a sample of 477 respondents, of whom 332 reported using cannabis at least once. Abstract. 94 participants, drawn from a parent sample of 1206 individuals who had previously taken part in an assessment of the factor structure of the Oxford-Liverpool Inventory of Feelings and Experiences (O-LIFE), were designated into groups differentiated in terms of low, moderate or high levels of positive schizotypy. All also completed the dissociative experiences scale (DES) and the Inventory of Suggestibility (IS). Our findings suggest that higher reported levels of dissociative experiences and higher suggestibility both independently predict higher scores of positive schizotypy, although suggestibility appears to be a stronger predictor than dissociation.

Both schizotypy and at-risk mental states (ARMS: prodromal states) define individuals at risk for psychotic symptoms. However, the relationship between the 2 is unclear. ARMS individuals are, by definition, help-seeking and therefore at greater risk. We tested whether high schizotypes and ARMS exist along the same continuum by examining maladaptive metacognitions and distress. Background. The literature on colorectal cancer (CRC) screening is contradictory regarding the impact of weight status on CRC screening. This study was intended to determine if CRC screening rates among 2005 National Health Interview Survey (NHIS) respondent racial/ethnic and gender subgroups were influenced by weight status. Methods. Univariable and multivariable logistic regression analyses were performed to determine if CRC screening use differed significantly among obese, overweight, and normal-weight individuals in race/ethnic and gender subgroups.
