Background:
Some α1-adrenoceptor antagonists possess anti-cancer actions that are independent of α1-adrenoceptors and the aim of these studies was to assess the relative cytotoxic potencies of α1-adrenoceptor antagonists and the mechanisms involved in these actions.
Methods:
PC-3 and LNCap human prostate cancer cells were exposed to α1-adrenoceptor antagonists (0.01-100μM) and cell survival assessed after 24-72hr. The levels of apoptosis, autophagy and stress related proteins were also determined.
Results:
The relative cytotoxic potency order was prazosin=doxazosin>terazosin=silodosin=alfuzosin>tamsulosin on both cell types, but LNCaP cells were significantly more sensitive to these effects than PC-3 cells. Prazosin and doxazosin increased levels of …

Background: Women diagnosed with atypical ductal hyperplasia (ADH) or atypical lobular hyperplasia (ALH) have a fivefold increased risk of developing breast cancer. Because ADH/ALH can be precursors or predictive markers of a subsequent clinically significant event (SCSE), i.e. atypia, in situ or invasive carcinoma, the clinical outcome for these patients ranges anywhere from remission to invasive malignancy. Background: A monoclonal antibody, trastuzumab targets the human epidermal growth factor receptor 2 (HER2) oncogene that is overexpressed in 25–30% of breast cancers. In combination with first-line therapy, trastuzumab resulted in significant improvement in survival outcomes for those with HER2-positive metastatic breast cancer. Due to its improvement in outcome and prolonged survival, trastuzumab has been established as standard of care in both adjuvant and metastatic settings. However, along with common adverse events, trastuzumab has been found to be associated with cardiotoxicity. Background: Culture shapes how people understand illness and death, but few studies examine whether acculturation influences patients’ end-of-life treatment preferences and medical care.
Methods and Findings: In this multi-site, prospective, longitudinal cohort study of terminally-ill cancer patients and their caregivers (n = 171 dyads), trained interviewers administered the United States Acculturation Scale (USAS). The USAS is a 19- item scale developed to assess the degree of ‘‘Americanization’’ in first generation or non-US born caregivers of terminally-ill cancer patients. Introduction: There is ongoing debate over banning electronic cigarette (e-cigarette) use (vaping) in public places. Many people perceive secondhand e-cigarette vapors (SHV) to be relatively harmless, which may affect their support for policies to restrict vaping in public places. Given that awareness of secondhand cigarette smoke risks predicts public support for clean air policies, we hypothesized that greater perceived harm of SHV to personal health would be associated with stronger support for vaping restrictions.

It is critical to understand patients' information use from the patient perspective, especially when patients are from different cultures and levels of health literacy. A cross-sectional survey supplemented with interviews of breast cancer survivors including both Latina and non-Latina women was undertaken. Subjects were classified as active information seekers, passive information receivers, and/or users of information. Subjects were further classified by stage of information use, progressing from unawareness or awareness of available information to use or non-use of information to make health decisions. MPNSTs are rare malignancies that are classically associated with pre-existing plexiform neurofibromas in neurofibromatosis type 1 (NF-1) patients, but also occur in association with radiation as well as sporadically in patients with no known risk factors. The typical presentation of sporadic MPNST is a new painless enlarging mass. The typical presentation of MPNST in an NF-1 patient is rapid enlargement or new onset of pain associated with a pre-existing plexiform neurofibroma. Although both MPNST and benign neurofibromas share in common the absence of neurofibromin function due to loss of both NF-1 alleles, malignant transformation to MPNST requires several additional aberrations, …

'Why I Feel Bad': Refinement On The Effects Of Prostate Cancer Upon Lifestyle Questionnaire And An Initial Exploration Of Its Links With Anxiety And Depression Among Prostate Cancer Patients, Christopher Sharpley, Vicki Bitsika, David Christie

Objective: To psychometrically refine a standardized scale for identifying those lifestyle changes that were most likely to contribute to anxiety and depression among prostate cancer (PCa) patients.
Methods: Three hundred and eighty-one PCa patients who had received their initial diagnosis between one and 96 months completed a survey of background variables, anxiety and depression inventories and the 36-item Effects of Prostate Cancer upon Lifestyle Questionnaire (EPCLQ).
Results: Levels of anxiety (24%) and depression (26%) were similar to those previously reported for PCa patients. The EPCLQ was shown to have satisfactory psychometric properties and significantly predicted anxiety and depression scores and …
