A Bioinformatics Approach To Understanding The Pathogenesis Of Ectopic Spine Calcification, Fang Chi Wang

This study used mice lacking equilibrative nucleoside transporter 1 (ENT1-/-) as a preclinical model to study pathogenesis of ectopic spine mineralization in diffuse idiopathic skeletal hyperostosis (DISH). We hypothesized that mineralization of the annulus fibrosus (AF), was driven by dysregulation of cellular processes and pathways associated with apoptosis, S100A9 proteins, the PI3K-Akt pathway, and lipid metabolism. Target pathways and processes were assessed using in situ localization and quantitative analyses. Metabolomic analysis of AF and plasma data was conducted to identify altered metabolites. Bovine in-vitro production(IVP) of embryos is a growing field for producing offspring with desirable genetics in the cattle industries. While IVP is effective, oocyte cytoplasmic maturation is compromised, and bovine oocytes matured in-vitro have reduced metabolic activity than those matured in-vivo. Mitochondria are the central unit of oocyte metabolism, producing ATP through OXPHOS. Photobiomodulation is a light treatment reported to improve metabolic activity. In five experiments, we measured the effects of photobiomodulation treatment with red LED at 16-(L-16) and 20 h(L-20) of bovine oocyte in-vitro maturation on subsequent embryonic development, mitochondrial activity and nuclear progression. Blood flow restriction (BFR) exercise has become an increasingly common modality due to evidence that it may yield positive muscle-related effects from low-intensity exercise. However, eccentric-only exercise in a multi-joint format has not yet been investigated in regard to the BFR influence on muscle recovery characteristics. The purpose of this study was to assess muscle recovery responses via isometric peak force (PF), soreness (i.e., VAS, PPT, PPD), and functional power (i.e., SLHD) following a 5-minute low-load multi-joint eccentric exercise bout with or without blood flow restriction. The nuclear hormone receptor (NHR), DAX-1 (dosage-sensitive sex reversal, adrenal hypoplasia critical region, on chromosome X, gene 1), is important in adrenal and gonadal development as well as steroidogenesis. It is encoded by the NR0B1 gene and functions mainly as a transcriptional repressor. Classified as an orphan receptor within the NHR superfamily, DAX-1 has been shown to inhibit other NHRs including estrogen receptor, androgen receptor and steroidogenic factor 1. DAX-1 is found to be underexpressed in breast and prostate cancers and, specifically in prostate cancer, is believed to be a transcriptional repressor of genes that are involved in epithelial-mesenchymal transition …

Defense of mucosal tissues from microbial infection and allergy is reliant on continual production of antibodies. The aryl hydrocarbon receptor (AhR) is known to regulate B cell development and is associated with suppression of systemic humoral immunity. Recent attention has been paid to the role of the AhR in altering expression of cell adhesion molecules (CAMs). B cells express CAMs and chemokine receptors to migrate around the body for localized secretion of antibodies. Chronic ankle instability (CAI) is a neurophysiologic deficit resulting in diverse sensorimotor impairments. Following acute ankle sprains, pain, mechanical instability, and joint deafferentation reduce sensory input to the central nervous system (CNS). In response, the CNS sends altered motor signals to lower extremity muscles. These CNS changes contribute to various neuromuscular impairments in CAI patients, the most common of which is reduced balance performance. Specifically, CAI patients struggle to modulate spinal reflex excitability of the soleus muscle when progressing from simpler to more complex balance tasks. This overreliance on spinal reflexes results in inconsistent activation of the ankle stabilizing muscle. …

The goal of this study is to determine how the heart and lungs respond from participation in Competitive Rifle with division one collegiate and intercollegiate club athletes. Exercise performance is widely believed to generally be maximal among young males. However, little research has been conducted to investigate the impact of sex or age on muscle performance. This study investigated the effects of sex and age on forelimb and all limb grip strength performance in wildtype and titin mutant mice. Titin is the largest known protein, with known involvement in muscle function. The mice in this study were Ttn∆112-158 with a 75% deletion to the PEVK region of titin. Active learning and required group work have been shown to increase student motivation in in-person and synchronous online courses, but little is known about their impact in online asynchronous settings. Context: Early serious illness conversations related to end-of-life and goals of care with seriously ill patients have been associated with improving patients' outcomes and quality of care (Bernacki et al., 2015).
