Hemostatic Efficacy Of Sheep-Derived Fibrin Glue For Liver Biopsy In Swine, Siriluck Luckanahasaporn, Theerawat Tharasanit, Passakorn Briksawan, Sumit Durongphongthron

Fibrin glue has been widely used for controlling hemorrhage and sealing tissue in surgery. The purpose of this study was to evaluate the hemostatic efficacy of sheep-derived fibrin glue. Six Marino sheep were used to collect concentrate fibrinogen preparation by the ammonium sulfate precipitation method. Crossbred pigs (n=6) were used to evaluate hemostatic efficacy by monitoring whole blood clotting time by the glass slide method, bleeding time and bleeding quantity during liver biopsy. Mean whole blood clotting time on a glass slide started at 201.1±90.47 and 4.43±3.73 seconds, and was completed at 447.83±63.77 and 31.93±4.28 seconds in control and treatment …

Classical swine fever virus (CSFV) contributes to economic loss of swine production in endemic countries. Serum neutralization and enzyme-linked immunosorbent assay (ELISA) are serological tests commonly used for monitoring CSFV antibody status with serum samples. This experiment evaluated the detection of CSFV antibody in oral fluid samples in commercial ELISA test kit by in vitro study of negative oral fluid mixed with serum of known CSFV serum neutralizing (SN) titer and in vivo oral fluid samples obtained from experimental animals. Porcine reproductive and respiratory syndrome (PRRS) has become a major swine disease worldwide. Relevant management strategies and diagnostic assays are of importance for PRRS virus (PRRSV) control. The objective of this study was to determine the use of oral fluids for PRRSV monitoring in endemically PRRSV-infected herds. PRRSV RNA and PRRSV-specific antibodies were monitored using oral fluid samples and serum samples in two conventional swine farms in Thailand (‘Farm A’, a one-site conventional system farm, and ‘Farm B’, a one-site P0-P1 segregation system farm) during farrowing to nursery periods. The aim of this study was to isolate antimicrobial-resistant bacteria from intensive commercial and backyard swine operations (farms) in Northern Thailand and to characterize antimicrobial usage among swine farmers. A total of 51 swine farms in Chiang Mai, Lampoon, and Nan provinces were visited from February to October 2011 to collect swine fecal samples for bacterial isolation (Salmonella spp., Escherichia coli, Enterococcus faecalis) and antimicrobial resistance testing. The impact of air pollution on airway cellular defense of pigs raised under backyard, semi-commercial and commercial conditions was investigated. Flame atomic absorption spectrometry was used to analyze Cadmium (Cd) and Lead (Pb) while manual cold vapor atomic absorption spectrometry was applied to evaluate Mercury (Hg) contents of dust particles from farms. Tests for viable counts of swine alveolar macrophages (SAM) that simulate animal responses to air pollutants, microbiological techniques for the recovery of bacterial cells and evaluation for cellular function were undertaken. Plasmid Profiles Of Multidrug-Resistant Escherichia Coli From Clinically Healthy Swine, Khin Khin Lay, Nisit Chansong, Rungtip Chuanchuen

A total of 145 Escherichia coli isolates originated from clinically-healthy pigs were analyzed for plasmidprofiles. All but one isolate (n=144) carried one to nine plasmids with size ranging from 0.7-16.2 Mda. Fifty-sixplasmid profiles were defined based on sizes and numbers of plasmids in each strain, of which the most commonpatterns were 15.8, 4, 3.3, 1.7, 1.3 Mda and 16.2, 15.8, 4, 3.3, 2.3, 1.7, 1.3, 1, 0.7 Mda. The results indicate widedistribution of plasmids among the commensal E. coli isolates originated from clinically-healthy pigs.

In January 2010, we examined the prevalence of Salmonella in the rectal swab of 66 swine on 8 farms and in 25 pork products at 6 meat retailers of Sa Kaew province. Salmonella was isolated from 3%(2/66) of swine rectal swab samples and 96%(24/25) of pork samples. In rectal swab samples, both S. Weltevreden and S. Dumfries were isolated from one sample, and S. Salbutamol residues in swine tissues and fluids were investigated. Tissue, serum and urine samples were collected from 33 pigs after 14 days of oral administration of 3 ppm salbutamol. The concentrations of salbutamol were determined by enzyme-linked immunosorbent assay and further confirmed by capillary electrophoresis. The highest urinary concentration of salbutamol was 145.12 ng/ml at 18 hour and remained detectable for 30 days after the last feeding. Salbutamol was undetectable in serum samples. The highest concentrations of salbutamol in liver, kidney, lung, heart, brain and muscle were 70.42 ng/g, 31.88 ng/g, 26.06 ng/g, 6.76 ng/g, 3.41 ng/g and 2.97 ng/g, …

The Contribution Of Active Efflux In Reduced Susceptibilities To Copper Sulfate And Zinc Chloride In Escherichia Coli Isolates From Swine, Chailai Koowatananukul, Nisit Chansong, Rungtip Chuanchuen

A hundred-eighty Escherichia coli isolates from swine were tested for minimal inhibitory concentrations (MICs) of 9 antimicrobials and two heavy metals copper sulfate and zinc chloride. All the E. coli isolates were resistant to at least one antibiotic and 175 (97.2%) isolates were multi-drug resistant. Forty antibiotic resistance patterns were identified, of which the most common pattern was AMP-CHP-CIP-ERY-GEN-STR-SUL-TET-TRI (13.3%). All the isolates formed one large population of susceptibility to copper sulfate and zinc chloride at the concentrations of 6.4-12.8 mM and 3.2-6.4 mM, respectively. Exposure to copper sulfate and zinc chloride at sublethal concentration did not change the susceptibility. …

The efficacy of microsatellite markers was evaluated in order to use in parentage control in swine. Genomic DNA from 80 samples were extracted, and amplified using 16 microsatellite markers (D00768, KVL9000, NLRIP0001, S0663, S0710, S0719, S0766, SJ859, SJ923, SJ924, SJ925, SJ926, SJ927, SJ929, X53085 and X63893) in each single polymerase chain reaction (PCR). The PCR products were analyzed using agarose and polyacrylamide gel electrophoresis (PAGE). The result showed that 15 microsatellite loci could be amplified except SJ925. A hundred twenty weaned crossbred pigs of Duroc Jersey and Landrace-Largewhite, weighing an average of 6.55 ± 0.62 kg were divided into three groups. All were aimed at elucidating the relationship between the duration of oestrus, the time of ovulation, the time of insemination and fertility. The objective of this review is to update information concerning the function of seminal plasma on the pig reproductive system, as well as its importance in artificial insemination (AI). It has been shown that seminal plasma advances ovulation in gilts by an average time of 10-14 hours, improves sperm motility, sustaines sperm viability during uterine transport, suppresses immune activities (chemotaxis of neutrophils), improved sperm transport, improves fertilisation rate, improves litter size in gilts and indirectly enhances fertility. The hormone estrogen, within the seminal plasma, causes a release of prostaglandins from the pigs endometrium to the utero-ovarian veins and lymphatic vessels. …

An outbreak of leptospirosis in a 2600 sow breeding herd, in which 5% of the sows aborted, was investigated. Serum samples from 11 aborted sows were examined for leptospira antibodies. Using a Microscopic Agglutination Test (MAT), containing only 12 serovars, no leptospira serovars were detected. However, after using a Microcapsule Agglutination Test (MCAT) kit, a further MAT using the additional three serovars (sejroe, hardjo and bratislava) revealed that the cause of abortion was Leptospira interrogan serovars bratislava and sejroe.
