Cadmium (Cd) is a non-essential trace element and its environmental concentrations are increasing due to human activities. Edible plants can accumulate high concentrations of Cd, which could be toxic to humans. Understanding how and where Cd is stored in plants is important for ensuring lower concentration of Cd in the food. In this thesis, the accumulation and distribution of Cd in three agricultural plants, namely lettuce (Lactuca sativa L.), barley (Hordeum vulgare L.) and radish (Raphanus sativus L.), were investigated with a focus on the potential mechanisms involved in the localization of Cd in the root. The Ras family of small GTPases (N-, H-, and K-Ras) is a group of important signaling mediators. Ras is frequently activated in some cancers, while others maintain low level activity to achieve optimal cell growth. In cells with endogenously low levels of active Ras, increasing Ras signaling through the ERK and p38 MAPK pathways can cause growth arrest or cell death. Ras requires prenylation – the addition of a 15-carbon (farnesyl) or 20-carbon (geranylgeranyl) group – to keep the protein anchored into membranes for effective signaling. The plastics that make up over 10% of human waste are synthesized with phthalate plasticizers which are utilized in plastics as an additive to improve durability and flexibility. Many phthalates have been identified as endocrine disrupting chemicals (EDCs) and studies have shown dramatic deleterious effects as a result of species exposure to growing numbers of EDCs in the environment. Northern Fulmars (Fulmarus glacialis) are a northern hemisphere seabird species that forages opportunistically, meaning that they take advantage of a wide variety of different food sources, including inadvertent consumption of plastic. Toxic Effects Of Copper, Cadmium, And Methoxychlor Shown By Neutral Red Retention Assay In Two Species Of Freshwater Molluscs, Nikolett Molnar, Peter Fong

We used neutral red retention assay in lysosomes of digestive gland cells as an indicator for stress effects by the environmental contaminants Cu, Cd, and the pesticide methoxychlor in two freshwater molluscs, the unionid mussel, Elliptio complanata (Lightfoot) and the ramshorn snail Helisoma trivolvis (Say). Mussels and snails were exposed for 7 and 14 days to Cu and Cd each at nominal concentrations of 2.5 μg/L, 5.0 μg/L, and 10.0 μg/L, and to methoxychlor concentrations of 1.0 μg/L, 10.0μg/L, and 100.0 μg/L. Cationic Amphiphilic Drug-Induced Autophagosome Accumulation Is Due To Autophagosome Sequestration Within Vimentin Intermediate Filament Networks Resulting In Prolonged Autophagosome Half-Life, Miriam Devorah Kleinman

Accumulations of autophagosomes and non-esterified cholesterol are

observed in several cell lines derived from lysosomal storage diseases,

including Niemann Pick Type C (NPC). The relationship between

autophagosome accumulation and lysosomal non-esterified cholesterol is

unclear. Exposure of murine hepatoma 1c1c7 cultures to the cationic

amphiphilic drugs (CADs) U18666A, imipramine and clozapine caused

lysosomal non-esterified cholesterol and autophagosome accumulation.

Measurement of LC3-II conversion in the presence of lysosomal inhibitors

bafilomycin A1 and NH4Cl, degradation of long-lived proteins, and

colocalization of GFP-LC3 and LAMP1 indicated an increase in

autophagosome synthesis without compensatory increase in clearance.

Targeting Α4 Integrin Containing Complexes In Multiple Myeloma Using Peptidomimetics, Michael Foster Emmons

In our previous work we demonstrated that the integrin antagonist, HYD1, induced necrotic cell death in myeloma cell lines in vitro and in vivo as a single agent. In order to further delineate biomarkers of response to HYD1 we developed an isogenic drug resistant variant named H929-60. We show that the acquisition of resistance towards HYD1 correlates with reduced expression of the cleaved α4 integrin subunit and beta 1 integrin. Moreover, we demonstrate that HYD1 interacts with α4 integrin in myeloma cells. The Department of Energy's (DOE) Savannah River Site (SRS) is a 777 km2 former nuclear weapon material production and current research facility located along the Savannah River in west-central South Carolina. In 1954, direct environmental discharges were released from "M-Area", a fuel and aluminum-clad Uranium (U) nuclear reactor target manufacturing facility through a drainage ditch into the Tims Branch-Steed Pond water system.
