The southern Appalachian Mountains have among the highest salamander diversity in the world, largely due to local speciation in the family Plethodontidae. Plethodontid salamanders (i.e., lungless salamanders) are particularly sensitive to habitat climate conditions due to their reliance on cutaneous respiration, and their immediate environmental conditions (microhabitat) likely influence their dispersion and activity more than the large-scale atmospheric conditions. The Northern Gray-cheeked salamander (Plethodon montanus) is restricted to high elevations in the Appalachian Mountains. Renal cell carcinoma (RCC) is the 8th most common cancer in the United States, with the clear cell variant (ccRCC) being the most prevalent. Over 14,000 people die every year to RCC, with rates continuing to increase with an aging general population. Patients suffering from metastatic RCC (mRCC) have extremely poor prognoses, with a 5-year survival of only 11.2%. Current treatment options include resection of primary lesions, tyrosine kinase inhibition (Sunitinib, Pazopanib), mTOR inhibition (Temsirolimus, Everolimus), and immune checkpoint inhibition (Nivolumab, Atezolizumab). The lack of approved vaccines, medications and treatment regimens has significantly contributed to the rapid spread of mosquito-borne viruses such as Dengue and Zika virus. Ever since the eighteenth century, experts have tried to tell farmers how to farm. The agricultural enlightenment in Europe marked the beginning of a long arc of new experts aiming to change agricultural knowledge and practice. This dissertation analyzes the pivotal period in the late nineteenth and early twentieth century in Germany and the United States when scientists, improvers, and market agents began to develop comprehensive ways to communicate agricultural innovation to farmers. A series of three oceanographic survey cruises were conducted in June, July, and August of 2019 in the northeastern Gulf of Maine and Bay of Fundy. Surface water samples were collected and analyzed for enumerations of cell densities of the dinoflagellate Alexandrium catenella in relation to cell densities of diatoms. Hydrographic profiles of temperature, salinity, and nutrients (silicate and nitrate) were also made at each station. Data were analyzed to determine if there was any statistically significant evidence of allelopathic interference imparted by diatoms that impede A. catenella. Sensory experiments require anesthesia so the animal is immobilized, however fish anesthetics have shown to depress sensory responses. Newer anesthetics may offer similar anesthetic relief, but differ in means of action so sensory responses may be unaffected. Propofol has been used intravenously on small elasmobranchs but may provide prolonged effects if used as an immersion anesthetic. Objectives of this study were 1. Determine appropriate concentration of anesthetic to minimize induction and recovery for animals anesthetized at a surgical plane of anesthesia and 2. Adipose tissue can be characterized as either being a white (energy storing) depot or a brown (energy expending) depot and both have been found to contain dense networks of neural innervation. This adipose nerve supply regulates numerous metabolic functions and likely plays an important role in the function of adipose blood vessels. Candida yeasts are common commensals that can cause mucosal disease and life-threatening systemic infections. While many of the components required for defense against Candida albicans infection are well established, questions remain about how various host cells at mucosal sites assess threats and coordinate defenses to prevent normally commensal organisms from becoming pathogenic. Using two Candida species, C. albicans and C. parapsilosis, which differ in their abilities to damage epithelial tissues, we used traditional methods (pathogen CFU, host survival, and host cytokine expression) combined with high-resolution intravital imaging of transparent zebrafish larvae to illuminate host-pathogen interactions at the cellular level …

Investigations of human biology and disease have been hindered by the use of animal models. The information obtained from such studies often results in clinically irrelevant results and drug trial failures. Additionally, several governing bodies have been formulating legislation to move away from animal models and toward more ethical and efficient testing platforms for drug discovery and cosmetic research. A unidimensional Rasch approach was used to explore whether the data collected through the National Survey of Adoptive Parents of 2007 (NSAP) for the well-being items represented a single latent construct and to establish a base model for comparison. A consecutive approach was then used as an exploratory tool to draw out potential multiple dimensions. Finally, multidimensional item response theory (MIRT) was used to confirm the results of the consecutive approach findings while comparing with the unidimensional baseline. Endothermic organisms such as mammals and birds rely on high energy budgets to regulate body temperature. Many studies have previously investigated the thermo-regulation of mammals under the lower critical temperature of the thermoneutral zone, yet our knowledge in determining the upper critical limits is still scarce. As an endotherm, lab mice (Mus musculus) are perfect models to determine the cost to maintain constant body temperature as ambient temperature increases. With the prevalence of neurodegenerative pathologies in our society today it is imperative that we begin to look at novel approaches to the underlying problem of dying neurons that are not replaced. Adult neural stem cells exist naturally and could potentially be manipulated into targeted repair of damaged brains, given substantial research. Congenital muscular dystrophy with megaconial myopathy (MDCMC) is an autosomal recessive disorder characterized by progressive muscle weakness and wasting. Megamitochondria in skeletal muscle biopsies and cognitive impairments in MDCMC patients are observations exclusive to this type of muscular dystrophy. The disease is caused by loss of function mutations in the choline kinase beta (CHKB) gene which results in dysfunction of the Kennedy pathway for the synthesis of phosphatidylcholine (PC). Patient‐derived xenografts (PDXs) generated by implanting human tumor tissue into a transplant compliant mouse host have been of increasingly importance to preclinical development and have been demonstrated to have advantages compared to cancer cell lines and cell‐line xenografts (CLX) for modeling therapeutic responses in cancer. Nevertheless, many open questions remain regarding the relationship between study design factors and classification of treatment response and the molecular fidelity of tumors passaged in PDXs relative to the original patient tumor(s). Vernal pools in the northeastern United States provide essential habitat for pool-breeding amphibians and provide resources for other forest-dwelling wildlife. These pools and pool-breeding amphibians in particular are threatened by land conversion associated with urbanization and urban-associated factors. The responses of these amphibians and of birds and mammals using vernal pools to intermediate levels of urban development are largely unknown. I used field observations and lab experiments to study the amphibians, birds, and mammals associated with vernal pools along an urban development gradient in greater Bangor, Maine.

Polymicrobial communities exist throughout the human body and include both fungi and bacteria. During disease, cross-kingdom interactions among bacteria, fungi, and/or the immune system can alter virulence and lead to complex polymicrobial infections. The fungus C. albicans is among the most commonly isolated fungi in the context of fungal-bacterial co-infections and is often accompanied by the bacterium P. aeruginosa at a variety of sites throughout the body including mucosal tissues such as the lung. In vitro, C. albicans and P. aeruginosa have a cyclic, bi-directional, and largely antagonistic relationship, but these interactions do not account for the role of the …

Most marine organisms are broadcast spawners, releasing their sperm and eggs into the water column. Methods of measuring in situ fertilization have proven successful with a few model species, which are reviewed in my introductory chapter. However, many commercially exploited species, such as the sea scallop Placopecten magellanicus, have been neglected. Sea scallop populations have greatly increased from fishing closures, but the mechanism behind this response is uncertain, particularly in regard to fertilization.
