Chronic kidney disease (CKD) progresses faster in males than females; however, the underlying mechanisms remain poorly understood. Sex differences in glomerular capillary morphology has been hypothesized to contribute, in part, to the increased susceptibility to hypertension-induced renal injury and CKD progression in males, but this has not been investigated. The goal of the present study was to assess glomerular capillary morphology in male vs. female rats with intact kidneys and after uninephrectomy (UNX). We hypothesized that glomerular capillary radii (RCAP) and length (LCAP) would be greater in male rats.

Maternal obesity has led to an increase in adverse offspring developmental outcomes and a greater risk for long-term metabolic diseases. Choline, a semi-essential nutrient, can be incorporated into phosphatidylcholine (PC) as well as sphingomyelin (SM) and donate its labile methyl group for the remethylation of homocysteine after choline is oxidized to betaine. Prenatal choline insufficiency has been related to maternal obesity and metabolic diseases, such as metabolic associated fatty liver disease (MAFLD). Choline may interact with maternal obesity to influence the programming offspring.

Ephemeroptera taxa are not frequently used in toxicity testing; however, some mayfly taxa may be more sensitive to aquatic pollutants than standard test organisms used to determine anthropogenic effects on aquatic ecosystems. Additionally, some standard test organisms are not native to the Appalachian region and may not be truly reflective of the effects on native organisms. With mayflies not being the typical test organism, there is not a great deal of literature on culturing methods for this organism. Ever since the eighteenth century, experts have tried to tell farmers how to farm. The agricultural enlightenment in Europe marked the beginning of a long arc of new experts aiming to change agricultural knowledge and practice. This dissertation analyzes the pivotal period in the late nineteenth and early twentieth century in Germany and the United States when scientists, improvers, and market agents began to develop comprehensive ways to communicate agricultural innovation to farmers. To secure animal welfare and maximum production, feed additives are often added to nursery pig diets to increase nutrition digestibility, enhance intestinal health, and prevent or mitigate disease, with the ultimate goal of improving growth performance. This thesis performed a longitudinal analysis using next-generation sequencing to investigate the dynamic changes of gut microbiota in weaned pigs fed commercial feed additives. Poultry nutritionists continue to identify methods to lower feed price without compromising broiler growth performance, processing yields, or animal well-being. A common approach is to reduce dietary crude protein (CP), the second most expensive component in a broiler diet. Subsequently, reducing CP will reduce the inclusion levels of intact protein sources (e.g., soybean meal [SBM]), which can be accomplished by supplementing individual amino acids (AA). The modern broiler is growing at a rapid rate generating tremendous amounts of heat. A sensitive Net Energy (NE) system is needed to measure body heat production (HP) generated primarily by daily maintenance and synthesis and degradation of myofibrillar and sarcoplasmic protein. The first two chapters present evaluation of the classic way to calculate NE versus a new methodology; the Arkansas NE (Ark NE) system, with birds from two genetic lines fed diets with different AA content or different ME content in two different environmental temperatures.

I tested the hypothesis that recombinant leptin protein can be introduced to zebrafish in vivo through non-invasive soaking in a solution containing the protein. One way to study various molecules’ effects in vivo is through intraperitoneal or intracerebroventricular injections during the embryonic or larval stage, which is invasive, difficult to administer, and can have a high mortality rate. 48 hours post fertilization (hpf) zebrafish were soaked in a His-tagged recombinant leptin protein solution at 10 nM and 100 nM concentrations (produced by Genscript). After soaking, zebrafish larvae were washed extensively to remove all recombinant protein on their exterior before homogenization. …

The prevalence of non-alcoholic fatty liver disease (NAFLD) in children has increased over the past decades, creating a need for animal models that recapitulate the features of the pediatric disease. Iberian pigs have a leptin-resistant phenotype characterized by hyperleptinemia, hyperphagia, and extreme adipogenesis. We hypothesized that neonatal Iberian pigs fed a high fat high-fructose (HFF) diet will develop a pattern of liver injury resembling pediatric NAFLD. Ulcerative colitis (UC) is a disabling inflammatory bowel disease. UC is characterized by chronic, relapsing inflammation of the colon and rectum. Current treatments such as monoclonal antibodies against TNF-α, IL-6, IL-12/p40, adhesion molecules, Janus kinases (JAK) inhibitors have side effects or lose their effects over time. Alternative approaches with fewer side effects for patients are needed. Montmorency tart cherries (Prunus cerasus) are a good source of anti-inflammatory flavonoids. Male Sprague-Dawley rats fed diets of enriched yeast-leavened biscuits had no discernible advantage over those fed enriched chemically-leavened biscuits in three gastric indices studied. Restraint-stress administered to half of each dietary group was ineffectual in producing gastric ulcers, but did produce gastric color and appearance changes in fifty-eight per cent of the group receiving the chemically-leavened diet.

Thiamin and riboflavin-retention analyses done on various baked products produced conflicting results. Yeast-leavened products retained less thiamin and riboflavin than chemically-leavened products, but which chemical leavening agent was best for vitamin retention was different for each vitamin.

An increase in dietary cholesterol and cholic acid will produce hypercholesterolemia in rats. For ten months, two groups of rats (six in each group) were fed commercial rat pellets (control group) or rat pellets plus cholesterol (1%) and cholic acid (0.25%) (experimental group). A blood sample from each animal was divided into three aliquots for three separate experiments on the erythrocytes: a membrane cholesterol and phospholipid determination, a 3-0-[14C] methylglucose (30MG) transport determination, and a 2,3-diphosphoglycerate (2,3 DPG) determination. The effects of morphine and codeine free-choice alcohol consumption in male Sprague-Dawley rats were studied. Intakes of a 10% alcohol solution in milliliters per 100 gm. of body weight were on recorded weekly. No difference was found in the alcohol intake of rats given either a morphine-supplemented diet (0.01% of the diet) or daily injection of morphine sulfate (2 mg./kg.); however, there was a significant lower intake of water (p < .05) in those rats fed the diet with morphine. Total fluid consumption of both groups given the morphine-supplemented diet and daily injection of morphine sulfate was decreased (p < .05). It was found that morphine did not affect the percentage of total fluid consumed as alcohol or the weight gain.

Two levels of codeine phosphate (10 mg./kg. and 20 mg./kg.) were injected subcutaneously into rats twice a day. It was observed that the codeine significantly decreased alcohol intake in rats …

The consumption of alcohol is a problem not only in this country (U.S.), but also in many other places in the world. Thus we find the Indians in Peru, Bolivia and other neighboring countries (Ecuador and northern part of Argentina) consuming alcohol to the point that they become alcoholics, in addition to this they also chew coca leaves, plant from which cocaine is extracted.

Effects Of Dietary Factors And Morphine Administration On Voluntary Alcohol Consumption And Dopamine Metabolism In Rats, Claudia Smithson

The effect of diet and dietary additives on voluntary alcohol consumption in male Sprague-Dawley rats was studied. Alcohol intake measured weekly and reported as ml of a 10% solution ingested per 100 grams body weight. A four week pre-experimental period was designed to determine individual tendencies to drink and thus to eliminate the high and low drinkers from the study. No difference was found in alcohol intake with either starch or sugar as the carbohydrate source; however, there was a greater weight gain ( p < .025) in those animals consuming sugar-based diets. Rats on poor-quality diets containing minimal amounts of vitamins and minerals continued to gradually increase alcohol consumption during the study. This diet, supplemented with coffee, caused the greatest significant increase in alcohol intake and the lowest weight gains. Addition of coffee to the diets significantly increased alcohol consumption (p< .005) but decreased growth rate (p < .0005) . However, when the poor quality diets were supplemented with vitamins and minerals, alcohol consumption was reduced by about 50% (p < .005) and weight gain was significantly increased (p < .0005).

The daily intraperitoneal administration of morphine sulfate significantly decreased alcohol intake (p < .02) and the per cent of total fluid consumed as alcohol (p < .02) but did not effect water consumption.

Studies were conducted utilizing albino rats to determine the relationship of voluntary alcohol consumption with various dietary additives and exposure to cigarette smoke in combination with poor quality diets. Male, Sprague-Dawley rats, from weanling age to 100 grams, were given the various experimental diets and a free fluid choice of ten per cent ethanol or tap water. Fluid intakes were recorded and analyzed statistically. Individual variation was common with a generally low level of alcohol consumption when rats were fed a poor quality diet of common United States foods. The high incidence of tooth decay, one of the most common diseases found in all age groups, gives a reason for the study of methods to prevent dental caries. Certain dietary factors, specifically carbohydrates, have been implicated in the decay process. Since diet influences both the oral and systemic environments in which the teeth develop, mature, age, and decay (1), many studies have been conducted to determine the effects of oral administration of different carbohydrates on dental caries.
