In the brain, HIV predominantly infects microglia/macrophages and astrocytes to a lesser extent. These cells form virus reservoirs with low levels of infection that are very hard to eradicate. Even though the use of cART increases survival rate in HIV patients, the virus persists as a chronic condition. cART is not able to effectively cross the BBB, control HIV replication, or attenuate inflammation in brain reservoirs. Therefore, the virus still causes neuronal dysfunction, pain-related pathology, and ultimately HAND. Despite the many governmental and medicinal restrictions created to combat the opioid epidemic in the United States, opioid abuse and overdose rates continue to rise. The development of an aptamer-based voltammetric sensor and biosensor is described in this dissertation. The aim was to develop a low-cost, sensitive, and specific aptamer-based sensor for on-site, label-free determination of codeine and fentanyl in biological fluids. To do this, the surfaces of screen-printed carbon electrodes (SPCE) were modified with gold nanoparticles (AuNPs), followed by the addition of single-stranded DNA aptamers. The dinoflagellate Karenia brevis, blooms annually in the Gulf of Mexico, producing a suite of neurotoxins known as the brevetoxins. The cellular toxin content of K. brevis, however, is highly variable between or even within strains. I investigated biochemical differences between high (KbHT) and low (KbLT) toxin producing cultures both derived from the Wilson strain, related to energy-dependent quenching (qE) by photosystem II, and the content of reduced thiols of the proteome. The coffee ring effect (CRE) phenomenon is originated from the nonuniform solvent evaporation of a sessile droplet deposited onto the flat substrate. Once the evaporation starts, the droplet suspended particles move outwards by the radial flow and concentrate in the edge region of the evaporating droplet, resulting in the formation of the so-called CRE stains. Chloroperoxidase (CPO), a member of the heme peroxidase family, has diverse catalytic activities toward a broad range of substrates. In addition to catalyzing halogenation reactions involved in the biosynthesis of halogen-containing compounds, CPO also catalyzes reactions typical of traditional heme peroxidases, catalases, and cytochrome P450 enzymes. Despite the powerful and versatile catalytic activity of CPO, its applications have been thwarted by the difficulty in regenerating the active enzyme and substrate (peroxide) induced protein inactivation. This study determined the effects of the copper water quality criterion (WQC) by the EPA on [1] swimming performance and [2] predator-prey interactions of the Sailfin Molly, across a salinity gradient. Fish acclimated to FW (0 ppt) and 8-ppt saltwater were exposed to 11.3 and 8.44 µg/L Cu for 96 h, respectively. At the end of the exposures, fish swimming performance was determined by using the critical swimming speed, Ucrit, the speed at which a fish cannot longer maintain position in the water column. Ochratoxin A (OTA) is a potent mycotoxin found in a wide range of agricultural products that has been linked to mitochondrial damage and renal disease. The standard methods for OTA analysis currently rely on the use of high-performance liquid chromatography (HPLC) coupled to fluorescence detection or mass spectrometry. Toward a highthroughput analysis of OTA, a single-stranded DNA aptamer, modified with a fluorophore, coupled to a complementary sequence, modified with a FRET-based quencher that dissociates in the presence of the target toxin, is proposed. This study analyses the spatial and temporal variability of chlorophyll-a in Chesapeake Bay; assesses the performance of Ocean Color 3M (OC3M) algorithm; and develops a novel algorithm to estimate chlorophyll-a for coastal shallow water. The OC3M algorithm yields an accurate estimate of chlorophyll-a concentration for deep ocean water (RMSE=0.016), but it failed to perform well in the coastal water system (RMSE=23.17) of Chesapeake Bay. A novel algorithm was developed which utilizes green and red bands of the Moderate Resolution Imaging Spectroradiometer (MODIS) sensor. The novel algorithm derived the chlorophyll-a concentration more accurately in Chesapeake Bay (RMSE=4.20) than the OC3M algorithm. …

Perfluoroalkyl substances (PFASs) are contaminants of emerging concern, and have been detected in drinking water, wildlife, humans, and the environment. Cyclodextrins (CDs), cyclic sugars composed of glucose monomers, are proposed as a potential remediation strategy. CDs can form host-guest complexes with hydrophobic molecules; this complexation could be capitalized on for PFAS removal and sequestration. Degraded pollinator habitat may have far-reaching consequences for recovery of imperiled flowering plant populations. Studies indicate that broad-spectrum insecticides used in mosquito abatement are detrimental to non-target invertebrates, including pollinators. A decline in efficient pollinators can reduce plant fitness by decreasing reproductive output and constraining genetic diversity, a challenge for rare plants.

Trace amounts of pharmaceuticals have been detected in water, from nanograms per liter to micrograms per liter, and have a negatively effect in the aquatic environment and an increased potential risk of drug poisoning for human and animals. In order to address the problem, drug degradation catalyzed by chloroperoxidase (CPO) has been investigated. An ecological risk assessment was conducted on the risk to fish of chemical contaminants detected in the habitat of Albula vulpes in South Florida, to evaluate whether contaminants may be a driver of declines in the recreational bonefish fishery. All available contaminant detection data from Biscayne Bay, Florida Bay, and the Florida Keys were compared to federal and state guidelines for aquatic health to identify Contaminants of Potential Ecological Concern (COPECS). Mangos (Mangifera indica L.) are tropical/subtropical fruits belonging to the plant family Anacardiaceae. Anthracnose is the most deleterious disease of mango both in the field and during postharvest handling. It is most commonly caused by the Colletotrichum gloeosporioides complex. Mangiferin, a xanthanoid compound found in at least twelve plant families worldwide (Luo et al., 2012), is present in large amounts of the leaves and edible mangos. As a consequence of continual exposure to arsenic, nearly every organism, from Escherichia coli to humans have evolved arsenic detoxification pathways. One of the pathways is extrusion of arsenic from inside the cells, thereby conferring resistance. The R773 arsRDABC operon in E. coli encodes an ArsAB efflux pump that confers resistance to arsenite. The central Everglades serve as a Wildlife Management Area and as a Water Conservation Area for the Miami-Fort Lauderdale metropolitan area. It is also home to over 22 nonnative freshwater fish species and carries restrictive fish consumption guidelines for Mercury. In my study, boat anglers and canal bank anglers were personally interviewed in the field, to research their awareness and perspectives of these potential environmental and health threats. This thesis analyses buckwheat as a cover crop in Florida. The study was designed to demonstrate: soil enrichment with nutrients, mycorrhizal arbuscular fungi interactions, growth in different soil types, temperature limitations in Florida, and economic benefits for farmers. Buckwheat was planted at the FIU organic garden (Miami, FL) in early November and harvested in middle December. After incorporation of buckwheat residues, soil analyses indicated the ability of buckwheat to enrich soil with major nutrients, in particular, phosphorus. Symbiosis with arbuscular mycorrhizal fungi increased inorganic phosphorus uptake and plant growth. Regression analysis on aboveground buckwheat biomass weight and soil characteristics showed …

The poisonous fruiting bodies of Amanita muscaria (L. ex Fr.) Pers, ex Hook, are harvested by rural inhabitants of Sanada Town, Japan. These mountain villagers consume beni-tengu-take as a local delicacy, despite its potential hallucinogenic effects. The Japanese use several methods to detoxify beni-tengu-take, but believe pickling the mushrooms to be the safest. Other methods of preparation include grilling and drying the mushrooms. I documented the preparation and consumption of each detoxification method through local interviews with Japanese informants. I then used ion-interaction rp-HPLC to quantify the hallucinogenic compounds, ibotenic acid and muscimol, and determined the efficacy of each traditional …
