Twenty-four virulent bacteriophages isolated from raw milk and whey samples were characterised by structural protein composition and bacteriophage DNA restriction endonuclease fragments. Twenty-four lactococcal bacteriophages were possessed among 24-31 total structural proteins with molecular masses of 6.5-208.9 kDa and 2-8 major structural proteins with molecular masses of 6.5-38 kDa. Lactococcal bacteriophages were classified as 5 different groups by total structural proteins and 4 different groups by major structural proteins. The genome sizes of bacteriophages were estimated between 19.6 and 39 kb. The effects of melatonin hormone on rainbow trout (Oncorhynchus mykiss) erythrocyte carbonic anhydrase enzyme purified by using Sepharose 4B-L tyrosine-sulfanylamide affinity gel chromatography were investigated in vitro and in vivo. Carbonic anhydrase from Oncorhynchus mykiss erythrocytes was purified with a specific activity of ~400 EU/mg protein and ~200-fold in a yield of ~20. Melatonin did not inhibit carbonic anhydrase enzyme in vitro. Carbonic anhydrase activity of the control group was determined as 1597.7 ± 429.0 EU/g hemoglobin in vivo. Morphometric And Electrophoretic Variation In Different Honeybee (Apis Mellifera L.) Populations, İrfan Kandemi̇r, Meral Kence, Aykut Kence

Honeybee colonies from Turkey, Naxcivan (Azerbaijan) and Austria were studied for morphometric and electrophoretic variation and for comparison among different subspecies. Out of six enzyme systems, four were found to be polymorphic with 14 allozymes. Discriminant function analysis of 10 morphometric variables resulted in a total of four groups. Cluster analysis of electrophoretic data showed a similar type of groupings. Among the four polymorphic loci, Mdh-1 showed the highest and Est-3 the lowest differentiation among populations.

This study was carried out to isolate Clostridium perfringens from chickens and to detect the gene encoding the alpha toxin produced by all types of C. perfringens by polymerase chain reaction (PCR). Intestinal contents of 160 slaughtered chickens from 8 different farms in Elazığ province were analyzed. C. perfringens was isolated from 8 (5%) of the samples. DNA samples extracted from suspected isolates grown on selective agar were amplified by PCR using a pair of primers derived from the alpha toxin gene. This study was undertaken to determine changes in the flora of lactic acid bacteria during the ripening of Şavak tulum cheese and to identify the major species playing an important role in the ripening process. Tulum cheese was produced from raw sheep milk using traditional methods and ripened in plastic containers at 4 °C for 90 days. Samples were taken on days 0, 15, 30, 60 and 90 and analyzed for isolation and identification of lactic acid bacteria. A total of 783 isolates of lactic acid bacteria were identified. Respiratory diseases are very common in poultry. Bordetella avium and Ornithobacterium rhinotracheale are among the major microorganisms responsible for these diseases. The study was conducted to investigate the seroprevalence of O. rhinotracheale and B. avium in Aydın. Two hundred seventy-six chicken and 360 turkey serum samples were collected from poultry with respiratory disease suspicion in poultry enterprises around the city and samples were then examined for the presence of B. avium and O. rhinotracheale by commercial ELISA kits. The effects of homeopathy on pseudopregnancy in bitches were investigated. Thirty clinically pseudopregnant bitches were used. Fifteen dogs were treated by homeopathy and the remaining animals received placebo treatment. Thuja D30 globules, which contain Thuja Occidentalis, were given orally (8 globules, 3 times a day) as a homeopathic agent. Clinical response was evaluated every 5 days. Maximum duration of the treatment was 3 weeks. Physical changes in mammary glands and behavioral response were evaluated during the treatment. The aim of this study was to determine the alpha naphthyl acetate esterase (ANAE) activity in the peripheral blood leukocytes in Angora rabbits. The heparinized blood samples obtained from 10 healthy adult Angora rabbits were used. ANAE stainings were applied at different pH values for the determination of ANAE activities. The ANAE positive reactions were observed in the T lymphocytes, monocytes, neutrophils, eosinophils and platelets. The ANAE positive lymphocyte rate was found to be 68.2%.

Changes in proximate composition and amino acid content of frozen (-18 °C) wild sea bass (Dicentrarchus labrax, L. 1758) fillets were investigated during 9 months of storage. Initial protein and lipid contents were 19.75% and 1.22%, respectively, and changed to 19.31% and 3.58% in the 9th month of storage. Amino acid analyses were performed at intervals in the initial, third, sixth and ninth months of storage. The initial ratio of essential/nonessential amino acid (g amino acid/16 g N) was 0.75. It is inevitable to encounter many diseases of ostriches in Turkey as a consequence of the rapid expansion of ostrich farming and flocks in this country. Aspergillus fumigatus was isolated from the lungs and air sacs of a 3-month-old, male, blue-neck ostrich. In postmortem examination, the ingestion of a large amount of gravel was noted. The accumulated gravel had led to an occlusion of the passage between the proventriculus and gizzard. This work appears to be the first report of aspergillosis and gastric impaction developing together and leading to death in an ostrich.

The organs of an aborted equine fetus were examined histopathologically and microbiologically. Diffuse, mild cellular infiltration of neutrophils, lymphocytes and plasma cells was observed in the lamina propria of the intestine. In addition, bacterial colonies were seen in the intestinal content, wall of intestine and liver. Streptococcus equi subsp. zooepidemicus was isolated in pure culture from all organs. This is the first report of the isolation of S. equi subsp. zooepidemicus from an aborted equine fetus in Turkey.

Vaccine-associated feline sarcoma (VAFS) with multiple recurrences was diagnosed in the specimens at the injection site of a Turkish Van cat that had been vaccinated four months previously. The original tumor consisted of two compartments each 2.5 cm and 3.5 cm in diameter. It recurred five times following surgery and local recurrences necessitated five more radical resections within two years. Euthanasia was performed when pleural effusion developed, but evidence of metastatic disease was not observed. Unilateral uterine prolapse that occurred immediately after parturition was diagnosed in a 15-month-old cat brought to the clinics of the Department of Obstetrics and Gynecology, Faculty of Veterinary Medicine, Ankara University. Depending on clinical and ultrasonographic examination findings, the other uterine horn was though to be invaginated into the prolapsed horn. To purpose of treatment, ovariohysterectomy was performed. During laparotomy, the non-pregnant right uterine horn was seen to be invaginated into the pregnant uterine horn.

Copper toxicosis (CT) is a common autosomal recessive disorder in dogs. Recently, the CT locus was closely linked to microsatellite marker C04107. Currently, the gene underlying CT is unknown. In contrast to that of many other animals, knowledge regarding the canine karyotype is quite sparse. Except for the X and Y, all the chromosomes are acrocentric. Peripheral blood was used in this study and high-resolution G-banding and FISH were performed on dog chromosomes. This study investigated the selectivity of a 40 mm mesh size polyethylene codend commonly used by Turkish demersal trawlers. Seasonal selectivity data were collected for annular sea bream (Diplodus annularis), which is the most abundant discard species in the catch composition in Gülbahçe Bay. Four sets of data were collected in spring (4-18 April 2002), summer (10-25 July 2002), autumn (26 September-2 October 2002) and winter (22-23 January 2003). Our objective was to investigate hepatobiliary functions in healthy dogs using ^{99m}Tc-mebrofenin hepatobiliary scintigraphy (HBS): 200 µCi/kg ^{99m}Tc-mebrofenin was injected intravenously into 12 healthy adult dogs. One-hour dynamic images and 2, 4 and 24 h static images were obtained. HBS was evaluated for 7 variables. Cardiac washout occurred within 3 min in all dogs. A photophenic area in the region of the gall bladder was seen in 3.03 ± 0.21 min. However, bile ducts were not identified as separate structures in any dogs. Mice were immunized 3 times intranasally (i.n.) at intervals of 2 weeks with a formalin-inactivated Sendai virus (SV) whole virus with and without whole formalin-inactivated Bordetella pertussis (Bp) as adjuvant. The antibody response to SV was measured using the hemagglutination inhibition (HI) test in serum, and the IgA and IgG antibodies were evaluated using enzyme linked immunosorbent assay in saliva and serum. It was demonstrated that repeating immunizations with SV plus Bp could induce HI serum antibodies, IgA and serum IgG antibodies. However, in mice immunized subcutaneously with SV alone, high serum IgG antibodies and HI titers were induced only. …

One hundred and fifty fish caught from Keban Dam Lake were examined for the presence of Listeria monocytogenes. A random amplified polymorphic DNA (RAPD) assay using a random primer was performed to detect genetic variations among L. monocytogenes isolates. The selective enrichment procedure followed by selective isolation was used for listeria isolation from fish intestines. Ten (6.6%) L. monocytogenes were isolated and identified from fish intestines. Two different band profiles were obtained by the RAPD analysis of the isolates. The effect of a bacteriocin-producing strain, Lactobacillus sakei Lb706, on the behavior of Listeria monocytogenes in rainbow trout fillets stored under vacuum at 4 ºC for 14 days and at 10 ºC for 5 days was investigated. At 4 ºC, growth of L. monocytogenes was significantly inhibited (P < 0.05) by L. sakei Lb706 during 10 days of storage while bacteriocin-negative Lb706-B did not affect the growth of L. monocytogenes during 14 days of storage. In the presence of the sakacin A-producing strain of L. sakei (Lb706), the growth of L. monocytogenes was significantly inhibited (P < 0.05) in the first 3 days of storage at 10 ºC, after which its count increased to 10^7 CFU/g. The control group and the group containing L. sakei Lb706-B showed similar growth patterns of L. monocytogenes. Significant increases (P < 0.05) were observed in all groups in terms of psychrotrophic bacteria counts. No significant (P > 0.05) changes in pH values during storage at 4 ºC and 10 ºC were determined. Consequently, the results of the present study showed that L. sakei Lb706 can be used as a protective culture in freshwater reared rainbow trout, especially for preventing possible contaminations of Gram-positive food-borne pathogenic bacteria such as L. …

Some biological characteristics and the stock size of the pike (Esox lucius L., 1758) population in Lake Karamık were investigated between March 2002 and February 2003. The percentages of males and females in the population were 67.2% and 32.8%, respectively. Although fish samples ranged in age group from I to VII and from 15 to 66 cm in fork length, most of them were in age groups I-II and had fork lengths of 20-29 cm. Lead is one of the ubiquitous environmental pollutants that induce a broad range of physiological and biochemical dysfunctions in animals. This study evaluated the effects of dietary lead exposure and ascorbic acid on performance, serum biochemical parameters, plasma malondialdehyde and lead accumulation in broiler chickens. The aim of the study was to determine the effects of low energy diets on the enzymatic antioxidant activities of ewes in late pregnancy. Thirty Chios ewes were used. Twenty pregnant ewes were divided into 2 groups of 10 ewes each (groups II and III), with 10 non-pregnant ewes being separated into a third group (group I). The ewes in groups I, II and III were fed rations containing 9.14 MJ ME/kg of dry matter (DM)-10.23% crude protein (CP), 10.20 MJ ME/kg of DM-15.04% CP, and 8.82 MJ ME/kg of DM-14.47% CP, respectively. Distribution Of Antigen Types Of Canine Parvovirus Type 2 In Dogs With Hemorrhagic Enteritis In Turkey, Zeki̇ Yilmaz, Annamaria Pratelli, Serhat Torun

The results of antigenic characterization of canine parvovirus type 2 (CPV2) isolated from the feces of 60 dogs with hemorrhagic enteritis are reported. Antigenic characterization was carried out by comparing the reactivity pattern of 4 monoclonal antibodies (mAbs) to CPV2 in the hemagglutination inhibition (HI) test. CPV2 was detected in 21 out of 60 dogs (35%) and antigenic typing was able to be performed for only 16 dogs with >=1:8 titers. Nine samples were characterized as CPV2a and 7 as CPV2b, indicating that the CPV2a variant was more common than CPV2b in dog populations in Bursa province in Turkey.

Rapid Evaluation Of Bacteriolysis Of Listeria Innocua And L. Welshimeri Strains Isolated From Foods, Fi̇gen Çeti̇nkaya, Recep Çibik

This study was performed to determine the bacteriolytic extent of 10 strains of Listeria innocua and 9 strains of L. welshimeri isolated from foods. Bacteriolysis was measured at 650 nm by spectrophotometer. For this purpose, late exponential phase cells were transferred into potassium phosphate buffer (100 mM, pH 7.0) and incubated at 37 ºC. While the bacteriolysis range of L. innocua strains was 48% to 76% after 48 h of incubation, L. welshimeri isolates exhibited broader bacteriolytic variabilities, from 10% to 67%. The livers and lungs of 5596 sheep (443 local, 473 Romanian and 4680 Australian) slaughtered in Amman Central Abattoir during November-December 1999 were examined in routine meat inspection procedures for helminths. Dictyocaulus filaria, hydatid cyst, Fasciola hepatica and Dicrocoelium dendriticum were recovered from the examined sheep with variable prevalences. Dicrocoelium dendriticum is reported from the livers of imported sheep in Jordan for the first time.

The sex distribution, growth in length and weight, growth rates and condition factors of Chub (Leuciscus cephalus (L., 1758)) in Karakaya dam lake were examined. Females made up 63.64% and males 36.37% (1.75:1) of 527 samples. The females and males were in the I-VI and I-V age groups, respectively. The reproductive properties of L. cephalus such as spawning period, age at attainment of maturity, egg diameter and ovarium weight were investigated. This study was carried out to determine the effect of single dose cadmium chloride (CdCl_2) administration on the possible development of oxidative stress by measuring malondialdehyde (MDA) levels in the liver, kidney and blood serum of male and female rats.
