The objective of this thesis was to meet growing demand for the development of environmental biomonitors that protect ecosystems and public health. To do this, I determined the potential of oyster shell as a bioindicator of cadmium (Cd) in the environment by determining the mode of Cd uptake and relationships between Cd concentrations in the environment, shell, and soft tissues of juvenile eastern oysters (Crassostrea virginica Gmelin). I performed a review of the literature on the ability of oyster shell to retain metal contaminants and the factors that could affect this process (Chapter 2). I then reared C. virginica …

Tidal wetlands are vital for buffering coastal settings from the threats of accelerated sea level rise and storms. Understanding the factors that are most influential for the maintenance and recovery of tidal wetlands after extreme events compounded by future accelerated sea level rise is of the utmost importance, yet this knowledge is not well established. Two tidal wetland schemas investigated in this dissertation are mangrove systems in Vieques, Puerto Rico (including robust lagoonal-mangrove forest systems and fringing mangrove forests), and salt marshes in New England. The rocky intertidal ecosystem is an important ecological and cultural aspect of the picturesque Maine coast, playing a vital role in not only Maine’s coastal ecosystem, but also to Maine’s economy. It’s distinct community structure along the sharp elevational gradient and the presence of daily stressors (wave action, heat, and desiccation), make the rocky intertidal ecosystem an important model ecosystem to monitor for effects from anthropogenic impacts. This project examines the development of a smart boat which could serve as a possible marine research apparatus. The smart boat consists of a miniature vessel containing a low-cost microcontroller to live stream a camera feed, GPS telemetry, and compass data through its own WiFi access point. The smart boat also has the potential for autonomous navigation. My project captivated the interest of several members of California Polytechnic State University, San Luis Obispo’s (Cal Poly SLO) Marine Science Department faculty, who proposed a variety of fascinating and valuable smart boat applications.

As plastic pollution increases, top marine predators such as marine mammals are becoming increasingly susceptible to plastic particles and their additives. Plastic particles have been found in gastrointestinal tracts and scat of many marine mammals, and quantifying plastic pollution in those that are pelagic can provide insight into plastic pollution in mesopelagic ecosystems that are just beginning to be analyzed. Adapting well-developed laboratory techniques for microplastic (MP) isolation (i.e. density separation and chemical digestion), I isolated MPs from the scat of the deepest diving pinniped, the northern elephant seal (NES), and found that100% of scat samples (n=11) contained high counts …

The presence of oil has been shown to affect the microbiomes of the water column, sediments, and organisms, both by altering the diversity and the composition of those microbial communities. Climate change has negatively altered seawater conditions, which may have severe implications for marine ecosystems. Mussels are susceptible to environmental changes since they are primary consumers and filter-feeding bivalves. Two species of particular interest to the West Coast of the U.S. are Mytilus galloprovincialis and M. trossulus. M. trossulus is native to the California Coast and was historically prevalent from Southern California to Washington. However, with increased shipping and rising seawater temperature, M. galloprovincialis, an invasive species from the Mediterranean, pushed the species range of M. trossulus poleward. Very little is known about how heat stress during larvae stages effect larvae survivorship, early coral recruit settlement, and later stage coral survivorship. We focused on determining how heat stress during larvae stages effected Montipora capitata survivorship over time. After thermally stressing larvae, we asked how many larvae survived the treatment, how the treatment affected settlement, how many larvae survived the heat treatment but did not settle, and later stage coral survivorship experienced residual effects from the heat stress treatment. Temperature can exert impacts on many processes in ectotherms. With global temperatures rising due to climate change, many ectothermic species may exhibit changes in growth rates and size at maturity, and these changes can have population-level effects. Predicting responses of species to climate change will require not only knowledge of thermal tolerance limits, but also effects of temperature change on growth rates and other life history parameters. For arthropods that exhibit discontinuous growth (i.e., molting), this includes both intermolt period and growth per molt. Elevated surface temperatures exacerbate the threat of hypoxia within coastal ecosystems. These two primary stressors likely interact as they elicit opposing physiological responses from marine organisms. Metabolic depression is typically associated with hypoxia, while metabolic rates increase with temperature. Moreover, physiological effects of combined stressors may not be additive. In light of increasing pressures from hypoxia, elevated ocean temperatures, and other stressors within coastal regions, studies need to examine effects of multiple stressors on physiology of coastal organisms.

The Pearson and Rosenberg (P-R) conceptual model of macrobenthic succession was used to assess the impact of hypoxia (dissolved oxygen [DO] ≤ 2 mg/L) on the macrobenthic community on the continental shelf of northern Gulf of Mexico for the first time. The model uses a stress-response relationship between environmental parameters and the macrobenthic community to determine the ecological condition of the benthic habitat. The ecological significance of dissolved oxygen in a benthic habitat is well understood. In addition, the annual recurrence of bottom-water hypoxia on the Louisiana/Texas shelf during summer months is well documented.

An increase in marine mammal stranding and die-off events has been observed along the California coast. The exact cause to explain for these recent events is unknown, but El Niño and harmful algal blooms are established sources for temporary decreases in marine mammal health. Although biomarkers are frequently used to assess sublethal effects of contaminants, a lack of mechanistic linkages to higher-level effects limits the predictive power of biomarkers. Bioenergetics has been proposed as a framework for linking cellular effects to whole-animal effects. We investigated sublethal effects of exposure to wastewater treatment facility effluent in freshwater mussels in situ, thereby capturing ecologically relevant exposure conditions. Our study focused on the energetic biomarker AMP-activated protein kinase (AMPK), while also considering more traditional biomarkers like heat shock proteins (HSP70), and antioxidant enzymes (i.e., superoxide dismutase (SOD), glutathione-S-transferase (GST)). As salinity increases the geochemical speciation of Cu is altered as a result of organic/inorganic complexation/competition. Such salinity changes may further challenge the osmoregulatory capabilities of euryhaline organisms. This chemical-biological interaction complicates the understanding of the impacts of Cu in estuarine waters. Populations of the gastropod Nucella lapillus are polymorphic for shell color, with light-colored shells predominating on warmer, wave-protected shores and dark-colored shells limited primarily to cooler, wave-exposed shores. During thermal stress, darker shells attain higher body temperatures than lighter shells. These results suggest that heat stress may determine field distribution patterns. However, there is currently little evidence of physiological consequences of thermal stress in these organisms. Following the guiding hypothesis that heat stress leads to cellular energy depletion, we explored whether the central energy regulator AMP-activated Protein Kinase (AMPK) is activated by heat stress. A study of the subtidal macrobenthos in Back Bay, Virginia was conducted to examine community structure in relation to sedimentary and water quality characteristics. Samples were collected in August and November of 1987 and February and May of 1988 at ten stations.

Three site groups and three temporal groups were identified by cluster analyses of species abundance. Species composition between site groups was relatively homogeneous and eight species accounted for most of the variation between site groups. A comparison of plots of the biological and physical data in discriminant space suggested that variations in the biological data between site groups …

This dissertation is from the Joint Program Degree from the College of William & Mary and University of Virginia and awarded by the University of Virginia.

The effects of petroleum hydrocarbons on salt marsh grasses and gastropods were examined under three different circumstances. In the first study reported, a small pocket marsh was repeatedly dosed with small amounts of a No. 2 fuel oil. The second study investigated the effects of a large accidental spill of No. 6 fuel oil. The third study involved single doses of a fresh and a weathered crude oil on artificially enclosed segments of marsh.
