Molecular Identification Of Bovine Papillomaviruses In Dairy And Beef Cattle: First Description Of Xi- And Epsilonpapillomavirus In Turkey, Veysel Soydal Ataseven, Özgür Kanat, Yaşar Ergün

In the current study, 23 papilloma/tumor-like samples obtained from cattle having clinical lesions and 9 blood samples collected from healthy-appearing cattle in Turkey were examined for bovine papillomavirus (BPV) DNA using the degenerate primers FAP59/64, and the different types of BPV were distinguished by type-specific primer sets. Furthermore, histopathological studies of papillomavirus were performed. A total of 7 BPV types (BPVs 1, 2, 3, 4, 6, 8, 9), including genera Deltapapillomavirus, Xipapillomavirus, and Epsilonpapillomavirus, were identified. Toxoplasmosis is one of the most important foodborne parasitic diseases of humans. In particular, sheep muscles are significant sources of infection in the transmission of toxoplasmosis. Carnivorism is the most important transmission route for human populations. The aim of this study was to detect the prevalence of Toxoplasma gondii tissue cysts in sheep meats in retail stories of Turkey. Both cattle and dogs were examined in modern and rural dairy farms that had a history of abortion over 5%. The blood samples were collected from 427 aborted cattle and the sera were tested using a commercial ELISA test kit. Additionally, a necropsy procedure was carried out on the fetuses and calves dead within 2 months after birth; the tissue samples were evaluated by histopathologic, immunoperoxidase, and PCR techniques. Skeletal muscles (tongue, masseter, leg, intercostal, and diaphragmatic muscles) and brain samples of 100 sheep at slaughter were analysed for the presence of T. gondii tissue cysts along with serum IgG titres. Two methods of isolation by percoll gradient centrifugation and tissue microarray (TMA) technique with immunoperoxidase staining were used. Seropositivity was detected in 88% (88/100) of sheep sera analysed by indirect fluorescent antibody test. Tissue cysts were observed in 46 (52.3%, 46/88) and 15 (17%, 15/88) of the seropositive sheep with the isolation technique and TMA and immunoperoxidase staining, respectively. Sera from 1133 Turkish horses were obtained and tested for equid herpesvirus 3 (EHV-3) antibodies using a sero-neutralisation test. The sample consisted of 420 privately owned working horses from 2 regions of Turkey, 428 brood horses from private stud farms and breeding stations, and 285 racehorses from private stud farms. Neutralising antibodies against EHV-3 were found in 287 (25.3%) of the 1133 analysed sera. The highest seroprevalence for EHV-3 was detected in the brood horses (51.2%), followed by the racehorses (10.2%), and the working horses (9.3%). This study was conducted between July 2011 and January 2012 in Gökçeada, Turkey, to determine the metazoan parasite fauna of brown meagre (Sciaena umbra). The metazoan parasite fauna of 53 brown meagre samples was investigated and a total of 7 parasite species [Anoiktostoma coronatum, Paracryptogonimus aloysiae (Digenea), Longicollum pagrosomi (Acanthocephala), Contracaecum fabri (Nematoda), Trachelobdella lubrica (Hirudinea), Nerocila bivittata, and praniza larvae (Isopoda)] were identified. It was determined that Paracryptogonimus aloysiae was the dominant parasite species of brown meagre with the prevalence of 66.04% in this region. Anoiktostoma coronatum and Paracryptogonimus aloysiae were reported for the first time in Turkey.

Coccidiosis is a common enteric disease of turkeys that is caused by protozoan parasites belonging to the genus Eimeria. There are about seven species of Eimeria that affect turkeys and of these E. adenoeides is the most pathogenic and commonly recognized. A serological investigation of equine infectious anaemia (EIA), equine viral arteritis (EAV), glanders, and dourine was conducted on the sera of 346 Turkish horses using a combination of tests in series (ELISA and agar gel immunodiffusion) for EIA, virus neutralisation for EAV, and complement fixation (CF) for glanders and dourine. Forty-nine sera showed anticomplementary reactions and were not assessable in the CF test for glanders and dourine. No positive samples were detected for EIA, dourine, and glanders. Archaeological excavations at the site of Yenikapı, the most important main station for Metro and Marmaray projects on the European side of İstanbul, were started in 2004 with the discovery of numerous shipwrecks, amphorae, and skeletal remains belonging to animals. The findings from the excavations conducted by the İstanbul Archaeological Museums of the General Directorate of Cultural Heritage and Museums proved that this site, known in the Ottoman period as 'Langa Bostanları', was actually the ancient harbor of Theodosius. This review presents a search of the literature about gastrointestinal nematodosis of small ruminants. The search was performed on 58 articles published on necropsied sheep and goats between 1937 and 2011 in Turkey. This review identified 43 nematode species belonging to sheep and goats, assessed the prevalence of infection in different regions of Turkey, and evaluated the effect of age, sex, race, nutrition, and seasonal condition. This is the first review that specifically evaluated the gastrointestinal nematodes of small ruminants for 74 annual periods in Turkey.

Three pseudoscorpions identified as Neobisium validum were found in a honeybee hive in Bursa Province of Turkey. They were collected from the bottom of the hive but no relationship between the pseudoscorpions and bees was observed. This case is a first report of pseudoscorpions in a honeybee hive from Turkey.

The goal of this research was to determine the extent of resistance that turkey roundworms, Ascaridia dissimilis, have developed to anti-parasitic chemicals used in commercial poultry operations. Roundworm infections in turkeys have resulted in monetary losses for the poultry industry for years, generally due to poor feed conversion. The infection itself is subclinical and many turkeys have a light to moderate worm burden. Since parasitisms are light, this leads to the infections being noticed only during processing. This study aimed to purify glutathione reductase (GR) from turkey liver and investigate some of its characteristic features. The purification procedure comprised 2 steps: homogenate preparation and 2',5'-ADP Sepharose 4B affinity gel chromatography. Thanks to the 2 consecutive procedures, the enzyme, having the specific activity of 606.67 EU mg protein^{-1}, was purified with a yield of 10.75% and 2476-fold, and, in order to control enzyme purity, SDS-PAGE was done. This study was performed to determine the efficacies of Lavandula officinalis, Foeniculum vulgare, and Laurus nobilis essential oils on the control of Varroa destructor (Acari: Varroidae), the most common parasite of honeybees (Apis mellifera L.). Experimental colonies from the province of Bursa, Turkey, were divided into 5 homogeneous groups of 8 hives each. The aim of this study was to identify ectoparasites of wild roe deer (Capreolus capreolus) living in natural areas of Samsun Province that were wounded by car accidents or hunters between 2007 and 2010. A total of 90 ticks, 60 lice, and 5 deer keds were collected from 17 animals. Five tick species (Ixodes ricinus, Rhipicephalus bursa, Rhipicephalus turanicus, Haemaphysalis punctata, and Haemaphysalis concinna), 1 louse species (Cervicola meyeri), and 1 deer ked species (Lipoptena cervi) were identified. Cervicola meyeri and L. cervi were identified for the first time in Turkey.

A Maedi-Visna virus (MVV) infection was diagnosed in a 10-month-old lamb from a Turkish flock. The animal suddenly showed nervous signs and died within 2 days. At necropsy, different pulmonary and nervous lesions were observed. A Coenurus cerebralis cyst was also present in the right-brain hemisphere. Severe nonpurulent meningoencephalitis and lymphoproliferative pneumonia were detected by histopathology. MVV proviral DNA was demonstrated by polymerase chain reaction (PCR) in formalin-fixed and paraffin-embedded samples collected from the brain and the lungs. A total of 19,679 adult ticks were collected from 443 cattle in 9 villages in the West Aegean region of Turkey over 2 years (June 2006-May 2008). Thirteen out of the 19,679 adult ticks were identified as Hyalomma rufipes (12 adult males and 1 female). The ticks were identified according to their characteristic dark coloured conscutum and scutum along with the presence of small to medium but uniform and numerous punctations. The maedi-visna virus (MVV) is distributed worldwide and, to date, no epidemiological surveys have been carried out using ELISA testing in Turkey. The aims of this study were as follows: i) to verify the diagnostic performance of a home-made ELISA test (HM-ELISA) using Turkish ovine sera in comparison with other commercially available ELISA tests, and ii) to perform a preliminary epidemiological survey in İstanbul province. Lung, trachea, and serum samples obtained from turkeys showing clinical signs of respiratory disease were examined for the presence of Bordetella avium infection (bordetellosis) using culture, enzyme-linked immunosorbent assay (ELISA), and polymerase chain reaction (PCR). A wild boar (Sus scrofa) about 2 years old was brought to our parasitology laboratory from Orhaneli region, Bursa province. It was examined for ectoparasites and 32 lice collected from the animal's head were inspected and identified as Haematopinus suis. Five of them were found to be as nymphs, whereas 27 were adults. This paper is the first report of H. suis found in Bursa province, Turkey.

Mycoplasma hyopneumoniae is the primary agent of enzootic pneumonia in pigs. The aim of this study was to investigate the occurrence of M. hyopneumoniae in pigs raised in Turkey. Samples were obtained from the lungs of 286 pigs, which had already been separated from the carcasses at slaughter. Lung tissues were homogenized in 5 ml of Friis medium, the suspensions were inoculated into Friis medium, and then incubated at 37 °C until they developed an acid color change or for 21 days. This study was carried out to determine the prevalence of helminth species in horses, donkeys, and mules in the Central Black Sea region, between March 2004 and July 2005. For this purpose, 140 faecal samples were taken from horses (n = 83), donkeys (n = 31), and mules (n = 26) in Samsun, Sinop, Ordu, Amasya, and Tokat provinces. Infection rates were 91.57% (76 of 83) in horses, 96.77% (30 of 31) in donkeys, and 96.15% (25 of 26) in mules. A prospective longitudinal observation study was carried out with 150 randomly selected dairy herds from among 1684 Turkish Dairy Breeding Association (TDBA) herds in Burdur, Konya, and Kırklareli provinces. The herds were stratified as small scale (less than 11 milking cows) and medium-large scale (more than 10 milking cows). The survey was conducted between October 2003 and September 2004, and each herd was visited at least once per month. The aim of the present study was to apply immunohistochemistry, using a commercially available monoclonal antibody, for the diagnosis of aspergillosis in adult turkeys. Eight adult turkeys were studied. The lungs revealed grossly a few yellowish-white granulomas. Histopathologically, the granulomas were distributed in the pulmonary parenchyma or occasionally located around the airways. A few fungal hyphae were detected within the necrotic debris of the granulomas in the lungs using routine fungal stains. The mite formerly known as Varroa jacobsoni is one of the major problems in beekeeping worldwide. Recently, it was shown that this pest is not V. jacobsoni, which chiefly infests the nests of Apis cerana in the Malaysia-Indonesia region, and the widespread pest was recognized with the new name, V. destructor. Morphometric analysis was performed on collected Varroa specimens from various regions in Turkey and identified as V. destructor.

The aim of this study was to investigate the seroepidemiology of equine influenza infection in Turkey, measured using the haemagglutination inhibition (HI) assay. Serum samples were obtained from 623 equids aged older than 4 years from 5 different geographical regions in Turkey during the years 2003-2005. Antibodies against equine influenza virus (EIV) were found in 194 (31.1%) of the 623 sera. Seropositivity rates were detected as 41.8% (171/409), 12.8% (11/86), and 9.4% (12/128) in horses, mules, and donkeys, respectively. Lyme disease is a zoonotic infection caused by the spirochete Borrelia burgdorferi, which is transmitted by ticks of the Ixodes species. This is the first clinical case report of Lyme disease in Turkey. The subject was a 2-year-old male Saint Bernard with recurrent shifting leg lameness that began at about 6 months of age. In sera samples, B. burgdorferi IgM, IgG, and Western Blot were found to be positive. Chronic borreliosis was diagnosed and a long-term amoxicillin treatment was recommended.
